Effect of Superoxide Dismutase and Cryoprotectants on Viability of Frozen-thawed Porcine Oocytes by Vitrification Method (Vitrification법에 의한 돼지 난자의 동결-응해 후 생존능력에 있어서 동해보호제와 Superoxide Dismutase의 영향)

  • This study was performed to investigate the effect of different cryoprotectants and superoxide dismutase(SOD) on viability of frozen-thawed oocytes by vitrification method in the pig. The proportions of oocytes matured to metaphase-I stage were higher in medium with ethylene glycol and DMSO(19.9%) than in medium with glycerol and DMSO(6.5%). When the oocytes were exposed in medium containing ethylene glycol, oocyte matured to prophase-I were not observed. On the other hand. significant differences were not observed between in medium with and without SOD( 1 unit/$m\ell$) during IVM of vitrified and thawed immature oocytes. However, the maturation rate from metaphase-I to metaphase-II were higher in medium with that than without SOD. The penetration rates after IVF of oocytes frozen-thawed were also higher in medium with that than without SOD. These results indicate that frozen-thawed oocytes treated with ethylene glycol and DMSO was more protective against freezing effect and that addition of 1 unit/$m\ell$ SOD in medium fur prevent of lipid peroxidation may play a positive role in improving of viability of frozen-thawed oocytes.

Effect of Bovine Follicular Fluid and Hormones on In Vitro Oocyte Fertilization and Development of Bovine Embryos (소의 난포액과 호르몬이 난포란의 체외수정 및 체외발달에 미치는 영향)

  • This study was carried out to determine the effect of bovine follicular fluid(bFF), hormones, and fetal bovine serum(FBS) supplemented in the medium on the in vitro fertilization and development of bovine embryos. The ovaries were obtained from a local abattoir and placed in physiological saline kept at 30~32˚C and brought to the laboratory within 3~4 hours. The oocytes and follicular fluid were collected by aspiration from visible follicles, and the oocytes of grades I on the basis of the morphology of cumulus cells attached and the homogeneity of cytoplasmic granules were selected and used for maturation. The basal media used for oocyte maturation, fertilization and embryo development in vitro were Ham' F-10, TALP and TCM-199, respectively. The hormones supplemented in maturation medium were consisted of 35 pg /ml FSH, 10 pg /ml LH and 1 pg/mi estradiol-l7$\beta$. The bFF collected from 5~9 mm follicles was centrifuged, filtered and inactivated by heat-treatment at 56˚C for 30 min. FBS also was inactivated with the same method and kept at -20˚C until use. The embryos were co-cultured with the monolayer of bovine oviductal epithelial cells at 39˚C under 5% $CO_2$ in air for 9 days. The results obtained were summarized as follows: The fertilization rate of oocytes was found 87.4% from 10% FBS and hormones treatment for IVM, and 37.1% of these TVF embryos were developed to blastocyst stage in 10% FBS groups. Compared with this control system, the fertilization rate was decreased significantly(P<0.05) in the maturation without either FBS or hormones. These IVF embryos were developed to morula stage at the similar rate, but to blastocyst at significantly(P<0.05) lower rate in the embryo culture with or without FBS supplementation. The fertilization rate(82.9%) in hormones and 10% inactivated bFF was similar with 10% FBS and hormone groups(87.4%), but decreased significantly(P<0.05) in 20 or 30% bFF (61.0 or 66.0%), respectively. In vitro developmental competence to blastocyst stage in 10% FBS and 20% inactivated bFF(37.1% and 31.4%) was higher than in 10 or 30% inactivated bFF(20.0 or 19.2%) or 10, 20 and 30% fresh bFF(19.1, 21.0 and 17.5%) The results indicated that the in vitro fertillzation and development rate of the embryos should be improved in 10% FBS or 20% inactivated culture system and 20% inactivated bFF might be available economically for bovine oocyte maturation and embryo culture instead of fetal bovine serum.

Use of a Xanthine-Xanthine Oxidase System on In Vitro Maturation and Fertilization in Pig

  • This study was undertaken to evaluate the effects of catalase using xanthine (X) - xanthine oxidase (XO) system on in vitro maturation and fertilization in pig. When follicular oocytes were cultured in maturation medium with X and/or XO, the maturation rates were not significantly different between in medium with and without catalase despite of different culture periods. However, significantly (P<0.05) higher maturation rates were obrained in culture with X-XO system. The rates of degenerated oocytes were increased with culture periods prolonged, and were significantly (P<0.05) higher in medium without than with catalase at 120 h of culture. On the other hand, the parthenogenetic oocytes were observed with high proportions at 72 h of culture, hut were not different in medium with and without catalase at various times of culture. In another experiment, the frozen-thawed boar spermatozoa treated with X-XO system for in vitro fertilization. The penetration rates were higher in medium with that than without catalase during the in vitro fertilization with, none (P<0.05), XO and X+XO. On the other hand, when sperm were treated with none, X, XO and X+XO, lipid peroxidation were higher in medium without that than with catalase. However, the changes in sperm penetration and lipid peroxidation showed opposite patterns. The sperm suspensions were also treated with X and/or XO for assay of sulfhydryl (-SH) group content. Under the above all conditions, sperm-SH group were higher detected In medium with that than without catalase. The activity of sperm binding to zona pellucida was also evaluated through binding to salt-stored porcine oocytes. In control group, sperm binding to zona pellucida were higher than in medium with X, XO and X+XO groups. No significant differences, however, were observed between medium with and without catalase. In conclusion, the exposure of follicular oocytes and spermatozoa to X-XO system may be caused stimulating in vitro maturation and fertilization in pig. Study on the Developmental Rate of In Vitro Cultured Cats Oocytes Recovered from Ovaries Collected at Different Stages of the Reproductive Cycle (번식주기의 단계별로 회수한 고양이 난자의 체외수정과 체외발생에 관한 연구)

  • The study was carried out to investigate the effects of morphology, reproductive cycle, incubation time and activation of oocytes in vitro maturation of cats oocytes and development of IVM/IVF embryos. The results were summarized as follows : 1. The fertilization and developmental rate of fresh and salts-stored oocytes with and whithout cumulus cells were 65.7%, 17.1% and 28.6%, 8.6% and 57.1%, 13.3%, 23.3%, 3.3%, respectively. The rate of oocytes with cumulus cells(13.3%∼65.7%) was higher than that of denuded oocytes(3.3%∼28.6%). 2. The fertilization and developmental rate of oocytes recovered from ovaries collected at different stages of the reproductive cycle were 68.9%, 44.4%, 48.9% and 17.8%, 8.9%, 12.8%, respectively. 3. The fertilization and developmental rate of oocytes in vitro cultured at different time of incubation(24, 36 and 48 h) were 66.7%, 46.7%, 48.9% and 17.8%, 11.1%, 8.5%, respectively. respectively. The rate of oocytes incubated 24 h(66.7%) was higher than that oocytes incubated 36 and 48 h(46.7%∼48.9%). 4. The fertilization and developmental rate of oocytes treated activation and non-activation oocytes were 57.4%, 31.4% and 22.9%, 11.4%, respectively. The rate of oocytes treated activation was higher than that oocyte treat non-activation.

  • The effects of in vitro maturation and sperm treatment condition on the in vitro fertilization (IVF) and developmental capacity of bovine oocytes were investigated and the development of embryos was compared under the 2 different co-culture system, with GC or BOEC. The cultured embryo to 16 cell or morula wre transferred into recipients or frozen by 2 different freezing method. The results obtained were summarized as follows; 1. In vitro maturation rates of vovine follicular oocytes cultrued in TCM199 with 10% FCS or ECS were 64.0% and 72.7%, but the case of addition of 10% FCS or ECS to TCM199 co-cultured with granulosa cells were 81.3% and 84.0%, respectively. IVM rate of three TCM199 added to granulosa cells was higher than that of media without granulosa cells. 2. When bovine follicular oocytes were matured in TCM199 with 10% FCS and GC and then fertilized in vitro by sperm treated with caffeine, embryo developments of bovine oocytes co-cultured with BOEC were 38.4% and 51.4%, respectively. But those of bovine oocytes co-cultured with GC were 52.2% by sperm treated with caffeine-heparin. 3. Cleavage rates of bovine oocytes cultured with 10% FCS alone and fertilized in vitro by sperm treated with caffeine-heparin was 33.0%. 4. When bovine follicular oocytes were matured in TCM199 with 10% FCS and GC, embryo developments of bovine ooctyes co-cultured with BOEC of GC were 46.0% and 50.2%, respectively. 5. When bovine follicular oocytes were matured in TCM199 with 10% ECS and GC, embryo developments co-cultured with BOEC or GC were 45.2% and 51.4%, respectively. 6. When Korean Native cow's follicular oocytes matured in TCM199 with 10% FCS and GC, embryo developed co-cultured with BOEC or GC were 45.2% and 51.4%, respectively. 6. When Korean Native cow's follicular oocytes matured in TCM199 with 10% FCS and GC, embryo developments of the bovine oocyte co-cultured with BOEC and GC were 41.8% and 60.1%. But with FCS 10% those of the bovine oocytes co-cultured with BOEC and GC were 42.0% and 48.4%, respectively. 7. When Holstein's follicular oocytes were matured in TCM199 with 10% ECS and GC, embryo developments fo the bovine oocytes co-cultured with BOEC and GC were 50.0% and 57.7%, but with ECS 10% those of the bovine oocytes co-cultured with BOEC and GC were 52.2% and 56.5%, respectively. 8. The viability of frozen-thawed embryos ranged from 60~80% and those of frozen-thawed embryos from vitrification was lower than that from conventional metiod. 9. The selected fresh embryos were transferred nonsurgically to 7 recipients but did not result in pregnancy.

Effects of Morphology, Reproductive Cycle, Incubation Time and Activation of Oocytes on Developmental Rate of Embryos Fertilized in vitro (난자의 형태, 번식주기, 배양시간 및 활성화 처리가 개 난자의 체외수정후 발생에 미치는 영향에 관한 연구)

  • The study was carried out to investigate the effects of morphology, reproductive cycle, incubation time and activation of oocytes in vitro maturation of canine oocytes and development of canine IVM/IVF embryos. The results were summarized as follows: 1. The developmental rates to 16 cells of fresh, salts and 4$^{\circ}C$-stored oocytes with and without cumulus cells were 14.3%, 5.0% and 7.5%, 2.8% and 5.7%, 0.0%, respectively. The rate of oocytes with cumulus cells(5.7%~14.3%) was higher than that of denuded oocytes(0.0%~5.0%). 2. The developmental rate to If cells of in vitro cultured oocytes recovered from ovaries collected at different stages of the reproductive cycle were 0.0%, 10.7%, 1.5%, respectively. 3. The developmental rate to 16 cells of fresh oocytes with cumulus cell cultured for 24, 32 and 48 hrs in $CO_2$ incubator were 0.0%, 5.3%, 11.8%, respectively. The rate of oocytes cultured for 48 hrs was higher than that oocytes cultured for 24 and 32 hrs. 4. The development to If cells treated activation and non-activation oocytes were 15.0%, 6.7%, respectively. The rate of oocytes treated activation was higher than that oocyte treat non-activation.

Effect of Collective Methods on the Collection Efficiency, Blastocyst and Pregnancy Rate after IVP Embryo Transfer in Hanwoo (한우 난소의 채란방법이 회수율, 배발달율 및 수태율에 미치는 영향)

  • This study was undertaken to access the effect of collection methods on the collection efficiency, blastocyst rate and pregnancy rate after IVP embryo transfer. The ovaries of Hanwoo were obtained from an abattoir and kept on 25 to $28^{\circ}C$ and transported to laboratory within 4 hrs. The oocytes were collected by aspiration of follicles $(2\~6\;mm)$ with or without slicing of ovaries after aspiration. The oocytes were matured in vitro (IVM) for 20 to 24 hrs in TCM-199 supplemented with $10\%$ fetal bovine serum at $39^{\circ}C$ under $5\%\;CO_{2}$ in air. Following routine IVM/IVF procedure, the oocytes and presumed zygotes were cultured for three day in CRlaa medium with BSA. The cumulus cells at 2 to 8-cell stage of embryos removed then the embryos and were cultured in CRlaa medium containing $10\%$ fetal bovine serum in $5\%\;CO_{2}$ at $39^{\circ}C$. The fresh blastocysts cultured for 7 to 9 days were transferred into recipients. The numbers of oocytes recovered form two different methods, the aspiration and slicing after aspiration, were compared to know what. The number of oocytes per ovary was 8.2 and 6.5 in aspiration combining slicing, and aspiration groups, respectively (p<0.05). The cleavage rate in aspiration method are significantly (p<0.05) high than those in slicing post aspiration $(27.9\%)$, and aspiration $(25.5\%)$. The pregnancy .ate in aspiration method $(62.5\%)$ was high than that in slicing method after aspiration $(54.4\%)$. The pregnancy rates of aspiration method and slicing method after aspiration in nullipara $(58.1\%\;vs\;68.2\%)$ was high than that in pluripara $(49.5\%\;vs\;53.2\%)$. The results obtained that the increased number of oocytes per ovary in slicing method after aspiration could be better than that in aspiration method. Pregnancy rate in aspiration method was slightly higher in than that in slicing method after aspiration.

Effects of Culture Media and Oxygen Concentration on In Vitro Development of Porcine IVM/IVE Embryos (배양액 및 산소농도가 돼지 체외수정란의 발달에 미치는 영향)

  • During in vitro culture of mammalian oocytes and embryos, the cells are exposed to the risks that cause cell injury or death. Numerous studies have been reported that the cell injury may be induced by the action of free radicals generated by auto-oxidation. This study was undertaken to investigate the optimal culture condition system for in vitro culture of porcine embryos. We first evaluated the effect of culture media on the porcine embryo development. NCSU-23 and PZM-5, culture medium tested, were failed to produce significant difference on the rate of blastocyst formation. In NCSU-23, the developmental rate was slightly higher than that in PZM-5. During in vitro maturation (IVM), fertilizaton (IVF), and culture (IVC) under 5 or 20% oxygen ($O_2$), the rates of cleavage and development were insignificantly different from each other under our culture condition (20% $O_2$, in NCSU-23), the mean cell number per blastocyst was $40{\pm}10$. These results showed that medium and $O_2$ concentration had no significant effect on the development of porcine embryos.

Effects of Development and Viability of Pig Oocytes Matured in Defined Medium Containing PVA, PVP and pFF (PVA, PVP 및 pFF를 첨가한 체외성숙 한정배지가 미성숙 돼지 난포란의 성숙과 배발달에 미치는 영향)

  • This study was conducted to develop a serum-free, defined medium of IVM of pig oocytes. The TCM-199 with supplemented with polyvinylalcohol(PVA), polyvinylpyrrollidone(PVP) and porcine follicular fluid(pFF) were used as basal medium. The effects of the these additives on the rates of maturity and development under in-vitro fertilization and in vitro culture were examined and subsequently considered on the possibilities be sustituted for the bovine serum albumin(BSA). Maturation rate of pig oocytes in IVM media containing PVA(82.4%), pFF(89.4%) and BSA(90.0%) were significantly higher(P<0.05) than that of PVP(78.6%). Cleavage rate after IVF of PVP(64%) was significantly lower(P<0.05) than these of PVA(73%), pFF(77%) and BSA(73%) supplements. in vitro development rates to morulae and blastocyst on PVP(54%) were also significantly lower(P<0.05) than these of the supplements of PVA(63%), pFF(69%) and BSA(65%). In comparison of maturation and fertilization rates of pig oocytes in each supplements, the maturity rates of PVA(82.4%), pFF(89.4%) and BSA(90.0%) were significantly lower(P<0.05) than that of PVP(72.4%) and while, the fertilization rates of pFF(87.1%) and BSA(89.1%) were significantly higher(P<0.05) than these of PVA(78.0%) and PVP(70.6%). It may be concluded that PVA and pFF can be substituted far BSA in medium for culturing pig oocytes; however, it may be considered that PVP were limited to for BSA in the in vitro culture of the embryos.

Effects of Equilibration and Dilution Methods on the Survival of Vitrified Bovine IVE Embryos (동결액의 평형방법과 희석방법이 초자화 동결된 소 체외수정란의 생존성에 미치는 영향)

  • This study was conducted to investigate the effects of equilibration and dilution methods on the survival rate of vitrified IVM-IVF bovine blastocysts. Vitrification solution was composed with 20% glycerol, 20% ethylene glycol, 3/8 M sucrose and 3/8 M dextrose in D-PBS supplemented with 20% FBS (GESD). Embryos were equilibrated in 1 of 3 methods: 3-step (El), 2-step (E2), or 1-step (E3), and after loading into 0.25-ml straws, were plunged into liquid nitrogen. After warming in water bath at 2$0^{\circ}C$, cryoprotectants were diluted in 1 of 3 methods: 1) D1(VS+1/2 M sucrose, 1/2 M sucrose and l/4 M sucrose), 2) D2 (1/2 M sucrose and 1/4 M sucrose), or 3) D3(1/2 M sucrose only). All procedures except warming were conducted at room temperature. Survival and hatching rates of blastocysts and expanded blastocysts following equilibration methods were 50 and 83.6%, and 27.8 and 67.3%, respectively in El, which were significantly higher (P〈0.01) than those of E2 (16.7 and 23.2%, and 7.4 and 12.5%, respectively) and 23 (0 and 3.7%, and 0 and 0%, respectively). Survival and hatching rates of expanded blastocysts were significantly (P〈0.01) higher than those of blastocysts in El. Survival rates of blastocysts and expanded blastocysts following dilution methods were 52% and 80.6% in D2, which were significantly higher (P〈0.05) than those of D1 (29.6 and 48.3%) and D3 (47.2 and 63.8%). Hatching rates of blastocysts were similar in D1, D2 and D3, however in expanded blastocysts, that of D2(61.3%) was significantly higher (P〈0.01) than that of D1(34.5%). Survival rates of expanded blastocysts in D1 and D2, and hatching rates in D2 and D3 were significantly higher(P〈0.01) than those of blastocysts. These results indicate that the viability of vitrified blastocysts was improved by the several steps of equilibration, and by 2-steps dilution after warming, independently of their stage of development. The results also indicated that the expanded blastocysts are more profitable to vitrification than blastocysts.
