천연감미료 스테비오사이드와 스테비올의 생체내, 시험관내 유전독성평가 (In vitro and In vivo Evaluation of Genotoxicity of Stevioside and Steviol, Natural Sweetner)

  • The standard operation procedure of mouse lymphoma L5178Y $tk^{+/-}-3.7.2C$ gene mutation assay (MOLY) has been regarded as a sensitive in vitro mammalian cell gene mutation assay that is capable of detecting clastogens as well as mutagens. Using MOLY, one of natural sweetner, stevioside (5mg/ml) and its aglycon, steviol ($340{\;}\mu\textrm{g}/ml$) were evaluated the mutagenicity. Stevioside and steviol did not induce mutagenicity in MOLY. On the other hand, stevioside (250mg/kg, B.W.) and steviol (200mg/kg, B.W.) were also evaluated their ability to induce micronuclei in regenerating hepatocytes and bone marrow cells of ddY mice. From these results, stevioside and steviol did not induce any mutagenic effect both MOLY and in vivo micronucleus test.

  • Artificial heart is divided pulsation style and nonpulsation style greatly according to flowing of blood. nonpulsation pump is advantage of miniaturization avaliable because it is simple and non-volumic-pump than pulsation pump. Non pulsation pump is derided axial flow style and centrifugal style accordig to rotating style. An axial flow blood pump can be made smaller than a centrifugal blood pump because of its higher specific speed. A hemolysis is an important factor for the development of an axial flow blood pump. It is difficult to identify the areas where hemolysis nun. Evaluation of hemolysis both in in vitro and in vivo require a long time and are costly. Computational fluid dynamics(CFD) analysis enables the engineer to predict hemolysis on a computer. The aims of this study is Computational fluid dynamics in the whole axial flow pump and to verify the accuracy of prediction results of CFD analysis compare with in vitro experimental results.

In vitro Evaluation of Antidermatophytic Activity of Egyptian Bee Propolis in Combination with Plant Essential Oils in Sheep Hoof Plate: An Experimental Model

  • Bee propolis ethanolic extract with some plant essential oils was investigated for its antidermatophytic properties. The tested plant essential oils included jasmine, clove, lemon, Arabian jasmine, mint, rosa, olive and basil. The antidermatophytic activity has been compared to Naftifine-HCl and Clotrimazole used for dermatophyte treatment. Experimental model has been tested using sheep hoof plate for the in vitro tests to stimulate human nails. Mint, clove and basil with 4 mg/ml of bee propolis have a comparable efficacy to those of Naftifine-HCl and Clotrimazole. There is a great necessity for new effective low price and safe antidermatophyte agents to avoid recurrent infection. Propolis synergistic could be of great importance with essential oils of plants in dermatophyte therapy.

리팜피신 마이크로펠렛의 제조에 관한 연구 (Preparation and In-Vitro Evaluation of Gelatin Micropellets Containing Rifampicin)

  • The sustained-release micropellets containing rifampicin were prepared by spray congealing micropelleting technique using gelatin as the embedding matrix, and hardened by treating with the formalin-isopropanol mixture. Dissolution of rifampicin from micropellets was significantly retarded, and greatly dependent on formalin concentration, hardening time and pH of the dissolution medium. It was found that this prolongation was more distinguished in pH 1.2 dissolution medium rather than pH 7.4, which might be attributed to the swelling characteristics of gelatin used in the dissolution medium. In-vitro dissolution kinetics indicated that the drug release followed the first-order process.

Preparation and Evaluation of Novel Fenofibrate-loaded Self-Microemulsifying Drug Delivery System (SMEDDS)

  • Fenofibrate has been used for many years to lower cholesterol levels and its pharmacokinetic profile is well understood. However, due to its low solubility in water, it has low bioavailability after oral administration. In order to improve the dissolution rate, fenofibrate was formulated into a self-microemulsifying drug delivery systems (SMEDDS). We used pseudo-ternary phase diagrams to evaluate the area of microemulsification, and an in vitro dissolution test was used to investigate the dissolution rate of fenofibrate. The optimized formulation for in vitro dissolution assessment consisted of Lauroglycol FCC (60%), Solutol HS 15 (27%), and Transcutol-P (13%). The mean droplet size of the oil phase in the microemulsion formed from the SMEDDS was about 130 nm. The dissolution rate of fenofibrate from SMEDDS was significantly higher than that of the reference tablet. Our studies suggested that the fenofibrate containing SMEDDS composition can effectively increase the solubility and oral bioavailability of poorly water-soluble drugs.

Determination of Water Content in Skin by using a FT Near Infrared Spectrometer

  • The water content of skin was determined using a FT near infrared (NIR) spectrometer. NIR diffuse reflectance spectra were collected from hairless mouse, in vitro, and from human inner arm, in vivo. It was found that the variation of NIR absorbance band 1450 nm from OH vibration of water and 1940 nm from the combination involving OH stretching and OH deformation, depending on the absolute water content of separated hairless mouse skin, in vitro, using the FT NIR spectrometer. Partial least squares regression (PLSR) was applied to develop a calibration model. The PLS model showed good correlation. For practical use of the evaluation of human skin moisture, the PLS model for human skin moisture was developed in vivo on the basis of the relative water content of stratum corneum from the conventional capacitance method. The PLS model predicted human skin moisture with a standard errors of prediction (SEP) of 3.98 at 1130-1830 nm range. These studies showed the possibility of a rapid and nondestructive skin moisture measurement using FT NIR spectrometer.

Computer-aided design/computer-aided manufacturing of hydroxyapatite scaffolds for bone reconstruction in jawbone atrophy: a systematic review and case report

  • Background: We reviewed the biological and mechanical properties of porous hydroxyapatite (HA) compared to other synthetic materials. Computer-aided design/computer-aided manufacturing (CAD/CAM) was also evaluated to estimate its efficacy with clinical and radiological assessments. Method: A systematic search of the electronic literature database of the National Library of Medicine (PubMed-MEDLINE) was performed for articles published in English between January 1985 and September 2013. The inclusion criteria were (1) histological evaluation of the biocompatibility and osteoconductivity of porous HA in vivo and in vitro, (2) evaluation of the mechanical properties of HA in relation to its porosity, (3) comparison of the biological and mechanical properties between several biomaterials, and (4) clinical and radiological evaluation of the precision of CAD/CAM techniques. Results: HA had excellent osteoconductivity and biocompatibility in vitro and in vivo compared to other biomaterials. HA grafts are suitable for milling and finishing, depending on the design. In computed tomography, porous HA is a more resorbable and more osteoconductive material than dense HA; however, its strength decreases exponentially with an increase in porosity. Conclusions: Mechanical tests showed that HA scaffolds with pore diameters ranging from 400 to $1200{\mu}m$ had compressive moduli and strength within the range of the human craniofacial trabecular bone. In conclusion, using CAD/CAM techniques for preparing HA scaffolds may increase graft stability and reduce surgical operating time.

  • 생체적용 센서 코팅 재료로서 polydimethylsiloxane (PDMS)를 선정하였으며 합성 및 성형과정에서 용출될 수 있는 잔류 독성 물질의 세포독성을 확인하고자 하였다. ISO 10993-5, Biological evaluation of medical devices-Part 5 : Tests for in vitro cytotoxicity (의료기기의 생물 안정성 평가-제5부: 세포 독성 시험-체외시험)를 통하여 세포 독성 평가를 실시하였다. 양성 대조군으로 organo-tin을 사용하였으며 음성 대조군으로 혈청이 포함되지 않은 RPMI 1640배지를 사용하였다. 고체 시료의 표면적에 대하여 $125{\mu}L/cm^2$가 되도록 혈청이 포함되지 않은 RPMI 1640 배지를 용기에 첨가하였으며 $38^{\circ}C$를 유지하며 일정시간 동안 추출하였다. 세포 독성 평가는 1) NIH 3T3 fibroblast 단일세포층을 형성한 후 추출물을 첨가하는 방법과 2) 세포와 함께 추출물을 넣어 배양하는 방법을 동시에 시행하였다. 세포 형태학적인 변화 관찰과 MTT (tetrazolium dye, 2-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide) 시험법에 의한 세포 활성 측정을 병행함으로써 고체 시료로부터 추출된 물질의 세포독성 여부와 고체시료의 표면에 대한 세포의 감응성도 함께 관찰할 수 있었다.

  • An in vitro and in vivo studies were conducted to evaluate the antibacterial efficacy of certain botanicals viz., rhizomes of turmeric (Curcuma longa) and leaves of amla (Phyllanthus emblica), asparagus (Asparagus racemosus), bael (Aegle marmelos), boerhavia (Boerhavia diffusa), garlic (Allium sativum) and basil (Oscimum basicilum) against bacterial pathogens viz., Staphylococcus sp., Bacillus sp. and Klebsiella cloacae, of silkworm, Bombyx mori. Asparagus and basil, amla and boerhavia, basil and bael at concentration of 20, 000 ppm showed higher antibacterial activity against Staphylococcus sp., Bacillus sp., K. cloacae respectively, both in vitro and in vivo studies.

In vitro Evaluation of AraC Prodrugs for Their Antiviral Activity

  • The araC prodrugs (1~5) carrying a special acyl group at 5'-Ο-or $N^4$-position were evaluated for in vitro antiviral activity against various human viruses. When tested against HSV-1 and HSV-2 cultured in the verso cells, the prodrugs exhibited slightly higher $ED_{50}$ values compared with one of the parent araC but showed more increased $CC_{50}$ values in all cases. Consequently the overall selectivity indexes of prodrugs were higher than that of arab. The prodrugs, except compound 5, exhibited very potent activity similar to that of araC ($ED_{50}$ about $0.12{\mu}g/mι$) when evaluated against another human DNA virus, cytomegarovirus. However, theses araC prodrugs were completely inactive against RNA viruses i.e. poliovirus and coxackie B3 virus at the concentration of 4250{\mu}g/mι.$
