비허온습만(脾虛蘊濕万)이 아토피 피부염 유발 NC/Nga 생쥐의 혈액내 면역 관련 인자에 미치는 영향 (Effects of BHOSB on Various Immunological Factors Related to Pathogenesis of Atopic Dermatitis in DNCB Treated NC/Nga Mice)

  • Atopic dermatitis is an allergic inflammatory skin disease caused by aberrant and overreactive immune responses including overactivation of $T_H2$ immune responses, high levels of IgE as well as proinflammatory cytokines and chemokines. We examined whether BHOSB, a traditional herbal medicine, has modulatory effects on various immunological factors related to pathogenesis of atopic dermatitis in ONCB treated NC/Nga mice. Oral administration of BHOSB at the concentration of 10.8 mg/mouse/day significantly inhibited the production of IgE compared with control, and the levels of IgG2a and IgG2b, but not IgG1, were also significantly reduced. Production of IL-6 and TNF-a was greatly decreased. The results from flowcytometry of peripheral blood mononuclear cells indicated that the percentages of C03+C069+ cells and C04+ were significantly decreased by BHOSB. Theses results suggested that BHOSB has suppressive effects on aberrant and overreactive immunological activities in ONCB-induced dermatitis mice of NC/Nga.

Effects of Brazilin on Induction of Immunological Tolerance by Sheep Red Blood Cells in C57BL/6 Female Mice

  • Brazilin was examined for its effects on the induction of immunological tolerance. Brazilin was administered to C57BL/6 female mice for 2 consecutive days before the immunization with high dose SRBC (109 cells) which can produce immunological tolerance. Delayed type hypersensitivity, IgM plaque forming cells, ConA induced IL-2 production and mitogen- or antigen-induced proliferation of lymphocytes were measured as evaluation parameters. Administration of brazilin prior to immunization could keep the DTH and IL-2 production almost optimaly immunized levels. Brazilin also inhibited the elevation of non-specific suppressor cell activity. ConA induced proliferation of splenocytes in high dose SRBC immunized mice was significantly decreased by pretreatment of brazilin. And this might be one of the reason for augmentation of DTH by brazilin. However, IgM plaque forming cells were not affected by the treatment of brazilin. These results indicate that brazilin prevents the induction of mmunological tolerance caused by high dose SRBC by suppressing the elevation of suppressor cell activity and by inhibiting the decrease in IL-2 production in C57BL/6 female mice.

아토피와 알러지에 대한 한의학적 약물치료의 혈액면역학적 개선효과 (Effects of Herbal Prescriptions on the Immunological Parameter in Patients with Atopic & Allergic Diseases; Clinical Observational Study)

  • The aim of this study was to investigate the effects of several herbal prescriptions in patients with allergy or atopic disease, such as atopic dermatitis, asthma and allergy rhinitis and others, on the blood immunological parameters and to verify the safety of long-term use of herbal medicine. Eighty one patients with allergy or atopy disease who taken herbal medicine at least for 2 months were compared with patients who taken same medicine with no allergy & atopy disease (n=14) and normal healthy subjects (n=22). According to the comparison of immunological parameters change, the data showed that herbal medicine decreased IgE (P=0.003), Eosinophil ratio (P<0.001) and count (P<0.001) of White Blood Cell (WBC) and did not affect to the liver cell enzymes in blood. This results indicated that herbal medicine decreased immune hypersensitivity and improved chronic inflammation related to blood immunological parameters of allergy or atopy diseases. In addition, herbal medicine seemed to be safe to the liver function for long-term use.

Immunostimulatory Effects of Silica Nanoparticles in Human Monocytes

  • Amorphous silica particles, whose applications are increasing in many biomedical fields, are known to be less toxic than crystalline silica. In this study, the inflammatory effects of amorphous silica nanoparticles were investigated using 30-nm amorphous silica nanoparticles and human peripheral blood mononuclear cells (PBMCs) or purified monocytes. As a result, production of IL-$1{\beta}$ and IL-8 were increased. In addition, the mitochondrial reactive oxygen species (ROS) was detected, which may lead to mitochondrial membrane disruption. Most importantly, inflammasome formation was observed. Therefore, these results provide immunological information about amorphous silica nanoparticles and suggest that amorphous silica nanoparticles can evoke innate immune reactions in human monocytes through production of IL-$1{\beta}$ and IL-8.

  • To enhance our understanding of immunological stimulating effects through the pathway by which nitric oxide (NO) activity and alkaline phosphatase(ALP) enzyme activity from the marine algae, Ulva lactuca, we have investigated NO activity by using mouse RAW264.7 cell line. And ALP enzyme activity performed by spleen of ICR mouse. The results showed that NO activity of the $H_2O$ fraction is the most effective than activities of other solvent fractions.

  • This study was performed to investigate the immunological side effects of chloramphenicol (CAP) on olive flounder, Paralichthys olivaceus. To investigate immunological effects on olive flounder, we determined the changes of chemiluminescence (CL) response of flounder kidney-derived leucocyte after the treatment of CAP in vivo and in vitro. The CL activity was significantly decreased in a dose-dependent manner during the treatment of CAP in vitro. Similarly, a dose-dependent reduction of CL response, although not significant, were observed during the treatment of CAP in vivo. The results suggest that CAP reduced the function of flounder phagocytosis in vivo and in vitro, indicating the immunosuppressive ability of CAP.

Chemotherapeutic Candidate Inducing Immunological Death of Human Tumor Cell Lines

  • The immunological death induction by EY-6 on the human tumor cell lines was screened. Human colon carcinoma (HCT15, HCT116), gastric carcinoma (MKN74, SNU668), and myeloma (KMS20, KMS26, KMS34) cells were died by EY-6 treatment with dose-dependent manner. CRT expression, a typical marker for the immunological death, was increased on the EY-6-treated colorectal and gastric cancer cells. Interestingly, the effects on the myeloma cell lines were complicated showing cell line dependent differential modulation. Cytokine secretion from the EY-6 treated tumor cells were dose and cell-dependent. IFN-${\gamma}$ and IL-12 secretion was increased in the treated cells (200% to over 1000% of non-treated control), except HCT116, SNU668 and KMS26 cells which their secretion was declined by EY-6. Data suggest the potential of EY-6 as a new type of immuno-chemotherapeutics inducing tumor-specific cell death. Further studies are planned to confirm the efficacy of EY-6 including in vivo study.

  • As a nanotechnology has been actively applied to the overall areas of scientific fields, it is necessary to understand the characteristic features, physical behaviors and the potential effects of exposure to nanomaterials and their toxicity. In this article we review the immunological influences induced by several nanomaterials and emphasize establishment of the animal models to estimate the impact of these nanomaterials on development of immunological diseases.

  • Beevenom immunotherapy(BVIT) in allergic patients is a well-established treatment modality for the prevention of systemic anaphylactic reactions caused by insect stings. BVIT is accompanied by increases in allergen-specific IgG, particularly the IgG4 isotype, which blocks not only IgE-dependent histamine release from basophils but also IgE-mediated antigen presentation to T cells. Inhibition of T cells after BVIT also involves decreased induction of the costimulatory molecule ICOS, which, in turn, seems to be dependent on the presence of IL-10, also associated with the inhibited status of T cells after BVIT. Suppression of T cells by IL-10 is an active process, which depends on the expression and participation of CD28. Immune tolerance in specific allergen immunotherapy might be a consequence of decreased Th2 or increased Th1 response of allergen specific T lymphocytes. BVIT shifted cytokine responses to allergen from a TH-2 to a TH-1 dominant pattern, suggesting direct effects on T cells. Many studies showed that severe side effects due to venom immunotherapy are rare. These results suggest that immunological changes after BVIT may be applied to be therapeutic alternative of general allergic diseases including beevenom allergy.

Regulation of Immunological Effect of Rubia cordifolia Extract and Associated MAPKs Pathway in RBL-2H3 Cell-line

  • Rubia cordifolia has been used to treat diseases for many years in China and India. Although the biological properties and major compounds of R. cordifolia have been extensively studied, the underlying mechanisms of its biological effects remain elusive. In terms of immunological effects, anti-inflammation effect of macrophage (Raw 264.7) simply has been reported. In this study, R. cordifolia was extracted in 70% ethanol and the extract did not affect to macrophage (Raw 264.7) pro-inflammation and T cell (Molt-4). However, in mast cell (RBL-2H3), it showed inhibition of degranulation. The inducing inhibitory effect on degranulation was related to concentration dependent variation in phosphorylation of ERK-1/2 and upregulating the JNK phosphorylation in RBL-2H3 cells. Based on these data, we concluded that R. cordifolia newly have anti-allergenic effects in RBL-2H3 and might be used as a therapeutic agent to treat or prevent allergic diseases such as asthma and atopic dermatitis.
