제대혈 유래 중간엽줄기세포에서 HLA의 발현과 Mixed Lymphocyte Reaction (Expression of HLA and Mixed Lymphocyte Reaction of Mesenchymal Stem Cells Derived from Human Umbilical Cord Blood)

  • In recent years, the mesenchymal stem cells (MSC) derived from various tissues have been widely tested for developing cell therapies, tissue repair and transplantation. Although there has been much interest in the immunomodulatory properties of MSC and their immunologic reactions following autologous, allogeneic and xenogenic transplantation of MSC in vivo, up to date, the expression of immunogenic markers, such as class I and II human leukocyte antigens (HLA), after differentiation of human umbilical cord blood (hUCB)-derived MSC has been poorly investigated and require extensive in vitro and in vivo testing. In this experiment, the expression of the HLA-ABC and HLA-DR on hUCB-derived MSC have been tested by immunocytochemical staining. The undifferentiated MSC were moderately stained for HLA-ABC but very weakly for HLA-DR. In order to investigate the inhibitory effect of allogeneic lymphocytes on proliferation of MSC, the MSC were cultured in the presence or absence of peripheral allogeneic lymphocytes stimulated with concanavalin A. The allogeneic lymphocytes did not significantly inhibit MSC proliferation. We conclude that hUCB-MSC expressed moderately class I HLA antigen while almost negatively class II HLA antigen. The MSC have an immunomodulatory effect which can suppress the allogeneic response of lymphocytes. These in vitro data suggest that allogeneic MSC derived from cord blood can be useful candidate for allogeneic cell therapy and transplantation without a major risk of rejection.

밀리타리스 동충하초(Cordyceps militaris) 에탄올 추출물의 면역억제 마우스 면역활성에 미치는 영향 (Immunomodulatory Activities of Ethanol Extract of Cordyceps militaris in Immunocompromised Mice)

  • 본 연구에서는 면역억제 동물모델에서 밀리타리스 동충하초 50% 에탄올 추출물의 면역력 증강 기능을 평가하였다. 이를 위하여 C57BL/6 마우스에 cyclophosphamide를 2회 복강주사 하여 면역력을 억제한 후, 밀리타리스 동충하초 추출물을 30, 100, 300 mg/kg 용량으로 12일간 경구투여 하였다. 마우스를 희생하여 몸무게 및 면역장기 무게, 비장세포의 증식, 비장세포의 cytokine 분비능, NK 세포 활성을 측정하였다. 그 결과, cyclophosphamide 투여에 의한 면역억제는 마우스의 몸무게와 간의 무게에 영향을 주지 않았으나 흉선의 무게는 감소시켰고 비장의 무게는 증가시켰다. 밀리타리스 동충하초 추출물 투여는 마우스의 몸무게 및 면역장기 무게에 영향을 주지 않았다. Cyclophosphamide 투여는 비장세포의 증식능을 감소시켰으며 밀리타리스 동충하초 추출물은 용량 의존적으로 비장세포 증식을 증가시켜 실험에 사용한 전 용량에서 비장세포의 유의적인 증식효과를 보였다. 비장세포의 cytokine 분비능을 측정한 결과, 밀리타리스 동충하초 추출물 투여는 IL-2, IL-12, IFN-${\gamma}$, TNF-${\alpha}$ 같은 Th1 cytokine의 분비를 대조군에 비해 유의적으로 증가시켰으나, IL-4와 IL-10 같은 Th2 cytokine의 분비에는 영향을 미치지 않았다. 또한 cyclophosphamide는 NK 세포의 활성을 정상군에 비하여 유의적으로 감소시켰으며, 밀리타리스 동충하초 추출물 투여는 cyclophosphamide에 의해 저하된 NK 세포 활성을 현저하게 증가시켰다. 이상의 결과를 종합해 볼 때, 밀리타리스 동충하초는 면역력이 억제된 상황에서 면역력을 증강시키며, 이러한 면역력 증강 효과는 체액성 면역보다 세포성 면역력 증강에 기인하는 것으로 보인다.

The activation of NLRP3 inflammasome potentiates the immunomodulatory abilities of mesenchymal stem cells in a murine colitis model

  • Inflammasomes are cytosolic, multiprotein complexes that act at the frontline of the immune responses by recognizing pathogen- or danger-associated molecular patterns or abnormal host molecules. Mesenchymal stem cells (MSCs) have been reported to possess multipotency to differentiate into various cell types and immunoregulatory effects. In this study, we investigated the expression and functional regulation of NLR Family Pyrin Domain Containing 3 (NLRP3) inflammasome in human umbilical cord blood-derived MSCs (hUCB-MSCs). hUCB-MSCs expressed inflammasome components that are necessary for its complex assembly. Interestingly, NLRP3 inflammasome activation suppressed the differentiation of hUCB-MSCs into osteoblasts, which was restored when the expression of adaptor proteins for inflammasome assembly was inhibited. Moreover, the suppressive effects of MSCs on T cell responses and the macrophage activation were augmented in response to NLRP3 activation. In vivo studies using colitic mice revealed that the protective abilities of hUCB-MSCs increased after NLRP3 stimulation. In conclusion, our findings suggest that the NLRP3 inflammasome components are expressed in hUCB-MSCs and its activation can regulate the differentiation capability and the immunomodulatory effects of hUCB-MSCs.

Immunomodulatory Properties of Lactobacillus plantarum NC8 Expressing an Anti-CD11c Single-Chain Fv Fragment

  • The lactic acid bacteria species Lactobacillus plantarum (L. plantarum) has been used extensively for vaccine delivery. Considering to the critical role of dendritic cells in stimulating host immune response, in this study, we constructed a novel CD11c-targeting L. plantarum strain with surface-displayed variable fragments of anti-CD11c, single-chain antibody (scFv-CD11c). The newly designed L. plantarum strain, named 409-aCD11c, could adhere and invade more efficiently to bone marrow-derived DCs (BMDCs) in vitro due to the specific interaction between scFv-CD11c and CD11c located on the surface of BMDCs. After incubation with BMDCs, the 409-aCD11c strain harboring a eukaryotic vector pValac-GFP could lead to more efficient expression of GFP compared with wild-type strains shown by flow cytometry analysis, indicating the enhanced translocation of pValac-GFP from L. plantarum to BMDCs. Similar results were also observed in an in vivo study, which showed that oral administration resulted in efficient expression of GFP in both Peyer's patches (PP) and mesenteric lymph nodes (MLNs) within 7 days after the last administration. In addition, the CD11c-targeting strain significantly promoted the differentiation and maturation of DCs, the differentiation of $IL-4^+$ and $IL-17A^+$ T helper (Th) cells in MLNs, as well as production of $B220^+$ $IgA^+$ B cells in the PP. In conclusion, this study developed a novel DC-targeting L. plantarum strain which could increase the ability to deliver eukaryotic expression plasmid to host cells, indicating a promising approach for vaccine study.

유근피 추출물이 대식세포 면역조절에 미치는 영향 (Immunomodulatory Activity of Water Extract of Ulmus macrocarpa in Macrophages)

  • 왕느릅나무(Ulmus macrocarpa)의 껍질을 말린 유근피는 오랫동안 부종, 감염 및 염증 제어의 목적으로 사용되어져 왔음에도 불구하고 잠재적 면역조절 효과에 관해서는 연구가 이루어진 바 없다. 본 연구에서는 전통 약용자원에서 새로운 면역기능 증가 신소재 발굴의 일환으로 유근피 열수 추출물의 면역 조절 효능을 RAW 264.7 대식세포 모델을 이용하여 조사하였다. 이를 위한 대식세포의 활성화 관련 지표로서 NO, TNF-α, IL-1β 및 IL-10의 생성량 변화를 조사하였다. 비록 유근피 추출물이 처리된 RAW 264.7 대식세포에서 IL-1β의 유의적인 유리는 관찰할 수 없었으나, NO, TNF-α 및 IL-10의 생성은 세포독성을 나타내지 않는 범위에서 유근피 추출물 처리 농도 의존적으로 증가되었으며, 이는 또한 iNOS, TNF-α 및 IL-10의 단백질 발현 증가와 연관되어 있었다. 아울러 유근피 추출물은 LPS에 의한 과도한 NO의 생성 억제능도 함유하고 있었으며, 유근피 추출물에 의한 대식세포의 활성화에는 NF-κB와 PI3K/Akt 및 MAPKs 등과 같은 면역 활성을 유도하는 신호전달계의 활성화가 연관되어 있음을 알 수 있었다. 따라서 본 연구의 결과는 유근피 추출물이 대식세포 활성화를 통한 면역 증강제로서의 개발 가능성이 매우 높음을 시사한다.

한방혼합액 APA-01의 면역 증강 효과 (Enhancement of Immune Response by New Herb Mixture, APA-01, in Mice)

  • APA-01, which is an aqueous extract of five Chinese herbs, is a modified formula of Huoxiang-Zhengqi-San. The effect of new herb extract on immune response was investigated. The parameter examined to assess apparent immune response of APA-01 in mice included changes of body weight, relative weight of immune organs, cell proliferation and cytokine gene expression. The body weight and relative weight of immune organs were not significantly changed among the tested groups. In the spleen cell prolijeration assay, APA-01 increased the cell proliferation in a dose-dependent manner. Methotrexate (MTX), an agent of immune suppression, inhibited the spleen cell proliferation (IC/sub 50/: 800㎍/㎖). However, APA-01 significantly inhibited the suppression of mouse spleen cell proliferation. Therefore, it seems that APA-01 has a reducing effect of immune suppression. Immunomodulatory effect of APA-01 was further investigated using reverse transcription polymerase chain reaction (RT-PCR) in mouse spleen cells. In RT-PCR test, APA-01 enhanced the expression of cyclooxygenase-2 (COX-2) mRNA in a dose-dependent manner. In spite of immune suppression by MTX, COX-2 mRNA was induced by co-treatment with APA-01. These results suggest that APA-01 stimulates the proliferation of spleen cells, regulates the expression of COX-2 mRNA, and accelerates the recovery of inhibition of spleen cell proliferation induced by MTX, thus providing the immunological basis for clinical benefit of APA-01.

오리로부터 분리한 Lactobacillus salivarius JWS 58과 Lactobacillus plantarum JWS 1354 균주의 면역활성효과 (Immuno-enhancing Effects of Lactobacillus salivarius JWS 58 and Lactobacillus plantarum JWS 1354 isolated from duck)

  • Lactobacillus salivarius JWS 58 (JWS 58) and Lactobacillus plantarum JWS 1354 (JWS 1354) are isolated from duck intestine and have ability to produce bacteriocin. The objective of this study was to evaluate the immunomodulatory effects of JWS 58 and JWS 1354. The nitric oxide (NO) and cytokines (IL-$1{\beta}$ and TNF-${\alpha}$) were measured in C57BL/6 mouse peritoneal macrophages to determine immune enhancing effects of JWS 58 and JWS 1354. A Listeria (L.) monocytogenes challenge mice model was used to evaluate immune enhancement ability of JWS 58 and JWS 1354 in vivo. The results showed that JWS 58 and JWS 1354 increased the production of NO or cytokines by peritoneal macrophages and that oral administration of viable probiotic strains in mice elicited the immuno-modulatory effect upon L. monocytogenes challenge. JWS 1354 showed stronger immune enhancing effects than JWS 58. Collectively, this study demonstrated that Lactobacillus strain JWS 58 and JWS 1354 possess immune enhancing effect. Furthermore, two stains are expected to use feed supplement to prevent diseases by pathogenic bacteria through releasing bacteriocin and enhancing host immune responses in animal.

  • Multiple periodontal pathogens sequentially colonize the subgingival niche during the conversion from gingivitis to destructive periodontal disease. An animal model of sequential immunization with key periodontal pathogens has been developed to determine whether T and B lymppocyte effector functions are skewed and fail to protect the host from pathogenic challenge. The present study was performed to evaluate immunomodulatory effect of exposure to Fusobacterium nucleatum(F. nucleatum) prior to Porphyromonas gingivalis(P. gingi - valis). Group 1(control) mice were immunized with phosphate-buffered saline, Group 2 were immunized with F. nucleatum prior to P. gingivalis, while Group 3 were immunized P. gingivalis alone. All the T cell clones derived from Group 2 demonstrated type 2 helper T cell clone(Th2 subsets), while those from Group 3 mice demonstrated Th1 subsets. Exposure of mice to F . nucleatum prior to P. gingivalis interfered with opsonophagocytosis function of sera against P. gingivalis. In adoptive T cell transfer experiments, in vivo protective capacity type 2 helper T cell clones(Th2) from Group 2 was significantly lower than type 1 helper T cell clones(Th1) from Group 3 against the lethal dose infection of P. gingivalis. Western blot analysis indicated the different pattern of recognition of P .gingivalis fimbrial proteins between sera from Group 2 and Group 3. In conclusion, these study suggest that colonization of the subgingival niche by F .nucleatum prior to the periodontal pathogen, P. gingivalis, modulates the host immune responses to P. gingivalis at humoral, cellular and molecular levels.

Immunoglobulin productivity assay를 이용(利用)한 홍삼투여(紅蔘投與) 실험동물(實驗動物)의 IgG, IgM, IgA 비교(比較) 연구(硏究) (The Comparative Study of IgG, IgM, and IgA in Laboratory Animal Administrated Red-ginseng, Using Immunoglobulin Productivity Assay)

  • Objective : The immune system is a complex of systems, all of which work together to clear infection from the body. In Korea, red ginsenghas been one of the herbs most widely used to enhance the immune system for thousand of years. More recently, red ginseng has been reported to have many positive effects on the immune system. The purpose of this study was evaluate the effects of Korean red ginseng and Chinese red ginseng on IgG, IgM, and IgA, using immunoglobulin productivity assay. Methods : Male SD rats were separated into 3 groups. We administered Korean red ginseng (KRG) to one group and Chinese red ginseng (CRG) to another, with normal saline for the Control group consecutively and orally for 3 months. The dose of red ginseng was 500mg per day, as a powder with soluble water. Immunoglobulin levels from spleen cell were estimated by ELISA kit. Results : In immunoglobulin productivity assay (cell), the IgG level of the KRG group significantly increased but there was no significant difference in the IgG of the CRG group. The IgM level of the KRG group significantly increased stimulated with PWM. When it was unstimulated, the level of IgM in KRG and CRG increased together. The IgA level of the KRG group significantly increased when it was stimulated with PWM and unstimulated. Conclusion : According to the above results, oral administration of red ginseng for 3 months is considered useful for immunomodulatory effect, and Korean red ginseng may be superior to Chinese red ginseng in that effect.

Dead Lactobacillus plantarum Stimulates and Skews Immune Responses toward T helper 1 and 17 Polarizations in RAW 264.7 Cells and Mouse Splenocytes

  • This study was undertaken to evaluate the immunomodulatory effect of dead nano-sized Lactobacillus plantarum (nLp) in RAW 264.7 cells and murine primary splenocytes. nLp is a dead, shrunken, processed form of L. plantarum nF1 isolated from kimchi (a traditional Korean fermented cabbage) and is less than 1 μm in size. It was found that nLp treatment stimulated nitric oxide (NO) production more in RAW 264.7 macrophages than pure live L. plantarum (pLp), and that the stimulatory properties were probably largely derived from its cell wall. In addition, nLp induced murine splenocyte proliferation more so than pLp; in particular, a high dose of nLp (1.0 × 1011 CFU/ml) stimulated proliferation as much as lipopolysaccharide at 2 μg/ml. Moreover, according to our cytokine profile results in splenocytes, nLp treatment promoted Th1 (TNF-α, IL-12 p70) responses rather than Th2 (IL-4, IL-5) responses and also increased Th17 (IL-6, IL-17A) responses. Thus, nLp stimulated NO release in RAW 264.7 cells and induced splenocyte proliferation more so than pLp and stimulated Th1 and Th17 cytokine production. These findings suggested that dead nLp has potential use as a functional food ingredient to improve the immune response, and especially as a means of inducing Th1/Th17 immune responses.
