Objective : The purpose of this study was to investigate the protective effects of Cheongsimondam-tang(COT) on the animal model of depression induced immobilization stress. Method 1) Male rats were used for this experiment. The subject were divided into 4 groups(1. normal 2. saline solution administered during immobilization stress treatment 3. COT of 100mg/kg administered 4. COT of 400mg/kg administered). 2) Immobilization stress was treated for 1 hours on day. During 2 days of immobilization stress treatment, they were executed forced swimming test, passive avoidance test, elevated plus maze test. Corticosterone in blood were measured Results 1) In EPM test, stress group showed significantly increased anxiety, COT groups showed significantly decreased anxiety. 2) In forced swimming test, COT groups did not show significantly decreased immobilization. 3) In passive avoidance test, stress group showed significantly decreased learning execution, COT groups showed significantly increased learning execution. 4) Stress group showed significantly increase in serum level of corticosterone, COT of 400mg/kg group showed significantly decreased serum level of corticosterone. Conclusion : These results suggest that Cheongsimondam-tang(COT) is effective in the treatment of depression.

To investigate the effect of Qi promoting-agents, clearing heat of deficiency type-agent, clearing heat of excessive type-agent, warming interior part of body-agent and Shihoieokgan-Tang against the immobilization stress, Cypri Rhizama, Auranti pericarpium, Aucklandiae Radix and Bupleuri Radix, Soyo-San, Hwangyeonhaedok-Tang, Geongangbuja-Tang and Shihoieokgan-Tang was administered intragastically 1hr prior to immobilizaiton stress and the change of corticosterone level has been measured by the flurometric method. The results obtained are as follows: 1. Qi promoting-agents(Cypri Rhizama, Auranti pericarpium, Aucklandiae Radix and Bupleuri Radix) showed mildly reducing effect or did not shown the reducing effect against the elevated coritocostereone level induced by the immobilization stress in the mice. 2. Hwangyeonhaedok-Tang, Gungangbuza-Tang, and Soyo-San did not show the reducing effect against the elevated corticostereone level induced by the immobilization stress in the mice. 3. Shihoieokgan-San was effective against the elevated corticosterone level induced by the immobililzation stress at 1g/kg, 3g/kg respectively. Consequently these data proved that Qi pomoting-agents as single herb(Cypri Rhizama, Auranti pericarpium, Aucklandiae Radix and Bupleuri Radix), clearing heat of deficiency type-agent(Soyo-San), warming interior part of the body(Gungangbuza-Tang), clearing heat of excessive type-agent(Hwangyeonhaedok-Tang) have no effect to decrease the elevated corticosterone level in mice exposed to the immobilization stress. Therefore our study revealed that Shihoieokgan-San which has the effects of herbs and formulae mentioned above decreased the elevated corticosterone level in mice exposed to the immobilization stress significantly.
This exoeriments were conducted to study anti-stress effects Samulanshintang on the weight change, ${\beta}-lipoprotein$, total cholesterol and triglyceride consentration of rats stressed by immobilization. The following results have been obtained : 1. Samulanshintang reduced significantly the body weight loss of rats stressed by immobilization. 2. Samulanshintang decreased significantly the serum cortisol level of rats stressed by immobilization. 3. Samulanshintang decreased significantly the serum ${\beta}-lipoprotein$ level of rats stressed by immobilization. 4. Samulanshintang decreased significantly the serum total cholesterol level of rats stressed by immobilization. 5. Samulanshintang decreased significantly the serum triglyceride level of rats stressed by immobilization. According to the above results, it is concluded that Samulanshintang has significant effect in reducing stress.
The effect of xylazine administration on plasma cortisol, prolactin, glucose and packed cell volume (PCV) responses to immobilization and heat stress was investigated. Immobilization of rats for 2 hours by ligation of the fore and hind legs strongly caused approximately two-fold increase in plasma cortisol and prolactin levels. Plasma glucose and PCV were not significantly changed. Pretreatment of immobilized rats with xylazine (20 mg/kg body weight i.m.) resulted in approximately 20% reduction in both plasma cortisol and prolactin concentrations. A marked hyperglycemia and increase in the PCV value was observed. On the other hand, rats exposed to acute heat stress ($40^{\circ}C$, and 60% relative humidity) for 2 hours, also developed two fold increase in both plasma cortisol and prolactin concentrations and the pretreatment with xylazine caused a 20% reduction in the levels of both hormones. Plasma glucose level was not significantly changed in heat stressed rats but it was markedly increased after pretreatment with xylazine. PCV was significantly incrcased under heat stress and pretreatment with xylazine induced a pronounced elevation in this value. It was suggested that stimulation of cortisol and prolactin secretion in response to immobilization or heat stress can be partially reduced by an alpha 2-adrenergic agonist.
Objectives : This study investigated the effects of electroacupuncture on memory, depression, and plasma stress hormone levels in rats that were under immobilization stress. Materials and Methods : The immobilization-only group was given two hours of immobilization stress for 10 consecutive days. The immobilization and high frequency EA group was given two hours of immobilization stress simultaneously with high frequency (100Hz) electroacupuncture stimulation on the right ST-36 (Zusanli) for 10 consecutive days. We conducted the inhibitory avoidance and forced swimming tests to recognize whether immobilization stress and electroacupuncture have effects on memory and depression. We collected blood samples from the tail of each rat at 30, 60, 90, and 120-minute intervals during the immobilization stress and EA stimulation to measure plasma concentrations of ACTH, corticosterone, melatonin, and norepinephrine induced by immobilization stress and electroacupuncture. Results : There was a significant effect of high frequency on the increase in anamnesis based on the result of the inhibitory avoidance test, but there was no significant effect of decreasing depression based on the result of the forced swim test. Also, there was no significant effect on the response indicated by stress hormones. Conclusions : 1. High frequency electroacupuncture (100Hz) improved anamnesis in immobilization stress states under the inhibitory avoidance task. 2. High frequency electroacupuncture (100Hz) did not reduce depression induced by immobilization stress under the forced swimming test. 3. High frequency electroacupuncture (100Hz) did not decrease stress hormones through blood sampling.
S.D.rats were fed with 3.4% tyrosine supplemented diet for 5 days. Tyrosine diet had no effects on brain NE and MHPG-SO4 levels in non-stressed rats. When these animals were given 3 hr-immobilization stress, they responded in a manner that coped better to the stress. This was measured by the increase in brain MHPG-SO4 indicating the increase in norepinephrine turnover by the stressed animals. When rats were stressed, fed basal or high-tyrosine diet, brain tyrosine concentration dropped more than 26% over the non-stress control animals. 3-hr immobilization stress also decreased brain NE levels. However, while the stress resulted in a significant decrease(p<0.05) of brain NE in basal diet, the decrease was not significant in high-TYR diet group. And as the stress index, serum corticosterone, glucose, and free fatty acid concentratons also were assayed. In this study, it was found that high-TYR diet prevented the stress-induced depletion of brain NE and suppressed the rise in serum corticosterone, glucose, and free fatty acid. These results suggest that high-TYR diet increases the coping ability of body to stress.
Objective: Stress is known to be an inhibitor of the reproductive hypothalamic-pituitary-gonadal (HPG) axis. However, the neural and molecular connections between stress and reproduction are not yet understood. It is well established that in both humans and rodents, kisspeptin (encoded by the kiss1 gene) is a strong stimulator of the HPG axis. In the present study we hypothesized that endocannabinoids, an important neuromodulatory system in the brain, can act on the HPG axis at the level of kiss1 expression to inhibit reproductive function under stress. Methods: Adult male Wistar rats were unilaterally implanted with an intracerebroventricular cannula. Afterwards, the animals were exposed to immobilization stress, with or without the presence of the cannabinoid CB1 receptor antagonist AM251 (1 ${\mu}g/rat$). Blood samples were collected through a retro-orbital plexus puncture before and after stress. Five hours after the stress, brain tissue was collected for reverse transcriptase-quantitative polymerase chain reaction measurements of kiss1 mRNA. Results: Immobilization stress (1 hour) resulted in a decrease in the serum luteinizing hormone concentration. Additionally, kiss1 gene expression was decreased in key hypothalamic nuclei that regulate gonadotrophin secretion, the medial preoptic area (mPOA), and to some extent the arcuate nucleus (ARC). A single central administration of AM251 was effective in blocking these inhibitory responses. Conclusion: These findings suggest that endocannabinoids mediate, at least in part, immobilization stress-induced inhibition of the reproductive system. Our data suggest that the connection between immobilization stress and the HPG axis is kiss1 expression in the mPOA rather than the ARC.

Each part was homogenated and its catecholamine and serotonin contents were measured with HPLC. In our study, stress mainly induced increase of concentration of neurotransmitters in brain, but had mild effect on other physical function of rats. GH inhibited stress induced changes of neurotransmitter content in brain.
