In vitro Organogenesis and Propagation of Heloniopsis orientalis Thunb

  • Heloniopsis orientalis (Liliaceae) is an important horticultural crop native to Korea. Under natural conditions, germination is poor and plant growth is delayed. Therefore, we have developed a vegetative propagation method to produce plants with vigorous growth characteristics via tissue culture. The regenerated shoots were then initiated directly from leaf explants on an MS medium containing either 0.5 to 2.0 mg/L 2,4-D or 1.0 to 3.0 mg/L BA. Healthy plantlets with adventitious roots were formed on the medium supplemented with 1.0 mg/L BA (81%). BA triggered callus initiation without caulogenesis or rhizogenesis, and callus formation was better on the half-strength MS medium than on the full-strength medium. This in vitro propagation protocol will be useful for conservation, as well as for mass propagation.

  • The experiment was conducted to obtain basic informations on the effects of key environments and fungicides on the sporulation, conidial germination and hitial growth of the hyphae of grape powdery mildew fungus Uncinula necator. Maximum sporulation occurred at RH 75~96% in vitro adiusted with sulfuric acid solution. Conidial germination and initial growth of the hyphae were better at 5% water agar than distilled water, and best at $26^{\circ}C$ and pH 5.0 of the substrates. Germination occurred equally well at light intensity of below 100 lux or dark condition, which was better at water agar supplemented with leaf extract 30% than pure water agar. The water agar supplemented with triazole fungicides reduced conidial germinations and initial growth of the hyphae signifcantly, in proportion to the increase in concentration of the fungicides, but the maguitude of reduction depended on the fungicides. Particularly in myclobutanil, reduction rate was very low as increased in concentration.

  • An efficient in vitro regeneration protocol for moth bean [Vigna aconitifolia (Jacq.) Marechal] via somatic embryogenesis has been developed. Embryogenic callus cultures were established from the cotyledonary node as explant on semi-solid Murashige and Skoog (MS) medium supplemented with $0.75mg\;1^{-1}$ 2,4-dichlorophenoxyacetic acid (2,4-D) and $1.5mg\;1^{-1}$ 6-benzylaminopurine (BA) and with various additives ($50mg\;1^{-1}$ ascorbic acid and $25mg\;1^{-1}$ each of adenine sulphate, citric acid and $_L-arginine$). Numerous somatic embryos differentiated on MS basal nutrient medium supplemented with $0.25mg\;1^{-1}$ 2,4-D and $0.5mg\;1^{-1}$ of kinetin (Kin). Sustained cell division resulted in the formation of cell aggregates, which progressed to the globular- and heart-shaped somatic embryos and then, if they differentiated properly, to the torpedo shape and cotyledonary stages. The transfer of embryos onto fresh MS basal medium containing $0.2mg\;1^{-1}$ BA and $2.0mg\;1^{-1}$ gibberellic acid enabled the embryos to achieve complete maturation and germination. More than 80% of somatic embryos were converted into true-to-type fertile plants. In vitro-regenerated plantlets with well-developed roots were successfully hardened in a greenhouse and established in soil.

  • The experiment was carried out in order to choose fungicides which are proper to be used in flowering time of fruit trees. The effect of fungicides on pollen germination and pollen tube growth was investigated in vitro. The results obtained are as follows : 1. Difolatan-Wp, Lime-sulfur, Captan-Wp and Bordeaux mixture greatly inhibited pollen germination and pollen tube growth. 2. The effect of Daconil-Wp and Zimaneb-Wp on pollen germination and pollen tube growth was not so great as that of Difolatan-Wp, Lime-sulfur, Captan-Wp or Bordeaux mixture. 3. Polyram-combi-Wp, Topsin-Wp and Benlate-Wp did not affect pollen germination but these fungicides slightly inhibited pollen tube growth. 4. Polyoxin-Wp significantly increased pollen tube growth but did not show promotive effect on pollen germinaiton.

The Effect of Ginseng Saponin Fraction on the Germination and Early Growth of Rice Seed (Oryza sativa L.) (인삼사포닌이 벼의 발아 및 초기 생장에 미치는 영향)

  • The effects of ginseng saponin on the germination and early growth of rice seeds (Oryza sativa L.) were investigated. The early growth (length) of test rice seeds which were rinsed for 60 hrs in $10^{-4}$% saponin solution prior to transplantation to water agar bed was about 20% faster than that of control seeds, It was also found that the amylase activity of the seeds was most active when the seeds were rinsed in 10-4% saponin solution for 48 hrs. In vitro investigation showed that the amylase activities were stimulated about 30% by the treatment of $10^{-5}$% saponin solution compared with control group. From the above results, it seems that the ginseng saponin might activate amylase of rice seed during germination, resulting in rapid growth of rice.

  • Optimized conditions for Agrobacterium-mediated lisianthus pollen transformation were adjusted using various factors such as temperature, pH and sucrose concentration. Pollen tube growth was successfully achieved in a medium (pollen germination medium; PGM) containing $7-15\%$ sucrose with pH in the range of 5.5-7.0 at temperature of $20-27^{\circ}C$. Lisianthus pollen was vacuum-infiltrated with Agrobacterium cell suspension for 20 min, and transformed pollen was confirmed by GUS histochemistry and Southern hybridization following RT-PCR. Transgenic pollen system may be utilized for establishing an area of plant transient expression systems based on the convenient pollen transformation procedure presented in here.

Antifungal Activity of 4',7-Dimethoxyisoflavone Against Some Fungi

  • The 4',7-dimethoxyisoflavone was isolated from the leaves of Albizzia lebbeck for the first time. This flavonoid showed antifungal activity against some plant pathogenic fungi tested in vitro, e.g., Alternaria melongenae, A. brassicicola, A. brassicae, Curvularia maculans, C. pallescens, C. lunata, Curvularia species, Colletotrichum species, Helminthosporium penniseti and H. speciferum. The sensitivity of different fungi to this chemical varied considerably. A. brassicae was most sensitive as complete inhibition of germination was observed in all the concentrations(100 to 1000 ppm) of the chemical. Similar effect on H. speciferum and Curvularia species was also recorded at 500 ppm, whereas H. penniseti did not germinate at 250 ppm. A. melongenae and A. brassicicola also did not germinate at 1000 ppm while 750 ppm was inhibitory to C. lunata and C. maculans. Germination in almost all fungi was significantly inhibited at each concentration in comparison to control except Curvularia sp. and H. speciferum. Use of 4',7-dimethoxyisoflavone to control some plant diseases under field conditions has been suggested.

  • Medicinal plants play important roles in controlling plant diseases as one of the safest and ecofriendly methods. These plants have been used in the form of crude extracts as well as active principles in vitro and under field conditions to control plant diseases. Among the active principles, alkaloids have shown significant antifungal activity. We have investigated the effect of two alkaloids viz., (-)-corydahnine and (-)-isocorypahnine isolated from Corydalis chaerophylla, against spore germination of some plant pathogenic and saprophytic fungal spores. Significant inhibition of spore germination at 100 ${\mu}g/ml$ was seen against Curvularia penniseti, Curvularia sp. and Colletotrichum gloeosporioides by (-)-corydahnine but (-)-isocorypalmine was also effective against fungi included in the experiment.

In Vitro Antifungal Activities of 13 Fungicides against Pepper Anthracnose Fungi (고추 탄저병균에 대한 13종 살균제의 살균 활성)

  • For the effective control of pepper anthracnose, in vitro antifungal activities of 13 commercial fungicides were tested on spore germination, mycelial growth, and sporulation of Colletotrichum gloeosporioides, anthracnose fungus. Among them, captan, chlorothalonil, dithianon, fluazinam and folpet completely inhibited the spore germination of C. gloeosporioides at the concentration of 0.8 ${\mu}g/ml$. They were followed by mancozeb and propineb, showing more than 80% inhibition of spore germination at 20 ${\mu}g/ml$. The mycelial growth of C. gloeosporioides was strongly inhibited by fluazinam and nuarimol. Except for nuarimol, most of the fungicides represented more antifungal activity on the spore germination than the mycelial growth of C. gloeosporioides. Azoxystrobin and metominostrobin, Strobilurin fungicides, were only moderately active against the spore germination and the mycelial growth of C. gloeosporioides, but they were effective antisporulant against C. gloeosporioides. From these results, control of pepper anthracnose will have achieved by preventive spray of the commercial fungicides.

Influence of Environmental Factors on Conidial Germination of Alternaria dauci (당근 검은잎마름병원균 Alternaria dauci의 포자 발아에 미치는 환경인자의 영향)

  • Effects of temperatures, relative humidity, pH, and dry periods on conidial germination of Alternaria dauci were evaluated under in vitro conditions. Conidial germination was accelerated at over 95% relative humidity in $15^{\circ}C$ to $25^{\circ}C$ condition. Conidial germination was rapidly reduced at 5 regardless of relative humidity conditions. More than 50% of the conidial germination were initiated within 2 h at $25^{\circ}C$ through pH 5 to 7. The highest conidial germination of A. dauci was on 0.2% of carrot leaf extract. Conidia could survive longer than 12 h, even though its germination decreased. After a 12 h dry period, around 10% of conidia revived and germinated when conidia were hydrated again. These results could be used as the useful information on conidial germination of A. dauci and ecology of Alternaria leaf blight.
