지속적인 생균제의 첨가가 돼지의 성장, 영양소 이용율, 혈중 요소태 질소 및 면역능력에 미치는 영향 (Effect of Continuous Feeding of Probiotics on Growth Performance, Nutrient Digestibility, Blood Urea Nitrogen and Immune Responses in Pigs)

  • 본 연구의 목적은 생균제의 지속적인 첨가에 의한 돼지에게 있어 성장능력, 영양소 소화율, 혈중 요소태 질소량 및 면역반응에 미치는 영향을 평가하기 위해 실시하였다. 처리는 대조구, 대조구 사료에 0.1%의 생균제 첨가구(P-O.I), 대조구 사료에 0.2%의 생균제 첨가구 (P-0.2)로 하였다. 사양실험에서는 21 일령에 이유한 총 60마리 의 돼지(펑균 체중 6.17 $\pm$ 0.45 kg)를 공사하여 성별 빛 체중에 따라 난괴법에 의해서 배치하였다. 옥수수 대두박 위주의 실험사료에 전 기간에 걸쳐 항생제는 첨가하지 않고 생균제만을 처리별로 첨가하였다. 0${\sim}$8주간의 사양설험에서는 처리간 일당증체량, 일당사료섭취량 및 사료효율에서 유의한 차이가 나타나지 않았다 하지만 9${\sim}$20주에서는 P-O.I과 P-0.2 처리구가 대조구에 비해 유의하게 일당증체량이 증가하였으며 (P<0.05) 첨가수준이 증가할수록 그 경향은 더욱 뚜렷하였다. 전체 사양 실험 기간동안 일당증체량, 일당사료섭취량 및 사료효율은 생균제 첨가구가 대조구에 비해 증가하는 경향을 보였으나 통계적으로 유의한 차이는 없었다. 생균제에 의한 영양소 소화율 및 질소 축적율을 조사하기 위해 자돈기와 육성기에 두번에 결쳐 대사실험을 실시하였다. 자돈기(평균 체중 17.93 $\pm$ 1.45kg )에서는 건물, 조단백질, 조지방의 소화율이 P-O.I 과 P-0.2 생균제 처리구에서 대조구에 비해 증가하였으며 (P<0.05), 조회분의 소화율은 P-0.2 처리구가 대조구에 비해서 유의하게 증가하였다. (P<0.05). 칼슐의 소화용은 P-0.2 처리구가 P-O.I 및 대조구에 비해 유의 하게 높았지만(P<0.05), 인의 소화율에서는 처리간 차이가 나타나지 않았다. 질소 축적율에서는 P-O.2와 P-O.1 생균제 처리구가 분내 질소 함량을 유의하게 낮추었으나( P<0.05), 뇨내 질소 함량 및 질소 축적율에는 영향을 미치지 않았다. 육성기(평균체중 41.80 $\pm$2.68kg)에서는 처리간 영양소 소화율 및 질소 축적율이 통계적으로 유의한 차이를 보이지 않았다. 그리고 혈중 요소태 질소농도 및 혈액내 액혈구, IgG, IgA의 함량도 전 기간에 걸쳐 처리간의 차이가 나타나지 않았다. 본 실험을 통하여 사료내 지속적인 생균제의 첨가는 돼지의 성장과 영양소 소화율을 향상 시킬 수 있을 것으로 사료된다.

유기황의 수준별 급여가 산란계의 생산성, 계란품질 및 세포성 면역능에 미치는 영향 (Effects of Dietary Organic Sulfur on Performance, Egg Quality and Cell-mediated Immune Response of Laying Hens)

  • 본 연구는 산란계 사료에 유기황의 수준별 첨가급여가 산란계의 생산성, 계란품질 및 세포성 면역능에 미치는 영향을 구명하고자 실시하였다. 31주령 로만브라운종에 분말 형태의 유기황을 기초사료에 0%, 0.1%, 0.2%, 0.4% 수준으로 첨가 급여하였으며, 처리구당 5반복 반복당 18수씩 전체 360수를 수용하여 24주간 사양실험을 실시하였다. 산란 수와 난중은 매일 조사하였고, 계란품질은 8주 간격으로 측정하였다. 난황지방산과 혈액은 산란계 42와 54주령에 분석하였으며, 계란의 황 함량, IL-2 및 CD4+와 CD8+은 사양실험 종료 후 분석하였다. 본 연구결과, 31~38주령에 산란율은 처리구 간에 차이가 없었지만, 39주령 이후 유기황의 급여수준에 따라 대조구와 차이를 보였으며, 47~54주령의 산란율은 0.4% 급여구에서 대조구에 비해 현저하게 증가하였다(P<0.05). 계란의 난백높이와 호우유닛은 유기황 급여구에서 대조구에 비하여 유의적으로 개선되었다(P<0.05). 계란의 난황 내 다중불포화지방산은 첨가수준에 따라 현저하게 증가하였다(P<0.05). 유기황 수준이 증대됨에 따라서 불포화 지방산은 증가하였고, 포화지방산은 감소되었으며, 이러한 현상은 54주령에서 더욱 현저하게 나타났다(P<0.05). 계란에서 황 함량은 유기황 첨가 수준에 증대함에 따라서 증가되었다(P<0.05). 또한 혈청 알부민은 42주령에서 유기황 급여구에서 유의적으로 증가하였고(P<0.05), AST는 감소하였다(P<0.05). 단백질은 42주령의 산란계에서 유기황 급여에 따라 현저히 증대되었고(P<0.05), HDL 콜레스테롤은 유기황 첨가수준이 높아짐에 따라 유의적으로 상승하였다(P<0.05). 비장과 혈중 IL-2 및 CD4+/CD8+도 유기황의 첨가수준에 따라 현저하게 증가하였다(P<0.05). 그러므로 산란계에 0.4% 유기황의 첨가급여로 생산성, 계란품질 및 세포성 면역능력은 개선되었다.

사육 밀도가 육계 생산성, 면역 수준 및 계육 품질에 미치는 영향 (Effects of Stocking Density on the Growth Performance, Immune Status and Breast Meat Quality of Broiler)

  • 본 연구는 육계의 사육 밀도가 사양 성적, 혈액 구성, 면역 및 계육 품질에 미치는 영향을 조사하기 위해 실시하였다. 육계(Ross 308)를 385수를 공시하여 4개의 사육 밀도(26, 22, 19, 16 birds/㎡)를 두어 완전 임의 배치하여 실험하였다. 사료와 물은 자유 급이하였다. 면역 글루불린(Ig) 및 corticosterone을 평가하였으며, 증체량, 혈액 성분 및 가슴육 품질을 분석하였다. 전체 사육 기간 동안 증체량, 사료 섭취량은 사육 밀도가 증가함에 따라 유의적으로 감소하였다(P<0.05). 그러나, 초기에는 사육 밀도가 증가할수록 사료 섭취량이 유의적으로 증가하였다(P<0.05). 백혈구, 적혈구, 혈소판 및 혈액 생화학 조성은 사육 밀도에 따라 유의적인 차이를 보이지 않았다. 스트레스 지표(H/L 비율 및 cortico- sterone), IgA, IgM은 사육 밀도간의 유의적인 차이를 보이지 않았으나, IgG는 사육 밀도가 증가할수록 유의적으로 증가하였다(P<0.05). 육계 가슴육 pH, 보수력, 가열감량은 사육 밀도 간의 유의적인 차이를 보이지 않았으나, 전단력은 사육 밀도 16 birds/㎡ 처리구에서 유의적으로 높은 값을 나타내었다(P<0.05). 결론적으로 고밀도에서 체중이 유의적으로 감소하였으며, 16 birds/㎡의 사육 밀도에서는 육계의 전단력과 육계의 IgG에 유의적인 차이를 나타내었다. 따라서 고밀도 사육은 지양해야 될 것으로 판단되며, 향후 다양한 연구가 필요할 것으로 사료된다.

저수온기(13-15℃) 치어기 참돔(Pagrus major) 사료 내 비타민 C 대체제로써의 감귤착즙박 및 발효감귤착즙박의 이용 가능성 (Dietary Supplementation of Citrus and Fermented Citrus By-product for Juvenile Red Seabream Pagrus major at Low Water Temperature)

  • This study compared the effects of dietary supplementation of citrus by-product (CBP) and CBP fermented with Bacillus subtilis (F-CBP) on growth performance, feed utilization, intestinal histology and innate immunity of red seabream Pagrus major with three commercial immune-boosting products. The six experimental diets were supplemented with L-ascorbyl-2-polyphosphate (LAPP; the control diet), CBP or F-CBP at a concentration of 100 mg vitamin C equivalent/kg diet or one of three commercial immune boosters. The Experimental diets were fed to triplicate groups of 17 fish (initial body weight, 116 g) for 8 weeks. The water temperature during the feeding trial was maintained at $13-15^{\circ}C$. Growth and feed utilization did not differ significantly among the six dietary treatments, nor did the phagocytic activity, superoxide dismutase or total immunoglobulin concentrations. However, myeloperoxidase activity was significantly higher in the CBP groups. For the intestinal histology, the intestine diameter, villi and enterocyte heights and number of goblet cells did not differ significantly among groups. Therefore, CBP or F-CBP can be used as a valuable eco-friendly byproduct in diets for fishes including red seabream to maintain their normal growth and health.

Characteristics of IEF Patterns and SDS-PAGE Results of Korean EPO Biosimilars

  • Erythropoietin (EPO) is mainly produced in kidney and stimulates erythropoiesis. The use of recombinant EPOs for doping is prohibited because of its performance enhancing effect. This study investigated whether biosimilar EPOs could be differentiated from endogenous one by iso-electro-focusing plus double blotting and SDS-PAGE for antidoping analysis. The established method was validated with positive control urine. The band patterns were reproducible and meet the criteria, which was made by world anti doping agency (WADA). Isoelectric focusing was conducted in pH range 2 to 6. Recormon (La Roche), Aropotin (Kunwha), Epokine (CJ Pharm Co.), Eporon (Dong-A), Espogen (LG Life Sciences), and Dynepo (Shire Pharmaceuticals) were detected in basic region. All biosimilars showed discriminative isoelectric profiles from endogenous EPO profiles, but they showed different band patterns with the reference one except Epokine (CJ Pharm Co.). Next, SDS-PAGE of biosimilar EPOs resulted in different molecular weight patterns which were distributed higher than endogenous EPO. Commercial immune assay kit as an immune affinity purification tool and immobilized antibody coated magnetic bead were tested for the purification and concentration of EPO from urinary matrix. The antibody-coated magnetic bead gave better purification yield. The IEF plus double blotting and SDS-PAGE with immunoaffinity purification method established can be used to discriminate biosimilar EPOs from endogenous EPO.

Preparation of Selenium-enriched Bifidobacterium Longum and its Effect on Tumor Growth and Immune Function of Tumor-Bearing Mice

  • In this study, we demonstrated selenium (Se) accumulation in Bifidobacterium longum strain (B. longum) and evaluated the effect of Se-enriched B. longum (Se-B. longum) on tumor growth and immune function in tumor-bearing mice. Analysis using high-performance liquid chromatography-inductively coupled plasma mass spectrometry (HPLC-ICP-MS) revealed that more than 99% of Se in Se-B. longum was organic, the main component of which was selenomethionine (SeMet). In the in vivo experiments, tumor-bearing mice (n=8) were orally administrated with different doses of Se-B. longum alone or combined with cyclophosphamide (CTX). The results showed that the middle and high dose of Se-B. longum significantly inhibited tumor growth. When Se-B. longum and CTX were combined, the antitumor effect was significantly enhanced and the survival time of tumor-bearing mice (n=12) was prolonged. Furthermore, compared with CTX alone, the combination of Se-B. longum and CTX stimulated the activity of natural killer (NK) cells and T lymphocytes, increasing the levels of interleukin-2 (IL-2) and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), and the leukocyte count of H22 tumor-bearing mice (n=12).

On-farm evaluation of dietary animal and plant proteins to replace fishmeal in sub-adult olive flounder Paralichthys olivaceus

  • Background: High demand and low supply of fishmeal due to overexploitation of fisheries resources have resulted in a dramatic increase in the price of this ingredient. Olive flounder (Paralichthys olivaceus) commercial feed contains approximately 60% fishmeal and limited success has been achieved in identifying sustainable alternative protein sources for this species. Methods: An on-farm feeding trial was conducted to compare a basal diet containing 65% as the control (CONT) with two experimental diets replacing 10% of fishmeal by animal protein (AP10) or 20% of fishmeal by animal and plant protein (APP20). Sub-adult olive flounder averaging 327 ± 9.3 g (mean±SD) were fed one of the three diets in triplicate groups for 16 weeks. Results: Weight gain, specific growth rate, feed efficiency, protein efficiency ratio, and survival were not significantly different among fish fed all the experimental diets (P > 0.05). Also, non-specific immune responses (superoxide dismutase and lysozyme activity), serum biochemical parameters, and intestinal villi length were not significantly different among fish fed all the experimental diets (P > 0.05). Conclusions: Therefore, based on growth performance, non-specific immune responses, serum biochemical parameters, and intestinal histology, dietary animal and plant protein mixtures could replace up to 20% of fishmeal in the diet of sub-adult olive flounder.

Isolation and Characterization of Proteoglycan Derived From Human Placenta and its Biological Activities

  • Chondroitin sulfates proteoglycans were isolated from human placenta. For the identification of enzymatic digestion products of isolated proteoglycan, strong anion exchange-high performance liquid chromatography (SAX-HPLC) was performed. By the action of chondroitin ABC and chondroitin B lyase, three unsaturated disaccharides 2-acetamide-2-deoxy-3-O-($\beta$-D-gluco-4-enepyranosyluronic acid)-D-galactose ($\delta$Di-OS), 2-acetamide-2-deoxy-3-O-($\beta$-D-gluco-4-enepyranosyluronic acid)-6-O-su lfo-D-galactose ($\delta$Di-6S) and 2-acetamide-2-deoxy-3-O-($\beta$-D-gl uco-4-enepyranosyluronic acid)-4-O-sulfo-D-galactose ($\delta$Di-4S) were produced from the human placenta proteoglycan. The anticoagulant activity of chondroitin sulfate proteoglycan was evaluated by activated partial thromboplastin time (aPTT) assay and thrombin time (TT) assay. The clotting times of aPTT and TT were increased from 72 to 144 sec and 19 to 27 sec, respectively. The Immune-modulating activity of chondroitin sulfate proteoglycan was examined by cell proliferation assay and these results suggest that it may play a role in suppression of the function of immune-related cells.

Differential expression patterns of gangliosides in the tissues and cells of NIH-mini pig kidneys

  • Gangliosides are a ubiquitous component of the membranes of mammalian cells that have been suggested to play important roles in various cell functions such as cell-cell interaction, adhesion, cell differentiation, growth control and signaling. However, the role that gangliosides play in the immune rejection response in xenotransplantation is not yet clearly understood. In this study, differential expression patterns of gangliosides in HEK293 (human embryonic kidney cells), PK15 (porcine kidney cells), NIH-kd (NIH-mini pig kidney cells, primary cultured) and the cortex, medulla and calyx of the NIH-mini pig kidney were investigated by high-performance thin-layer chromatography (HPTLC). The results revealed that HEK293, PK15 and NIH-kd contained GM3, GM2 and GD3 as major gangliosides. Moreover, GM3, which are the gangliosides of NIH-kd, were expressed at higher levels than HEK293 and PK15. Especially, GT1b were expressed in HEK293 and NIH-kd but not in PK15. Finally, GM1 and GD1a were expressed in NIH-kd, but not in HEK293 or PK15. These results suggest that differential expression patterns of gangliosides from HEK293, PK15 and NIH-kd are related to the immune rejection response in xenotransplantation.

Effects of enzymatically hydrolyzed fish by-products in diet of juvenile rainbow trout (Oncorhynchus mykiss)

  • Five experimental diets were formulated to evaluate the effects of dietary enzymatically hydrolyzed tuna by-product on growth, non-specific immune responses, and hematology of juvenile rainbow trout (Oncorhynchus mykiss). A basal diet with 50% of fishmeal was used as control (CON) and four other diets replaced 12.5% ($TBB_{12.5}$), 25% ($TBB_{25}$), 37.5% ($TBB_{37.5}$), and 50% ($TBB_{50}$) of fish meal in the CON diet. Juvenile rainbow trout ($4.87{\pm}0.05g$) were randomly distributed into 15 tanks (50 L) and fed 3-4% of wet body weight two times a day. At the end of 7 weeks of feeding trial, weight gain, specific growth rate, feed efficiency, and protein efficiency ratio of fish fed CON diet were significantly higher than those of fish fed $TB_{50}$ diet (P < 0.05). But there were no significant differences among fish fed CON, $TBB_{12.5}$, $TBB_{25}$, and $TBB_{37.5}$ diets (P > 0.05). There were no significant differences in GPT levels among fish fed CON, $TBB_{12.5}$, $TBB_{25}$, and $TBB_{37.5}$ diets. Also, there were no significant differences in lysozyme, superoxide dismutase, glucose, and total protein levels in all experimental diet (P > 0.05). The broken-line analysis indicated that the minimum dietary level of enzymatically hydrolyzed tuna by-product to replace fishmeal could be 29.7% in rainbow trout. These results indicated that the optimum level of dietary enzymatically hydrolyzed tuna by-product could replace greater than 29.7% but less than 37.5% of fishmeal in juvenile rainbow trout diet.
