A 7-year-old, female snow goose (Anser caerulescens hyperboreus) with history of decreased activity for 2 month died in Daejeon Zoo Land in September 2006. At necropsy, granulomatous pneumonia and hepatomegaly with multiple cysts were observed. Small masses were found in the spleen. Microscopically, fibrinous pneumonia distributed in most of the lung lobe with pulmonary edema and congestion. Especially, granulomatous inflammation with numerous multinucleated giant cells was observed around the dilated bronchi. To confirm the diagnosis, acid-fast (Ziehl-Neelsen method) and periodic acid-Schiff (PAS) staining was performed. Acid-fast staining showed red bacterial colony indicating tuberculosis. PAS staining was also positive enough to diagnose aspergillus spp. co-infection that was an opportunistic fungi occurring in immuno-compromised animals. Based on the above results, we confirmed that the case submitted was diagnosed as avian tuberculosis.
Expressions of cellular adhesion molecules (CAMs) are closely related to the formation of early atherosclerosis, an age-dependent vascular disorder. However. previous research provided only limited and conflicted reports on age-related alterations of CAMs' expressions and even much less is known the modulation of CAMs by calorie restriction (CR), In this study, expression of vascular cell adhesion molecule-1 (VCAM-1), intercellular adhesion molecule-1 (ICAM-1), E-selectin, P-selectin and platelet/endothelial cell adhesion molecule-1 (PECAM-1) in aorta and kidney were investigated by western blot and immuno-histochemical stain utilizing ad libitum (AL) and CR rat. Nocardia, aerobic members of the order of Actinomycetaceae, produces infections in human lung. Nocardial infection is associated with underlying diseases of immuno-suppression or treatment with corticosteroid. It is difficult to detect Nocardia by sputum examination or histologic sections and it has rarely been diagnosed by fine needle aspiration of the lung. We describe a case of pulmonary nocardiosis in a 72 year-old man, diagnosed by fine needle aspiration, which was confirmed by culture of aspirates. The aspirates showed neutrophil-predominant inflammatory cells with microorganisms demonstrated by Gomori methenamine silver and Gram stain. The organisms had characteristic long blanching filamentous structures. The lesions on chest X-ray were in resolution with antimicrobial therapy.
Objectives : The aim of this study was to investigate the asthma-suppressive and immuno-regulatory effect of Notopterygii Rhizoma(NR) extract on OVA(ovalbumin)-induced asthma in mice. Methods : C57BL/6 mice out of all the experimental groups, except the Normal group and the NRI group, were sensitized and challenged with OVA. C57BL/6 mice were exposed to OVA three times a week for 12 weeks and analyzed by flow cytometer, ELISA, H&E stain. Results : The concentrations of IL-4, IL-5, IL-13, IgE in serum of the OVA-NRII group decreased significantly compared with those of the OVA-Control group. The number of $Gr-1^+/CD11b^+$, $CCR3^+$, $CD3^+/CD19^+$, $CD3e^+/CD69^+$cells in the OVA-NRI group decreased significantly compared with those of the OVA-Control group. The collagen accumulation in the lung sections of the OVA-NRII group decredased signi- ficantly compared with that of the OVA-Control group. Conclusions : These results suggest that Notopterygii Rhizoma(NR) would be a effective candidate for herbal-originated anti-asthmatic drug. However, this drug should be further studied for characterization of the accurate action and underlying mechanism using variant disease model in the future.
Objectives: Dojeckjiyu-tang has been used to treat Hwaseol & Jeokri. The object of this study is examination of the treatment effect of Dojeckjiyu-tang for ulcerative colitis of the mouse descending colon. Methods and Materials : Twenty-one rats were divided into 3 groups and treated as follows: the control group was untreated mice. The UCE group was ulcerative colitis elicited mice. The DJT group was Dojeckjiyu-tang treated mice after ulcerative colitis elicitation. The groups were examined with common morphology, paneth cells in intestinal crypt, absorptive cells and goblet cells in epithelium, cell division in mucose, COX-1 as mucosal protector, COX-2 (which appears to play an important role in inflammation), IL-2R-, ICMA-1-inducing cellular immuno-chainreaction, and the distribution of apoptotic cells. Results: 1. The morphology of colonic mucosa from UCE mice: the disappearance of epithelium and intestinal propria in hemorrhagic erosions were seen, but in the morphology of colonic mucosa from DJT-treated mice, the configuration of epithelium and intestinal propria were the same as normal. 2. The distribution of goblet cells and absorptive cells with microvilli in intestinal propria from UCE mice: a noticeable decrease of goblet cells and absorptive cells with microvilli were seen, but with the distribution of goblet cells and absorptive cells with microvilli in intestinal propria from DJT -treated mice, the configuration of goblet cells and absorptive cells with microvilli were the same as normal. 3. The immunohistochemical stain for BrdD in colonic mucosa and COX-1 in lamina propria from UCE mice: BrdU positive cells and COX-1 positive cells in the region of hemorrhagic erosion disappeared, but in the immunohistochemical stain for BrdU in colonic mucosa and COX-1 in lamina propria from DIT-treated mice, BrdU positive cells and COX-1 positive cells were seen. 4. The immunohistochemical stain for COX-2 in lamina propria, IL-2R-in lamina propria, intestinal propria and submucosa and ICMA-1 in intestinal propria and submucosa from DCE mice: a noticeable increase COX-2, IL-2R-, ICMA-1 positive cells were seen, but in the immunohistochemical stain for COX-2 in lamina propria, IL-2R-in lamina propria, intestinal propria and submucosa and ICMA-1 in intestinal propria and submucosa from DJT-treated mice, a numerical decrease of COX-2, IL-2R-, ICMA-1 positive cells was observed. 5. The distribution of apoptotic cells in epithelium and lamina propria from UCE mice: a noticeable increase of apoptotic cells in region of hemorrhagic erosion was seen, but in the distribution of apoptotic cells in epithelium and lamina propria from DJT-treated mice, a remarkable decrease of apoptotic cells was seen. Conclusions: According to the above results, Dojeckjiyu-tang has a moderate effect on ulcerative colitis in descending colon.
Although Mycobacterium tuberculosis complex strains remain responsible for the majority of diseases caused by mycobacterial infections worldwide, the increase in HIV (human immuno deficiency virus) infections has allowed for the emergence of other non-tuberculous mycobacteria as clinically significant pathogens. M. tuberculosis was detected by two-tube nested polymerase chain reaction (PCR) and non-tuberculous mycobacteria was detected by PCR-restriction fragment length polymorphism (RFLP) with Msp I. Result of niacin test is equal to result of two-tube nested PCR after culture for M. tuberculosis. In this study, acid fast bacilli stain (AFB. stain) >2+ case, Detection of Mycobacteria is similar to result before culture and after culture. AFB. stain <1+ case, result of mycobacteria is distinguished. Conclusionly, these results suggest that identification of mycobacteria must go side by side both culture and PCR for more fast and accuracy.
Pneumocystis carinli is an established cause of pulmonary infections in immuno-compromised hosts. Several cytoiogical stains, such as Papanicolaou, Gomori methenamine sliver(GMS) and Diff-Quik have been used for detection of the organism, but occasionally can be laborious and, due to a degree of nonspecificity, may be misleading. We evaluated the diagnostic utility of immunocytochenmical stains that recognize P. carinii in bornchoalveolar lavage from experimentally Induced P. carinii pneumonia rats(n=15). In audition to routine stains for diagnosis by morphologic recognition of P. carinii on Papanicolaou, GMS and Diff-Quik stains, bronchoalveolar lavage samples were reacted with immunocytochemical stains using monoclonal antibodies(MAB) 092 and 902. In bronchoalveolar lavage P. carinii organisms were detected In 9 of 10 cases(90%) using each MAB 092 and 902, whereas GMS and Diff-Quik stains demonstrated P. carinii in 13(86%) and 11(73%) of 15 cases respectively. In lung tissue specimens(n=15) P. carinii organisms were well identified on GMS stain and immunohistochemical stains using MAB 092 and 902 in ail cases. We believe that the immunocytochemical staining using MAB 092 and/or 902 is a very useful and diagnostic tool In addition to GMS and Diff-Qulk stain to detect P. carinii organisms in bronchoalveolar lavage.
This study was designed to investigate the distributions of the positive cells in rat spleens by two monoclonal antibodies of MT1 and MB2. The spleens of immature 10 rats (Sprague Duwely, approximately 200gm) were collected and paraffin-embedded sections of spleens were stained with immunohistochemical methods. Higher proportions of MT1-positive cell number in spleens were ordered as marginal zone(8.5~18.1%), red pulp(2.1~8.8%) and periarterial lymphoid sheath(0~1.6%) in white pulp, and those of MB2-positive cell number are ordered as the central area of the periarterial lymphoid sheath(100%), red pulp(29.1~45.0%), marginal zone(15.2~30.4%), and peripheral area of periarterial lymphoid sheath(2.3~3.5%). The positive cells by MB2 are more numerous in number than by MT1. The above results were concluded that the positive cells by above two monoclonal antibodies were scattered throughout the red pulp and marginal zone, but in the central area of the periarterial lymphoid sheath, the MB2-positive cells only were present.
Background: This study was undertaken to detect the patterns of ciliogenesis in newborn rats trachea. Materials and Methods: The experimental animals(Sprague-Dawley strain) were divided five groups, one day, two day, three day, five day and seven day newborns as experimental groups. To obtain differential distribution of $\alpha$-tubulins in the ciliated cells and patterns of ciliogenesis, we used immunohistochemical stain method with mouse anti $\alpha$-tubulin monoclonal antibody as the primary antibody and gold particles conjugated goat anti mouse IgG as the secondary antibody. And we observed the specimens by electron microscope (Hitach-600 Model). Results : 1) The distribution of the $\alpha$ -tubulin reactions in apical zone was slightly decreased from three day after birth. 2) From 5th day after birth, the decreasing number of gold particles in intermediate zone was remarkable. 3) On the comparison with the other zones, the number of gold particles in the Golgi zone for seven days showed no statistical difference. Conclusion : The ciliogenesis of the tracheal ciliated cells in early newborn rat were made via centriolar and acentriolar pathways, in late groups, from five day after birth, the major ciliogenesis pattern might be centriolar pathway.
The gene encoding human serum albumin (HSA) was cloned from human liver cDNA library by PCR. The HSA cDNA in size of 2,176 bp, including 1,830 bp of open reading frame, was cloned into the plasmid carried with the 5'flanking sequence of goat $\beta$-casein gene (-4,044 to +2,025 bp) to get a tissue specific expression vector in mammary gland named pGB562/HSA (12.5 kb). A 9.6 kb DNA fragment in which the sequence is in order of goat $\beta$-casein gene regulatory sequence, HSA cDNA and SV40 polyadenylation signals was isolated from the pGB562/HSA by SacI and DraIII cutting, and used to microinject into the pronuclei of mouse fertilized eggs to produce transgenic mice. Three transgenic mice (2 female and 1 male) were identified by PCR and dot Southern blot analysis. The copy numbers of integrated transgene were more than 10 copies in line #21 and #26 as well as over 50 copies in line #31 of transgenic mice. HSA protein collected from the milk of lactating transgenic mice was confirmed by immuno-detection of Western and slot blot. The concentrations of HSA in the milk were from 0.05 to 0.4 mg/ml. An obvious antigen and antibody conjugate could be observed in immunohistochemical stain of mammary gland tissue from lactating day 11 of HSA transgenic mice. The transmission of transgene and its expression was recognized according to the results of RT-PCR and sequences analyses of their progeny.
