Type II Isopentenyl Diphosphate Isomerase로서 Synechocystis sp. PCC6803의 sll1556의 작용 특성 (Functional Characterization of sll1556 of Synechocystis sp. PCC6803 as Type II Isopentenyl Diphosphate Isomerase)

  • Synechocystis sp. PCC6803의 type II Isopentenyl diphosphate isomerase gene(sll1556, Syidi2)의 특성을 살펴보기 위하여 ${\Delta}idi$인 E. coli $DH5{\alpha}$를 제작하고, 이 균주에서 cloning하고 발현시켰다. 라이코펜 합성 유전자들(crtE, crtB, and crtI)과 mevalonate pathway 유전자들(MvK1, MvK2, Mvd)를 함유한 ${\Delta}idi$ E. coli $DH5{\alpha}$ 균주를 mevalonate가 함유된 LB 배지에서 배양하면 mevalonate pathway 유전자들을 함유한 E. coli $DH5{\alpha}$균주는 mevalonate에 의해 생성된 isopentenyl diphosphate의 독성에 의해 매우 느린 성장을 보였다. 라이코펜 합성유전자들과 mevalonate 합성유전자들을 함유한 ${\Delta}idi$ E. coli $DH5{\alpha}$ 균주에 Syidi2를 도입한 결과, mevalonate가 함유된 LB배지에서 균체의 성장이 완전히 회복되었으며, 라이코펜이 합성되었음을 나타내는 붉은 균락이 형성되었다. 이에 따라, SyIdi1과 ECidi를 도입하여 비교한 결과, 라이코펜 합성 유전자들과 mevalonate pathway 유전자들을 함유한 ${\Delta}idi$ E. coli $DH5{\alpha}$ 균주 자체와 SyIdi1을 도입한 라이코펜 합성 유전자들과 mevalonate pathway 유전자들을 함유한 ${\Delta}idi$ E. coli $DH5{\alpha}$ 균주는 IPP의 독성에 의해 성장이 매우 느렸으나, SyIdi2, RSidi, HPidi, 및 ECidi를 함유하고 있는 ${\Delta}idi$ E. coli $DH5{\alpha}$ 균주는 균체의 성장과 라이코펜의 합성을 완전히 회복하였으며, 그중 가장 우수한 라이코펜 생합성 결과를 나타낸 것은 pSUP-LYCSyIdi2로서 균체당 라이코펜 생성량이 control 대비 2.8배이었다.

  • Starvation에 의해서 down-regulation 되는 유전자 10개를 얻었다(Comt, RGN, Scd1, Temt, Idi1, Fabp5, Car3, Cyp2c70, Pinx1, Poldip3). 이들은 starvation에 의한 대사변화의 대부분은 liver와 관련된 것으로 볼 수 있다. Starvation중에 물 공급은 암수 동일하게 apoptosis, autophage, endoplasmic reticulum quality control (ERQC)유도에 영향을 미치지 않았다. 이 같이 starvation에 의해서 down-regulation되는 유전자발현조절이 liver에 국한된 것이라기보다는 개체의 항상성유지에 관련 많은 pathway가 관련되어있는 것으로 판단된다. 장기간의 간혈 starvation은 glucose소비가 많은 brain과 면역기능조절에 중요한 thymus의 정상기능에 영향을 미칠 수 있는 것으로 보인다. 유전자 Scd1의 경우는 ♀보다가 ♂이 민감한 반응을 보이는 것으로 보아 ♀/♂의 성 특이적인 대사에 starvation과 NaCl이 밀접한 관계가 있는 것으로 보인다. Starvation시 물 공급도 중요하지만 개체의 항상성유지에 NaCl공급이 중요하다는 결과를 얻었다.

Development of a Transformation System for the Medicinal Fungus Sanghuangporus baumii and Acquisition of High-Value Strain

  • To further explore the molecular mechanism of triterpenoid biosynthesis and acquire high-value strain of Sanghuangporus baumii, the Agrobacterium tumefaciens-mediated transformation (ATMT) system was studied. The key triterpenoid biosynthesis-associated gene isopentenyl diphosphate isomerase (IDI) was transformed into S. baumii by ATMT system. Then, the qRT-PCR technique was used to analyze gene transcript level, and the widely targeted metabolomics was used to investigate individual triterpenoid content. Total triterpenoid content and anti-oxidant activity were determined by spectrophotometer. In this study, we for the first time established an efficient ATMT system and transferred the IDI gene into S. baumii. Relative to the wild-type (WT) strain, the IDI-transformant (IT) strain showed significantly higher transcript levels of IDI and total triterpenoid content. We then investigated individual triterpenoids in S. baumii, which led to the identification of 10 distinct triterpenoids. The contents of individual triterpenoids produced by the IT2 strain were 1.76-10.03 times higher than those produced by the WT strain. The triterpenoid production showed a significant positive correlation with the IDI gene expression. Besides, IT2 strain showed better anti-oxidant activity. The findings provide valuable information about the biosynthetic pathway of triterpenoids and provide a strategy for cultivating high-value S. baumii strains.

시뮬레이션을 이용한 대기행렬 네트워크 도착과정의 변동성함수에 관한 연구 (A Simulation Study on the Variability Function of the Arrival Process in Queueing Networks)

  • 본 연구에서는 대기행렬네트워크 성과측정 방법 중의 한 가지로서 널리 이용되는 분해법의 구성요소로 제안된 변동성 함수 의 이론적 근거를 살펴보고 성과척도 측정의 정확도 제고를 위하여 회귀분석을 통한 변동성 함수의 모수추정 개선방안을 제안하고자 한다. 이를 위하여 변동성이 높은 도착과정과 서비스 과정이 포함된 직렬 대기행렬 네트워크에서의 이탈과정의 자동상관계수 함수를 추정하여 분해법에 사용할 수 있는 방안을 알아본다.

CHARACTERISTICS OF PERFORMANCE AND EXHAUST EMISSION OF DIESEL ENGINES BY CHANGES IN FUEL PROPERTIES AND APPLICATION OF EGR

  • In this study, the potential use of oxygenated fuels such as ethylene glycol mono-normal butyl ether (EGBE) was investigated in an attempt to reduce the emission of exhaust smoke from diesel engines. Effects of the combustion method on exhaust emission of DI and IDI diesel engines were also examined. Since EGBE is composed of approximately 27.1% oxygen, this is one of several potential oxygenated fuels that could reduce the smoke content of exhaust gas. EGBE blended fuels have been proven to reduce smoke emission remarkably compared to the conventional commercial fuels. The test was conducted with single and four cylinder, four stroke, DI and IDI diesel engines. The study showed that a simultaneous reduction of smoke and NOx emission could be achieved by the combination of oxygenated blend fuels and the cooled EGR method in both DI and IDI diesel engines. It was also found that a reduction rate of exhaust emission in a DI engine was larger than an IDI diesel engine.

Identification of simvastatin-regulated targets associated with JNK activation in DU145 human prostate cancer cell death signaling

  • The results of this study show that c-Jun N-terminal kinase (JNK) activation was associated with the enhancement of docetaxel-induced cytotoxicity by simvastatin in DU145 human prostate cancer cells. To better understand the basic molecular mechanisms, we investigated simvastatin-regulated targets during simvastatin-induced cell death in DU145 cells using two-dimensional (2D) proteomic analysis. Thus, vimentin, Ras-related protein Rab-1B (RAB1B), cytoplasmic hydroxymethylglutaryl-CoA synthase (cHMGCS), thioredoxin domain-containing protein 5 (TXNDC5), heterogeneous nuclear ribonucleoprotein K (hnRNP K), N-myc downstream-regulated gene 1 (NDRG1), and isopentenyl-diphosphate Delta-isomerase 1 (IDI1) protein spots were identified as simvastatin-regulated targets involved in DU145 cell death signaling pathways. Moreover, the JNK inhibitor SP600125 significantly inhibited the upregulation of NDRG1 and IDI protein levels by combination treatment of docetaxel and simvastatin. These results suggest that NDRG1 and IDI could at least play an important role in DU145 cell death signaling as simvastatinregulated targets associated with JNK activation.

Dot-immunoassay를 이용한 Aujeszky's disease virus항원 및 항체 검출 (Detection of antibodies and antigens of Aujeszky's disease virus using dot-immunoassay)

  • To establish more specific and simple diagnostic methods for detection of the antibodies and antigens of Aujeszky's disease virus(ADV), we designed indirect dot-immunoassay(IDI) and double sandwich dotimmunoassay(DSDI) using the solid phases of nitrocellolose paper and polystyrene plate. The diagnostic efficacy of these methods was investigated. As the sensitivity of IDI was tested by various virus concentration, the specimens with the virus titer above $10^{4.0}TCID_{50}/0.2ml$ showed positive reaction, but that below $10^{1.0}TCID_{50}/ml$ revealed negative. Tonsil emulsion at the virus titer of $10^{4.5}TCID_{50}/0.2ml$ showed the highest sensitivity as diluted by 1/100. In detection of ADV antigens from the various tissues of the rats and pigs infected with ADV, IDI using monoclonal antibody showed the higher specificity as compared with IDI using polyclonal antibody and virus isolation method. The efficacy of the DSDI for detection of ADV antibody was compared with other tests. The sensitivity of DSDI was higher than virus neutralization(VN) and agar gel immunodiffusion test(AGID). Meanwhile, specificity of DSDI was lower than AGID, but similar to IDEA. In comparison with VN test, DSDI showed 96.9% agreement to VN test that is the highest of three tests. In general, application of polyclonal antibody in both tests caused the higher sensitivity but the lower specificty.

Alterations of Antioxidant Status and Mitochondrial Succinate Dehydrogenase Activity in the Liver of Wistar Strain Albino Rats Treated with by Ethanol Extracts of Annona senegalensis Pers (Annonaceae) Stem Bark

  • Numerous ethnomedicinal uses have been attributed to different parts of Annona senegalensis (ASE), including its uses as food and food additives. The present study investigated toxicological and antioxidant effects of 28 days administration of ethanol extracts of ASE stem bark to Wistar strain albino rats. Acute toxicity test was done to determine lethal dose in Wistar rats while sub-acute toxicity test was conducted on rats divided into four groups (A - control, B - 50 mg/kg, C - 100 mg/kg, D - 150 mg/kg, respectively and treated for 28 days. Oxidative stress markers in liver and kidney as well as hepatic succinate dehydrogenase activity in the mitochondrial and post mitochondrial fractions (PMF) were evaluated. The $LD_{50}$ value of ASE was > 2,000 mg/kg. White blood cell counts gradually increased, but red blood cell counts and haematocrits level decreased significantly (p < 0.05) by about 50%. Liver enzymes in the serum and mitochondrial succinate dehydrogenase activity increased significantly (p < 0.05). Superoxide dismutase and catalase activities also increased in liver mitochondria and PMF while malondialdehyde (MDA) and reduced glutathione levels increased only in the PMF. Furthermore, only MDA levels increased significantly in the kidney after 28 days extract administration. Histopathological examination showed hepatic necrosis and no obvious signs of nephrotoxicity. Anona senegalensis is relatively safe, but prolonged ingestion could induce oxidative stress and impair ATP synthesis through the modulation of the activity of mitochondrial succinate dehydrogenase.

바이오디젤유를 사용하는 간접분사식 디젤기관의 내구 및 배기 특성 (The Durability and Exhaust Emission Characteristics of an IDI Diesel Engine Using Biodiesel Fuel)

  • To evaluate the durability characteristics of in-direct injection diesel engine using BDF 20(a blend of 20% biodiesel fuel and 80% diesel fuel in volume), an IDI diesel engine used to commercial vehicle was operated on BDF 20 for 300 hours. Engine dynamometer testing was completed at regularly scheduled intervals to investigate the combustion characteristics, engine performance and exhaust emissions. The engine performance and exhaust emissions were sampled at 1 hour interval for analysis. From the results, the combustion variations such as the combustion maximum pressure($P_{max}$) and the crank angle at which this maximum pressure occurs(${\Theta}_{Pmax}$) were not appeared during long-time dynamometer testing. Also, BSFC with BDF 20 resulted in lower than with diesel fuel. The peak pressure with BDF 20 was higher than that with diesel fuel due to the oxygen content in BDF. And, BDF 20 resulted in lower emissions of carbon monoxide, carbon dioxide, and smoke emissions with a little increase of oxides of nitrogen than diesel fuel. It was concluded that there was no unusual deterioration of the engine, or any unusual change in exhaust emissions during the durability test of an IDI diesel engine using BDF 20.

간접분사식 디젤기관에서 바이오디젤연료의 연소 특성 (The Combustion Characteristics of Biodiesel Fuel as an Alternative Fuel for IDI Diesel Engine)

  • Recently, lots of researchers have been attracted to develope various alternative fuels and to use renewable fuels as a solution of environmental problems from automobile. The use of biodiesel fuel is an effective way of substituting diesel fuel in the long nun. It is a domestically produced, renewable fuel that can be manufactured from vegetable oils, used vegetable oils, or animal fats. In this study, the usability of biodiesel fuel derived from rice ban oil, one of the oxygenated fuels as an alternative fuel for diesel engines was investigated in IDI diesel engine. Emissions were characterized with neat biodiesel fuel and with a blend of biodiesel fuel and conventional diesel fuel. Since the biodiesel fuel includes oxygen of about 11%, it could influence the combustion process strongly. So, the use of biodiesel fuel resulted in lower emissions of carbon monoxide and smoke emissions with some increase in emissions of oxides of nitrogen. It is concluded that biodiesel fuel can be utilized effectively as a renewable fuel for IDI diesel engine.
