Plant-derived PAP proteins fused to immunoglobulin A and M Fc domains induce anti-prostate cancer immune response in mice

  • In this study, recombinant Fc-fused Prostate acid phosphatase (PAP) proteins were produced in transgenic plants. PAP was fused to immunoglobulin (Ig) A and M Fc domain (PAP-IgA Fc and PAP-IgM Fc), which were tagged to the ER retention sequence KDEL to generate PAP-IgA FcK and PAP-IgM FcK. Agrobacterium-mediated transformation was performed to produce transgenic tobacco plants expressing four recombinant proteins. Genomic PCR and RT-PCR analyses confirmed the transgene insertion and mRNA transcription of PAP-IgA Fc, PAP-IgM Fc, PAP-IgA FcK, and PAP-IgM FcK in tobacco plant leaves. Western blot confirmed the expression of PAP-IgA Fc, PAP-IgM Fc, PAP-IgA FcK, and PAP-IgM FcK proteins. SEC-HPLC and Bio-TEM analyses were performed to confirm the size and shape of the plant-derived recombinant PAP-Fc fusion proteins. In mice experiments, the plant-derived IgA and IgM Fc fused proteins induced production of total IgGs including IgG1 against PAP. This result suggests that IgA and IgM Fc fusion can be applied to produce recombinant PAP proteins as a prostate cancer vaccine in plant expression system.

A quantitative study of the transfer of colostral immunoglobulins to the newborn Korean native calf (한우 송아지의 초유섭취에 의한 수동면역 획득 상태)

  • The levels of the serum and colostral total protein and immunoglobulins of the Korean native cows immediately after parturition and colostrum-conferred passive immune status of the Korean native calves were studied, and the results obtained were summerized as follows: The mean total protein, total immunoglobulin, IgG, IgM, and IgA concentrations in sera of 10 Korean native cows immediately after parturition were $6.8{\pm}0.4g/dl$, $24.39{\pm}3.41mg/ml$, $19.49{\pm}2.70mg/ml$, $4.56{\pm}1.14mh/ml$, and $0.35{\pm}0.08mg/ml$ respectively. And the mean total protein, total immunoglobulin, IgG, IgM, and IgA concentrations in colostrum were $15.0{\pm}1.1g/dl$, $116.13{\pm}23.07mg/ml$, $101.51{\pm}22.59mg/ml$, $9.46{\pm}1.99mg/ml$, and $5.17{\pm}1.59mg/ml$ respectively. The mean concentrations of total protein and immunoglobulins in colostrum were 2.1 times to 14.8 times higher than those in serum. The mean concentrations of total protein, total immunoglobuln, IgG, IgM, and IgA in sera from 211 Korean native calves at 2 days old were $6.7{\pm}0.9g/dl$, $35.74{\pm}9.56mg/ml$, $29.06{\pm}8.07mg/ml$, $4.91{\pm}2.99mg/ml$, and $1,9{\pm}0.99mg/ml$ respectively. Although the calves had been suckled the dam's colostrum, each of calves was considerably various in levels of serum total protein and immunoglobulins and 16.1 percent of the calves were in a state of hypoglobulinemia.

  • Noninfectious aortitis, inflammatory abdominal periaortitis, and idiopathic retroperitoneal fibrosis are chronic inflammatory diseases with unclear causes. Recent studies have shown that some cases of aortitis are associated with immunoglobulin G4 (IgG4)-related systemic disease. Herein, we report a case of IgG4-related aortitis (IgG4-RA) that was diagnosed after surgery. Our patient was a 46-year-old man who had experienced abdominal pain for several weeks. Preoperative evaluations revealed an area of aortitis on the infrarenal aorta. He underwent surgery, and histological examination resulted in a diagnosis of IgG4-RA.

Effects of Curcuma aromatica or inositol monophosphate supplementation on growth performance and immune status of lactating sows and piglets

  • The aim of the present study was to investigate the influences of Curcuma aromatica or inositol monophosphate supplementation on body weight of sows at different stages, feed intake, backfat thickness of sows at different stages, body weight of piglets at different stages, and immunoglobulin G (IgG) concentration in sow blood and milk. Eighteen crossbred (Landrace × Yorkshire) sows (249.9 ± 3.2 kg) and their litters were used in a 28-day feeding trial to observe the effects of Curcuma aromatica or inositol monophosphate as dietary supplements on performance and IgG concentration of blood and milk in lactating sows and piglets. The dietary treatments comprised a control corn-soybean-based basal diet (CON); control diet + Curcuma aromatica at 0.5% (CA), and control diet + inositol monophosphate at 0.10% (IMP). Sow body weight at different stages, average daily feed intake, and sow backfat thickness at different stages were not affected in all three treatment groups. The body weight of piglets at weaning and average daily gain of piglets born to sows from the IMP group showed significant improvement compared to piglets of sows from the CA treatment group. Treatment had no effect on the IgG levels in blood and milk. In conclusion, supplementation of 0.5% CA or 0.10% IMP in sows has no effect on growth performance and IgG in sows and piglets compared with the control diet.

  • Immunoglobulin G4-related disease (IgG4-RD) is an immune-mediated fibro-inflammatory disorder characterized by specific pathological findings and elevated serum IgG4 level. IgG4-RD in the stomach is rare, and occasionally diagnosed as gastric subepithelial tumor (SET) by endoscopy or computed tomography scan. Two female patients in the age group of 40-50 years were diagnosed with 4 cm sized gastric SET. One underwent laparoscopic gastric wedge resection. Another one had a history of subtotal gastrectomy for early gastric cancer and idiopathic thrombocytopenic purpura with oral steroids administration. She underwent a completion total gastrectomy with splenectomy for the gastric SET and ITP. The pathology showed storiform fibrosis, and IgG4 was positive in immunohistochemistry (IHC) stain. IgG4-RD is known as a medical disease that could be treated with oral steroids. The difficulty in preoperative diagnosis of the disease occasionally causes unnecessary gastric resection. Thus, preoperative diagnostic methods for IgG4-RD such as deep biopsy with IHC stain or magnetic resonance imaging are needed.

Seroprevalence of Dengue Virus Antibody in Korea (한국인에서 뎅기바이러스 항체의 혈청 유병률 연구)

  • Purpose: The number of dengue fever cases is rising due to increasing overseas travel. Vaccination makes severe dengue fever in seronegative individuals after vaccination when they exposure to wild-type dengue virus. We investigated the seroepidemiology of the dengue virus for monitoring of Korean dengue virus immunity and establishing the prevention of dengue infection. Methods: The study was based on 446 residual sera collected from 98 infants (2 months to 1 year old), 152 adolescents (13 to 19 years old), 90 adults (20 to 50 years old), and 106 elderly participants (more than 65 years old) for other studies. Antibody levels for dengue virus immunoglobulin G (IgG) in each age group were measured using an enzyme-linked immunosorbent assay (ELISA). For each dengue virus IgG positive or equivocal result, an IgG ELISA was performed for Japanese encephalitis virus. Results: Of the 446 serum samples, only 1 (0.2%) adolescent had a positive result from the dengue IgG antibody test. In the dengue virus IgG antibody test, 14 (3.1%) samples showed equivocal results (10 adolescents and 4 elderly). In the 1 positive case of dengue virus IgG, the Japanese encephalitis IgG test was also positive. In the 14 equivocal cases of dengue virus IgG, there were 6 positive, 3 equivocal, and 5 negative of Japanese encephalitis IgG. Conclusions: The seroprevalence rate of dengue virus was very low in Koreans. This study provides important data for establishing the policy for preventive measures of dengue fever. It will be necessary to continuously monitor for dengue virus immunity.

Identification of the Chicken Meat Allergens (닭고기 중 알레르기 유발성분의 동정)

  • The chicken meat has been reported as one of the food causing allergic reactions predominantly to Korean. At present, several in vitro tests for immunoglobulinG (IgG)-mediated as well as IgE-mediated food allergy are available. 13 clinically chicken meat-allergic patients were investigated together with 4control subjects for identification of chicken meat-specific reactivity by ELISA. Also, protein profile and IgE, IgGtotal and IgG4-reacting allergens were detected by means of sodium dodecyl sulfate-polyacrylamide gel electro-phoresis (SDS-PAGE)and immunoblotting. Chicken meat extracts were prepared as raw, heated, heat and simulated gastric fluid (SGF) treated samples to characterize the stability of allergen to physicochemical treatment. SDS-PAGE revealed 9~200 kDa bands. And in immunoblotting 7 sera were identified most major bands between 10 and 78 kDa. In case of IgE, six proteins (17, 26, 35, 40, 78 kDa) were predominant in heat-treated extract, and the one (35 kDa) was present in SGF-treated preparations. In case of IgG$_{total}$ and IgG4, most of them showed a patters simmilar to IgE. There were significant differences (P<0.05) in IgE, IgG$_{total}$ , IgG4 Abs to chicken meat between the allergic and control subjects in ELISA. In addition, the concentration of IgG4Abs in the challenge-positive subjects was significantly higher than that of control subjects. It is considered that the specific IgE response to chicken meat was rarely prevalent to Koreans. However, the specific IgG4 response play an important role in the development of allergic symptoms.

  • Egg yolk immunoglobulin (IgY) is much widely used in medical fields, but its use in serology of plant viruses is much limited. We produced an IgY against pepper mild mottle virus (PMMoV) and applied it to several serological tests. Polyclonal antibodies were obtained from the egg yolk of chicken immunized with a total of 2mg of purified PMMoV over 2 months. The titers of antibodies were measured with the ring-test over six months after the first injection. The highest.titers of IgY was 1/2,560 at 2 months after the first injection. Approximately 60-80 mg of IgY were obtained from one egg yolk. Using the IgY, 1ng/ml of purified PMMoV was detected with the indirect ELISA. Gelrite gel double diffusion test, ELISA and tissue immuno-binding assay employing IgY gave similar sensitivity and specificity to those of IgG developed in rabbit. Therefore, the IgY which can be obtained in large quantities from a chicken, might be useful for the antibody production and the serology of plant viruses.

The Effects of the Low Power Wavelengths of Helium-Neon Infra- Red Laser on the Changes of the Serum Immunoglobulin Concentration in Burn Rats (저 에너지 레이저 파장이 화상쥐 혈청면역물질 농도 변화에 미치는 영향)

  • This studies were to investigate the effects of low power wavelengths Helium-Neon Infra-Red(He-Ne 1R) laser on the changes of the serum immunoblobulin(Ig) concentrations in bum rats, The thirty Spraque-Dawley adult male rats were assigned to the 5 groups: the experimental groups(3). the bum control group(1) and the control group(1). There was made three degree burn by the 290 mW IR on the back oreach rats, from 3 days after being burned. the experimental laser groups were irradiated low power wavelengths(292Hz- 1168Hz, 4672Hz) He-Ne IR laser far 5 minutes every day during the 7days. The results were as follows; 1. The concentrations of immunoglobulin G(Ig G) in serum of all experimental groups on the treated with the low power wavelengths of the He-Ne IR laser during the 5 minutes far 7 days were significantly lower than those of bum groups, but those 1168 Hz group were significantly higher than those of the 292 Hz and 4672 Hz groups. 2. There were significantly decreased the change of the level of immunoglobulin M(Ig M) in serum of all experimental groups on the treated with 5 minutes laser fer 7 days to the burn control group. and were significantly increased on treated with more higher wavelengths groups to those of lower groups. 3. The concentrations immunoglobulin E(Ig E) in serum of the 292Hz wavelengths groups were significantly lower than those in bum control group. As above results. the changes of the immunoglobulin in serum levels on the healing process have meaningful effected with the low power wavelengths role on the treated with He-Ne IR laser.

A Case of Immunoglobulin G4-Related Disease Presenting as a Pleural Mass

  • Immunoglobulin G4 (IgG4)-related disease is a newly recognized condition characterized by fibroinflammatory lesions with dense lymphoplasmacytic infiltration, storiform-type fibrosis and obliterative phlebitis. The pathogenesis is not fully understood but multiple immune-mediated mechanisms are believed to contribute. This rare disease can involve various organs and pleural involvement is even rarer. We report a case of IgG4-related disease involving pleura. A 66-year-old man presented with cough and sputum production for a week. Chest radiography revealed consolidation and a pleural mass at right hemithorax. Treatment with antibiotics resolved the consolidation and respiratory symptoms disappeared, but the pleural mass was unchanged. Video-assisted thoracoscopic surgery was performed. Histopathology revealed dense lymphoplasmacytic infiltration and storiform fibrosis with numerous IgG4-bearing plasma cells. The serum IgG4 level was also elevated. Further examination ruled out the involvement of any other organ. The patient was discharged without further treatment and there is no evidence of recurrence to date.
