Changes in growth performance, nutrient digestibility, immune blood profiles, fecal microbial and fecal gas emission of growing pigs in response to zinc aspartic acid chelate

  • Objective: This study was conducted to investigate the effect of zinc aspartic acid chelate (Zn-ASP) on growth performance, nutrient digestibility, blood profiles, fecal microbial and fecal gas emission in growing pigs. Methods: A total of 160 crossbred ([Landrace×Yorkshire]×Duroc) growing pigs with an initial body weight (BW) of 25.56±2.22 kg were used in a 6-wk trial. Pigs were randomly allocated into 1 of 4 treatments according to their sex and BW (8 replicates with 2 gilts and 3 barrows per replication pen). Treatments were as follows: i) CON, basal diet, ii) TRT1, CON+0.1% Zn-ASP, iii) TRT2, CON+0.2% Zn-ASP, and iv) TRT3, CON+0.3% Zn-ASP. Pens were assigned in a randomized complete block design to compensate for known position effects in the experimental facility. Results: In the current study, BW, average daily gain, and gain:feed ratio showed significant improvement as dietary Zn-ASP increased (p<0.05) in growing pigs. Apparent total tract digestibility (ATTD) of dry matter was increased linearly (p<0.05) in pigs fed with Zn-ASP diets. A linear effect (p<0.05) was detected for the Zn concentration in blood with the increasing levels of Zn-ASP supplementation. Lactic acid bacteria and coliform bacteria were affected linearly (p<0.05) in pigs fed with Zn-ASP diets. However, no significant differences were observed in the ATTD of nitrogen, energy and Zn. And dietary Zn-ASP supplementation did not affect fecal ammonia, hydrogen sulfide and total mercaptans emissions in growing pigs. Conclusion: In conclusion, dietary supplementation with Zn-ASP of diet exerted beneficial effects on the growth performance, nutrient digestibility, blood profiles and fecal microbes in growing pigs.

모돈 사료 내 Portulaca oleracea L.의 첨가가 번식성적, 혈액성상, 면역반응 및 분 중 미생물에 미치는 영향 (Effects of Portulaca oleracea L. Supplementation on Reproductive Performance, Blood Profiles, Immune Response and Fecal Microflora in Multiparous Sows)

  • 본 실험은 모돈 사료 내 Portulaca oleracea L.(POL)의 첨가가 번식성적, 혈액성상, 면역반응 및 분 중 미생물에 미치는 영향을 구명하기 위해 수행하였다. 본 실험을 위해 총 20두의 모돈(Landrace×Yorkshire; 229.2±1.15 kg)을 공시하였으며, 개시체중에 기반하여, 2처리 10반복, 반복당 1두씩 완전임의 배치하였다. 실험 처리구는 옥수수-대두박 기초사료를 대조구로 하여 기초사료 내 POL을 0.05% 첨가한 처리구가 포함되었으며, 분만 직후부터 포유기간 동안 수행하였다. 사료 내 POL을 첨가하였을 때, 모돈의 체중, 등지방두께 및 번식성적에서는 유의적인 차이가 나타나지 않았다(p>0.05). 혈액성상에서 사료 내 POL의 첨가하였을 때, 모돈의 림프구가 유의적으로 감소하는 것으로 나타났다(p<0.05). 또한, 면역반응에서 사료 내 POL을 첨가하였을 때 포유모돈의 혈 중 IL-1β의 수치는 유의적으로 감소하였으며(p<0.05), IgG는 증가하는 경향이 관찰되었다(p=0.051). 분 중 미생물에서는 POL 처리구에서 포유모돈의 coliforms이 감소하는 경향을 확인하였다(p=0.063). 본 실험의 결과들을 종합해보면, 사료 내 POL의 첨가는 번식성적에 부정적인 영향을 미치지 않으며, 모돈의 혈액성상, 면역반응 및 분 중 미생물에 긍정적인 효과를 미치는 것을 시사한다.

  • 마늘의 주요 성분인 allicin투여 후 유도되는 사람 말초혈액의 단핵구의 유전자 발현에 미치는allicin의 효과를 규명하였다. DNA microarray를 이용하여, allicin이 chemokines, cytokine, 면역관련 유전자 및 신호전달 관련 유전자의 발현을 유도하는 것을 확인하였다. 반대로 allicin은 Th1 type의 획득면역 관련 유전자의 발현을 억제하였다. 염증세포에 있어서 allicin은 억제효과 및 자극 효과를 동시에 보여주었다. 이는 allicin이 휴지기 세포에서 먼저 증가시킨 특정 유전자의 발현을 이후에 감소시키는 결과를 보여주는 것으로 positive와 negative 효과를 발휘하는 새로운 기전을 제시하는 것이다. Allicin에 대한 광범위하고 새로운 관심을 고려해 볼 때 본 연구에서 보여주는 많은 유전자의 발현 양상은 좀 더 특정적이고 효과적인 치료법을 고안하는 데 유용할 것이다.

IMMUNIES, a unique polyherbal extract, exhibits antiproliferative activity and improves tumor-bearing canine patients: a pilot study

  • Dog owners seek treatment when their pets develop cancer. IMMUNIES is traditional herbal medicine-based figment made of 10 natural herbs, designed to maintain host immune function. The major component of IMMUNIES is Dendropanax morbiferus. This clinical pilot study monitored the toxicity and efficacy of IMMUNIES. Four senile dogs with spontaneously occurring mammary and liver cancers were enrolled in this study and treated orally daily for 3 months, and their blood/urine biochemical profiles were examined each month. IMMUNIES was well tolerated during the treatment period. Blood urea nitrogen, creatinine, alanine aminotransferase, alkaline phosphatase, and C-reactive protein levels decreased in all four dogs, whereas red blood cells and hematocrit were within the normal range. IMMUNIES also changed the expression of several molecular targets in the anticancer pathway, such as pro-NAG-1, p53, and cyclin D1. Although the tumors did not completely respond to IMMUNIES, the biochemical profiles and clinical examination showed a stabilized cancer status for 3 months. Thus, IMMUNIES was found to be safe and well-tolerated in the dosage range tested and exhibited cancer antiproliferative activity in canine cancer. Future studies should address other potential benefits of IMMUNIES, including correlative assessments of immune function, quality of life, and owner satisfaction.

Effects of Pre-conditioning dose on the Immune Kinetics and Cytokine Production in the Leukocytes Infiltrating GVHD Tissues after MHC-matched Transplantation

  • Background: Graft-versus-host disease (GVHD) is a huddle for success of hematopoietic stem cell transplantation. In this study, effects of irradiation dose on immune kinetics of GVHD were investigated using B6 ${\rightarrow}$ BALB.B system, a mouse model for GVHD after MHC-matched allogeneic transplantation. Methods: BALB.B mice were transplanted with bone marrow and spleen cells from C57BL/6 mice after irradiation with different doses. Leukocytes residing in the peripheral blood and target organs were collected periodically from the GVHD hosts for analysis of chimerism formation and immune kinetics along the GVHD development via flow cytometry. Myeloid cells were tested for production of IL-17 via flow cytometry. Results: Pre-conditioning of BALB.B hosts with 900 cGy and 400 cGy resulted in different chimerism of leukocytes from the blood and affected survival of GVHD hosts. Profiles of leukocytes infiltrating GVHD target organs, rather than profiles of peripheral blood leukocytes (PBLs), were significantly influenced by irradiation dose. Proportions of IL-17 producing cells in the infiltrating $Gr-1^+$ or $Mac-1^+$ cells were higher in the GVHD hosts with high does irradiation than those with low dose irradiation. Conclusion: Pre-conditioning dose affected tissue infiltration of leukocytes and cytokine production by myeloid cells in the target organs.

Whole-Blood Gene-Expression Profiles of Cows Infected with Mycobacterium avium subsp. paratuberculosis Reveal Changes in Immune Response and Lipid Metabolism

  • Mycobacterium avium subsp. paratuberculosis (MAP) is the causative agent of Johne's disease, a chronic debilitating disease affecting ruminants worldwide. In the present study, we aimed to determine the major gene networks and pathways underlying the immune response to MAP infection using whole-blood cells, as well as provide the potential transcriptional markers for identifying the status of MAP infection. We analyzed the transcriptional profiles of whole-blood cells of cattle identified and grouped according to the presence of MAP-specific antibodies and the MAP shed by them. The grouping was based on the results obtained by ELISA and PCR analyses as follows: i) Test1 group: MAP-negative results obtained by ELISA and positive results obtained by PCR; ii) Test2 group: MAP-positive results obtained by ELISA and negative results obtained by PCR; iii) Test3 group: MAP-positive results obtained by ELISA and positive results obtained by PCR; iv) uninfected control: MAP-negative results obtained both by ELISA and PCR analysis. The results showed down-regulated production and metabolism of reactive oxygen species in the Test1 group, activation of pathways related to the host-defense response against MAP (LXR/RXR activation and complement system) in the Test2 and Test3 groups, and anti-inflammatory response (activation of IL-10 signaling pathway) only in the Test3 group. Our data indicate a balanced response that serves the immune-limiting mechanism while the host-defense responses are progressing.

Temporal expression profiling of long noncoding RNA and mRNA in the peripheral blood during porcine development

  • Objective: We investigated the temporal expression profiles of long noncoding RNA (lncRNA) and mRNA in the peripheral blood of pigs during development and identified the lncRNAs that are related to the blood-based immune system. Methods: Peripheral blood samples were obtained from the pigs at 0, 7, 28, and 180 days and 2 years of age. RNA sequencing was performed to survey the lncRNA and mRNA transcriptomes in the samples. Short time-series expression miner (STEM) was used to show temporal expression patterns in the mRNAs and lncRNAs. Gene ontology and Kyoto encyclopedia of genes and genomes analyses were performed to assess the genes' biological relevance. To predict the functions of the identified lncRNAs, we extracted mRNAs that were nearby loci and highly correlated with the lncRNAs. Results: In total of 5,946 lncRNA and 12,354 mRNA transcripts were identified among the samples. STEM showed that most lncRNAs and mRNAs had similar temporal expression patterns during development, indicating the expressional correlation and functional relatedness between them. The five stages were divided into two classes: the suckling period and the late developmental stage. Most genes were expressed at low level during the suckling period, but at higher level during the late stages. Expression of several T-cell-related genes increased continuously during the suckling period, indicating that these genes are crucial for establishing the adaptive immune system in piglets at this stage. Notably, lncRNA TCONS-00086451 may promote blood-based immune system development by upregulating nuclear factor of activated T-cells cytoplasmic 2 expression. Conclusion: This study provides a catalog of porcine peripheral blood-related lncRNAs and mRNAs and reveals the characteristics and temporal expression profiles of these lncRNAs and mRNAs during peripheral blood development from the newborn to adult stages in pigs.

고콜레스테롤혈증 여자 노인에서 항산화 물질 복용에 따른 혈중 지질 농도와 면역능의 변화 (The Change of Lipid Metabolism and Immune Function Caused by Antioxidant Material in the Hypercholesterolemic Elderly Women in Korea)

  • This study was performed to examine the change of lipid metabolism and immune function caused by antioxidant material in hypercholesterolemic elderly women (serum total cholesterol $\geq$200 mg/dI). The subjects were 51 elderly women aged over 60 yrs. They were divided into antioxidant nutrients complex group (n = 25) and spirulina group (n= 26). Antioxidant nutrients complex (1 capsule/day) and spirulina (7.5 mg/day) were used for intervention for 8weeks. All the subjects were fully informed the purpose of study and gave written consents to participate in this study. Dietary intakes, anthropometric indices and blood assessment for lipid, immune function and antioxidant status were measured before and after supplementation. Either antioxidant nutrients complex or spirulina supplementation for 8weeks resulted in improved antioxidant status evidence by increased TAS (total antioxidant status) and decreased TB-ARS (thiobarbituric acid reactive substance) . This intervention led to decreased serum levels of total cholesterol, LDL-cholesterol, oxLDL, apolipoprotein B, IL-6 and IL-6 production by peripheral blood lymphocyte. In conclusion, the lipid profiles, immune function and antioxidant capacity were improved after either antioxidant nutrient complex or spirulina supplementation for hypercholesterolemic women. Therefore, improving antioxidant status using supplemen-tation could provide means of controlling cardiovascular disease in Korean elderly people.

Expression Profiles of Immune-related Genes in Fluoxetine-treated Human Mononuclear Cells by cDNA Microarray

  • To investigate the effect of fluoxetine, one of selective serotonin reuptake inhibitors (SSRIs), on the immune system, human peripheral blood mononuclear cells (PBMC) were treated with fluoxetine $(10^{-7}\;M)$ for 24 h, and immune-related genes were analyzed by cDNA microarray. Expression of the immunerelated genes such as CD107b (LAMP-2), CD47 receptor (thrombospondin receptor), CD5 antigen-like (scavenger receptor cysteine rich family), copine III (CPNE3), interleukin (IL)-18 (interferon-gammainducing factor), integrin alpha 4 (CD49d), integrin alpha L subunit (CD11a), IL-3 receptor alpha subunit, L apoferritin, and small inducible cytokine subfamily A (Cys-Cys) member 13 (SCYA13) was induced by fluoxetine. This result suggests that fluoxetine may affect the immune system, and provides fundamental data for the involvement of SSRIs on immunoregulation.

이유자돈 사료 내 동애등에(Hermetia illucens)의 첨가가 분변지수, 혈액성상, 면역반응 및 소장형태에 미치는 영향 (Effects of Hermetia Illucens Supplementation on Fecal Score, Blood Profiles, Immune Response and Small Intestinal Morphology in Weaned Pigs)

  • 본 실험은 사료 내 동애등에(HI, Hermetia illucens)의 첨가가 이유자돈의 분변지수, 혈액성상, 면역반응 및 소장형태에 미치는 영향을 구명하기 위해 수행하였다. 본 실험을 위해 총 24두의 이유자돈(Landrace×Yorkshire×Duroc; 5.86±0.13 kg)을 공시하였으며, 개시체중에 기반하여, 4처리 6반복, 반복당 1두씩 완전임의 배치하였다. 실험기간은 0~14일간 진행하였다. 실험 처리구는 옥수수-대두박 기초사료를 대조구로 하여 기초사료 내 HI를 1, 2 및 3% 첨가한 처리구가 포함되었다. 혈액성상에서 사료 내 HI의 첨가수준이 증가함에 따라 혈중 단핵구(p<0.01)와 호산구(p<0.05)가 유의적으로 증가하는 것으로 나타났으며, 적혈구는 선형적으로 감소하는 경향이 관찰되었다(p=0.08). 한편, 면역반응에서 혈 중 TNF-α의 수치는 사료 내 HI의 첨가수준이 증가함에 따라 선형적으로 감소하는 경향이 관찰되었으며(p=0.067), 분변지수 또한 HI의 첨가수준이 증가함에 따라 선형적으로 감소하는 경향이 관찰되었다(p=0.058). 소장형태에서 십이지장의 융모 높이는 HI의 첨가수준이 증가할수록 개선되는 유의적인 효과가 나타났다(p<0.05). 본 실험의 결과들을 종합해 보면, 사료 내 HI 첨가는 이유자돈의 설사저감, 면역반응 및 소장형태에 긍정적인 효과를 미치는 것을 시사한다.
