Effects of Astragalus Polysaccharides, Achyranthes bidentata Polysaccharides, and Acantbepanax senticosus Saponin on the Performance and Immunity in Weaned Pigs

  • Two trials were conducted to study the effects of two Chinese herbal polysaccharides, Astragalus polysaccharides (APS) and Achyranthes bidentata polysaccharides (ABPS), and one Chinese herbal saponin, Acantbepanax senticosus saponin (ASS), on the immunity and growth performance of weaned pigs. Experiment 1 was a 14-day growth assay, in which 32 weaned pigs were randomly allocated to one of four dietary treatments: i) 0.05% talcum powder control; ii) 0.05% APS; iii) 0.05% mixture of APS and ASS in a 1:1 ratio by weight; and iv) 0.05% mixture of APS, ASS, and ABPS in a ratio of 1:1:1 by weight. Blood samples were collected on day 14 to determine plasma parameters. Feed intake, body weight gain, and feed efficiency were also determined. Experiment 2 was a 21-day immunity assay, in which 16 weaned pigs were randomly allotted to one of two dietary treatments: i) 0.05% talcum powder control; and ii) 0.05% mixture of APS and ASS in a 1:1 ratio by weight. On day 21, pigs were challenged with lipopolysaccharide (LPS) and 3 h later blood samples were collected and analyzed for lymphocyte proliferation as well as interleukin 6 (IL-6), insulin-like growth factor 1 (IGF-1), growth hormone (GH), and cortisol levels. In Experiment 1, feeding Chinese herbal polysaccharides and saponin increased growth performance of the pigs. The effects of the mixture of APS and ASS were especially notable, as there was a significant improvement in growth performance compared with the control (p<0.05). The plasma concentration of immunoglobulin G (IgG), nitric oxide (NO), and nitric oxide synthase (NOS) were increased in all treatments groups, with the mixture of APS and ASS increasing the level of IgG and NOS significantly (p<0.05), compared with the control. There was no difference in the NO level between the control and treatment groups (p>0.05). In Experiment 2, Chinese herbal polysaccharides and saponin showed immunostimulating effects. The level of cortisol, GH, and IGF-I were significantly increased (p>0.05), and the level of IL-6 showed a significant decrease (p<0.05) in the APS and ASS treatment after the LPS challenge. The mixture of APS and ASS could stimulate the blood lymphocyte proliferation significantly whether the LPS was injected or not (p<0.05). These results show that Chinese herbal extracts can improve growth performance and stimulate immunity of weaned pigs. A mixture of APS and ASS, compared with APS alone, could be a new kind of immunostimulant for weaned pigs, which could result in greater positive effects on their growth performance and immunity.

  • The purpose of this study was to investigate the effect of nutritional status on the cell-mediated and humoral immunity in female college students. The nutritional status of twenty subjects was determined by six-days food records, anthropometric measurements, and biochemical assessments of serum nutrients. Cell-mediated and humoral immunity of the subjects was analyzed by in vivo and in vitro assessments. The results were summerized as follows : First, The average daily energy intake was 1437Kcal(CHO : PRO : FAT = 61:13:26), which corresponds to 71.9% of RDA. 셀레늄함유 청보리 사일리지급여가 육성기 거세한우의 생산성 및 혈액성상에 미치는 영향 (Influences of Feeding Seleniferous Whole Crop Barley Silage on Growth Performance and Blood Characteristics in Growing Hanwoo Steers)

  • 본 연구는 다양한 수준의 셀레늄함유 청보리 사일리지를 육성기 거세한우에 급여 시 성장특성 및 혈액성상에 미치는 영향을 조사하였다. 시험은 육성기 거세한우 20두 (개시체중 $208.8{\pm}17.9$ kg)를 공시하였고, 처리구는 셀레늄수준과 형태에 따라 4처리구 (셀레늄시비 및 비시비 청보리 사일리지이용 0.1 mg/kg 셀레늄 (대조구), 0.3 mg/kg 셀레늄 (T1), 0.9 mg/kg 셀레늄(T2) 및 무기셀레늄 0.9 mg/kg 셀레늄 (T3)로 나누어 처리구당 5두씩 배치하여, 90일간 사양시험을 실시하였다. 각 시험사료는 셀레늄함유 및 일반 청보리를 조합하여 배합비의 30%를 대체하였고, 무기셀레늄구 (T3)는 대조구사료에 0.9 mg/kg의 셀레늄수준에 상응하는 양의 sodium selenite를 첨가하였다. 사료내 조단백질 및 에너지함량은 처리구간 동일하였다. 사료 내 셀레늄의 수준 및 급여형태에 따라 사료섭취량과 증체에는 영향을 미치지 않았고, 혈중 포도당 농도는 대조구가 T1과 T2구 보다 유의하게 낮았다 (p<0.05). 혈중 총지질농도는 셀레늄수준이 증가할수록 유의하게 낮아져, T2구와 T3구가 대조구 보다 유의하게 낮았다 (p<0.05). LDL-콜레스테롤은 셀레늄급여구가 대조구보다 유의하게 낮았다 (p<0.05). 혈중 중성지방 또한 T2구가 대조구보다 유의하게 낮았다 (p<0.05). 혈중 면역글로불린농도는 셀레늄의 급여로 전반적으로 상승하는 경향을 보였고, 사료 내 같은 셀레늄수준에서 T3구보다는 T2구가 높은 양상을 나타내었다. 혈중 셀레늄농도는 셀레늄함유 청보리 급여수준이 높아짐에 따라 유의하게 증가였으나 (p<0.05), T3구는 대조구와 유의한 차이가 나타나지 않았다. 이상의 결과에서 육성한우에 대한 셀레늄함유 청보리급여는 육성우의 당 및 지질대사에 관여하는 것으로 나타났고, 특히 혈중 총지질 및 LDL-콜레스테롤의 농도를 감소시켰다. 또한 무기셀레늄보다는 청보리 내 존재하는 셀레늄이 장관내에서 더 잘 흡수되는 것으로 나타났고, 이는 육성기 및 어린 성장단계에 있는 거세한우의 면역력을 향상시키는데 도움이 될 수 있을 것으로 판단된다.

한국인 수유부의 수유초기 이행유의 모유성분 분석과 영아의 섭취량 추정 연구 (Studies of nutrient composition of transitional human milk and estimated intake of nutrients by breast-fed infants in Korean mothers)

  • 본 연구는 분만 후 5일에서 15일 사이에 분비되는 이행유의 성분을 분석하여 모유의 전반적인 영양소의 함량을 측정하고, 모유영양아의 하루 섭취량을 추정하여 2010년 한국인 영양섭취기준과 비교함으로써 국내 모유영양아의 영양 섭취의 적절성을 파악하고자 하였다. 연구 대상은 정상 만삭아를 출산하고 서울경기 지역 소재 산후조리원에서 회복중인 100명의 산모들이었다. 이들은 연구의 취지를 이해하고 참여 동의한 뒤 개인의 특성에 관한 설문지를 작성하고 모유 샘플을 제공하였다. 수유부들의 나이는 평균 $32.01{\pm}4.08$세였고, 신장은 평균 $161.93{\pm}4.79cm$, 만삭 체중은 $65.92{\pm}8.10kg$이었다. 모유 성분 분석 결과 모유 내 에너지 농도는 $59.99{\pm}8.01kcal/dL$, 단백질의 농도는 $1.47{\pm}0.27g/dL$, 지방의 농도는 $2.88{\pm}0.89g/dL$, 탄수화물의 농도는 $6.72{\pm}0.22g/dL$이었다. 지방산의 경우 ${\omega}-6$ 계열의 리놀레산과 아라키돈산의 모유 속 농도는 각각 $181.44{\pm}96.41mg/dL$, $28.15{\pm}8.89mg/dL$이었고, ${\omega}-3$ 계열의 리놀렌산, DHA의 모유 속 농도는 각각 $5.67{\pm}1.86mg/dL$, $5.74{\pm}2.57mg/dL$ 로 나타났다. 모유 속 비타민 A, 비타민 D, 비타민 E의 농도는 각각 $28.95{\pm}17.50{\mu}g/L$, $23.09{\pm}11.16ng/mL$, $7.37{\pm}15.43mg/L$이었으며, 비타민 C, 비타민 $B_1$, 비타민 $B_2$, 비타민 $B_{12}$, 그리고 엽산의 모유 속 농도는 $30.22{\pm}18.43mg/L$, $75.14{\pm}209.61{\mu}g/L$, $617.82{\pm}267.79{\mu}g/L$, $637.74{\pm}271.92pg/mL$, $5.16{\pm}2.58ng/mL$이었다. 무기질 중 칼슘의 모유 속 농도는 $20.71{\pm}3.34mg/dL$, 철의 모유 속 농도는 $5.86{\pm}8.61mg/L$, 칼륨의 모유 속 농도는 $66.71{\pm}10.35mg/dL$, 나트륨의 모유 속 농도는 $27.72{\pm}10.16mg/dL$이었으며, 아연과 구리는 모유 속에 각 $0.44{\pm}0.41mg/dL$, $70.48{\pm}30.41{\mu}g/dL$의 농도로 함유되어 있었다. 면역성분인 IgA의 모유 속 농도는 $61.85{\pm}31.97mg/dL$, total IgE의 모유 속 농도는 $2.35{\pm}0.93IU/mL$이었다. 영아의 하루 섭취량을 추정한 결과에 의하면 에너지 지방, 탄수화물, 비타민 A, 비타민 C, 비타민 $B_1$, 엽산, 칼슘의 하루 섭취 추정량은 이행유 섭취량으로 조정된 2010년 한국인 영양섭취기준의 충분섭취량에 미치지 못하였으나, 단백질, 비타민 D, 비타민 E, 비타민 $B_2$, 비타민 $B_{12}$, 철, 칼륨, 나트륨, 아연, 구리 등은 이행유 섭취량으로 조정된 2010년 한국인 영양섭취기준의 충분 섭취량을 충족하였다. 대부분의 영양소는 추정 섭취량이 이행유 섭취량으로 조정된 2010년 한국인 영양섭취기준과 비교하여 대체적으로 비슷하였으나 비타민 $B_1$, 엽산, 칼슘, 비타민 D, 구리 등의 영양소는 이행유 섭취량으로 조정된 2010년 한국인 영양섭취기준과 다소 차이가 있었다. 이러한 결과는 한국인 모유의 영양성분과 관련 있는 요소를 파악하는 후속 연구의 필요성을 시사하며, 한국인 모유의 성분함량과 섭취량에 관한 지속적인 연구는 모유의 질적 향상을 꾀하고 영아의 성장발달을 도모하기 위한 방안을 마련하는 기초자료로 활용될 수 있을 것이다.

Helicobacter pylori Infection is a Risk Factor for Iron-Deficiency Anemia in Korean High School Girls

  • This study investigated the relationship of Helicobacter pylori (H. pylori) infection to iron-deficiency anemia in high school girls. Four hundred and fourty-five subjects resided in Ulsan City in Korea were evaluated by questionnaire or blood analysis for serum indicators of iron status, daily nutrient intakes, symptoms of anemia, and H. pylori IgG antibody status. In H. pylori infected subjects, total energy intake of was 1534.14±350.81 kcal (73.0% of the Korean RDA), the iron intake of subjects was 11.38±3.90 mg (71.1% of Korean RDA) and calcium intake was 467.63±175.96 mg (58.3% of the Korean RDA). Carbohydrate (p < 0.05), vitamin A (p < 0.05), vitamin C (p < 0.01) intakes were significantly lower in infected subjects than in non-infected subjects. The prevalence of anemia (hemoglobin (Hb) < 12 g/dL.) in H. pylori infected subjects was 22.7% as compared to 14.7% in non-infected subjects. The mean serum ferritin of infected subjects (21.71±21.50 ng/mL) was significantly lower than in non-infected subjects (p < 0.05) (as was the mean Hb concentration (12.54±1.51 g/dL) and hematocrit (37.99±3.64%)). Total iron binding capacity (TIBC) of H. pylori infected subjects (449.09±78.23 ㎍/dL) was significantly higher than that of non-infected subjects (432.99+76.15 ㎍/dL) (P < 0.05). The levels of iron, red blood cell (RBC) and transferrin saturation were similar in infected and non-infected subjects. Two symptoms of anemia, 'pale face (p < 0.01)' and 'decreased ability to concentrate (p < 0.005)', were significantly higher in H. pylori infected subjects than in non-infected subjects. High school girls are known to be more vulnerable to iron deficiency than other age groups, and this research demonstrates that infection with H. pylori increases their risk of iron-deficiency anemia.

Comparison of Single and Blend Acidifiers as Alternative to Antibiotics on Growth Performance, Fecal Microflora, and Humoral Immunity in Weaned Piglets

  • The banning of the use of antibiotics as feed additive has accelerated investigations of alternative feed additives in animal production. This experiment investigated the effect of pure citric acid or acidifier blend supplementation as substitute for antibiotic growth promoters on growth performance, fecal microbial count, and humoral immunity in weaned piglets challenged with Salmonella enterica serover Typhimurium and Escherichia coli KCTC 2571. A total of 60 newly weaned piglets (crossbred, 28-d-old; average 8 kg initial weight) were randomly assigned to four dietary treatments in a completely randomized design. Dietary treatments included NC (negative control; basal diet), PC (positive control; basal diet+0.002% apramycin), T1 (basal diet+0.5% pure citric acid), and T2 (basal diet+0.4% acidifier blend). All piglets were orally challenged with 5 mL of culture fluid containing $2.3{\times}10^8$ cfu/mL of E. coli KCTC 2571 and $5.9{\times}10^8$ cfu/mL of S. typhimurium at the beginning of the experiment. The PC group showed the highest ADG and ADFI, whereas gain:feed was improved in the PC and T1 group (p<0.05). All dietary treatments showed significant reduction in fecal counts of Salmonella and E. coli, compared to NC (p<0.05), with PC being better than T1 and T2. Significant elevation in fecal Lactobacillus spp. counts was shown by treatments with T1, T2, and PC, whereas Bacillus spp. counts were increased by treatment with T1 and T2 compared to NC and PC diet (p<0.05). Serum IgG concentration was increased by T1 diet (p<0.05), whereas IgM and IgA were not significantly affected by any of the dietary treatments (p>0.05). From these above results, it can be concluded that, as alternatives to antibiotics dietary acidification with pure citric acid or acidifiers blend did not fully ameliorate the negative effects of microbial challenges in respect of growth performance and microbial environment, however improved immunity suggested further research with different dose levels.

L-arginine and N-carbamoylglutamic acid supplementation enhance young rabbit growth and immunity by regulating intestinal microbial community

  • Objective: An experiment was conducted to determine the effects of L-arginine (L-Arg) and N-carbamoylglutamic acid (NCG) on the growth, metabolism, immunity and community of cecal bacterial flora of weanling and young rabbits. Methods: Eighteen normal-grade male weanling Japanese White rabbits (JWR) were selected and randomly divided into 6 groups with or without L-Arg and NCG supplementation. The whole feeding process was divided into weanling stage (day 37 to 65) and young stage (day 66 to 85). The effects of L-Arg and NCG on the growth, metabolism, immunity and development of the ileum and jejunum were compared via nutrient metabolism experiments and histological assessment. The different communities of cecal bacterial flora affected by L-Arg and NCG were assessed using high-throughput sequencing technology and bioinformatics analysis. Results: The addition of L-Arg and NCG enhanced the growth of weanling and young rabbit by increasing the nitrogen metabolism, protein efficiency ratio, and biological value, as well as feed intake and daily weight gain. Both L-Arg and NCG increased the concentration of immunoglobulin A (IgA), IgM, and IgG. NCG was superior to L-Arg in promoting intestinal villus development by increasing villus height, villus height/crypt depth index, and reducing the crypt depth. The effects of L-Arg and NCG on the cecal bacterial flora were mainly concentrated in different genera, including Parabacteroides, Roseburia, dgA-11_gut_group, Alistipes, Bacteroides, and Ruminococcaceae_UCG-005. These bacteria function mainly in amino acid transport and metabolism, energy production and conversion, lipid transport and metabolism, recombination and repair, cell cycle control, cell division, and cell motility. Conclusion: L-Arg and NCG can promote the growth and immunity of weanling and young JWR, as well as effecting the jejunum and ileum villi. L-Arg and NCG have different effects in the promotion of nutrient utilization, relieving inflammation and enhancing adaptability through regulating microbial community.

Effects of essential oil coated with glycerol monolaurate on growth performance, intestinal morphology, and serum profiles in weaned piglets

  • Objective: This study aimed to investigate the effects of essential oil coated with glycerol monolaurate (GML) on the growth performance, intestinal morphology, and serum profiles of weaned piglets. Methods: A total of 144 weaned piglets (Duroc×[Landrace×Yorkshire], average weight 8.07±3.33 kg) were randomly assigned to three groups with six replicate pens and eight piglets per pen: i) CON: a corn-soybean basal diet; ii) LEG: with 1,000 mg/kg essential oil coated with GML; and iii) HEG: with 2,000 mg/kg essential oil coated with GML. Results: Results showed that average daily gain was increased (p<0.05) linearly by essential oil coated with GML supplementation on day 14 to 28 and day 0 to 28 compared with the CON group. Dietary supplementation with HEG increased (p<0.05) total antioxidant capacity and catalase activity on day 14, and immunoglobulin A (IgA) and IgM concentration on day 28 and tended to increase IgG on day 28. In addition, the crypt depth in the jejunum was reduced (p<0.05), and villus height and villus height/crypt depth in the ileum were increased (p<0.05) in the HEG group compared with the CON group. Moreover, lower (p<0.05) concentrations of tumor necrosis factor-α, interferon-γ, interleukin-1β (IL-1β), IL-8, and IL-10 were observed in the jejunum of piglets supplemented with HEG compared with the CON group. In addition, dietary HEG tended to decrease IL-6 level in the jejunum of piglets compared with the CON group. Conclusion: Dietary essential oil coated with GML can improve growth performance of weaned piglets. Moreover, supplementing 2,000 mg/kg essential oil coated with GML was demonstrated to improve antioxidant ability, and intestinal morphology, and reduce jejunal inflammatory factor levels.

General Pharmacology of $^{13}C$-Urea Powder Preparation in ${Helikat}^{TM}$

  • The pre-mixed $_{13}C$-urea powder preparation in ${Helikit}_{TM}$ for test of Helicobacter pylori was evaluated for pharmacological properties. The oral doses of the preparation used in mice were 30-fold as compared to human doses. The results obtained in the present study demonstrate that spontaneous movement, hexobarbital-induced hypnosis, rotarod performance, body temperature, acetic acid-induced writhing syndrome, chemical and electroshock convulsion, pupil size and intestinal propulsion had not been affected at the oral doses of 230, 700 and 2100 mg/kg in mice. The blood pressure was slightly elevated as given intravenously in rats at a dose of 5 mg/kg of the preparation, but respiration was not influenced at the dose. In isolated guinea pig ileum and rat fundus preparation, the preparation at a concentration of $1{\times}10^{4}$ g/ml neither caused any direct effect nor inhibited the contraction produced by acetylcholine, histamine or 5-hydroxytryptamine. These results reported here provide evidence that pre-mixed $^{13}C$ 13/C-urea powder preparation is free of general pharmacological properties performed in oral administration.

Effect of golden needle mushroom (Flammulina velutipes) stem waste on laying performance, calcium utilization, immune response and serum immunity at early phase of production

  • Objective: This experiment was conducted to evaluate the effects of golden needle mushroom (Flammulina velutipes) stem waste (FVW), on organic eggs production, calcium utilization, antibody response, serum immunoglobulin, and serum cytokine concentration at early phase of production in laying hens. Methods: A total 210, 19 weeks old aged ISA Brown layers were randomly assigned into 5 equal treatment groups, with 7 replications of 6 hens each. Dietary treatment included a standard basal diet as control; antibiotic (0.05% flavomycin); 2% FVW; 4% FVW; and 6% FVW. The experimental duration was 10 weeks. Results: There was no significant differences (p>0.05) on hen day egg production, egg weight, egg mass, feed intake, and feed conversion ratio (FCR) among experimental groups. Unmarketable eggs were significantly lower (p<0.05) both in 4% FVW and 6% FVW fed groups than control group. The calcium retention and calcium in egg shell deposition were significantly higher (p<0.05) in FVW inclusion groups than control and antibiotic groups. Antibody titers against Newcastle diseases were significantly higher (p<0.05) in 6% FVW fed group (except combined with 4% FVW at day 147) and infectious bronchitis were significantly higher (p<0.05) in FVW fed groups (except 2% FVW and 4% FVW at day 161) than control and antibiotic groups. Serum immunoglobulin sIgA was significantly higher (p<0.05) in all levels of FVW and IgG was significantly higher (p<0.05) in 4% FVW than control and antibiotic groups. Serum cytokine concentration interleukin-2 (IL-2) was significantly higher (p<0.05) in 6% FVW; IL-6 and tumor necrotic $factor-{\alpha}$ were significantly higher (p<0.05) both in 4% FVW and 6% FVW than control and antibiotic groups; IL-4 was significantly higher (p<0.05) in antibiotic, 2% FVW and 4% FVW fed groups than control. Conclusion: F. velutipes mushroom waste can be used as a novel substitute for antibiotic for organic egg production and sound health status in laying hens.
