IN SITU RUMINAL DEGRADATION KINETICS OF FORAGES AND FEED BYPRODUCTS IN MALE NILI-RAVI BUFFALO CALVES

  • The rate and extent of digestion of dietary carbohydrates has a tremendous impact on ruminal fermentation and the productivity of the animals. The objective of the study was to determine the dry matter (DM) and neutral detergent fiber (NDF) degradabilities and rate and extent of feed byproducts (cotton seed cake, wheat bran), legumes [berseem (Egyptian clover), lucern (Medicago sativa), cowpeas (Vigna sinensis)], grasses [maize (Zea mays), millet (Panicum miliaceum), sorghum (Sorghum vulgare)] and wheat straw in ruminally fistulated male buffalo calves. By using nylon bags, 10 grams sample was exposed to the ruminal fermentation for 0, 1, 2, 4, 6, 10, 16, 24, 36, 48 and 96 hours. Dry matter and NDF degradability was measured at 48 hours. Extent of DM and NDF disappearance was determined at each time point. Rates of disappearance of DM and NDF were determined by regressing the natural logarithm of the percentage of original DM and NDF remaining in the bags between 1 and 96 hours. The dry matter digestibility (DMD) of the feed byproducts (FBP) and legume forages when incubated in the rumen of male buffalo calves were greater (p < 0.05) than grasses. Extent of digestion followed similar pattern as DMD. Rate of DMD was higher in FBP than in legumes and was the lowest in the wheat straw. The NDF degradability (NDFD) of FBP, legumes and grasses did not differ, however, wheat straw had the lowest NDFD from all the feeds tested. The lowest NDFD of wheat straw may have been due to the depressing effect of lignin on fiber digestion. The FBP and legumes had higher (p < 0.05) rates and lower extents of NDF digestion than grasses.

Metabolisable Energy, In situ Rumen Degradation and In vitro Fermentation Characteristics of Linted Cottonseed Hulls, Delinted Cottonseed Hulls and Cottonseed Linter Residue

  • Dietary supplementation with conventional linted cottonseed hulls (LCSH) is a common practice in livestock production all over the world. However, supplementation with mechanically delinted cottonseed hulls (DCSH) and cottonseed linter residue (CLR) is uncommon. Cottonseed by-products, including LCSH, DCSH and CLR, were assessed by chemical analysis, an in situ nylon bag technique, an in vitro cumulative gas production technique and in vitro enzyme procedure. The crude protein (CP) content of CLR (302 g/kg dry matter (DM)) was approximately 3 times that of LCSH and 5 times that of DCSH. The crude fat content was approximately 3 times higher in CLR (269 g/kg DM) than in LCSH and 4 times higher than in DCSH. Neutral detergent fibre (311 g/kg DM) and acid detergent fibre (243 g/kg DM) contents of CLR were less than half those of DCSH or LCSH. Metabolisable energy, estimated by in vitro gas production and chemical analyses, ranked as follows: CLR (12.69 kJ/kg DM)>LCSH (7.32 kJ/kg DM)>DCSH (5.82 kJ/kg DM). The in situ degradation trial showed that the highest values of effective degradability of DM and CP were obtained for CLR (p<0.05). The in vitro disappearance of ruminal DM ranked as follows: CLR>LCSH>DCSH (p<0.05). The lowest digestibility was observed for DCSH with a two-step in vitro digestion procedure (p<0.05). The potential gas production in the batch cultures did not differ for any of the three cottonseed by-product feeds. The highest concentration of total volatile fatty acids was observed in CLR after a 72 h incubation (p<0.05). The molar portions of methane were similar between all three treatments, with an average gas production of 22% (molar). The CLR contained a higher level of CP than did LCSH and DCSH, and CLR fermentation produced more propionate. The DCSH and LCSH had more NDF and ADF, which fermented into greater amounts of acetate.

Ruminal Degradability of Tropical Feeds and Their Potential Use in Ruminant Diets

  • The objective of this study was to determine the degradability of cassava chip (CC), cassava waste (CW), yellow sweet potato (YP), white sweet potato (WP), purple sweet potato (PP), corn meal (CM), and rice bran (RB) using in situ technique. Two ruminally fistulated steers with an average weight of $303{\pm}10kg$ were used to determine in situ degradabilities of DM and OM. Seven feed sources were weighted in nylon bags ($38{\mu}m$ pore size) and incubated ruminally for 1, 2, 4, 6, 8, 12, 24, and 48 h. The results showed that asymptote (a+b) and effective degradability (ED) of DM of energy sources ranked from the highest to the lowest; CC, YP, WP, PP, RB, CW, and CM (99.3, 92.5; 97.6, 87.9; 97.5, 87.9; 97.2, 87.8; 87.5, 63.6; 78.6, 63.0 and 81.7; 59.3, respectively) and for OM asymptote (a+b) and effective degradability (ED) were similar to those of degradation of DM (99.4, 93.4; 98.8, 89.8; 98.5, 89.4; 98.4, 88.1; 92.4, 65.8; 85.1, 66.9 and 83.6, 63.3, respectively). It was concluded that disappearance characteristic of CC was the highest and it may potentially facilitate the achievement of optimal ruminal availability of energy: protein especially with NPN for microbial protein synthesis.

The Effect of Yerba Mate (Ilex Paraguariensis) Supplementation on Nutrient Degradability in Dairy Cows: An In sacco and In vitro Study

  • This study was carried out to investigate the effects of Yerba Mate (YM) supplementation on nutrients' degradation, in vitro dry matter disappearance, gas production and rumen ammonia concentration. Three rumen-fistulated Holstein Friesian cows were used for the in situ incubations and provided rumen liquor for in vitro incubations. The inclusion of YM in a control diet (pasture+pellets) affected some in sacco degradation parameters. YM supplementation decreased the effective degradability and degradation rate of pasture crude protein (CP), and it seems to slow down the degradation of pasture neutral detergent fiber. A significant increase of degradation of pasture acid detergent fiber (ADF) was detected after YM inclusion in the control diet. YM supplementation reduced in vitro gas production of pasture and ammonia concentration of pellets. The addition of YM in ruminant diet could decrease ammonia production and increase protein availability for productive purposes. The moderate presence of tannins in YM could have affected the degradation kinetics of pasture CP and ADF and the ammonia production of pellets.

Apoptosis of bursal lymphoid cells in chickens experimentally infected with IBDV(SH/92)

  • Chickens at 3-weeks of age were inoculated with a highly virulent strain (SH/92) of Infectious Bursal Disease Virus(IBDV) through ocular and cloacal routes. The infected chickens were killed at 6, 12, 24, 48, 72, and 96 hrs post inoculation (PI) and Bursa of Fabricius(BF) were collected. The sizes of bursal follicules in infected chickens decreased at 48 to 96 hrs PI. Histologically the cellular changes were first evident at 12 hrs PI and characterized by condensation of nuclear chromatin of bursal lymphocytes indicating apoptosis. By 24 hrs PI apoptotic lymphocytes dramatically increased. In addition infiltration of heterophils were also seen in the follicles and in the interfollicular connective tissues. At 48 hrs PI, cystic cavities were observed in the follicles. As the infection advanced the bursal follicles showed atrophy accompanied by disappearance of heterophils and reduction in number of lymphocytes in the cystic cavities which was replaced by proteineous materials. The nuclei of most affected lymphocyte stained positively with the in situ end labeling for apoptosis. Electron microscopy showed viral particles with crystalline array in the lymphocytes of BF infected with IBOV. These results indicated that SH/92 IBDV infection in chickens caused increased apoptosis in the BF.

  • A study on the absorption mechanism of cefixime(CF), an oral ${\alpha}-amino$ group deficient cephalosporin antibiotic, has been undertaken through the rat jejunum and nasal cavity using an in situ simultaneous perfusion technique developed in our laboratory. CF was well absorbed in the jejunum and nasal cavity of rats at pH 5.0, but not at pH 7.0. CF absorption was studied over four orders of magnitude in concentration to determine saturability. Disappearance of CF in the perfusate followed first-order kinetics at all tested concentrations. The apparent first-order absorption rate constant was found to be dependent on the concentration over the range of $0.1\;mM{\sim}3\;mM$ in the jejunum and nasal cavity of rats. Inhibitors were added to determine the competitive inhibition of CF absorption. The presence of L-tyrosine, L-phenylalanine, alanine-alanine, glycine-glycine and cefadroxil produced the significant inhibition of CF absorption in the nasal cavity and jejunum. However, there was no evidence of the inhibition in the presence of cefazolin. In addition, The CF absorption in the nasal cavity and jejunum was inhibited significantly by ouabain and 2,4-dinitrophenol(DNP). This study suggested that CF is absorbed across the rat nasal cavity and jejunum by carrier-mediated transport mechanism and energy consuming system.

A Study in the neighbouring sea variation of Cheju and Influence of China Coastal Water by Topex/Poseidon Altimeter Data and in-situ Salinity Data

  • Appearance and disappearance of the China Coastal Waters(CCW) in the neighbouring sea of Cheju Island was very different yearly but usually appeared strongly in summer. At this time, sea level and salinity were varied in this area by the influence of the CCW. Satellite data(T/P;Topex/Poseidon) and Salinity (NFRID;National Fisheries Research and Development Institute) were used from 1993 to 2001. We compared with TG data of NOR I and TIP data in the observed station(33 31'N, 12632'E). Coefficient of correlation was 0.6~0.8 with the exception of 1993 and 1995. And variations of salinity was higher than $32.00\%_{\circ}$ in the southwestern part of Cheju Island and the southern part of the South Sea of Korea during June-October and SLA(Sea level Anomaly) was 10-11cm. Salinity of the southeastern part was higher than those of the southwestern part and SLA was 12~13cm because of the influence of Tsushima Current.

Ruminal Solubility of Trace Elements from Selected Philippine Forages

  • The ruminal solubility of copper (Cu), manganese (Mn), molybdenum (Mo), and zinc (Zn) in eight Philippine forages was studied. These forages were: 1) grasses: paragrass (Brachiaria mutica (Forsk.) Stapf), stargrass (Cynodon plectostachyum Pilger), and napiergrass (Pennisetum purpureum Schumach.); 2) creeping legumes: calopogonium or calopo (Calopogonium muconoides Desv.) and centrocema (Centrocema pubescens Benth.); 3) tree lequmes: gliricidia or kakawate (Gliricidia sepium (Jacq.) Walp.), leucaena or ipil-ipil (Leucaena leucocephala (Lam.) de Wit.), and sesbania or katuray (Sesbania grandiflora (L.) Poir). Nylon bags with samples were incubated for 0, 3, 6, 12, 24, 48 and 72h in three rumen cannulated sheep fed with timothy hay (Phleum pratense L.) at 2% body weight/d. The 0-h bags were washed with deionized water. There were species differences (p < 0.05) on the different solubilities of trace elements. At O-h incubation, the trend of solubility was Mo (54%) > Zn (43%) > Cu (38.7%) > Mn (29.5%). At 3-h incubation, all the elements except Mn had an average solubility above 50%. Combining particulate passage rate (1.9%/h) and various trace elements disappearance rates, the effective ruminal solubilization (ERS) of trace elements were computed. The ERS across species ranged from 44.6 to 89.9% for Cu, 29.9 to 84% for Mn, 66 to 95.1% for Mo, and 30.1 to 82.3% for Zn resulting to a trend of Mo > Zn > Cu > Mn.

Replacing Concentrate with Wheat Straw Treated with Urea Molasses and Ensiled with Manure: Effects on Ruminal Characteristics, In situ Digestion Kinetics and Nitrogen Metabolism of Nili-Ravi Buffalo Bulls

  • To evaluate the effects of replacing concentrate with urea molasses treated fermented wheat straw (FWS) ensiled with cattle manure (CM) on ruminal characteristics, in situ digestion kinetics and nitrogen (N) metabolism was studied in Nili Ravi cannulated buffalo bulls in a $4{\times}4$ Latin Square Design. Wheat straw treated with urea (4%) and molasses (6%) was ensiled with cattle manure (CM) (70:30) and fermented for 40 days. Four iso-nitrogenous and iso-caloric diets were formulated. In the FWS0, FWS10, FWS20 and FWS30 diets 0, 10, 20 and 30% of the concentrate was replaced with FWS, respectively. Daily intake by bulls was restricted to 1.5% dry matter (DM) of body weight. Ruminal ammonia nitrogen concentration was greater (p<0.05) in bulls fed FWS diet than for those fed FWS0 diet at 3, 6, 9 and 12 h post-parandial. Bulls fed FWS 20 and FWS 30 diets had higher ruminal pH at 3 and 6 h post-parandial than bulls fed FWS10 and FWS0. Ruminal total volatile fatty acid (VFA) concentrations 3 h post-parandial were greater (p<0.05) in bulls fed FWS0 than those fed FWS diets. However ruminal VFA tended to increase at 6, 9 and 12 h post-parandial as the level of FWS increased. In situ ruminal DM and neutral detergent fiber (NDF) degradation, rates of disappearance and extent of digestion were higher (p<0.05) for bulls fed FWS30 diet than those fed FWS0. Ruminal DM and NDF lag time tended to decrease (p<0.05) as FWS concentration in the diet increased. Feed intake, nitrogen intake, N-balance and blood urea-N did not differ (p>0.05) in buffalo bulls fed different diets. Wheat straw treated with urea and molasses and ensiled with CM enhanced the nutritive value of wheat straw and improved nutrient utilization in buffalo bulls when up to 30% of the concentrate was replaced with FWS; no adverse effects on ruminal characteristics and nutrients digestibilities were detected.

주요 조사료원과 쑥, 녹차의 반추위 건물 및 조단백질 소화율에 대한 분석 (Analysis of Ruminal Dry Matter and Crude Protein Digestibility on Major Roughage, Wormwood and Green Tea)

  • 본 연구는 국내 주요 조사료(rice straw, timothy and alfalfa) 및 쑥과 녹차의 반추위 소화율과 물리적 구조를 비교하기 위해 in vitro 와 in situ 실험을 실시하였다. 각 사료별 3, 6, 9, 12, 18, 24, 36, 48 및 72 시간 동안 배양하였고, in vitro 실험에서는 가스 발생량, 미생물 성장량, pH를 측정하였다. 가스발생량은 배양시간이 경과할 수록 모든 시간대에서 증가하였고(p<0.05), 녹차에서의 가스발생량이 가장 낮았다. 미생물 성장량은 배양시간이 경과할수록 모든 처리구에서 증가하는 경향이었으나, 유의차는 나타나지 않았고(p<0.05), 쑥과 녹차의 미생물 성장량이 다른 처리구에 비해 낮게 나타났다(p<0.05). 반추위내 pH는 배양시간 경과할 수록 감소하였고, 티모시의 pH가 다른 사료에 비해 가장 낮았으며, 볏짚의 pH가 다른 사료에 비해 높았다(p<0.05). In situ 실험에서 모든 시간대 녹차의 건물소화율(DM; Dry Matter) 및 조단백 소화율(CP; Crude Protein)은 가장 높았다(p<0.05). 또한 반추위내 통과 속도 4% 쑥에서 건물소화율이 가장 높았고, 알팔파는 조단백 소화율이 가장 높았다(p<0.05). 녹차와 쑥의 표면과 기공을 주사전자현미경(SEM; scanning electron microscope)으로 관찰한 결과, 쑥 표면에는 미생물이 존재하지 않아 반추위 내 영양소 소화율이 낮은 것으로 사료되고, 녹차는 기공에 미생물이 관찰 되었다. In vitro 및 in situ 실험의 결과를 통하여 반추동물의 사료원료로 잠재적인 가치가 있을 것으로 사료된다.
