Establishment of the Optimum Culture Conditions for Mozzarella Cheese manufacturing by Streptococcus macedonicus LC743 with Immunomodulating Activity (면역 활성능을 가진 Streptococcus macedonicus LC743을 이용한 모짜렐라 치즈 제조의 최적 배양조건 확립)

  • This study aimed to establish the optimum culture conditions for Mozzarella cheese by using Streptococcus macedonicus LC743, a strain selected for its immunomodulatory activity. For process optimization, 1.0% and 2.0% strain was inoculated and incubated at $32^{\circ}C$ and $35^{\circ}C$, respectively. The general components, bacterial count, total solids, yields, and immunomodulatory activity of the Mozzarella cheese were investigated. When the strain was inoculated at 2.0% and incubated at $32^{\circ}C$, the product quality and immunomodulatory activity was optimal and required minimal processing time. Therefore, 2.0% of S. macedonicus LC743 starter culture was added to milk at $32^{\circ}C$, after pasteurization at $63^{\circ}C$ for 30 min, and agitated for 4~5 min after addition of $230{\mu}L/kg$ of rennet. Curd was made by setting the milk 25~35 min after addition of 0.01~0.02% calcium chloride. The curd was cut at 0.1~0.12% acidity (81 min later) and after heating the cheese to up to $43^{\circ}C$. Whey was removed at an acidity of 0.17~0.18% by agitation for 53 min. Next, cheddaring for 210 min up to an acidity of 0.6~0.65%, stretching at $72{\sim}75^{\circ}C$, and molding at $65{\sim}70^{\circ}C$ were performed, and the product was allowed to cool down to $5^{\circ}C$. Salting was done with a solution of $18{\sim}20^{\circ}B{\acute{e}}$ at $12^{\circ}C$ for 20 min and drying occurred at 80~90% relative humidity at $10^{\circ}C$ for 2~3 days.

Comparison of biological activity according to extracting solvents of four Acanthopanax root bark (추출 용매에 따른 오갈피속 근피의 생리활성 기능 탐색 및 비교)

  • The biological activities of water, ethanol and 50% ethanol extracts from Acanthopanax root bark were compared. 94% of Hep3B cell growth was inhibited by adding 1.0g/L of 50% ethanol extracts from A. senticosus root bark. It was also showed that above 90% of A549 cell growth was inhibited by adding 1.0g/L of 50% ethanol extracts. The 50% ethanol extracts of A. sessiliflorum root bark showed that the extracts selectivity were from 1.5 to 3.4 by adding all samples. For screening immunomodulating activities, Jurkat(T-cell) was showed that the cell growth and viability were more increased and activited 275% by adding the 50% ethanol extracts from A. senticosus root bark. The result of anti-mutagenicity of 50% ethanol extracts of A. senticosus root bark was most effective than any other samples. The enhancement of glutathione-S-transferase activity was increased 241% by adding 1.0g/L 1 : 1 extracts of A. senticosus root bark. 72% of oxidation was inhibited by adding 1.0g/L of 50% ethanol extracts from A. senticosus root bark.

Effect of Hizikia fusiforme Water Extracts on Splenocyte Proliferation and Cytokine Production in Mice (톳 열수 추출물이 마우스 비장세포 증식증과 염증성 사이토카인 (IL-1β, IL-6, TNF-α) 분비에 미치는 영향)

  • Hizikia fusiforme (seaweed fusiforme) has long been used as a food source mainly in Korea and Japan. This study was performed to evaluate the immunomodulative effects of Hizikia fusiforme in mice. Hizikia fusiforme water extracts (0, 50, and 500 mg/kg b.w.) were orally administrated into the mice every other day, for four weeks. The proliferation of splenocytes, as well as the levels of proinflammatory cytokines (IL-$1{\beta}$, IL-6, and TNF-${\alpha}$) secreted by activated macrophages were measured. Splenocyte proliferation was enhanced in the experimental groups compared to that of the control group. Also, the mice with Hizikia fusiforme water extracts supplementation in both concentrations showed increased levels of cytokine production by activated peritoneal macrophages compared to those in the control group. The highest levels of cytokine (IL-$1{\beta}$, IL-6, TNF-${\alpha}$) production were observed in the 50 mg/kg b.w. supplementation group stimulated by LPS for all three cytokines. The results of this study showed that the supplementation of Hizikia fusiforme water extracts may enhance the immune function by regulating the splenocytes proliferation and the cytokine production by activated macrophages. Further studies are needed to identify the stimulative and immunomodulating components of Hizikia fusiforme.

Antitumor and immunomodulatory effects of a hot water extract of Phellinus baumii using Sarcoma-180 in the mouse (Pheflinus baumii 자실체 추출물의 Sarcoma-180 에 대한 항암 및 면역효과)

  • A hot water extract(HWE-P4) was separated from the fruit bodies of PMO-P4, and its antitumor and immunomodulatory activities against sarcoma-180 in ICR mice were investigated. The internal transcribed spacer(ITS) regions from PMO-P4 were amplified using polymerase chain reaction(PCR) and sequenced. The results revealed that PMO-P4 was belong to the Phellinus baumii. When oral administration at the dose of 160mg/kg/day in the mice until the end of the experiment with 2 week's pre-feeding of the HWE-P4, the survival rate of the mice was 152% for 50days after the inoculation of sarcoma-180 and the suppression rate of the tumor growth was 35.3%(p<0.05) for 28 days after inoculation of sarcoma-180. The HWE-P4 increased 71.4% of the CD4/CD8 ratio and 5-fold of the expression of CD25(IL-2 receptor chain) compared with the control. From these results, the antitumor activity of HWE-P4 is exerted through its immunomodulating activity on the host's immune system.

Immuno-potentiating and Antitumor Effects against Mouse Sarcoma 180 by Crude Polysaccharides Extracted from Fruiting Body of Russula rosacea (장미무당버섯(Russula rosacea)의 자실체에서 추출한 조다당류의 생쥐 Sarcoma 180에 대한 면역증강 및 항암 효과)

  • Russula rosacea, one of edible and medicinal mushroom belonging Agaricales of Basidiomycota, has been known to have good inhibitory effects on sarcoma 180 and Ehrlich carcinoma of mice. Neutral saline soluble (0.9% NaCl), hot water soluble and methanol soluble substances (hereinafter referred to Fr. NaCl, Fr. HW and Fr. MeOH, respectively) were extracted from fruiting body of the mushroom. In vitro cytotoxicity tests, crude polysaccharides were not cytotoxic against cancer cell lines such as Sarcoma 180, HepG2, HT-29 and NIH3T3 at the concentration of $10{\sim}2000\;{\mu}g/ml$. Intraperitoneal injection with crude polysaccharides exhibited life prolongation effect of $21.4{\sim}45%$ in mice previously inoculated with Sarcoma 180. Fr. HW improved the immuno-potentiation activity of B lymphocyte by increasing the alkaline phosphatase activity by 6.8 fold compared with control at the concentration of $500\;{\mu}g/ml$. In case of Fr. NaCl, the numbers of peritoneal exudate cells and circulating leukocytes were increased by 6 and 2.6 folds at the concentration of 50 mg/kg, respectively. Therefore, the antitumor effect against mice Sarcoma 180 by Russula rosacea could be due to immunomodulating activity.

Isolation of Immunomodulatory Antitumor Active Polysaccharide (RGAP) from Red Ginseng By-Product and Its Physico-chemical Properties (홍삼추출잔사로부터 항암면역조절 활성을 보여주는 홍삼산성다당체(RGAP)의 분리 및 이화학적 특성)

  • This study was carried out in order to develop the method for isolation of red ginseng acidic polysaccharide (RGAP) haying immunomodulating antitumor activity from red ginseng by-product. The red ginseng by-product was obtained from red ginseng residues produced in processing of red ginseng ethanol extract. The yield of RGAP isolated by ultrafiltration was 20.9%. The active substance (GFP) was purified by DEAE-sepharose column chromatography RGAP induced nitric oxide (NO) exhibited tumoricidal activities against P8l5 (mastocytoma) tumor cells. Acid-hydrolyzed RGAP fragments were shown four to five spots. These sopts showed the same R$_{f}$ values with sugars designated as rhamnose, glucose, glactose and glucuronic acid. Some physico-chemical properties of RGAP were investigated. pH and dry reduction content at 105$^{\circ}C$ were 4.74 and 4.72%, respectively. Crude protein, ash and Pb contents were 3.30%, 4.74% and 2.30 ppm. These results suggest that we will be able to produce RGAP from red ginseng by-product by ultrafiltration in a large scale.e.

Effects of Plant Water Extract Codonopsis Lanceolatae on Mouse Immune Cell Activation Ex Vivo (더덕 물 추출물의 경구 투여가 마우스 면역 세포 활성에 미치는 효과)

  • Codonopsis lanceolatae has been used as one of the traditional remedies as well as food source. However, few studies on their immunomodulating effects have been reported. We previously reported that ex vivo supplementation of Codonopsis lanceolatae water extracts enhanced splenocyte proliferation compared to the control group. In order to elucidate its ex vivo effect, six to seven week old balb/c mice were fed ad libitum on a chow diet and water extracts of Codonopsis lanceolatae were orally administrated every other day for four weeks at two different concentrations (50 and 500 mg/kg B.W). After preparing the single cell suspension, the proliferation of splenocytes was determined by MTT (3- [4,5-dimethylthiazol-2-y] -2,5-diphenyl terazolium bromide) assay. The production of cytokine (IL-1${\beta}$, IL-6, TNF-${\alpha}$), secreted by macrophages stimulated with LPS or not, was detected by ELISA assay using a cytokine kit. After 48 hrs of incubation with the mitogen (ConA or LPS) stimulation, the mice splenocyte proliferation in experimental group was statistically increased at two different concentrations than that in control group. The cytokines production was more significantly enhanced at the lower supplementation (500 mg/kg B.W.) group rather than higher concentration (500 mg/kg B.W.) compared to the control group. The results of this study may suggest that the supplementation of water extract of plant mixture could regulate the immune function by increasing the splenocyte proliferation and enhance the immune function through regulating cytokine production capacity by activated macrophages in mice.

Anti-metastatic Effect on Cancer cell and Immune System Activation by Orally Administered Boyanghwano-tang (보양환오탕(補陽還五湯) 경구투여 후 면역 활성화에 의한 암 전이 억제 효과)

  • Objectives: This study was designed to investigate intestinal immune system activation and anti-metastatic effect on cancer cells by orally administered extracts of Boyanghwano-tang. Methods: To observe immunomodulating effects of Boyanghwano-tang on Peyer's patch cells, we measured cytokines GM-CSF, IL-4. In addition to observing effects of Boyanghwano-tang on hematopoiesis, we measured proliferation of bone marrow cells mediated by Peyer's patch cells in vitro. IgA induction activated in intestinal content and serum was measured to observe the effect of orally administered Boyanghwano-tang on mucosal immune system. After administering ovalbumin (OVA) with Boyanghwano-tang, Proliferation of Peyer's patch cell was measured to investigate gut immunostimulatory effect. Anti-metastatic experiments were conducted in vivo mouse model by using colon 26-M3.1 carcinoma cell. Results: The amounts of GM-CSF and IL-4 in the culture supernatant of Peyer's patch cells were significantly increased compared to the control group. The proliferation of bone marrow cell was significantly up-regualted with Boyanghwano-tang. These results indicate that oral administration of Boyanghwano-tang enhances the secretion of hematopoietic growth factors such as GM-CSF and IL-4 from Peyer's patch cells, and these cytokines also act on modulator of bone marrow cell proliferation. After orally administering OVA with Boyanghwano-tang, IgA induction and Proliferation of peyer's patch cell was up-regulated with Boyanghwano-tang. These results means orally administered Boyanghwano-tang activates intestinal immune system and has an inhibitory effect on tumor metastasis. In addition, We found that orally administered Boyanghwano-tang significantly inhibited tumor metastasis in vivo. Conclusions: Orally administered Boyanghwano-tang appears to have considerable activity on the anti-metastasis by activation of immune system.

Activation of Immune System & Antimetastatic Effects of Ojeok-san by Oral Administration (오적산(五積散) 경구투여에 의한 면역활성과 종양 전이 억제 효과)

Methods: Cell viability of Ojeok-san was tested with colon 26-M3.1 carcinoma cells and Peyer's patch cells in vitro. Antimetastatic experiments were conducted in vivo mouse model by using colon 26-M3.1 carcinoma cell. To observe immunomodulating effects of Ojeok-san on Peyer's patch cells, we measured interleukin (IL)-4, GM-CSF. In addition to observing effects of Ojeok-san on hematopoiesis, we measured proliferation of bone marrow cells mediated by Peyer's patch cells in vitro. IgA induction activated in serum and intestinal content was measured to observe the effect of orally administered Ojeok-san on mucosal immune system. After administering Ovalbumin (OVA) with Ojeok-san, Proliferation of Peyer's patch cell was measured to investigate gut immunostimulatory effect. Results: in vitro cytotoxicity analysis, the inhibitory concentration $(IC)_{50}$ of the colon 26-M3.1 carcinoma cell was $890{\mu}g/ml$. $IC_{50}$ of the Peyer's patch cells with LPS was $990{\mu}g/ml$. We found that orally administered Ojeok-san significantly inhibited tumor metastasis in vivo. In addition, the amounts of IL-4 and GM-CSF in the culture supernatant of Peyer's patch cells were significantly increased compared to the control group. Immunological Activity of Solvent Fractions from $Epimedium$ $koreanum$ Nakai (삼지구엽초 용매별 분획 추출물의 면역관련 활성)

  • $Epimedium$ $koreanum$ Nakai is a wild medicinal plant commonly consumed in South Korea due to its beneficial health effects. In this study, the antimutagenic and immunological activities of $E.$ $koreanum$ Nakai extracts were investigated for their use in food. In the immunomodulating activity, the effects of $E.$ $koreanum$ Nakai on the B cell (Rhamos) and T cell (Molt-4) were investigated. The results showed that the growth and viability of the B and T cells were increased and activated more in the ethylacetate (1.35 and 1.48 times) and water fraction (1.30 and 1.40 times), respectively. In the Ames test, none of the fractions produced a mutagenic effect on $Salmonella$. $typhimurium$ TA98 and TA 100. The ethylacetate fraction showed a strong antimutagenic effect (98%) on and a high butanol fraction (84%) of B(${\alpha}$)P in $S.$ $typhimurium$ TA98 and TA100, respectively. In 4-nitroquinoline-1-oxide (4NQO), all the solvent fractions showed an over 70% antimutagenic effect, except for the chloroform extract. Especially, ethylacetate and butanol showed strong inhibition of the mutagenic effects (80 and 90%) on 4NQO in $S.$ $typhimurium$ TA98 and TA100, respectively. These results provide preliminary data for the development of $E.$ $koreanum$ Nakai as an edible food material.
