발효마늘 추출물의 항혈전 및 심혈관개선 효과 (The Effects of Anti-Thrombotic Activities and Cardiovascular Improvement of Fermented Garlic Extracts)

  • 이 연구의 목적은 발효 마늘 추출물의 항혈전 활성 및 심혈관개선 효과를 조사하고자 하였다. 심혈관 질환(CVD)의 발생률은 선진국에서 빠르게 증가하고 있으며, CVD는 현재 이환율과 사망률의 주요 원인을 나타내고 있다. 천연물 및 대체의학은 CVD의 위험을 감소시키는 것으로 알려져 있다. 마늘은 항염증제, 항산화제 및 항혈소판 활동을 위해 전 세계적으로 사용되는 약용식물이다. 우리는 발효 과정에서 생산된 생리활성 화합물의 생체 이용률이 증가함에 따라 이들 제품의 발효 제제가 발효되지 않은 형태보다 강력한 항혈소판 효과를 가질 수 있다는 가설을 세웠다. 따라서, 본 연구자는 쥐 혈소판으로부터 표준 광투과 응집 측정법 및 생체외 과립 분비를 사용하여 시험관내 및 생체외 혈소판 응집 분석을 통해 이들 화합물을 비교 분석 하였다. 발효된 조제물이 시험관내 및 생체외 양쪽에서 혈소판 응집이 보다 강력하고 유의적인 억제를 발휘한다는 것을 발견 하였다. 마찬가지로, 조밀한 혈소판 과립으로부터의 ATP 방출은 발효되지 않은 조제-처리된 쥐의 혈소판과 비교하여 발효된 조제-처리된 쥐 혈소판에서 유의하게 억제되었다. 결론적으로 발효 과정이 발효중에 생산된 활성 화합물의 생체 이용률이 증가한 결과 시험관내 및 생체외에서 혈소판 기능에 더 강력한 효과를 발휘한다고 확인 할 수 있었다. 따라서 발효 제품은 혈소판 관련 CVD에 대한 강력한 치료제로서 항혈소판 및 항혈전제로 사용될 수 있다.

Shear Stress and Atherosclerosis

  • Hemodynamic shear stress, the frictional force acting on vascular endothelial cells, is crucial for endothelial homeostasis under normal physiological conditions. When discussing blood flow effects on various forms of endothelial (dys)function, one considers two flow patterns: steady laminar flow and disturbed flow because endothelial cells respond differently to these flow types both in vivo and in vitro. Laminar flow which exerts steady laminar shear stress is atheroprotective while disturbed flow creates an atheroprone environment. Emerging evidence has provided new insights into the cellular mechanisms of flowdependent regulation of vascular function that leads to cardiovascular events such as atherosclerosis, atherothrombosis, and myocardial infarction. In order to study effects of shear stress and different types of flow, various models have been used. In this review, we will summarize our current views on how disturbed flow-mediated signaling pathways are involved in the development of atherosclerosis.

Water soluble tomato concentrate regulates platelet function via the mitogen-activated protein kinase pathway

  • Tomato extract has been shown to exert antiplatelet activity in vitro and to change platelet function ex vivo, but with limitations. In this study, antiplatelet activity of water soluble tomato concentrate (Fruitflow I) and dry water soluble tomato concentrate (Fruitflow II) was investigated using rat platelets. Aggregation was induced by collagen and adenosine diphosphate and granule-secretion, $[Ca^{2+}]_i$, thromboxane B2, cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP) levels were examined. The activation of integrin ${\alpha}_{IIb}{\beta}_3$ and phosphorylation of signaling molecules, including mitogen-activated protein kinase (MAPK) and PI3K/Akt, were investigated by flow cytometry and immunoblotting, respectively. Prothrombin time (PT) and activated partial thromboplastin time (aPTT) were examined. Moreover, in vivo thrombus weight was tested by an arteriovenous shunt model. Fruitflow I and Fruitflow II significantly inhibited agonist induced platelet aggregation, adenosine triphosphate and serotonin release, $[Ca^{2+}]_i$, and thromboxane B2 concentration, while having no effect on cAMP and cGMP levels. Integrin ${\alpha}_{IIb}{\beta}_3$ activation was also significantly decreased. Moreover, both concentrates reduced phosphorylation of MAPK pathway factors such as ERK, JNK, P38, and PI3K/Akt. In vivo thrombus formation was also inhibited. Taken together, these concentrates have the potential for ethnomedicinal applications to prevent cardiovascular ailments and can be used as functional foods.

Polar Body: Indicator of Oocyte's Maturation, Have Any Function on Oocyte?

  • Polar body was usually used as a determinant of oocyte's maturation. Polar body morphology could reflect the embryo quality and implantation competence. This review only focuses on morphology of the first polar body and embryo developmental rate in the presence or absence of polar body. However, it is very difficult to describe whether polar body has any effects on embryo development in vitro or in vivo. Further intensive research is needed to determine its function on embryo development.

  • 가바성 억제 신경세포는 해마의 정상적인 기능을 조절하는데 중요한 역할을 하며 해마 병변을 유발하는 중요한 요소이다. 본 연구는 in vivo 실험법을 사용하여 해마 CA1 영역에서의 전기 생리학적 반응을 측정함으로써 가바성 신경세포의 기능을 분석하고 이를 슬라이스 실험법과 비교하고자 하였다. Fimbria-fonix 전기자극시 전형적인 population spike가 나타났고 $10{\sim}M$ bicuculline 존재하에서는 전기자극에 의해 burst 형태의 population spike가 나타났다. Population spike의 크기는 자극 강도에 비례하였으며 그 숫자도 bicuculline 전극사용시와 같이 동일한 양상을 보였다. CA1 영역의 흥분성 수준을 측정하기 위해 paired-pulse 자극을 하였는데 짧은 자극 간격에서 억제성 반응을 보였고 burst형태의 afterdischarge를 나타내었다. CA1 영역에서 in vivo실험법을 사용한 가바성 신경세포반응의 결과는 추체세포의 흥분성 조절을 효과적으로 분석할 수 있으며 in vitro 실험법에 비해 기능적 평가가 더욱 이상적임을 알 수 있다.

  • Ginseng saponin from different part of Ginseng and 7 kinds of ginsenosides effect on immnotuncition of young and old mice. There have up-regulation effect in vivo and in vitro experiments for low function of old mice, but not up-regulation effect at normal immuno-function of young mice. That means "Adaptogens" of Ginseng saponin. Not appositions effect at Pg1 and Rb1 and have find, that effect relations with molecular weight of ginsenosides.senosides.

Study of the cartilage matrix production-promoting effect of chicken leg extract and identification of the active ingredient

  • BACKGROUND/OBJECTIVES: Osteoarthritis (OA) is a major public health issue in Japan and other countries, and foods that prevent or treat OA are in strong demand. Proteins and peptides in chicken meat and bones are known for being rich in functional and nutritional ingredients for the improvement of osteoporosis. We speculated that chicken legs, a food consumed in many regions of the world, may also contain such ingredients. In this study, we aim to (i) evaluate the effect of chicken leg extract (CLE) on the promotion of cartilage matrix production and (ii) identify the active ingredient in CLE that contributes to this function. MATERIALS/METHODS: Artificial CLE digest was prepared, and the acid mucopolysaccharide production-promoting activity of the CLE digest was evaluated by alcian blue staining of ATDC5 cells. CLE was orally administered to rabbits with burr holes in the knee joint of the femur, and the degree of regeneration of cartilage matrix was evaluated. Furthermore, we investigated orally administered CLE-derived peptides in human plasma using LC-MS. From measuring the acid mucopolysaccharide production-promotion activity of these peptides, a molecule considered to be an active ingredient in the CLE digest was identified. RESULTS: CLE digest promoted acid mucopolysaccharide production and facilitated regeneration of cartilage matrix in in vitro and in vivo experiments. Four peptides including phenylalanyl-hydroxyproline (Phe-Hyp) were detected as CLE-derived peptides in human plasma. The effect of CLE was inferred to be due to Phe-Hyp, which was confirmed to be present in the CLE digest. CONCLUSIONS: It was shown that CLE stimulated the production of articular cartilage matrix both in vitro and in vivo, and that CLE could be an effective food for preventing or treating OA. Furthermore, only Phe-Hyp was confirmed as the active compound in the CLE digest, suggesting that the activity of CLE was due to Phe-Hyp.

Ginsenoside Rd alleviates mouse acute renal ischemia/reperfusion injury by modulating macrophage phenotype

  • Background: Ginsenoside Rd (GSRd), a main component of the root of Panax ginseng, exhibits anti-inflammation functions and decreases infarct size in many injuries and ischemia diseases such as focal cerebral ischemia. M1 Macrophages are regarded as one of the key inflammatory cells having functions for disease progression. Methods: To investigate the effect of GSRd on renal ischemia/reperfusion injury (IRI) and macrophage functional status, and their regulatory role on mouse polarized macrophages in vitro, GSRd (10-100 mg/kg) and vehicle were applied to mice 30 min before renal IRI modeling. Renal functions were reflected by blood serum creatinine and blood urea nitrogen level and histopathological examination. M1 polarized macrophages infiltration was identified by flow cytometry analysis and immunofluorescence staining with $CD11b^+$, $iNOS^+$/interleukin-12/tumor necrosis factor-${\alpha}$ labeling. For the in vitro study, GSRd ($10-100{\mu}g/mL$) and vehicle were added in the culture medium of M1 macrophages to assess their regulatory function on polarization phenotype. Results: In vivo data showed a protective role of GSRd at 50 mg/kg on Day 3. Serum level of serum creatinine and blood urea nitrogen significantly dropped compared with other groups. Reduced renal tissue damage and M1 macrophage infiltration showed on hematoxylin-eosin staining and flow cytometry and immunofluorescence staining confirmed this improvement. With GSRd administration, in vitro cultured M1 macrophages secreted less inflammatory cytokines such as interleukin-12 and tumor necrosis factor-${\alpha}$. Furthermore, macrophage polarization-related pancake-like morphology gradually changed along with increasing concentration of GSRd in the medium. Conclusion: These findings demonstrate that GSRd possess a protective function against renal ischemia/reperfusion injury via downregulating M1 macrophage polarization.

  • Numerous investigators have studied various activities of natural products and have found that they have not only nutritional effects, but also beneficial properties to cure various diseases and to maintain good health. Job's Tear(Yul-Moo) is a grass crop that has long been used in traditional medicine and as a nourishing food. Although its mechanism of action remains unclear, Job's Tear has been reported to exhibit anti-inflammatory, stomachic, anti-allergic, and anti-spastic effects and has been used in China for the treatment of warts, rheumatism, and neuralgia. Previous results in our laboratory demonstrated that the ethanol extract and the water extract of Job's Tear exerted an immune regulatory function on mice cells in vitro. The present study was performed to investigate the ex vivo effect of Job's Tear on immune function. Seven to eight weeks old mice(Balb/c) were fed chow diet ad libitum and water extract of Job's Tear was administered orally every other day for four weeks at two different concentrations(50 and 500mg/kg B.W.). Splenocytes proliferation with mitogen stimulation with Con A and LPS was enhanced at 50 mg/kg B.W. of Job's Tear compared to those of the control group. The results of this ex vivo study showed that proliferation of splenocytes and macrophage activation were seen in the mice orally administrated 50 mg/kg B.W. of Job's Tear water extracts. In conclusion, this study suggests that Job's Tear extracts may enhance immune function by regulating splenocyte proliferation and the cytokine prodution capacity of activated macrophages in mice.

TAGLN2-mediated actin stabilization at the immunological synapse: implication for cytotoxic T cell control of target cells

  • Actin dynamics is critical for the formation and sustainment of the immunological synapse (IS) during T cell interaction with antigen-presenting cells (APC). Thus, many actin regulating proteins are involved in spatial and temporal actin remodeling at the IS. However, little is known whether or how actin stabilizing protein controls IS and the consequent T cell functions. TAGLN2 − an actin-binding protein predominantly expressed in T cells − displays a novel function to stabilize cortical F-actin, thereby augmenting F-actin contents at the IS, and acquiring leukocyte function-associated antigen-1 activation following T cell activation. TAGLN2 also competes with cofilin to protect F-actin in vitro and in vivo. During cytotoxic T cell interaction with cancer cells, the expression level of TAGLN2 at the IS correlates with the T cell adhesion to target cancer cells and production of lytic granules such as granzyme B and perforin, thus expressing cytotoxic T cell function. These findings identify a novel function for TAGLN2 as an actin stabilizing protein that is essential for stable immunological synapse formation, thereby regulating T cell immunity.
