  • 위염과 위궤양을 일으키는 Helicobacter pylori는 그 감염 환자에서 강한 항체형성을 유도하는 urease를 생산한다. 식물체를 이용하여 H. pylori에 대한 식용백신 모델을 제조하기 위하여 H. pylori의 urease 유전자를 지니고 있는 pH808 plasmid로부터 750bp의 ureA DNA를 PCR에 의해서 증폭한 후 이를 담배식물에서 발현이 되도록 조작하였다. 재분화된 형질전환 식물체로부터 ureA 유전자의 도입과 mRNA발현 및 단백질합성을 분석하였다. CaMV 35S promoter에 의한 ureA 유전자의 발현은 SDS polyacrylamide 전기영동 및 immunoblot실험에서 박테리아로부터의 재조합 단백질과 같은 크기의 30 kDA UreA 단백질이 생산되었음을 확인할 수 있었다.

  • 저자들은 발열, 기침, 객담을 주소로 입원한 24세 남환의 임상증상, 이학적 검사, 혈청학적 및 면역학적, 유전자 검사상 X연관 무감마글로불린혈증으로 진단된 예를 관찰하였기에 문헌 고찰과 함께 보고 하는 바이다. 반복적인 감염을 보이는 젊은 성인 환자에서 유전자 이상(Btk gene mutation)에 의한 면역결핍질환(X-linked agammaglobulinemia)의 가능성을 꼭 의심해 보아야 한다.

Deoxynivalenol- and zearalenone-contaminated feeds alter gene expression profiles in the livers of piglets

  • Objective: The Fusarium mycotoxins of deoxynivalenol (DON) and zerolenone (ZEN) cause health hazards for both humans and farm animals. Therefore, the main intention of this study was to reveal DON and ZEN effects on the mRNA expression of pro-inflammatory cytokines and other immune related genes in the liver of piglets. Methods: In the present study, 15 six-week-old piglets were randomly assigned to the following three different dietary treatments for 4 weeks: control diet, diet containing 8 mg DON/kg feed, and diet containing 0.8 mg ZEN/kg feed. After 4 weeks, liver samples were collected and sequenced using RNA-Seq to investigate the effects of the mycotoxins on genes and gene networks associated with the immune systems of the piglets. Results: Our analysis identified a total of 249 differentially expressed genes (DEGs), which included 99 upregulated and 150 downregulated genes in both the DON and ZEN dietary treatment groups. After biological pathway analysis, the DEGs were determined to be significantly enriched in gene ontology terms associated with many biological pathways, including immune response and cellular and metabolic processes. Consistent with inflammatory stimulation due to the mycotoxin-contaminated diet, the following Kyoto encyclopedia of genes and genomes pathways, which were related to disease and immune responses, were found to be enriched in the DEGs: allograft rejection pathway, cell adhesion molecules, graft-versus-host disease, autoimmune thyroid disease (AITD), type I diabetes mellitus, human T-cell leukemia lymphoma virus infection, and viral carcinogenesis. Genome-wide expression analysis revealed that DON and ZEN treatments downregulated the expression of the majority of the DEGs that were associated with inflammatory cytokines (interleukin 10 receptor, beta, chemokine [C-X-C motif] ligand 9), proliferation (insulin-like growth factor 1, major facilitator superfamily domain containing 2A, insulin-like growth factor binding protein 2, lipase G, and salt inducible kinase 1), and other immune response networks (paired immunoglobulin-like type 2 receptor beta, Src-like-adaptor-1 [SLA1], SLA3, SLA5, SLA7, claudin 4, nicotinamide N-methyltransferase, thyrotropin-releasing hormone degrading enzyme, ubiquitin D, histone $H_2B$ type 1, and serum amyloid A). Conclusion: In summary, our results demonstrated that high concentrations DON and ZEN disrupt immune-related processes in the liver.

Marine-derived Ca-Mg complex influences lipid and glucose metabolism, serum metabolites, colostrum profile, and stress hormone in sows over four-parity periods

  • Minerals is required small amounts among various nutrients, but it has a significant impact on sow longevity and reproduction performance. This study was carried out to see the beneficial effects of marine-derived Ca-Mg complex on the reproductive performance of sows during four-parity periods. Seventy-two gilts ([Yorkshire × Landrace] × Duroc), with an average body weight of 181 kg, were randomly allocated to three groups; CON (basal diet), 0.3LC (CON - MgO - 0.3% limestone + 0.4% Ca-Mg complex), and 0.7LC (CON - MgO - 0.7% limestone + 0.4% Ca-Mg complex). During parity 3 and 4, the expression level of SCD gene was lower in the umbilical cord of piglets born to 0.3LC and 0.7LC sows compared with the CON sows. During parity 2, 3 and 4, SLC2A2 and FABP4 gene expressions were higher in the umbilical cord of piglets born to 0.7LC sows and the placenta of sows from 0.3LC groups, respectively. Ca-Mg complex increased (p < 0.05) Ca and Mg concentrations in sows and their piglets' serum as well as in colostrum regardless of parities. The serum vitamin D concentration was higher (p < 0.05) in their first parity, whereas serum prolactin and estrogen concentrations were higher (p < 0.05) during the fourth and third parity, respectively. The growth hormone concentrations were higher (p < 0.05) in the piglets born to sows during the first and second parity. The fat and immunoglobulin A (IgA) concentrations in colostrum were higher (p < 0.05) during the third and fourth parity, respectively. A reduction (p < 0.05) in salivary cortisol, epinephrine, and norepinephrine concentrations was observed in 0.3LC and 0.7LC sow groups compared with CON after farrowing regardless of parity, however before farrowing, a reduction in norepinephrine was observed. Before farrowing, the epinephrine and norepinephrine concentrations were higher (p < 0.05) during the first and second parity. After farrowing, the concentration of these hormones was higher during the second parity. Taken together, sows' parity and dietary Ca-Mg complex supplementation influenced serum metabolites, colostrum nutrients, stress hormones as well as the gene expressions related to lipid and glucose metabolism.

Investigation of PCR-RFLPs within Major Histocompatibility Complex B-G Genes Using Two Restriction Enzymes in Eight Breeds of Chinese Indigenous Chickens

  • New polymorphism of major histocompatibility complex B-G genes was investigated by amplification and digestion of a 401bp fragment including intron 1 and exon 2 using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique with two restriction enzymes of Msp I and Tas I in eight breeds of Chinese indigenous chickens and one exotic breed. In the fragment region of the gene, three novel single nucleotide polymorphisms (SNPs) were detected at the two restriction sites. We found the transition of two nucleotides of A294G and T295C occurred at Tas I restriction site, and consequently led to a non-synonymous substitution of asparagine into serine at position 54 within the deduced amino acid sequence of immunoglobulin variable-region-like domain encoded by the exon 2 of B-G gene. It was observed at rare frequency that a single mutation of A294G occurring at the site, also caused an identical substitution of amino acid, asparagine 54-to-serine, to that we described previously. And the transversion of G319C at Msp I site led to a non-synonymous substitution, glutamine 62-to-histidine. The new alleles and allele frequencies identified by the PCR-RFLP method with the two enzymes were characterized, of which the allele A and B frequencies at Msp I and Tas I loci were given disequilibrium distribution either in the eight Chinese local breeds or in the exotic breed. By comparison, allele A at Msp I locus tended to be dominant, while, the allele B at Tas I locus tended to be dominant in all of the breeds analyzed. In Tibetan chickens, the preliminary association analysis revealed that no significant difference was observed between the different genotypes identified at the Msp I and Tas I loci and the laying performance traits, respectively.

RAG-1 and IgM Genes, Markers for Early Development of the Immune System in Olive Flounder, Paralichthys olivaceus

  • Fish larvae are immediately exposed to microbes from hatching to maturation of their lymphoid organs, therefore effective innate mechanisms is very important for survival. However, the knowledge of the development of immune system in fish is limited and in demand now. In vertebrates, recombination-activating gene 1 (RAG-1) and immunoglobulin M (IgM) have been considered as very useful markers of the physiological maturity of the immune system. In this study, the expression of the both genes was assessed throughout the early developmental stages of olive flounder larvae (5-55 dph) and used as markers to follow the development of immune system. RAG-1 and IgM mRNA expression was detectable at 5 dph and remained so until 55 dph. These patterns of expression may suggest that the olive flounder start to develop its function around 5 dph. Tissue distribution was found that both genes mRNAs are only expressed in the immune-related organ such as spleen, kidney and gill. The early detection of IgM mRNA led to the investigation of its presence in oocytes. Both RAG-1 and IgM mRNA transcripts were detected in unfertilized oocytes, suggesting that they are maternally transferred. The biological significance of such a phenomenon remains to be investigated.

배 에탄올 추출물이 난황에 의하여 유발된 생쥐의 천식에 미치는 영향 (Effects of pear ethanol extract on asthma induced by ovalbumin in mice)

  • Objectives : In the theory of Korean medicine, Pear has long been considered to protect throat, bronchus and lung. Pear has been believed to remove sputum in Korean people. This study was conducted to investigate the effect of pear ethanol extract (PEE) on asthma induced by ovalbumin in mice. Methods : We investigated the effects of PEE on airway hyperresponsiveness to methacholine, production levels of ovalbumin (OVA) specific total immunoglobulin G (IgG), IgG1, IgG2a and IgE in serum and histopathological changes of lung tissues in asthamtic mice. Results : PEE decreased airway hyperresponsiveness to methacholine significantly compared to non-treated asthmatic mice (P<0.05). Level of OVA specific IgE in serum was lowered by oral administration of PEE effectively (P<0.05). In histopathological observation, administration of PEE reduced infiltration of immune cells into lung tissue. Conclusion : These results suggest that pear has anti-asthmaitc action and related mechanims are involved in anti-inflammatory action such as reducing level of OVA specific IgE and immune cell infiltration.

항체 : 치료제로서의 부활 (Resurrection of antibody as a therapeutic drug)

  • Currently 18 monoclonal antibodies were approved by FDA for inj ection into humans for therapeutic or diagnostic purpose. And 146 clinical trials are under way to evaluate the efficacy of monoclonal antibodies as anti-cancer agents, which comprise 9 % of clinical trials in cancer therapy field. When considering a lot of disappointment and worries existed in this field during the past 15 years, this boom could be called as resurrection. Antibodies have several merits over small molecule drug. First of all it is easier and faster in development, as proper immunization of the target proteins usually raises good antibody response. The side effects of antibodies are more likely to be checked out in immunohistomchemical staining of whole human tissues. Antibody has better pharmacokinetics, which means a longer half-life. And it is non-toxic as it is purely a "natural drug. Vast array of methods was developed to get the recombinant antibodies to be used as drug. The mice with human immunoglobulin genes were generated. Fully human antibodies can be developed in fast and easy way from these mice through immunization. These mice could make even human monoclonal antibodies against any human antigen like albumin. The concept of combinatorial library was also actively adopted for this purpose. Specific antibodies can be screened out from phage, mRNA, ribosomal library displaying recombinant antibodies like single chain Fvs or Fabs. Then the coding genes of these specific antibodies are obtained from the selected protein-gene units, and used for industrial scale production. Both $na\ddot{i}ve$ and immunized libraries are proved to be effective for this purpose. In post-map arena, antibodies are receiving another spotlight as molecular probes against numerous targets screened out from functional genomics or proteomics. Actually many of these antibodies used for this purpose are already human ones. Through alliance of these two actively growing research areas, antibody would play a central role in target discovery and drug development.

  • Objective: Bee Venom(BV) has been used for various kinds of inflammatory or painful conditions in Oriental Medicine clinics, and there publishes reports on its therapeutic effects and the probable mechanism of those therapeutic effects, where CDs and cytokines plays important role. This study investigated the influences of bee venom on the expressions of CDs and cytokines of HMC cell line Methods: In this study we analysed the expression profile of HMC cell line treated with BV of 10-2ug/ml in relation to that of HMC cell line treated with vehicle by way of CD/cytokine microarray hybridization with 342 genes on it. Results: There were no upregulated genes by more than 3 fold, while there showed some downregulated genes by less than 1/3 fold as follows: colony stimulating factor 2, CD122, IL-7, CD112, TNF-alpha, CD138, CD166, TGFbetaR2, CD42b, CD62L, CD111, interleukin 10 receptor alpha, colony stimulating factor 1(macrophage), CD38 antigen(p45), CD121a, CD33 antigen(gp67), colony stimulating factor 1 receptor, B cell linker protein (SLP65) mRNA, CD94, alanyl(membrane) aminopeptidase, immunoglobulin(CD79A) binding protein 1, CD205, CD241, CD207, CDw121b, integrin alpha L(CD11a), integrin beta 1(CD29), CD91, CD42b. Conclusions: Bee venom treatment induced downregulation of some CDs or cytokines including $TNF-{\alpha}$. IL-1R with its possible implication in an antiinflammatory action of BV. Further research on expression profile changes induced by BV treatment is expected.

조기발병형 치주염의 표현형적 소집단의 IgG Subclass에 대한 연구 (The IgG subclass responses in the phenotypic subsets of the early-onset periodontitis)

  • 본 연구는 조기발병형 치주염의 서로 다른 4가지 표현형에 있어서 Porphyromonas gingivalis(Pg) 381과 Actinobacillus actinomycetemcomitans(Aa) Y4에 대한 상승된 IgG subclass의 양상을 평가하기 위해 시행하였다. Subform I(distinctive localized juvenile periodontitis pattern)에서 3명 subform II(post juvenile periodontitis pattern)에서 19명, subform III (localized but rapidly progressing pattern)에서 15명, subform IV(distinctive rapidly progressing periodontitis pattern)에서 15명의 환자를 조사하여 Pg에 대한 그들의 total IgG level과 각각의 IgG subclass level 및 Aa에 대한 IgG level을 검사했다. Pg에 대한 total IgG level은 subform II와 IV보다 subform I과 III에서 훨씬 높게 나타났다. IgG3 level이 subform I과 IV사이에서 현저한 차이가 있다는 점을 제외하고는, 다른 IgG subclass level에서 subform 사이에 아무런 차이가 없었다. Pg에 대한 IgG subclass는 single class 혹은 다양한 group에서 상승되어 나타났으며, IgG1+2+4가 가장 흔하게 발견되었고, 다음으로 IgG4 단독, IgG2 단독, IgG2+4, IgG2+3+4의 순으로 발견되었다. IgG2와 IgG4가 빈번히 상승되어 발견되었는데, 특히 severe form(subform III & IV)에서 그러했다. 뿐만 아니라, IgG level은 subform II, III, IV와 일치하여 점차적으로 증가하였고, 반면에 IgG1/IgG4 ratio는 그와 일치하여 감소되었다. 이러한 ratio의 감소는 단백질성의 오래된 항원의 과부하로 인해 immunoglobulin gene의 전환을 가능하게 한다는 것을 나타내고 있다. Aa에 대한 IgG2 level은 다른 유형보다 subform I에서 상당히 높았다. Pg에 대한 IgG2 levels이 subform I의 국소 부위에서 발생하는 disease activity와 밀접한 관련이 있으며, Aa의 경우에는 이러한 관련성이 나타나지 않았다. Pg에 대한 IgG2 level은 18-25세에서 훨씬 높은 동시에 26-35세에서는 감소했으며 결국 30대 후반에서는 더 높은 수치로 되돌아갔다. 이러한 결과는 Pg에 대한 IgG2 및 IgG responsiveness (single 혹은 combined)가 EOP의 severe form의 발달에 중요하게 작용하며 IgG2 levels은 IgG1/IgG4 ratio와 더불어 EOP의 localized type이 generalized type으로 계속 진행하는 것을 조절하는 역할을 하는 것으로 보인다는 것을 강하게 시사하였다.
