Sex ratio and conception rates of fresh/vitrified embryos at different developmental stages by ovum pick up in Hanwoo cows

  • Embryo transfer plays a crucial role in enhancing the breeding value of livestock; it has been applied in Hanwoo cattle, which is a popular breed for beef production in Korea. Both in vivo-derived (IVD) and in vitro-produced (IVP) embryos are used for this purpose; however, IVP embryos have been preferred recently owing to advancements in ovum pick-up (OPU) technology and genomic selection. Despite technological advancements, comprehensive data on large-scale OPU/IVEP/embryo transfer in Hanwoo cows are lacking. In this study, 16 elite Hanwoo donor cows were selected on the basis of specific criteria. Oocytes were retrieved from 241 cows using OPU. The collected cumulus-oocyte complexes (COCs) were matured, fertilized, and cultured in vitro to produce transferable embryos. Embryos were classified according to their developmental stage and then transferred to 675 recipient cows. A total of 3,317 COCs were collected, with an average of 13.76 COCs per cow. The number of transferable embryos produced per cow was 3.7. Hanwoo OPU-derived IVP embryos exhibited a higher production yield than the global average, indicating a stable IVEP environment. Both fresh and frozen IVP embryos yielded similar conception rates; hence, the use of vitrified-thawed embryos in transfer plans feasible. However, frozen-thawed embryos at Stage 7 had a lower conception rate than those at earlier stages. There was no significant difference between the conception rates of sexually mature heifers and postpartum cows used as recipients. The male-to-female offspring ratio increased as the developmental stage progressed. Seasonal effects on conception rates were not observed; however, higher abortion rates and a higher proportion of male offspring were observed during winter. This study provides valuable data for the Korean embryo transfer industry, enabling more strategic growth of the domestic Hanwoo embryo industry.

  • In vitro produced (IVP) embryos produced by in vitro fertilization (IVF) often exhibit wide variations in developmental competence and viability, considerably more than are exhibited by embryos that develop in vivo. These anomalies in IVP embryos may be due to heterogeneity of oocyte quality, suboptimal culture conditions, disturbances in gene expression, or most likely a combination of these factors (Ho et al., 1994; Roth et al., 1994; McKiernan and Bavister, 1998; Hasler, 1998; Schramm and Bavister, 1999; Doherty et al., 2000; Hyttel et al., 2000; Niemann and Wrenzycki, 2000; Wrenzycki et al., 2001). In research studies or in clinical applications with domesticated animals, cats, non-human primates and humans, oocytes used for IVF are usually collected from a heterogeneous cohort of ovarian follicles that include oocytes which normally might not be ovulated and/or are deficient in developmental competence.   • The removal of impulsive noise terms which occur in interventricular pressure (IVP) signals of a Korean-type total artificial heart is essential for estimation of atrial pressure change. We compared various order statistic filters and conclude that median filter with sidelength L = 1 is the most appropriate filter for IVP signals in the perspectives of operation cost, detail preserving (peak value), and waveform.

액상 정액의 보존 기간이 정자 기능 및 정자염색질 구조 분석에 미치는 영향과 인공수정 분만율과의 상관관계 (Effect of Storage Times on Sperm Function, Sperm Chromatin Structure Assay (SCSA) and Correlations Between Fertility and SCSA in Boars)

  • 본 연구는 인공수정 센터에서 이용되고 있는 종모돈의 인공수정 분만율의 차이를 기준으로 하여 정액의 보존 일에 따른 정자의 생존율, CASA, HOST, IVP 및 SCSA를 조사하여 이들 정자의 분석법들과 인공수정 분만율과의 상관관계를 조사하여 종모돈의 수정 능력을 예측할 수 있는 정액 평가법을 개발하고자 실시하였다. 종모돈의 액상정액을 대상으로 CASA, HOST, IVP 및 SCSA 기법을 적용한 정액의 평가 결과는 다음과 같았다. 정액의 보존 일별 HOST 결과는 보존 일이 경과함에 따라 유의적으로 감소하였고(P<0.05), IVP 결과 난자 당 침투 정자 수는 보존 3일째에 유의적으로 감소하였다(P<0.05). 보존 0일과 6일에 HOST는 분만율이 80% 이상인 종모돈 군이 70% 미만인 종모돈 군 보다 유의적으로 높았고, 3일에서는 80% 이상인 종모돈 군이 다른 종모돈 군에 비하여 높았다(P<0.05). IVP는 보존 0일에 분만율이 80% 이상인 종모돈 군이 70% 미만인 종모돈 군보다 난자당 침투 정자 수가 유의적으로 많았다(P<0.05). %Red는 보존 0일과 3일에는 분만율이 80% 이상과 70% 미만인 종모돈 군 간에 유의적인 차이를 보였으나 보존 6일에서는 모든 종모돈 군간에 유의적인 차이가 있었다(P<0.05). %Red(r=0.79, P<0.01 ; r=0.86, P<0.01 ; r=0.88, P<0.01)는 정액의 보존 일이 증가함에 따라 분만율과의 부의 상관 계수가 증가하였다. 이상의 결과를 종합해 보면 SCSA는 정액의 수정 능력을 평가하는데 있어서 매우 유용한 방법이며, 정액의 보존 일은 여러 가지 정자 기능과 SCSA 결과에 크게 영향을 주는 것으로 나타났다. 정액의 수정 능력을 평가함에 있어서 정액의 보존 일령을 고려하고 정자 기능 검사와 SCSA를 병행하여 실시할 경우 종모돈의 수정 능력 평가 시 정확성을 더욱 높일 수 있는 유용한 방법이 될 수 있을 것으로 사료된다.

  • In this paper, we established the general comparison principles for IVP of impulsive differential equations with time variables, which strictly extend and improve the precious comparison results obtained by V. Lakes. et.al . and S.K.Kaul([3]-[7]). Whit the general comparison results, we constructed the monotone iterative sequences of solution for IVP of such equations which converges the maximal and minimal and minimal solutions , respectively.

정액종류 및 배양조건에 따른 체외수정란의 생산 및 동결보존의 효율에 미치는 영향 (Effect of Semen Sources and Culture System on Efficiency of IVP Embryo Production and Cryopreservation)

  • The objective of this study was to optimize the selection of sperm sources, optimal culture systems and vitrification method depends on sperm sources. The oocytes were inseminated with either KPN 105, 114, 191, SNU 101, 102, 103 or epididymis and then embryos inseminated were cultured in oviductal cell co-culture or HECM-6 as defined me dium. The blastocysts produced were pooled according to sperm sources as KPN, SNU or epididymis and then vitrified by OPP vitrification method. The results obtained were as follows: 1. The cleavage(86.2 or 84.7%) and development rates to blastocyst (30.6 or 32.0%) were not significantly different between oviductal cell co-culture or HECM-6 culture systems(P<0.05). 2. To determine the optimal sperm sources for using IVF in this system, cleavage rates in KPN 191 and SNU 101 (74.2, 55.8%) were significantly lower rather than those in KPN 105, 114, SNU 102, 103 or epididymis (86.7, 85.1, 89.8, 85.5 or 81.2%), but development rates to blastocyst in KPN 114, SNU 103 or epididymis sperm (30.0, 33.0 or 28.6%) were significantly higher rater than those in KPN 105, 191, SNU 101, 102(21.4, 15.4, 14.9 or 25.4%), respectively (P<0.05). 3. The blastocysts produced were pooled according to sperm sources as KPN, SNU or epididymis and then vitrified by OPP vitrification method. The survival rates were not significantly different among sperm sources (89.6%: 43/48 ; 90.1%: 46/51 ; 83.3% : 20/24). These results obtained indicate that the defined medium, HECM-6, could be use to produce of IVP bovine embryos. Since the frozen semen must be required to maintain of unvariation data in IVP embryo production system, KPN 114 and SNU 103 produced in our laboratory were useful for this purpose. The blastocysts produced by different sperm sources as KPN, SNU or epididymis were vitrified by OPP vitrification method and survived very high rates. The OPP vitrification method could be susceptibility to use of IVP bovine blastocyst embryos.

  • This study was carried out to investigate the effect of biological factors on the in vitro production(IVP) of bovine oocytes for development of simple culture methods and medium. Oocytes from the slaughterhouse ovaries were matured and fertilized using general protocol and this study was examined if there were necessary to co-culture, media change, media type and embryo density. This results were as follows: 1. The development rate according to co-culture with cumulus cells and non co-culture as drop culture was not significantly different in cleavage (68.9 vs 71.7%), 8-cell stage (41.2 vs 44.1%) and blastocyst stage (12.2 vs 13.8%), respectively (p<0.05) 2. The blastocyst development rates in YS and CRIaa were higher than that in TCM199 (12.4, 10.4$ vs 3.7%), but the cleavage (69.0, 77.8 and 61.0%) and 8-cell stage (31.7, 37.0 and 35.7%) development accoring to YS, TCM199 and CRIaa ws not significantly different, respectively (p<0.05). 3. There was no significantly different in cleavage (62.6, 59.5 and 61.2%), 8-cell(34.7, 37.9 and 34.0%) and blastocyst (9.5, 11.6 and 12.8%) development among medium change time as control, Group I and Group II, respectively (p<0.05). 4. Blastocyst formation of 8-cell stage according to embryo density was not significantly different in 1, 10 and 25 embryos (27.3, 22.5 and 34.0%), respectively (p<0.05). These results indicated that simple culture system could reduce bovine IVP embryos as drop culture as non co-culture system, high density embryo (25 embryos/50 $\mu$1 drop). YS defined medium and no medium change for whole culture period, although other biological factors need to examine in order to produce efficient IVP bovine embryos.

  • 형상분리법을 이용하여 폐 PCBs로부터 금속성분을 회수하는 연구가 수행되었다. 기판에 붙어있는 모든 전자부품을 제거한 뒤 절단기 및 cutting mill을 사용하여 -3mm이하로 조쇄하였다. 이 조쇄물을 충격형 분쇄기로 분쇄하여 금속성분들을 단체 분리하였으며 햄머의 회전속도가 분쇄물의 특성에 미치는 영향을 조사하였다. 경사진동판을 사용하는 형상분리법을 이용하여 분쇄된 PCBs로부터 금속성분들을 분리 회수하는 실험을 행하였으며 경사진동판의 진동강도 및 경사각이 분리효율에 미치는 영향을 검토하였다. 경사진동판의 최적 조업조건은 진동간도, Kv=1.40 그리고 진동각, $ heta$=$10^{\circ}$이었다. 본 실험조건에서 회수율과 품위가 각각 90% 이상인 금속성분의 분리회수가 가능하였다.

Effect of Warming Temperature on the Viability of Bovine IVP Blastocyst Cryopreserved by Glass Micropipette (GMP) Vitrification

  • The purpose of this study was to investigate the warming temperature and exposed time on the post-thaw survival rate and viability of bovine blastocyst cryopreserved by GMP vitrification. Groups of three bovine IVP blastocysts were sequentially placed into vitrification solution before being loaded into the GMP straws and immersed into LN$_2$within 20 to 25 sec. The warming rate was increased 2 times of warming temperature for improvement of post-thaw survival rates. The frozen embryos were warmed either at 35 or 70$^{\circ}C$ for 1 or 2 sec and then diluted in sucrose solution. Post-thaw blastocysts were serially washed in 0.25 and 0.15 M sucrose in holding medium (HM: TCM199 supplemented with 10% FCS) and TCM-199 for each 5 min, respectively, and then cultured in TCM199 for 24 h. The rate of re-expanded blastocyst was significantly different fer 35 and 70$^{\circ}C$ warming temporature (76.4 vs. 89.3%; P<0.05). The rate of re-expanded blastocyst at 70$^{\circ}C$ for 1 sec was significantly higher than that for 2 sec (91.1 vs. 70.9%; P<0.05). The number of nuclei counted were significantly different among control, 35 and 70$^{\circ}C$ (121${\pm}$8.5 vs. 104${\pm}$11.7 vs. 114${\pm}$10.3; P<0.05). These results indicated that the increasing of warming rate can provide high survival rates of bovine IVP blastocysts. Especially, the best viability of post-thaw blastocyst could be thaw at 70$^{\circ}C$ for 1 sec. The warming temperature and exposed time far warming was considered to be limiting factors to the viability of bovine IVP embryos. he purpose of this study was to investigate the warming temperature and expose.
