Effect of Different Feed Additives on Growth Performance and Blood Profiles of Korean Hanwoo Calves

  • This experiment was conducted on 60 Hanwoo calves comprising five feed additive groups, with 12 calves in each group, to determine the effects of additives at pre- and post-weaning on growth performance and blood profile. The groups were control, antibiotic (Neomycin 110 ppm), illite (2%), fermented green tea probiotics (FGTP, 0.5%), and mixed additives (FGTP 0.25%, illite 1% and licorice 0.1%). The calves were offered experimental pellet feeds ad libitum and after one month were supplied with imported timothy hay. They moved freely within the group and suckled their mother' milk during the pre-weaning stage (birth to 3 months) and were separated from their dam during the post-weaning stage (4-5 months). During the pre-weaning stage, the highest average daily gain (ADG) was recorded in the antibiotic- and mixed additive-fed groups followed by FGTP, control and illite groups. In the post-weaning stage, significantly higher total weight gain and ADG were recorded in both the FGTP and mixed additive groups compared to the other groups (p<0.05). Feed efficiency of mixed additive- and illite-fed calves were almost similar with antibiotic-fed calves compared to the other two groups, but the ADG was lowest in illite-fed calves during the pre-weaning stage. In contrast, post-weaning calves fed FGTP and mixed additives showed better feed efficiency. The values of hematological indices, differential leukocyte count, blood proteins and immunoglobulin among the additive-fed calves were not significantly different (p>0.05), although hemoglobin and hematocrit values were lower in FGTP compared to control, but similar in mixed additive and antibiotic groups. These results indicate no detrimental effects of feed additives on the blood profile of calves at both pre- and post-weaning age. Serum albumin in post-weaning calves of all feed additive groups were similar but significantly lower (p<0.05) than in the control group. Post-weaning, IgM was significantly lower (p<0.05) in illite-fed calves compared to other treatment groups, but there was no difference at pre-weaning. Considering all factors, the mixed feed additives and FGTP can be the replacement feed formula for antibiotic for Hanwoo beef calf production, especially when used post- weaning.

Effects of Oral Administration of Difructose Anhydride III on Selected Health and Blood Parameters of Group-housed Japanese Black Calves during the Preweaning Period

  • Two field studies were conducted to determine the efficacy of difructose anhydride III (DFA III) as a supplement in colostrum replacer (CR) for improving the general health status (judged on the basis of incidence of enteritis, bronchitis, and pneumonia) of group-housed suckling Japanese Black calves. In a preliminary study, CR supplemented with DFA III (6 g) was orally administered within 24 h of calving to eight individually reared calves fed colostrum (DFA III group) (Exp. 1). Subsequently, CR supplemented with DFA III (6 g) was orally administered twice within 2 and 12 h of calving to four calves (DFA III group) that were not fed colostrum (Exp. 2). In both experiments, the health status of the calves was assessed during the preweaning period. In Exp. 2, hematological and blood-chemistry parameters were analyzed 24 h after the second administration of CR and at 1 wk and 1 month after calving. The results were compared between the DFA III and control groups (without DFA III supplementation; Exp. 1: n = 10, Exp. 2: n = 4). In Exp. 1, the number of calves requiring medications for the treatment of enteritis, bronchitis, and pneumonia during the preweaning period was significantly (p<0.05) lower in the DFA III group than in the control group. In Exp. 2, supplementation of DFA III did not influence the gain in body weight of calves during the pre-weaning period. Calves in the DFA III group tended to require medications for a shorter duration than those in the control group (DFA III: 10.3 d/calf, control: 21.3 d/calf; p = 0.07). Significant differences (p<0.05) in the level of mean corpuscular hemoglobin, total protein, total cholesterol, and immunoglobulin (Ig)G were observed between the DFA III and control groups. These differences probably reflect the health and nutritional status of the calves. Additionally, the serum iron and lactoferrin concentrations at 24 h and 1 wk after calving, respectively, differed significantly between the 2 groups. These concentrations might reflect the incidence of infections after calving. The present study revealed that the administration of DFA III as a CR supplement may prevent diseases in group-housed calves during the pre-weaning period.

감맥대조탕이 DNCB로 유발된 생쥐의 아토피피부염에 미치는 영향 (Effects of Gammakdaejo-tang(GMD) on DNCB induced Atopic Dermatitis in Mice)

  • Objectives : Atopic dermatitis is a chronic inflammatory skin disease with frequent relapses. This study was to investigate the effects of Gammakdaejo-tang(GMD) in DNCB induced atopic dermatitis mice. Methods : The study was divided into five comparion groups. 2,4-dinitrochlorobenzene(DNCB) solution was applied to Nc/Nga mice to induce atopic dermatitis, followed by normal group, negative control group with distilled water, positive control group with Dexamethasione and GMD 200mg/kg or 400mg/kg. The control group was orally administered 200㎕ once daily for 4 weeks. Visual skin condition, Immunoglobulin E, Histamine, Cytokine, Immune cells, Tissue biomarkers were observed. Results : As a result of the dermatitis score evaluation, it was confirmed that the GMD-administered group improved symptoms compared to the negative control group. As a result of measuring IgE, the GMD-administered group significantly decreased compared to the negative control group. As a result of measuring Histamine, GMD group except 200mg/kg of GMD significantly decreased compared to negative control group. As a result of measuring cytokine, GMD 200mg/kg significantly reduced IL-1β, IL-6 and TNF-α compared to the negative control. 400mg/kg significantly reduced IL-1β, IL-4, IL-5, IL-6, IL-10, TNF-α and significantly increased IL-2, IFNγ. As a result of confirming the immune cells, all experimental groups showed no difference in basophil, GMD group significantly reduced monocyte and eosinophil compared to negative control group, and GMD 400mg/kg group significantly reduced white blood cell and neutrophil. And significantly increased lymphocytes. As a result of measuring the gene expression level, all GMD group significantly increased TGF-β1 compared with the negative control group, and filaggrin, VEGF and EGF were significantly increased in GMD 400mg/kg group. Epidermis, dermis thickness, and eosinophil infiltration were found to be decreased in all GMD groups compared with the negative control group. Conclusions : GMD is effective in atopic dermatitis by reducing imbalance of immune response of T cells (Th1 / Th2) and reducing skin tissue damage and inflammatory response.

폐 한방슬러지로 배양한 팽이버섯(Flammulina velutipes) 추출물의 면역세포 활성화 효과 (Effect of Flammulina velutipes Extracts Cultivated with Oriental Herbal Plants on the Activation of Immune Cells)

  • 일반적인 버섯 배지(미강 10%), 폐 한방슬러지를 10% 첨가한 배지, 그리고 미강 10%와 폐 한방슬러지 10%를 혼합한 배지에서 재배한 팽이버섯의 면역기능 강화효과를 알아보기 위해 물 추출물과 에탄올 추출물로 분리하여 실험하였다. 그 중 폐 한방슬러지 10%를 첨가한 배지에서 재배한 팽이버섯의 에탄올 추출물을 첨가하였을 때, 비장세포의 증식을 크게 유도하였으며 또한 IL-6, TNF-${\alpha}$, IFN-${\gamma}$ 분비를 유도하였다. 그리고 B세포의 증식반응과 면역글로불린 생산도 증가하였으며, 대식세포주의 일산화질소 분비와 IL-6, TNF-${\alpha}$, GM-CSF 생산도 증가하였다. 또한 복수암 유발 종양세포를 이용한 항종양 효과를 측정 한 결과 대조군의 평균 21.1일보다 실험군은 24.5일로 16.1%의 수명 연장효과가 나타났다. 따라서 폐 한방슬러지를 이용하면 수입에 의존하고 있는 버섯 재배 배지 원료를 절약할 수 있으며 면역세포조절 기능을 강화시키는 기능성 버섯을 얻을 수 있다고 생각한다.

미립자 응집반응을 이용한 C-reactive Protein의 면역 측정 법에 관한 연구 (Microparticle-based Immunonassay for the Detection of C-reactive Protein in Serum)

  • 환자의 복수와 늑막액으로부터 p-diazonium phenylphosphorylcholine(DPPC) coupled Separose-4B affinity chromatography와 hydroxylapatite chromatography를 실시하여 C-reactive protein (CRP)를 분리, 정제하였다. 정제된 CRP를 토끼에게 면역화하여 항혈청을 얻고 affinity chromatography를 하여 면역항체(IgG)를 분리하였다. 분리된 면역항체를 미립자에 감작시킨 후 미립자 응집반응에 의하여 3분내에 CRP를 측정할 수 있는 간이 면역 측정법을 개발하였다. 본 연구에서 개발된 CRP측정법의 검출범위는 0.5~20mg/㎗이며, 임상 시험 결과 0.7~2.9mg/㎗에서는 강한 응집 반응을, 5.O~l3.2mg/㎗에서는 약한 응집반응을 보였고 28mg/dl이상에서는 항원 과잉으로 인한(zone of Ag excess phenomenon) 위음성을 나타냈다. 74명의 환자 혈청을 대상으로 CRP의 농도를 조사한 결과 평균치는 3.8mg/dl이었으며 대부분의 환자에서는 10mg/dl 이하의 농도로 존재하였다. 그러므로 1차 판정시 음성을 나타낸 시료라도 혈청을 5~10배정도 희석하여 재분석한다면 오차없이 CRP 를 검출할 수 있었다. 환자 혈청을 검체로 하여 본 연구에서 개발한 면역측정법과 현재 수입 시판 중인 프랑스의 B사 제품과 일본의 I사 제품을 비교한 결과 좋은 상관관계를 보였다. 이와 같은 평가 분석을 통하여 볼 때 본 연구에서 개발한 간이 면역 측정법은 사용이 비교적 간편하며 신빙성이 있어 CRP를 스크리닝하는데 효과적임을 알 수 있었다.

  • 목적: 인간 파르보바이러스 B19 감염은 무증상에서 생명을 위협하는 합병증에 이르기까지 광범위하고 다양한 증상을 나타낸다. 우리는 파르보바이러스 B19 감염의 지역사회 내 집단발생 시기 동안, 파르보바이러스 B19 감염의 다양한 임상 양상에 대해 조사하였다. 방법: 2017년 12월부터 2019년 4월까지 부산대학교 어린이병원에서 파르보바이러스 B19 면역 글로불린 M (IgM) 또는 중합효소 연쇄 반응(polymerase chain reaction, PCR)에 의해 검출된 파르보바이러스 B19 DNA를 가진 소아의 임상적 특징을 후향적 차트 분석을 통해 알아보았다. 결과: 24명의 어린이가 파르보바이러스 B19 감염으로 진단되었다. 24명의 환자 중 12명(50%)은 레이스 형태 발진이 있었고, 24명의 환자 중 4명(16.7%)은 점상출혈 발진, 24명의 환자 중 2명(8.3%)은 무병소 발열로 진단되었다. 24명 중 6명의 환자(25%)는 골수무형성위기가 발생하였고, 이 중 3명은 이전에 유전 구형적혈구증, 3명은 이전에 급성 림프모구 백혈병으로 진단되었던 환자이다. 결론: 감염홍반 외에도 파르보바이러스 B19 감염의 임상적 특징은 다양한 종류의 발진과 무병소 발열으로 나타날 수 있다. 또한, 혈액학적 증상으로 중성구 감소증, 골수무형성 위기와 같이 발생할 수 있다.

위암발증 원인균에 대한 생활습관 및 영양섭취에 있어서 농촌지역 주민조사 (Dietary Factors and Life Style Affecting the Causal Infection on the Gastric Cancer of Adults in Rural Korea)

  • 위암발증 원인을 밝히기 위하여 농촌지역 일반 주민들의 Helicobater pylori의 감염율과 생활습관 및 영양섭취 실태와의 상관관계를 검토하기 위하여 강원도 춘천시 인근 농촌 지역 주민들을 조사하였다. 조사 대상자는 40대 이상인 지역 주민 183명으로 남자 81명, 여자 102명에 대하여 설문조사와 식사량 조사, 혈액 생화학적 분석 및 혈청 ELISA법에 의한 Helicobater pylori의 감염 정도를 분석하였다. 그 결과를 요약하면 다음과 같다. 1. 조사 대상자의 학력은 74.3%가 국졸이하였고, 월수입은 80.3%가 100만원 이하였다. 2. 조사 대상자의 직업은 75.3%가 농업에 종사하였으며, 가족형태는 62.8%가 핵가족이었다. 3. 출산 자녀수는 75.9%가 2명에서 5명의 자녀를 출산하였고, 음주자는 46.4%이며 매일 술을 마시는 사람이 12%였다. 흡연자는 21.9%였으며, 비흡연자는 78.1%였다. 4. 평균신장은 157.4cm였고 여자의 평균 신장은 151.9cm, 남자의 평균 신장은 164.4cm였고 남녀간에 유의차가 있었다(p<0.001). 5. Helicobater pylori의 감염율은 총 조사 대상자 중 64.5%였으며, 남자가 65.4%, 여자가 63.7%였다. 감염율은 흡연자, 학력 수준이 낮을수록, 월수입이 적을수록, 출산 자녀수가 많을수록, 50대의 연령에서 많은 것으로 나타났다. 6. Helicobater pylori의 감염 음성자와 양성자간의 영양소 섭취실태를 보면 음성자가 에너지, 총지질(p<0.05), 동물성 지질(p<0.05), 철분 섭취량(p<0.001), 비타민 A의 섭취량(p<0.01), 비타민 C 섭취량(p<0.05)에 있어서 유의적으로 높은 섭취량을 나타내었다. 그러나 칼슘 섭취량은 음성인자가 낮았다.

A novel BTK gene mutation, c.82delC (p.Arg28 Alafs*5), in a Korean family with X-linked agammaglobulinemia

  • X-linked agammaglobulinemia (XLA) is a hereditary humoral immunodeficiency that results from Bruton's tyrosine kinase (BTK ) gene mutations. These mutations cause defects in B-cell development, resulting in the virtual absence of these lymphocytes from the peripheral circulation. Consequently, this absence leads to a profound deficiency of lg all isotypes, and an increased susceptibility to encapsulated bacterial infections. A 15-month-old Korean boy presented with recurrent sinusitis and otitis media after 6 months of age, and had a family history of 2 maternal uncles with XLA. Laboratory tests revealed a profound deficiency of Ig isotypes, and a decreased count of $CD19^+$ B cells in the peripheral circulation. Based on his family history and our laboratory test results, he was diagnosed with XLA. We performed BTK gene analysis of peripheral blood samples obtained from family members to confirm the diagnosis. Mutational analysis revealed a novel hemizygous frameshift mutation (c.82delC, p.Arg28Alafs*5), in the BTK gene. His mother and maternal grandmother were heterozygous carriers of this mutation and his two maternal uncles were hemizygous at the same position. After XLA diagnosis, intravenous immunoglobulin (400 mg/kg, monthly) treatment was initiated; recurrent sinusitis and otitis media were subsequently brought under control. To our knowledge, this is the first reported case of a Korean pedigree with a novel mutation in the BTK gene.

매생이 열수추출물의 면역 및 항암 활성 (Immunostimulating and Anticancer Activities of Hot Water Extract from Capsosiphon fulvescens)

  • 본 연구에서는 매생이 열수추출물의 항암 및 면역 활성을 알아보기 위하여 적정 추출 및 정제조건, 당 조성, 암세포독성, sarcoma-180 성장저지 효과, 백혈구수 변화, 보체계 활성, 장관 면역 활성, 경구독성 등을 검토하였다. 매생이 보체계 활성물질은 $100^{\circ}C$의 물로 3시간 동안 추출하여 4배량의 주정으로 24시간 침전시킨 다음 투석 및 한외여과하여 분자량 300kDa 이상의 고분자 물질을 분리정제 하였으며 당 조성은 xylose 19.01%, fucose 15.29%, mannose 4.23%, galactose 7.92%로 나타났다. In vitro에서 매생이 보체계 활성물질의 인체 암세포에 대한 독성은 고농도($10-30{\mu}g/ml$)에서만 경미하게 나타났으며, in vivo시험에서는 매생이 추출물의 투여로 마우스 고형암의 성장이 대조군에 비하여 40.13-59.42%가 저해되었다. 그러나 수명 연장율은 실험군간에 통계적 유의차가 없었다. 정제한 매생이 추출물을 마우스의 복강에 투여한 결과, 혈중 백혈구수가 대조군에 비하여 20mg/kg 투여군 39%, 50mg/kg 투여군 70%, 100mg/kg 투여군 83%가 증가하는 것으로 나타났으며, 면역과 관련이 있는 간장, 비장, 흉선의 중량이 증가하였다. 매생이 추출물을 마우스에 경구투여하고 분변으로 배출되는 IgA 항체의 양을 측정한 결과, 대조군이 $2,092{\pm}123.0{\mu}g/mg$인 데에 비하여 매생이 추출물 투여군은 $2,454{\pm}113.8-2,670{\pm}133.1{\mu}g/mg$로 높게 나타났다. 매생이로부터 추출한 면역증강물질은 마우스에 대한 급성독성을 측정한 결과, 체중 1kg당 2,000mg 투여 시 까지도 독성을 나타내지 않아 인체에도 무해할 것으로 사료되었다.

Korean Red Ginseng improves atopic dermatitis-like skin lesions by suppressing expression of proinflammatory cytokines and chemokines in vivo and in vitro

  • Background: The prevalence of allergic inflammatory diseases such as atopic dermatitis (AD), asthma, and allergic rhinitis worldwide has increased and complete recovery is difficult. Korean Red Ginseng, which is the heat-processed root of Panax ginseng Meyer, is widely and frequently used as a traditional medicine in East Asia. In this study, we investigated whether Korean Red Ginseng water extract (RGE) regulates the expression of proinflammatory cytokines and chemokines via the mitogen-activated protein kinases (MAPKs)/nuclear factor kappa B ($NF-{\kappa}B$) pathway in allergic inflammation. Methods: Compound 48/80-induced anaphylactic shock and 1-fluoro-2,4-dinitrobenzene (DNFB)-induced AD-like skin lesion mice models were used to investigate the antiallergic effects of RGE. Human keratinocytes (HaCaT cells) and human mast cells (HMC-1) were also used to clarify the effects of RGE on the expression of proinflammatory cytokines and chemokines. Results: Anaphylactic shock and DNFB-induced AD-like skin lesions were attenuated by RGE administration through reduction of serum immunoglobulin E (IgE) and interleukin (IL)-6 levels in mouse models. RGE also reduced the production of proinflammatory cytokines including $IL-1{\beta}$, IL-6, and IL-8, and expression of chemokines such as IL-8, thymus and activation-regulated chemokine (TARC), and macrophage-derived chemokine (MDC) in HaCaT cells. Additionally, RGE decreased the release of tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), $IL-1{\beta}$, IL-6, and IL-8 as well as expressions of chemokines including macro-phage inflammatory protein $(MIP)-1{\alpha}$, $MIP-1{\beta}$, regulated on activation, normal T cell expressed and secreted (RANTES), monocyte chemoattractant protein (MCP)-1, and IL-8 in HMC-1 cells. Furthermore, our data demonstrated that these inhibitory effects occurred through blockage of the MAPK and $NF-{\kappa}B$ pathway. Conclusion: RGE may be a useful therapeutic agent for the treatment of allergic inflammatory diseases such as AD-like dermatitis.
