Surface Analysis and Electrical Properties for Complex with Concentration of Metal ion in LB Ultra-thin Films Using IMI-O Polymer (IMI-O 고분자 LB막의 금속 이온의 착체 농도에 따른 전기특성 및 표면분석)

  • We fabricated an IMI-O polymer containing an imidazole group that could form a complex structure between the monolayer and the metal ions at the air-water interface. Also, the surface analysis and the electrical properties of metal ion complex of Langmuir-Blodgett (LB) films were investigated by using $\pi$-A isotherms. Atomoic force microscopy (AFM), current-voltage (I-V) measurements. In the $\pi$-A isotherms the molecular area was expanded with $Fe^{3+}$ concentration increase. It is considered that the expansion of molecular area is due to electrostatic repulsion between the polymer chains and hydrophobic increase of ionic strength. In the I-V characteristics, it is found that the limiting area has effects on the change of conductivity. And, the dielectric relaxation time decreased for increase of the $Fe^{3+}$ concentration.

Anisotropic Conductive Film (ACF) Prepared from Epoxy/Rubber Resins and Its Fabrication and Reliability for LCD

  • A thermoset type anisotropic conductive adhesive film (ACAF) comprising epoxy resin and natural butyl rubber (NBR) as the binder, micro-encapsulated imidazole as the curing agent, and Ni/Au coated polymer bead as a conductive particle has been studied. These films have been prepared to respond to requirements such as improved contact resistance, current status less of than 60 ${\mu}m$ and reliability. These films can also be used for connection between the ITO glass for LCD panel and the flexible circuit board. The curing conditions for the connection were 40, 20 and 15 seconds at 150, 170 and 190 $^{\circ}C$, respectively. The initial contact resistance and adhesion strength were 0.5 ${\Omega}/square$ and 0.4 kg/cm under the condition of 30 kgf/$^{cm}^2}$, respectively. After completing one thousand thermal shock cycling tests between -15 $^{\circ}C$ and 100 $^{\circ}C$, the contact resistance was maintained below 0.7 ${\Omega}/square$. Durability against high temperature (80$^{\circ}C$) and high humidity (85 % RH) was also tested to confirm long-term stability (1000 hrs) of the conduction.

Study on Preparation and Drug Release of Sulconazole Nitrate Gels (질산술코나졸겔의 제조 및 약물방출에 관한 연구)

  • Sulconazole nitrate(SCN), an imidazole derivative which has been effective in the treatment of dermatophytosis, tinea versicolor and candidiasis, was formulated as a gel containing drug, poloxamer 407, ethanol and propylene glycol. The resulting SCN gels were evaluated with respect to their viscosity, drug release rate, skin permeation rate. The apparent viscosity of SCN gel increased in proportion to poloxamer 407, drug and propylene glycol concentration. In case ethanol was added, the apparent viscosity decreased. The drug release rate of SCN gel increased in proportion to temperature and ethanol concentration. But the drug release rate decreased as the concentration of poloxamer 407 increased. The increase of drug concentration induced nonlinear increase of drug release rate. When propylene glycol was added at the level of 10%, the drug release rate increased but from 15% it decreased. The skin permeation rate decreased in high concentration of poloxamer 407. The skin permeation rate of SCN gel containing 15% ethanol increased about twice than that of gel without ethanol. The increase of drug concentration induced nonlinear increase of skin permeation rate. When propylene glycol was added at the level of 10%, the skin permeation rate increased but from 15% it decreased.

NMR Studies of Zinc-binding Luteinizing Hormone Releasing Hormone

  • Luteinizing Hormone Releasing Hormone(LHRH) is a decapeptide neurotransmitter known to be regulated by metal ions in the hyperthalamus. Zn-binding LHRH complex was systhesized, and zinc-LHRH complex was studied to understand what kinds of structural modifications would be critical in the LHRH releasing mechanism. Both nonexchangeable and exchangeable $^1H-NMR$ signal assignments were accomplished by pH-dependent and COSY NMR experiments. In addition, $^1H-NMR$ chemical shift changes of a-proton and peptide NH NMR signals at different pH condition, and $^1H-NMR$ signal differences between metal free and metallo-LHRH complex was monitored. NMR signals exhibit that primary metal-binding sites are nitrogens donor of imidazole ring and Arg, and peptide oxygen of Pro-His in the sequence. Structure obtained in this study has a cyclic conformation which is similar to that of energy minimized, and exhibits a specific a-helical turn with residue numbers $(2{\sim}7)$ out of 10 amino acids.

CROX (Cluster Regulation of RUNX) as a Potential Novel Therapeutic Approach

  • Comprehensive inhibition of RUNX1, RUNX2, and RUNX3 led to marked cell suppression compared with inhibition of RUNX1 alone, clarifying that the RUNX family members are important for proliferation and maintenance of diverse cancers, and "cluster regulation of RUNX (CROX)" is a very effective strategy to suppress cancer cells. Recent studies reported by us and other groups suggested that wild-type RUNX1 is needed for survival and proliferation of certain types of leukemia, lung cancer, gastric cancer, etc. and for their one of metastatic target sites such as born marrow endothelial niche, suggesting that RUNX1 often functions oncogenic manners in cancer cells. In this review, we describe the significance and paradoxical requirement of RUNX1 tumor suppressor in leukemia and even solid cancers based on recent our findings such as "genetic compensation of RUNX family transcription factors (the compensation mechanism for the total level of RUNX family protein expression)", "RUNX1 inhibition-induced inhibitory effects on leukemia cells and on solid cancers through p53 activation", and "autonomous feedback loop of RUNX1-p53-CBFB in acute myeloid leukemia cells". Taken together, these findings identify a crucial role for the RUNX cluster in the maintenance and progression of cancers and suggest that modulation of the RUNX cluster using the pyrrole-imidazole polyamide gene-switch technology is a potential novel therapeutic approach to control cancers.

Metalloporphyrin Catalyzed Olefin Epoxidation and Molecular Orbital Study (Metalloporphyrin의 Olefin Epoxidation과 분자궤도함수론적 고찰)

  • Product yields were determined for the reaction of styrene with sodium hypochlorite (NaOCl) in CH$_2$Cl$_2$ with various substituted manganese porphyrin complexes as catalysts. In the presence of the electron withdrawing group and ortho-substituted manganese porphyrin complexes, reaction rate and epoxide selectivity are increased. Also reaction rate and epoxide selectivity are largely increased by the presence of imidazole which behaves as axial ligand of the manganese porphyrin complexes. By the kinetic study with Michealis-Menten equation, the factor significantly affected to catalytic ability is $K_m$ value. A large binding affinity consists with the low $K_m$. With theoretical analysis by EHMO calculation, the results are in good agreement with experimental data.

The Crystal and Molecular Structure of 1-(3-Chloro-2-hydroxypropyl)-2-methyl-5-nitroimidazole (Ornidazole), $C_7H_{10}CIN_3O_3$

  • Ornidazole, C7H10ClN3O3, crystallizes in the triclinic, space group P1^, with a=13.605(2), b=14.054(1), c=8.913(5) Å, α=71.59(2), β=78.73(2), γ=64.86(1)°, μ=3.26 cm-1, Dc=1.499 g/cm3, Dm=1.497g/cm3, F(000)=684, and z=6. Intensities for 2693 unique reflections were measured on a CAD4 diffractometer with graphite-monochromated Mo-Kα radiation. The structure was solved by direct method and refined by block-diagonal least squares to a final R of 0.081 (Rw=0.047) for 1952 reflections with Fo>3σ (Fo). The asymmetric unit contains three independent molecules of the title compound. The bond lengths and bond angles are comparable with the values found in the other nitro-substituted compounds. The nitro groups are rotated (6.9°, 6.6°, 2.6° for the three independent molecule, respectively) about the C-N axes from the imidazole planes. The crystal structures are linked by two intermolecular hydrogen bonds of O-H---N type and one intermolecular hydrogen bond of O-H---O type.

Optically Active and Organosoluble Poly(amide-imide)s Derived from N,N'-(Pyromellitoyl)bis-L-histidine and Various Diamines: Synthesis and Characterization

  • An optically active diacid containing the L-histidine moiety was prepared by reacting pyromellitic dianhydride (1,2,4,5-benzenetetracarboxylic acid 1,2,4,5-dianhydride) 1 with L-histidine 2 in acetic acid, and was polymerized with several aromatic diamines 5a-g to obtain a new series of optically active poly(amide-imide)s (PAIs) using two different methods, such as direct polycondensation in a medium consisting of N-methyl-2-pyrrolidone (NMP)/triphenyl phosphite (TPP)/calcium chloride ($CaCl_2$)/pyridine (Py) and direct polycondensation in a tosyl chloride (TsCl)/pyridine (Py)/N,N-dimethylformamide (DMF) system as a condensation agent. The resulting new polymers 6a-g with inherent viscosity was obtained in good yield. The polymers were readily soluble in polar organic solvents, such as N,N-dimethyacetamide (DMAc), N,N-dimethyformamide (DMF), and dimethyl sulfoxide (DMSO). The obtained polymers were characterized by FTIR, specific rotation, elemental analysis as well as $^1$H-NMR spectroscopy and gel permeation chromatography (GPC). The thermal stability of the resulting PAIs was evaluated with thermogravimetric analysis techniques under a nitrogen atmosphere.

Characterization of Radiation Fabricated Imidazole Based Hydrogel (이미다졸 기반 수화겔의 방사선 합성 및 특성 평가)

  • Metronidazole (MD), which is used as an antibiotic, is largely used as an oral and skin application agent, and has inhibiting effect on the production of the fungus causing malodor. However, the maximum drug inclusion concentration is 0.75% of skin ointment in commercially available. In this study, hydrogels containing high concentration of metronidazole were prepared by using radiation crosslinking technique based on biocompatible polymers, and release characteristics and antimicrobial properties were evaluated. This study was preliminary carried out to investigate whether it could be used effectively as antibacterial dressing materials.

  • This study was undertaken to investigate the direct effect of three different kinds of steroid to Trichophyton(T.) mentagrophytes growth in vitro. A loopful of T. mentagrophytes was inoculated into Sabouraud's media and Sabouraud's media containing various concentrations of dexamethasone (0.04%-0.00004%) prednisolone (0.25%-0.00025%), triamcinolone acetonide (0.2%-0.0002%), Sabouraud's media containing imidazole nitrate (econasole nitrate) 10 micro gm. per cc. of media, and Sabouraud's media containing econazole nitrate $10{\mu}g$ per cc. of media of various dexamethasone concentrations (0.04%-0.00004%). After inoculation they were left for two weeks at room temparature and the colony diameter were compared between Sabouraud's media and Sabouraud's media containing various concentrations, of three different steroids, Sabouraud's media containing econazole nitrate and Sabouraud's media containing econazole nitrate of various dexamethasone concentrations. For complementary study, hanging slide culture were done between Sabouraud's media and Sabouraud's media containing various dexamethasone concentrations to compare the degree of branching of hyphae and sporulation serially. The study results were as follows: 1. There is no significant difference in growth rate of T. mentcgrophytes between Sabouraud's media and Sabouraud's media containing various concentrations of three different steroids. 2. There is no significant difference in growth rate of T. metagrophytes between Sabouraud's media containing econazole nitrate 10 micro gm per cc. of media and Sabouraud's media containing econazole nitrate 10 micro gm per cc. of media of various dexamethasone concentrations. 3. In hanging slide culture, there is no significant difference in branching of hyphae and sporulation between Sabouraud's media and Sabouraud's media containing various dexamethasone concentrations.
