This study examined the chemical composition of A. blasiliensis and the chemical structural properties of an immuno-stimulating polysaccharide. The amino acids, free sugars, and organic acids by HPLC and fatty acids by GC were analyzed. The immuno-stimulating substance from A. blasiliensis was extracted with hot water and purified by ethanol precipitation. It underwent ion exchange chromatography on DEAE-cellulose and gel filtration on Toyopearl HW 65F. Through GP-HPLC, the substance was found to be homogeneous. Its chemical structure was determined by $^{13}C-NMR$. Fatty acids, organic acids, and sugar alcohol composition consisted exclusively of linoleic acid, fumaric acid and mannitol, respectively. The amino acids were mainly glutamic acid, glycine, and arginine. By $^{13}C-NMR$ analysis, the immuno-stimulating substance was identified as ${\beta}-(1{\rightarrow}3)\;(1{\rightarrow}6)$-glucan, composed of a backbone with $(1{\rightarrow}3)$-linked D-glucopyranosyl residues branching a $(1{\rightarrow}6)$-linked D-glucopyranosyl residue. The ${\beta}$-glucan from A. blasiliensis showed pronounced immuno-stimulating activity on the antibody-production ability of B-lymphocytes by the hemolytic suspension assay. In these results, A. blasiliensis was estimated to have potent pharmacological properties and potential nutritional values.

Objectives : This experimental study was designed to investigate the effects of Acanthopanax, sessiliflorus SEEM extracts following gamma-ray irradiation on immuno-stimulating and anti-tumor activity in terms of proliferation of tumor cells, thymocytes, splenocytes, and NO production from peritoneal macrophages in mice. Methods: 10AS and 100 AS were the bark powders of Acanthopanax sessiliflorus $S_{EEM}$ stems which were exposed in 10 kGy or 100 kGy of electron beam respectively. Results : Treatment with either 10AS or 100AS increased proliferation rates of thymocytes and splenocytes significantly, and treatment with l0AS also decreased proliferation rates of tumor cells significantly. Treatment with either l0AS or l00AS promoted NO production from peritoneal macrophages significantly. Conclusion : These results suggested that AS has direct inhibition effect of tumor growth and immuno-stimulating activity. In conclusion, we demonstrate that AS could be used to treat cancer patient as complementary or alternative medicine to typical anti-cancer medication.
Both ethanol and water extracts of unossified velvet antler were found to exhibit a significant immuno-stimulating activity as measured by carbon clearance test in mice, a remarkable anti-fatigue effect in weight-loaded forced swimming performance in mice, a significant anti-stress effect on immobilization in rats. The antler extracts also showed a weak but significant anti-thrombotic activity. These findings are indicative of adaptogenic properties of antlers and their normalizing effects during stressful condition.
A potent immuno-stimulating activity was detected from the watersoluble and ethanol-precipitated crude extract (AG-1) of the root of Angelica gigas Nakai. The crude extract was fractionated into two fractions, an acidic AG-2 and a neutral AG-3 fraction by DEAE-cellulose adsorption. The two fractions contained polysaccharides of which M.W. were 10 Kdal and >2,000 Kdal respectively, proteins, and various inorganic components. The immunostimulating activities of two fractions were not reduced by proteinase K, acid or alkali treatment. The polysaccharides obtained from the root of A. gigas were mainly composed of arabinose, galactose, and galacturonic acid. These results indicated that immuno- stimulating components of the root of A. gigas was a kind of pectic polysaccharides or arabinogalactans.
Low molecular weight molecules and high molecular weight substances were found to have anti-tumor and immuno-modulating activity. Previously polysaccharides have been received much attention because of adhesives, food additives or animal foods (Whistler et al., 1976). In effort of developing new anti-tumor substances with low toxicity, numerous polysaccharides from yeast, algae, bacteria, higher plants and especially fungi have been investigated for anti-tumor and immuno-modulating activities. Thus the high molecular weight molecule was reported to have anti-tumor activity through host mediated immunity. In this brief article, attention will be paid to polysaccharides which is especially fungal origin.

To produce functional and high nutritional chungkook-jang, an immuno-stimulating and rapid growing bacterium was isolated from one of Korean traditional food, chungkook-jang. The isolated bacterium was identified as Bacilpumilus and deposited in Korean Collection for Type Culture (KCTC 10461BP). The chungkook-jang fermented by JB-l has good taste and pleasant smell with high content of free amino acids, and the water extract of chungkook-jang showed 410% of immuno-stimulation activity at concentration of 2 mg/ml. The rapid fermentation in 16 h is achieved by inoculation of 20 ml culture broth of JB-l into 6 kg cooked soybean in commercial scale at $40^{\circ}C$.
In recent studies on cefodizime, it may potentially have the capability of stimulating chemotactic activity of neutrophils and monocytes as well as the strong immuno-modulator. In turn, infection can result in a drastic change of mediators. which lead to initiate an immune response in an indirect way. With this backgrounds, we have studied to see if cefodizime can be a potential substance to induce an immunological function in dendritic cells and peritoneal macrophages. (omitted)
In this study, polysaccharides were isolated from Korean persimmon vinegar to characterize the polysaccharides existing as soluble forms within traditional Korean fermented beverages, and their immuno-stimulating activities were examined. Three successive chromatographies were used to purify the main polysaccharide in the persimmon vinegar, PV-1b-I, to homogeneity from the crude polysaccharide (PV-0). The molecular mass of PV-1b-I was estimated as 110 kDa and it contained significant proportions of mannose (46.8%), galactose (28.5%) and arabinose (19.1%). PV-1b-I strongly reacted with ${\beta}$-glucosyl Yariv reagent, suggesting the presence of an arabino-3,6-galactan moiety. PV-1b-I also induced high levels of macrophage activation and mitogenicity on murine splenocytes in vitro. The intravenous administration of PV-1b-I significantly augmented NK cytotoxicity against YAC-1 tumor cells. PV-1b-I also showed potent anticomplementary activity in a dose-dependent manner. Finally, C3 activation products were identified by crossed immunoelectrophoresis using anti-human C3 and the anti-complementary activity of PV-1b-I under $Ca^{2+}$-free conditions, suggesting that this PV-1b-I causes complementary activations via both alternative and classical pathways. From these results, one can conclude that Korean persimmon vinegar contains select polysaccharides in addition to healthy components, and these polysaccharides appear to provide immuno-stimulating activities beneficial to human health.
In order to develop new physiologically active polysaccharides from persimmon leaves, two different crude polysaccharides were prepared using hot water (PLW-0) and pectinase digestion (PLE-0) and their immuno-stimulating activities were estimated. PLW-0 and PLE-0 showed similar sugar compositions with 15 different sugars, including rarely observed sugars in general polysaccharides such as 2-O-methyl-fucose, 2-O-methyl-xylose, apiose, aceric acid, 3-deoxy-D-manno-2-octulosonic acid, and 3-deoxy-D-lyxo-2-heptulosaric acid, but the uronic acid content of PLE-0 was lower than that of PLW-0 caused by pectinase treatment. Both PLW-0 and PLE-0 showed potent anti-complementary activity in a dose-dependent manner which was similar to a known immuno-stimulating polysaccharide, PSK, from Coriolus versicolor. The activity of PLE-0 at a low concentration ($100{\mu}g/m{\ell}$) was higher than that of PLW-0. In an in vitro cytotoxicity analysis, PLW-0 and PLE-0 (up to $1,000{\mu}g/m{\ell}$) did not affect the growth of peritoneal macrophages and Colon 26-M3.1 carcinoma cells. In contrast, they enhanced lymphocyte proliferation activity. Peritoneal macrophages stimulated with PLW-0 and PLE-0 produced various cytokines, such as IL-6 and IL-12. However, PLE-0 was more effective on the cytokine production. Intravenous administration of PLW-0 and PLE-0 significantly augmented natural killer (NK) cell cytotoxicity against Yac-1 tumor cells 3 days after the treatment of polysaccharide fractions. But NK cells obtained from the PLE-treated group showed higher tumoricidal activity even at a low dose of $40{\mu}g$/mouse. In experimental lung metastasis of Colon 26-M3.1 carcinoma cells, prophylactic administration of PLW-0 and PLE-0 significantly inhibited lung metastasis in a dose-dependent manner and PLE-0 was more effective on the inhibition of cancer metasasis. The results lead us to conclude that the pectinase-treated process is indispensable to preparing polysaccharides with higher immune-stimulating activity from persimmon leaves.
Polysaccharides isolated from Korean black raspberry wine were examined for their chemical properties and immuno-modulating activities. The molecular mass of RB-1b-I, the main polysaccharide in black raspberry wine, was estimated as 180 kDa and it contained a significant proportion of mannose (76.8%) and 4 different minor component sugars such as galactose (15.8%), arabinose (3.8%), glucose (2.6%) and rhamnose (1.2%). This indicated that RB-1b-I was mainly present as a mannan, which had originated from the cell walls of fermenting yeasts. On the other hand, RB-1b-I induced high levels of macrophage activation as well as mitogenicity regarding murine splenocytes in vitro. The intravenous administration of RB-1b-I significantly augmented NK cytotoxicity against YAC-1 tumor cells. RB-1b-I also showed potent anti-complementary activity in a dose-dependent manner via both alternative and classical pathways. Results indicated that Korean black raspberry wine contains peculiar polysaccharides which provide beneficial immuno-stimulating activities for human health.
