Anti-inflammatory glucocorticoid (GC) derivatives have been clinically used in immune-malfunctional diseases for their immunosuppressive activity. However, there is still a lack of knowledge on the relationship between anti-inflammatory and immunosuppressive activities. In order to compare immunosuppressive activities with the known anti-inflammatory activities of the GC derivatives, eight clinically used GC derivatives including hydrocortisone, prednislone, 6$\alpha$-methyl prednisolone, triamcinolone, dexamethasone, betamethasone, triamcinolone acetonide and fluocinolone acetonide were selected, and lymphoblastosis inhibition and plaque-forming cell (PFC) response in mice were studied as immunological parameters. In Con A-induced lymphoblatosis inhibition invitro, all derivatives showed potent inhibition $IC_{50}$ values of the derivaties except methyl prednisolone and triamcinolone were less than $10^{-7}$M and good dose dependency was obtained. This result was well correlated with that of their anti-inflammatory potencies obtained. This result was well correlated with that of their anti-inflammatory potencies and their receptor binding affinities. However, in PFC response, consistent result were not obtained. Total numbers of PFCs per spleen were decreased by some derivatives, but numbers of PFCs per $10^6$ cells were not decreased by systemic administration of but numbers of PFCs per $10^6$ cells were not decreased by systemic administration of GC at the dose of 0.05 mg/mouse. Furthermore, at the dose of 0.1 mg/mouse, numbers of PFCs per $10^6$ cells were found to be increased, although total PFCs per spleen were decreased.

Objectives: The Toxoplasma gondii (T. gondii) is an intracellular opportunistic protozoan parasite that infects approximately one-third of the human population worldwide. Honey has long been used for treatment of many diseases in folk medicine. Honey has exhibited significant anthelmintic, nematicidal and anti-protozoal activities. This study was conducted to investigate the immunological patterns in rats infected with T. gondii who were treated orally with supplemented 15% Capparis spinosa honey (Saudi Arabia) for a period of 28 days. Methods: Immunoglobulin M, immunoglobulin G, and cytokines were detected by using enzyme-linked immunosorbent assays (ELISAs). In addition, the mortality and the morbidity rates were assessed. Results: Oral administration of Capparis spinosa honey as a natural food additive was experimentally shown to increase the antibody titer; furthermore, compared with the rats in the control group, the levels of the sera cytokines ($IFN-{\gamma}$, IL-1 and IL-6) were consistently higher at day 7 post-infection in the infected rats treated with oral supplements of Capparis spinosa honey. Conclusion: Orally administered supplements of Capparis spinosa honey increased both the antibody titer and the cytokines ($IFN-{\gamma}$, IL-1 and IL-6) levels in rats infected with T. gondii.
Immunological activities of Ginseng Radix water extract (GE) were compared with theirs of Insamhodo-tang water extract (IHT). GE and IHT (500 mg/kg) were administered p.o. twice a day for 5 days to C57BL/6 mice, respectively. IHT decreased the cell viability of murine thymocytes and splenocytes, but increased the population of $CD4^+$ cells in thymocytes, and the population of $Thy1^+$ cells and $CD4^+$ cells in splenocytes in contrast to GE. In addition, IHT decreased the phagocytic activity of peritoneal macrophages, but increased the production of nitric oxide from peritoneal macrophages in contrast to GE. These results suggest that IHT decrease a specific- and nonspecific immune reaction in comparison with GE.
After examining and analysing the medicinal herbs of fifty-three experimental papers, we studied their effects on immediate tumors in specific cancers. We did not study the influence on the life span of general cancerous cells. We looked to see if the combined usage of medicinal herbs and anticancer agents inhibited the tumor cell's growth. The serum test and blood cell count test showed if the medicinal herbs inhibited the side effects of the anticancer agent. The test showed that more than 80 percent of used medicinal herbs, brought anticancer activities. However, anticancer experimental studies using medicinal herbs have draw-backs. First, it is difficult to choose a prescription using the standards of Oriental Medicine because we are testing a mouse not a man. Second, because we only observed the indirect effect on the whole physiological regulation caused by the synergic effects of the complex prescription, we are not able to understand the detailed mechanism of the herbs. Therefore; if the anticancer effect of the herbs is proved by the experiment, we need to research the concrete medical action of medicinal herbs and the immunological analysis of herbal medicines on the body.
Objectives : This experiment was conducted to evaluate the inhibitory effects against lung metastasis and promotion of splenocytes by water-soluble components from five mushrooms extracts (WEFM): Garnoderma frondosa, Corious versicolor, Codyceps militaris, Hericium erinaceus and Lentinula edodes. Methods : Colon 26-L5 carcinoma cells were injected through the tail vein to induce lung metastatic cancer. Changes in weight of lung were observed and cytokine level was analyzed to evaluate immunological changes. Results : Oral administration of WEFM resulted in a significant inhibition of lung metastasis after intravenous injection of colon 26-L5 cells in a dose-dependent manner. There was also a significant increase in T cell and B cell mitogenic stimuli and production of IFN-g by splenocytes stimulated with Con A compared to untreated controls. Conclusion : WEFM may have anti-tumor activities via Th1-type dominant immune responses.
The gastrointestinal exposome represents the integration of all xenobiotic components and host-derived endogenous components affecting the host health, disease progression and ultimately clinical outcomes during the lifespan. The human gut microbiome as a dynamic exposome of commensalism continuously interacts with other exogenous exposome as well as host sentineling components including the immune and neuroendocrine circuit. The composition and diversity of the microbiome are established on the basis of the luminal environment (physical, chemical and biological exposome) and host surveillance at each part of the gastrointestinal lining. Whereas the chemical exposome derived from nutrients and other xenobiotics can influence the dynamics of microbiome community (the stability, diversity, or resilience), the microbiomes reciprocally alter the bioavailability and activities of the chemical exposome in the mucosa. In particular, xenobiotic metabolites by the gut microbial enzymes can be either beneficial or detrimental to the host health although xenobiotics can alter the composition and diversity of the gut microbiome. The integration of the mucosal crosstalk in the exposome determines the fate of microbiome community and host response to the etiologic factors of disease. Therefore, the network between microbiome and other mucosal exposome would provide new insights into the clinical intervention against the mucosal or systemic disorders via regulation of the gut-associated immunological, metabolic, or neuroendocrine system.
The present study was conducted to evaluate the effect of Yongdamsagantang (YST) on the regulatory mechanism of cytokines and nitric oxide (NO) for the immunological activities in RAW 264.7 cells. After the treatment of YST water extract, cell viability was measured by MTT assay, and NO production was monitored by measuring the nitrite content in culture medium. Inducible nitric oxide synthase (iNOS) and phospholylation of inhibitor of nuclear factor kappa B alpha ($p-I{\kappa}B{\alpha}$) were determined by Immunoblot analysis, and levels of cytokine were analyzed by sandwich immunoassays. Results provided evidences that YST inhibited the production of NO. iNOS, and interleukin-6, and the activation of $p-I{\kappa}B{\alpha}$ in RAW 264.7 cells activated with lipopolysaccharide. These findings showed that YST could have some anti-inflammatory effects which might play a role in therapy in Gram-negative bacterial infections.
Bio-Q is a modified prescription with the activities of supplementing Qi and blood in human body. In the present study, immunomodulatory effect of Bio-Q was examined. After oral administration of Bio-Q for 7 days to Balb/c mice, splenocytes were isolated and immunological experiments were performed. Bio-Q significantly increased the proliferation of splenocytes exposed to concanavalin A (Con A), while it did not in case of lipopolysaccharide (LPS) stimulation. Bio-Q also significantly increased CD3/CD19, CD4/CDB and NK cells by flow cytometric analysis. In addition, Bio-Q significantly enhanced the level of $INF-\gamma$ in splenocytes, but not $TNF-\alpha$ by ELISA. These results strongly suggest that Bio-Q has immunomodulatory activity through the regulation of T cell mediated immune pathway.
Immunological activities of the combined-administration of Acanthopanacis Cortex and Lycii Cortex Radicis were examined in BALB/C mice. The 40% ethyl alcohol extract of Acanthopanacis Corter (AE) or the 40% ethyl alcohol extract of Acanthopanacis Coriex and Lycii Cortex Radicis (ALE) were administered p.o. once a day for 7 days, respectively. AE did not affect the viability of thymocytes, but ALE decreased the viability of thymocytes. ALE enhanced the viability of splenocytes increased by AE. Also, AE enhanced the population of cytotoxic T cell in thymocytes, and ALE enhanced the population of helper T cell compared with AE. Furthermore, AE increased the population of $Thy1^+$ cells in splenocytes, and increased the population of splenic $CD4^+$ cells. In addition, ALE enhanced the phagocytic activity which was decreased by AE ALE decreased the production of nitric oxide increased by AE in peritoneal macrophages. These results suggest that ALE enhance an immune-regulative action of AE.
The inhibitory effects of ethanol extract of Radex Angelica uchiyamana, on LPS- or IFN- plus LPS-induced production of NO, and $TNF-{\alpha}$, and expression of iNOS and COX-2 were investigated in the activated RAW 264.7 cells. This extract significantly inhibited the production of NO and $TNF-{\alpha}$, and suppressed the expression of iNOS and COX-2 in a dose-dependent manner. These results show that ethanol extract of Radex Angelica uchiyamana may explain some known biological activities including anti-inflammatory effect, and is of considerable benefit in the treatment for NO and pro-inflammatory cytokine overproduction related immunological diseases.
