The Soluble Form of the Cellular Prion Protein Enhances Phagocytic Activity and Cytokine Production by Human Monocytes Via Activation of ERK and $NF-{\kappa}B$

  • The $PrP^C$ is expressed in many types of immune cells including monocytes and macrophages, however, its function in immune regulation remains to be elucidated. In the present study, we examined a role for $PrP^C$ in regulation of monocyte function. Specifically, the effect of a soluble form of $PrP^C$ was studied in human monocytes. A recombinant fusion protein of soluble human $PrP^C$ fused with the Fc portion of human IgG1 (designated as soluble $PrP^C$-Fc) bound to the cell surface of monocytes, induced differentiation to macrophage-like cells, and enhanced adherence and phagocytic activity. In addition, soluble $PrP^C$-Fc stimulated monocytes to produce pro-inflammatory cytokines such as $TNF-{\alpha}$, $IL-1{\beta}$, and IL-6. Both ERK and $NF-{\kappa}B$ signaling pathways were activated in soluble $PrP^C$-treated monocytes, and inhibitors of either pathway abrogated monocyte adherence and cytokine production. Taken together, we conclude that soluble $PrP^C$-Fc enhanced adherence, phagocytosis, and cytokine production of monocytes via activation of the ERK and $NF-{\kappa}B$ signaling pathways.

  • We propose a new method for recognizing the activities of daily living(ADL) based on the state-dependent motion analysis using 3-axial accelerometers and a glove type RFID reader. Two accelerometers are used for the classification of 5 body states based on the decision tree. Classification of the instrumental activities is performed based on the hand interaction with an object ID using an accelerometer and a RFID reader. Object-dependent hand movements are classified into 5 categories in advance and final decision combines the body state and the instrumental activities. Experiment shows that the suggested hierarchical motion analysis provides accuracy rate of over 90% for all 20 ADLs.

Prevalence of Human Papillomavirus Infection in Women in South Korea -Incidence of Positive HPV DNA and anti-VLPs in Residents of Busan City- (한국인 일반 여성의 HPV 감염 유병율 -부산지역 일반 여성에서의 HPV DNA 및 항 VLPs 항체 양성 빈도 -)

  • To investigate a population-based survey of the prevalence of human papillomavirus (HPV) infection in South Korea, we performed Papanicolaou smears and tests for HPV DNA and anti-HPV antibody detection in 909 sexually active general women (age range; 20-74 years, median 44 years) who were randomly selected residents from S district of Busan City. The presence of DNA of 36 different HPV types was detected by means of a GP 5+/6+ primer-mediated PCR enzyme immunoassay in cervical exfoliated cells, and IgG antibodies against L1 virus-like particles (anti-VLPs) of 5 HPV types 16, 18, 31, 33, and 58 were tested by means of enzyme linked immunoassay. The incidence of cytologic abnormality was 5.2% in Pap smear. The positive rate of HPV DNA was 10.4%, high in young women younger than 35 years old and proportionally increased according to the cytologic grades. The most often found HPV type was HPV 70, followed by HPV 16 and 33, and high-risk HPV types were more frequent in women younger than 35 years old. The most common HPV type in abnormal cytologic smears was HPV 16, followed by HPV 58 and 66. Anti-VLPs was positive in 19.7% and the frequent anti-VLPs type was against HPV 18, followed by HPV 31 and 16. The concordance between the markers for each specific HPV type was noted in 10 women and HPV 16 was the most frequent one. The incidence of multiple HPV infection was 18.9% and that of multiple anti-VLPs antibodies was 31%. Among 103 self-reported virgins, 4.9% had anti-VLP antibodies.

Comparison of TIA with ELISA for circulating antibody detection in clonorchiasis (간흡충증에 있어서 항체검출을 위한 Enzyme-linked Immunosorbent Assay와 Thin Layer Immunoassay의 비교)

  • A comparison was made of a new serological method, thin layer immunoassay (TIA), and an established method, enzyme-linked immunosorbent assay (ELISA), in the detection and quantiacation of antibodies in clonorchiasis. Saline extract of Iyophilized Clonorchis sinensis adult worm was used as antigen, and TIA by the method of Elwing et at. (1976) and ELISA by Voller et at. (1974) were performed. Using sera from known clonorchiasis cases,100% of the sera tested were Positive by TIA and 88.35 by ELISA. TIA produced false positive results in 14 out of 36 cases, which were 10 amoebiasis cases, 16 paragonimiasis cases and 10 healthy controls. ELISA. however, produced a small number of false positives, 7 out of 55 cases. There was correlation between Immunoglobulin G level in sera and ELISA value (correlation coefficient, 0.69), whereas no correlation between Immunoglobulin G level and TIA result. The Performance of TIA and ELISA was not correlated in the results using homologous antigen.

  • Neurognathostomiasis is a severe form of human gnathostomiasis which can lead to disease and death. Diagnosis of neurognathostomiasis is made presumptively by using clinical manifestations. Immunoblotting, which recognizes antigenic components of molecular mass 21 kDa and 24 kDa in larval extracts of Gnathostoma spinigerum (Gs 21/24), has high sensitivity and specificity for diagnosis of neurognathostomiasis. However, only very small amounts of the Gs 21/24 antigens can be prepared from parasites harvested from natural or experimental animals. To overcome this problem, we recently produced a recombinant matrix metalloproteinase (rMMP) protein from G. spinigerum. In this study, we evaluated this rMMP alongside the Gs 21/24 antigens for serodiagnosis of human neurognathostomiasis. We studied sera from 40 patients from Srinagarind Hospital, Khon Kaen University, Thailand, with clinical criteria consistent with those of neurognathostomiasis, and sera from 30 healthy control adults from Thailand. All sera were tested for specific IgG antibodies against both G. spinigerum crude larval extract and rMMP protein using immunoblot analysis. The sensitivity and specificity for both antigenic preparations were all 100%. These results show that G. spinigerum rMMP protein can be used as an alternative diagnostic antigen, in place of larval extract, for serodiagnosis of neurognathostomiasis.

Subacute toxicological study of PG102, a water-soluble extract derived from Actinidia arguta, in SD rats (다래(Actinidia arguta) 열매의 열수 추출물, PG102의 4주 반복 경구투여에 대한 안전성 연구)

  • It was previously found that PG102, a water-soluble extract derived from Actinidia arguta, was able to modulate Th1/Th2 pathways and suppress IgE production resulting in dramatic amelioration of atopic dermatitis in NC/Nga mouse and hairless rat models. In order to evaluate the subacute toxicity of PG102, female and male SD rats were daily fed with various doses of PG102 for 4 weeks. Six week old SD rats were randomly divided into 4 groups and orally administrated with 100-, 300-, and 1,000- mg/kg of PG102 as well as the vehicle only. At the end of the study, no significant differences in the body and organ weights were observed between control and treated rats of both genders. Hematological and blood chemical analysis showed little differences between the animal groups. Neither gross abnormalities nor histopathological changes were found. PG102 produced little or no subacute toxicity and could be used as a safe nutraceutical for the treatment of individuals with allergic diseases including atopic dermatitis.

Comparison of Anti-allergenic Activities of Various Polyphenols in Cell Assays

  • The inhibitory effects of 25 polyphenols against in vitro allergic reactions were compared using biochemical and cell assays. Three polyphenols including curcumin, gallic acid, and quercetin suppressed the release of $\beta$-hexosaminidase from ionophore A23187-stimulated RBL-2H3 cells more effectively (>50% inhibition at $100{\mu}M$ concentration). They were found to have potencies in suppressing the release of histamine not only from ionophore A23187-, but also from immunoglobulin E (IgE)-stimulated RBL-2H3 cells. Moreover, such suppressive effects of the three polyphenols were also observed in A23187 plus PMA-costimulated rat peritoneal mast cells. The extent of inhibition were quantified as the respective polyphenol concentration that inhibit 50% ($IC_{50}$) of $\beta$-hexosaminidase or histamine release, showing an inhibition tendency with decreasing order of curcumin>gallic acid>quercetin. Down-regulation of $Ca^{2+}$ influx was suggested as the cause of the inhibition of $\beta$-hexosaminidase and histamine releases in these cells. The immune process inhibition was confirmed by the observed reduction in the gene expressions and release of pro-inflammatory cytokine tumor necrosis factor (TNF)-$\alpha$, interleukin (IL)-$1\beta$, and IL-4, due probably to antioxidant activity of the polyphenols. These findings illustrate that curcumin, gallic acid, and quercetin may be beneficial against allergic inflammatory diseases.

Wideband Stacked Microstrip Antenna with Rectangular and Triangular Parasitic Patches for 860MHz Band (직사각형 및 삼각형 기생패치를 이용한 860MHz 대역 광대역 적층 마이크로스트립 안테나)

  • A wideband stacked patch antenna with parasitic elements, rectangular and triangle shaped patches, is proposed. Two different shaped parasitic elements are placed in the above of main rectangular microstrip patch antenna in order to achieve wide bandwidth for 860 MHz band. Coupling between the main patch and parasitic patches is realized by thick air gap. The gap and locations of parasitic patches are found to be the main factor of the wideband impedance matching. The proposed antenna is designed and fabricated on a ground plane with small size of $119mm{\times}109mm$ for application of compact transceivers. The fabricated antenna on an FR4 substrate shows that the minimum measured return loss is below -11.68dB at 824 MHz and an impedance band of 818~919 MHz(11.7%) at 10dB return loss level. The measured radiation patterns are similar to those of a conventional patch antenna with maximum gain of 2.11 dBi at 824 MHz.

Seroprevalence and Risk Factors of Toxoplasma gondii Infection among Cat Sitters in Korea

  • The seroprevalence of human toxoplasmosis has been increasing in Korea, and it is controversial whether cats are an important infection source or not. This study was performed to evaluate the seroprevalence of Toxoplasma gondii infection in a high risk group (cat sitters) and to determine the possible importance of cats as an infection source in Korea. Risk factors, including the age, sex, and diet of cat sitters, their contact experience and contact frequency with stray cats, and origin, number, and outdoor activity of their pet cats, were analyzed using structured questionnaires. A total of 673 serum samples from people who have frequent contact with cats (high risk group) and 1,114 samples from general people (low risk group) were examined for specific IgG antibodies against T. gondii by ELISA. The results revealed that the overall seroprevalence of T. gondii infection was 7.4% (n=1,787). The seroprevalence among low risk group was 8.0% (89/1,114), whereas that among high risk group was rather lower 6.4% (43/673), though this difference was statistically not significant (P=0.211). Among the risk factors, only the outdoor activity of pet cats was important; people having cats with outdoor activities revealed 2 times higher seroprevalence than people having cats with only indoor activities (P=0.027). In conclusion, the seroprevalence of T. gondii was not significantly different between the high risk group and low risk group, and the importance of cats as a source of infection in Korea is questionable.

Inhibitory Effects of Black-red Ginseng Extracts on Allergic Inflammation In Vitro and In Vivo (In Vitro 및 In Vivo 알러지성 염증반응에 대한 흑홍삼의 억제효과)

  • Black-red ginseng (BRGP) exhibits more potent biological activities than white and red ginseng. However, the effect of BRGP on allergic inflammation has not been studied extensively. In this study, we attempted to evaluate the effects of BRGP on allergic inflammation in vitro and in vivo. The effects of BRGP on pro-inflammatory mediators in phorbol myristate acetate (PMA) and A23187-stimulated human mast cells (HMC-1) and on trimellitic anhydride (TMA)-induced atopic dermatitis-like skin lesions in BALB/c mice were evaluated. BRGP suppressed the expression of $TNF-{\alpha}$, IL-6 and IL-8 from HMC-1 stimulated with PMA and A23187. Furthermore, the oral administration of BRGP markedly significantly reduced apparent severity of atopic dermatitis-like skin lesions, ear swelling, lymph node weight gains and serum IgE levels induced by TMA in mice. BRGP treatment also ameliorated epidermal hyperplasia and infiltration of inflammatory cells including mast cells. Taken together, BRGP possesses significant anti-atopic efficacy, suggesting that it could be used as a potential therapeutic agent for allergic inflammatory diseases, including atopic dermatitis.
