Variations of Immunoglobulins in Colostrum and Immune Milk as Affected by Antigen Releasing Devices

  • This work was conducted to examine the variation of immunoglobulins (Igs) in serum, immune milk, normal milk and colostrum upon implantation of a new Antigen Releasing Device (ARD). The core of each ARD housed an immunostimulating complex (ISCOM) that was made of adjuvant Quil A and type XIII lipase from a Pseudomonas sp. Each ARD was coated with polylactic acid, known as polylactide, that controls antigen release. Twenty lactating Chinese Holstein cows were divided into 2 groups (n = 10): test group and control group. All cows in the test group were implanted with a single injection in the right iliac lymph node with 3 types of ARDs, which were designed to release the antigens at d 0, 14 and 28 post-implantation. Blood and milk samples were collected from both groups, and colostrum samples were also collected from other post-partum cows in the same farm. Concentrations of $IgG_1$, IgA and IgM in whey and serum were measured by sandwich ELISA. The results showed that the $IgG_1$, IgA and IgM concentrations in serum and whey from the test group were higher than from the control group. Among the three Igs measured, the $IgG_1$ concentration in serum was significantly higher at d 40 after ARD implantation, and the $IgG_1$ concentration in whey peaked at d 9, 17 and 30, which corresponded with release of the antigen. Based on Pearson's correlation between Ig concentration and production parameters, IgA concentration in normal milk was positively correlated with lactation period, which reflected IgA changes during the lactation period in immune milk. In colostrum, $IgG_1$, IgA and IgM decreased abruptly from d 0 to 3, and then decreased slightly. In conclusion, serum $IgG_1$ concentration can be affected by controlled release of the ARD, while whey IgA levels are primarily affected by lactation period. These results may be useful in future studies designed to regulate concentrations of Igs in immune milk.

  • The stabilizing effect of fructooligosanharide (FO) on hen's egg yolk immunoglobulin (yIgG) by heat and acid was investigated. The heat stability of yIgG at 70∼80$^{\circ}C$ was enhanced in a concentration-dependent manner by adding 0∼50% (w/v) FO to a yIgG solution. Acid-induced inactivation of yIgG was also suppressed in a concentration-dependent relationship by addition of FO. Addition of 50% FO almost completely stabilized yIgG at pH 3. The remarkable stablizing effect of FO on yIgG may enhance the use of yIgG as functional food ingredients.

봉독의 항독소(IgY)생산을 위한 실험적 연구 (An Experimental Study on Production of Egg Yolk Antibody(IgY) against Bee Venom)

  • This study was carried out for production of neutral antibody to bee venom $(anti-phospholipase\;A_2IgY)$. Hen layings were injected repeatedly with bee venom and phospholipase $A_2$ with Freund's adjuvant. Specific antibody in egg yolk from immunized hen laying was separated, and purified, also immunological characteristics of anti phospholipase $A_2\;IgY$ was invested. The results were summarized as follows: 1. Phospholipase $A_2$ was showed single band at molecular weight 17,000 in SDS-PAGE and bee venom was showed two band at molecular weight 17,000 and under molecular weight 6,500 in SDS-PAGE. 2. During 70 days after hen immunized with bee venom and phospholipase $A_2$, antibodies(anti-bee venom IgY) to bee venom were showed poor ELISA value in egg yolk, but antibodies$(anti-Phospholipase\;A_2IgY)$ to phospholipase $A_2$ in egg yolk were increased ELISA value from 8 days or 15 days and found maximum ELISA value at 42 days. Also after booster at 49 days, ELISA value of anti Phospholipase $A_2\;IgY$ in egg yolk was supported at optical density(O.D) 1.0 level, continuously. 3. Titer of phospholipase $A_2\;IgY$ was showed 1: 32,000. 4. In double immunodiffusion test to phospholipase $A_2$ after double dilution of anti-phospholipase $A_2\;IgY$, only precipitation line was made in 1:1 dilution well of anti-Phospholipase $A_2\;IgY$. But In immunodiffusion test to anti-phospholipase $A_2\;IgY$ after double dilution of phospholipase $A_2$, Precipitation line to 250ul/ml well of phospholipase $A_2$ was showed. In double immunodiffusion test to bee venom(1mg/ml) after double dilution anti-phospholipase $A_2\;IgY$, all well without 1:32 dilution well were showed strong precipitation line. 5. In dot bloting test to anti-phospholipase $A_2\;IgY$ after diluting bee venom(0.5mg/ml), dot bloting color was showed clearly to $1/100(5{\mu}g/ml)$ in bee venom.

A Study on the Effect of Conjugated Linoleic Acid on Immunosuppressed Mice with Corticoids

  • This study was performed to investigate the immunopotentiative effects of conjugated linoleic acid on mice immunosuppressed by administratin corticoids. Mice were divided into four groups of 8 mice. Two groups (C, CP) were given diet supplemented with 1% linoleic acid (CLA) and the other two groups (L, LP) were given diet supplemented with 1% linoleic acid (LA) instead of CLA. Prednison was administered to two groups (CP, LP) for immune depression. After feeding diets for 3 weeks containing PDS injection for last 1 week. Serum and gut lumen lavage were taken. Measurement of total Ig were executed using sandwich ELISA. Serum levels of IgA, IgG, and IgM showed some trend which groups fed with CLA were higher than groups fed with LA while IgE was reduced in those fed the CLA intake, and groups administered with PDS were lower than groups administered with saline. However, no significant differences were seen in the proportion of total immunoglobulin in serum. In case of secretory IgA, Group C and CP were significantly higher than group L and LP. Especially between CP and LP, it can be seen effects of CLA. In addition that the CLA treated group weighted a significantly lower level than the one's that have not been treated with CLA. These result support the view that CLA potentiate the immune response and prevent immune depression caused by administrating of corticoids. In conclusion, CLA produced a situation favorable for immunopotentiative effects. Thus, the clinical application of CLA is warranted.

탄광부진폐증자의 혈청 면역단백 농도에 관한 연구 (Serum Immunoglobulin Levels in Coal Workers' Pneumoconiosis)

  • Serum immunoglobulin(Ig)A, IgG, IgM, levels were measured in 99 coal workers' pneumoconiosis (CWP) patients and 12 healthy coal workers and 9 non-miners to compare with each group by the radiological categories, its complications and working period in coal mine. Serum were measured by nephelometry, The findings were as follows: 1) Serum IgA levels were significantly different between three groups of CWP patient, healthy coal worker and non-miner ($mean{\pm}standard$ deviation: $226.4{\pm}87.7,\;221.3{\pm}45.1,\;170.1{\pm}65.7$ respectively). 2) There were no significant differences of Ig levels among radiological categories of CWP. 3) There were no significant differences of Ig levels among simple pneumoconiosis and its complicated disorders. 4) The three Ig levels were slightly increased in the group of mining years less than 20 years (IgA: r=0.1869, p<0.10 IgG: r=0.2902, p<0.05 IgM: r=0.2889, p<0.05).

  • According to The Comparative study on the IgA Nephropathy and hematuria, the results were as follows. 1. IgA nephropathy is included in category of the hematuria the part in deficiency of liver-kidney and damp-heat in oriental medicine. 2. The symptoms of hematuia is changes in color of urine-bloody, dark brown, or rusty colored-without pain in oriental medicine. 3. It is occured IgA nephropathy due to vacuity exhaustion, fidgetiness, internal damage, imfection. 4. For the medical prescriptions due to IgA nephropathy are used Sogyeeumja, $Dayebunch\check{o}ngeum$(大分淸飮), Dangguieum(當歸飮), $Ch\check{o}ngjangtang$(淸腸湯), Silyungtang(柴岺湯), Kamisagunjatang(加味四君子湯) as hematuria of symtom of IgA nephropathy.

버섯첨가 두부의 면역활성에 미치는 영향 (The Effects of Tofu (Soybean curd) Containing Mushrooms on the Immune Activities)

  • 본 연구는 버섯성분을 함유한 두부의 면역조절물질로의 가치를 평가하고자 수행하였다. 표고버섯(Letino edodes)과 민자주방망이버섯(Lepista nuda)의 자실체와 균사체를 두유에 첨가하고 응고제로 $CaSO_4{\cdot}2H_2O$ 또는 젖산발효추출물을 사용하여 두부를 만들었다. 두부와 버섯성분이 함유된 두부의 영양성분을 분석하였다. Sandwich ELISA를 이용하여 비장세포 배양액에서의 IFN-${\gamma}$와 IL-4및 TNF-${\alpha}$농도를 분석하였으며, 또한 혈장과 비장세포 배양액에서 IgG1, IgG2a 및 IgE등의 농도를 측정하였다. 조단백과 조지질은 버섯성분을 함유한 두부에서 감소되었으나, 인은 유의적으로 증가하였다. IFN-${\gamma}$는 자실체나 두부를 투여한 생쥐에서 유의적으로 감소하였으며, IL-4는 표고버섯 자실체가 함유된 두부를 투여한 생쥐에서 크게 감소하였다. TNF-${\alpha}$는 표고버섯 성분을 함유한 두부를 투여한 생쥐에서 유의적으로 증가하였다. 혈장의 IgG1은 거의 모든 실험군에서 증가되었으나, IgG2a는 대조군에 비해 큰 변화가 없었다. 비장임파구 배양액에서 측정한 IgG1 와 IgG2a의 농도는 자실체나 균사체를 첨가한 두부의 대부분의 그룹들에서 현저하게 높게 나타났다. IgE의 혈장농도는 두부나 표고버섯을 투여한 생쥐에서 크게 증가하였으나 버섯 성분을 첨가한 두부 투여 생쥐그룹에서는 그 생성이 억제되었다. 이러한 결과들은 버섯 성분을 함유한 두부들이 면역활성에 영향을 준다는 것을 나타내며, 면역활성의 관점에서 두부와 표고버섯의 혼합물은 개발가치가 있는 것으로 사료된다.

소아에서 관찰된 IgG-associated mesangial glomerulonephritis 2례 (Two Cases of IgG-associated Mesangial Glomerulonephritis in Children)

  • IgG 신병증(IgG-associated mesangial glomerulonephritis)은 전세계적으로 드물게 보고되고 있는 새로운 원발성 사구체 질환으로 1993년 Sato 등에 의해 처음으로 보고되었고 이후 1994년 Yoshikawa 등에 의해 소아에서의 10례가 보고되었다. 주로 단백뇨를 주소로 하며 현미경적 혈뇨를 동반하기도 한다. 이 질환은 조직 면역 형광 검사상 사구체 간질에 IgG의 침착을 보이는 사구체 신염이다. 이 질환은 초기에는 양성 경과를 취하는 것으로 알려졌으나 2002년 Fakhouri 등은 대부분은 양성 경과를 보이지만 일부는 말기 신부전증으로 진행함을 관찰할 수 있었으며 1례에서는 신장이식 후 이식신에서 IgG 신병증이 재발된 예도 보고 하였다. 본 증례에서는 한명은 신증후군이 동반되었고 한명은 무증상 현미경적 혈뇨만을 보였다. 이들 모두 면역 형광 현미경 소견상 사구체 간질에 IgG 침착이 주로 보였고 IgM, 보체의 침착도 보였다. 또한 전자현미경 소견상 사구체간질에 고밀도 전자 침착이 보였다. 신생검에서 면역 현미경 소견과 전자 현미경 소견으로 진단된 IgG 신병증 소아환자 2례를 경험하고 국내에서는 아직 보고된 예가 없기에 문헌 고찰과 함께 증례를 보고하는 바이다.

폐흡충(Paragonimus westermani) 감염백서에서의 혈청내 IgE 항체가의 변동 (Serum IgE levels in rats infected with Parugonimus westermani)

  • 폐츱충(Paragonimus westermani) 감염시 탈낭유충은 숙주내 여러조직을 거친 후 폐에 기생하게 되는데 이때 조직반응과 더불어 혈액내 호산구와 IgE 항체가의 변등이 일어난다. 이 실험에서는 백서에 폐흡충을 감염시키고 혈청내 총 IgE와 특이 IgG 항체값을 시간 경과별로 측정하여 감염후 혈청내 항체가의 변증을 관찰하였다. 폐흡충 퍼낭유충 20개를 백서에 경구 감염시키고 시기별(0,1,2,3,4,0,8주)로 IgE의 변동을 avidin-biotin을 이용하여 효소표식 면역검사법 (enzyme-linked immunosorbent assay: ELISA)으로 측정하였으며 IgG는 통상적인 ELISA 방법을 이용하여 측정하였다. 실험군의 혈청내 총 IgE치는 감염 3주와 3주 후에 흡광도가 각각 0.18±0.042, 0.18±0.081이었으나, 4주에 0.28±0.151로 급격히 중가하었으며 8주까지 지속적인 상승(0,43±0,055)을 보였다. 반면 대조군의 경우 전 실험기간을 통해 0.07∼0.12로 낮은 항체가가 유지되었다. 또한 실험군의 혈청내 IgG 항체는 감염 3주 후에 흡광토 0.20±0,032로 증가하기 시작하여 8주에 0.31±0.067을 보였으나 대조군에 비해 크게 증가되지 않았다. 이상의 결과로 보아 비호적숙주인 백서에서 폐흡충의 감염이 IgE 및 IgG 항체를 증가시킴을 알 수 있었다.

Newly Identified TLR9 Stimulant, M6-395 Is a Potent Polyclonal Activator for Murine B Cells

  • Background: Toll-like receptors (TLRs) have been extensively studied in recent years. However, functions of these molecules in murine B cell biology are largely unknown. A TLR4 stimulant, LPS is well known as a powerful polyclonal activator for murine B cells. Methods: In this study, we explored the effect of a murine TLR9 stimulant, M6-395 (a synthetic CpG ODNs) on B cell proliferation and Ig production. Results: First, M6-395 was much more potent than LPS in augmenting B cell proliferation. As for Ig expression, M6-395 facilitated the expression of both TGF-${\beta}1$-induced germ line transcript ${\alpha}$ ($GLT{\alpha}$) and IL-4-induced $GLT{\gamma}1$ as levels as those by LPS and Pam3CSK4 (TLR1/2 agonist) : a certain Ig GLT expression is regarded as an indicative of the corresponding isotype switching recombination. However, IgA and IgG1 secretion patterns were quite different--these Ig isotype secretions by M6-395 were much less than those by LPS and Pam3CSK4. Moreover, the increase of IgA and IgG1 production by LPS and Pam3CSK4 was virtually abrogated by M6-395. The same was true for the secretion of IgG3. We found that this unexpected phenomena provoked by M6-395 is attributed, at least in part, to its excessive mitogenic nature. Conclusion: Taken together, these results suggest that M6-395 can act as a murine polyclonal activator but its strong mitogenic activity is unfavorable to Ig isotype switching.
