갑상선기능이 정상인 성인에서 골다공증과 갑상선호르몬의 관련성 (The association of Osteoporosis and Thyroid Hormone in euthyroid adults)

  • 본 연구는 일부 종합검진 수검자들을 대상으로 골다공증이 갑상선호르몬에 미치는 영향을 검토하고자 2012년 1월부터 12월까지 G시의 일개 종합병원 건강검진센터에서 종합건강검진을 받았던 20세 이상의 지역주민 1,117명(남자 636, 여자 481)을 분석대상으로 하였다. 연구결과에서 연령과 성별을 보정하였을 때, 갑상선자극호르몬(thyroid stimulating hormone)에 대한 평균값은 정상군(${\geq}-1g/cm^2$)이 $1.61{\pm}0.07{\mu}IU/m{\ell}$, 골감소증군(-1 >, ${\geq}-2.5g/cm^2$)이 $1.82{\pm}0.08{\mu}IU/m{\ell}$, 골다공증군(< $-2.5g/cm^2$)이 $3.14{\pm}0.27{\mu}IU/m{\ell}$로 T-score가 감소할수록 증가하였다(p<0.001). 또한 성별과 FBS를 보정하였을 때, 유리타이록신(free thyroxine)에 대한 평균값은 정상군이 $1.30{\pm}0.01ng/d{\ell}$, 골감소증군이 $1.22{\pm}0.01ng/d{\ell}$, 골다공증군이 $1.13{\pm}0.04ng/d{\ell}$로 T-score가 감소할수록 감소하였다(p<0.001). 결론적으로 갑상선기능이 정상인 성인에서 T-score의 감소는 갑상선 자극호르몬(thyroid stimulating hormone)를 증가시키고, 유리 타이록신(free thyroxine)를 감소시킨다.

  • Antigenic potential of recombinant human growth hormone (LBD-007), a newly developed drug for growth hormone deficiency, was investigated in mice and guinea pigs. 1. Mice showed production of antibodies against LBD-009 (1.5IU/kg) with aluminum hydroxide gel(alum) as an adjuvant, Judaged by the heterologous anaphylaxis (PCA) test using rats. On the other hand, antibodies against ovalbumin (OVA) inoculated with alum were definitely detected. 2. In the studies with guinea pigs, both the inoculation of LBD-009 (0.15IU/kg-1.5IU/kg) only and of LBD-009 with complete Freund's adjuvant (CFA) as an adjuvant did produce weak positive reactions in homologous passive cutaneous anaphylaxis (PCA). On the other hand, the inoculation of ovalbumin with complete Freund's adjuvant (CFA) produced positive reaction in PCA.

Effect of alcohol dehydrogenase 1C (ADH1C) genotype on vitamin A restriction and marbling in Korean native steers

  • Objective: This work was to find the correlation of alcohol dehydrogenase 1C (ADH1C) genotype with vitamin A reduction and carcass traits during the vitamin A restriction period. Methods: In study 1, 60 Korean native steers were fed a diet (890 IU/kg) with 8,000 IU and 0 IU of supplemental premix vitamin A/kg of dry matter (DM) for control and treatment group, respectively. The levels of serum vitamin A were analyzed through high preparative performance liquid chromatography, and the ADH1C genotype was analyzed based on polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP; 78.1% TT type, 21.9% TC type); however, CC type was not found. Then, the interaction between ADH1C and carcass traits on the vitamin A restriction was investigated in study 2. A total of 136 Korean native steers were fed a diet that included 930 IU/kg vitamin A of DM. Results: Serum vitamin A in treatment was reduced to 112.4 IU/dL in steers with TT type of ADH1C, while for steers with TC type the concentration of serum vitamin A was dropped to 79.5 IU/dL (p<0.1) in study 1. This showed that TC type had the potential to lower serum vitamin A concentration during vitamin A restriction compared to TT type. In study 2 we found that eye muscle area, marbling and carcass weight in Korean native steers with TC type were higher than in steers with TT type (p<0.05). Conclusion: The interaction between vitamin A restriction and TC type of ADH1C gene could have the potential of increasing the marbling in Korean native steers. These results indicated that steers with TC type of the ADH1C gene were more sensitive to the change of serum vitamin A than TT types. Furthermore, this finding has the potential to enable a higher marbling score under the condition of vitamin A restriction in Korean native steers.

  • The hematologic values and serum chemistry were examined healthy male 12 Pere David's Deer and 8 Wapiti at the Farmland Zoological Garden. In wapiti, the blood values collected from horn and jugular vein were compared. The results were as follows: 1. 태반성 성선자극호르몬(HCG) 처리에 의한 독가시치, Siganus canaliculatus의 배란유도 (Induced Ovulation in rabbitfish, siganus canaliculatus, with Human chorionic Gonadotropin (HCG))

  • Successful produciton of seedlings for marine fish species are highly influenced by the aquisiton of sufficient number of good-quality eggs on required time. human chorionic gonadotropin has been used to induce successful ovulation for the purpose. Rabbitfish, siganus canaliculatus, is inhabited along the coast of Cheju Province and treated valuably for raw fish and fish roasted with seasoning. Female wild-captured rabbitfish (314 to 279 g in body weigth) were injected into intraperitoneal space with human chorionic gonadotropin (HCG) of 5,00,1,000, and 1,500 IU/kg body weight during spawning season of July 14 (trial 1), and July 27 (trial 2). Test fish in trial 1 were injected two times, first on July 14 and second on July 21 with one weekinterval because of no ovulation symptoms, but just one time in trial 2. All females injected both 1,000 and 1,500 IU/kg body weight ovulated in all trials. Among 500 IU/kg injection treatments, just one female ovulated in trial 1 showing 25% of ovulation rate, but none ovulated in trial 2. Ovulation was not occurred in control goups during test period. Time to reach ovulation after injection was 172 to 270 hours in trial 1 and 77 to 132 hours in trial 2, showing shorter as spawning season approached. Generally, ovulation of smaller females were late than that of larger ones. Gonadosomatic index (GSI) and pseudo-gonadosomatic index (Pseudo-GSI) were ranged 22 to 33% and 19 to 28%, respectively. One female spawned 406,200 to 1,032,000 eggs, and obtainable eggs per 100g of body weight were calculated by 130,000 to 190,000. Spawning rates were comparatively high by 96.0 to 98.4%. Rates to reach embryo-formation and hatching were higher in 500 high by 96.0 to 98.4%. Rates to reach embryo-formation and hatching were higher in 500 and 1,000 IU/kg treatments than in 1,500 IU/kg treatment. The result of present study demontrates that HCG treatment during spawning season could become very useful for ovulation of rabbitfish and the suitable dosage was suggested as 1,000 IU/kg of body weight.

Administration of vitamin D3 by injection or drinking water alters serum 25-hydroxycholecalciferol concentrations of nursery pigs

  • Objective: Two experiments were conducted to evaluate vitamin $D_3$ administration to nursery pigs by injection or in drinking water on serum 25-hydroxycholecalciferol ($25-OHD_3$) concentrations. Methods: At weaning, 51 pigs (27 and 24 pigs in experiments 1 and 2, respectively) were allotted to vitamin $D_3$ treatments. Treatments in experiment 1 were: i) control (CON), no vitamin administration beyond that in the diet, ii) intramuscular (IM) injection of 40,000 IU of vitamin $D_3$ at weaning, and iii) water administration, 5,493 IU of vitamin $D_3/L$ drinking water for 14 d post-weaning. Treatments in experiment 2 were: i) control (CON), no vitamin administration, and ii) water administration, 92 IU of $d-{\alpha}-tocopherol$ and 5,493 IU of vitamin $D_3/L$ drinking water for 28 d post-weaning. The lightest 2 pigs within each pen were IM injected with an additional 1,000 IU of $d-{\alpha}-tocopherol$, 100,000 IU of retinyl palmitate, and 100,000 IU of vitamin $D_3$. Results: In both experiments, serum $25-OHD_3$ was changed after vitamin $D_3$ administration (p<0.05). In experiment 1, injection and water groups had greater values than CON group through d 35 and 21 post-administration, respectively (p<0.05). In experiment 2, serum values peaked at d 3 post-administration in the injection groups regardless of water treatments (p<0.05) whereas CON and water-only groups had peaks at d 14 and 28 post-administration, respectively (p<0.05). Even though the injection groups had greater serum $25-OHD_3$ concentrations than the non-injection groups through d 7 post-administration regardless of water treatments (p<0.05), the water-only group had greater values than the injection-only group from d 21 post-administration onward (p<0.05). Conclusion: Serum $25-OHD_3$ concentrations in pigs increased either by vitamin $D_3$ injection or drinking water administration. Although a single vitamin $D_3$ injection enhanced serum $25-OHD_3$ concentrations greater than water administration in the initial period post-administration, a continuous supply of vitamin $D_3$ via drinking water could maintain higher serum values than the single injection.

산업용배지 사용에 의한 영지버섯 균주 WK-003균사체 액체 배양으로부터 생산된 세포외 다당체의 간 보호 효과 (Hepatoprotective Effect of Exo-polysaccharide Produced from Submerged Mycelial Culture of Ganoderma lucidum WK-003 by Using Industrial Grade Medium)

  • 영지버섯(Ganoderma lucidum WK-003) 균사체의 액체 배양에 의하여 세포외 생물고분자 물질의 산업적 생산을 위한 최적 조건을 검토하였다. 세포외 고분자물질 생산을 위한 산업용배지의 최적 농도는 molasses 5%와 CSL 2.5%에서 균사체 생장(12.4 g/l)과 균체외 다당체 생산(3.6g/l)이 최대를 나타났으며, 5 l 발효조 배양시에는 균사체 생장이 합성배지(8.1g/l)일때가 산업용배지(7.3 g/l)보다 높게 나타났으나, 균체외 다당체 생산은 합성배지(7.2 g/l)보다 산업용 배지(11.2 g/l)일 때가 더 높게 나타났다. 간 보호 효과는 합성배지와 최적화된 산업용 배지에서 생산된 균체외 다당체를 $CCl_4$ 독성이 유발된 rat에 경구 투여하였을 때 주요 지표를 나타내는 GPT활성은 산업용배지에서 생산된 균체외 다당체 인 경우 704IU/L에서 356IU/L로 낮추었으며, 합성배지에서 생산된 균체외 다당체는 704IU/L에서 349IU/L로 낮춘 결과를 보였다. 따라서 합성배지와 산업용 배지에서 생산된 균체외 다당체의 간 보호 작용은 거의 차이가 없는 것으로 나타났다.

  • 한국재래산양(韓國在來山羊)의 번식형질(繁殖形質)을 조사(調査)하고, 아울러 70일령(日令)부터 190일령(日令)까지 매(每)15일(日) 간격(間隔)으로 혈청(血淸) 성(性) hormone의 농도변화(濃度變化)를 조사(調査)하였다. 한국재래산양(韓國在來山羊)의 성성숙일령(性成熟日令)과 체중(體重)은 183.6일(日)과 14.3 kg이었고. 발정시기(發情時期)와 발정지속기간(發情持續期間)은 20.3일(日)과 36.7시간(時間)이었으며, 임신기간(姙娠期間), 복당산자수(腹當産仔數) 및 생시체중(生時體重)은 각각(各各) 148.4일(日), 1.4두(頭) 및 1.8 kg이었다. 혈청(血淸) LH의 수준(水準)은 70일령(日令)에서 3.93mIU/ml로 최고치(最高値)였고, 일령(日令)이 증가(增加)할수록 감소(減少)되는 경향(傾向) 이어서 190일령(日令)에는 1.21mIU/ml로 최소치(最少値)였다. FSH는 모든 관찰시간(觀察時間)에서 1.25 mIU/ml 以下의 수준(水準)이었다. Prolactin은 85일령(日令)에 3.09 ng/ml로 최소치(最少値)였고, 175일령(日令)에 4.65 ng/ml로 최고치(最高値)를 나타냈다. Estradiol-$17{\beta}$의 수준(水準)은 일령(日令)이 증가(增加)할수록 증가(增加)되는 경향(傾向)이어서 190일령(日令)에서 7.95 pg/ml로 최고치(最高値)를 나타냈다. Progesterone은 모든 관찰시간(觀察時間)에서 1.0 ng/ml 이하(以下)로서 매우 낮은 수준(水準)을 유지(維持)하였다.

능성어, Epinephelus septemfasciatus의 번식생물학적 연구 I. HCG 처리에 의한 배란유도 (Reproductive Bioligy of the Sevenband Grouper, Epinephelus septemfasciatus I. The Effect of HCG on Ovulation Induction)

  • 이 실험은 능성어의 친어관리와 종묘생산을 위한 연구의 일환으로 HCG 처리에 의한 난의 성숙과 배란유도를 알아보고자 행하였다. 제주대학교 해양연구소와 한라수산에서 1996년 6월에서 8월에 걸쳐 300-400 g, 1,500-2,000 g, 2,500-4,000 g 되는 능성어를 대상으로 하여 HCG를 500-1,000 IU/kg 체중농도로 2-3회 처리하였다. 이들의 생식소 발달은 일반적인 paraffin 절편법에 의하여 haematoxylin과 eosin으로 비교염색하여 조사하였다. 체중 300-400 g, 1,500-2,000 g, 2,500-3,200 g 되는 능성어에 HCG를 500-1,000 IU/kg 체중농도로 2-3회 처리한 결과 생식소는 난원세포군, 20~$30\mu$m 내외의 어린 난모세포를 가지고 있었다. 그러나 체중 3,400-4,000 g 되는 능성어에 HCG 1,000 IU/kg 농도로 2회 처리한 결과 난경 ~$800\mu$m되는 건강한 난과 퇴행란이 배란되었다. 체중 4,000 g 개체에서 배란된 난수는 4,252개 였으며, 이 중에서 건강한 난은 20.8%, 퇴행란은 79.2%였다. 그리고 생식소내에는 배란 흔적과 함께 난경 ~$50\mu$m전후의 주변인기 난모세포, 20~$30\mu$m의 어린 난모세포들이 차지하고 있다.

닭의 혈청 Creatine phosphokinase (CPK) 총 활성 및 CPK isoenzymes분획에 관한 연구 (Studies on the Serum Total Creatine Phosphokinase (CPK) Activities and CPK Isosenzymes Fractions of Chicken)

  • Clinically healthy 66 female Maniker breed chicken(22 of 2-week-old : group A, 23 of 8-week-old: group B and 21 of 27-week-old: group C) and 66 female Manina breed chicken (21 of 2-week-old : group D, 22 of 8-week-old : group E and 23 of 27-week-old : group F) were examined to establish physiological basic data on serum total Creatine phosphokinase (CPK) activities and CPK isoenzymes fractions. The results obtained were summarized as follows; 1. Serum total CPK activities were $1,088{\pm}254.0\;IU\;/{\ell},$ $1,454{\pm}337.2\;IU\;/{\ell}$ and $1,440{\pm}526.3\;IU\;/{\ell}$ in gorup A, group B and group C of Maniker breed, respectively. Group B and group C showed higher values than that of group A (P<0.01) meaning high values nth aging. 2. Serum total CPK activities were $1,676{\pm}420.5\;IU\;/ {\ell},$ $1,007{\pm}283.1\;IU\;/{\ell}$ and $862{\pm}294.5\;IU/{\ell}$ in group D, group E and group F of Manina breed, respectively. Group D showed the highest value among groups (P<0.01) and Manina breed showed the low values of serum total CPK activities with aging. 3. Manina breed at 2-week-old and Maniker breed at 8-week-old and 27-week-old showed significant high values of total serum CPK activities in breed differance (P<0.01) 4. In the pattern of serum CPK isoenzyme fractions, group A and group B were high with decreasing order of $CK_3>CK_2>CK_1$ and group C was high with decreasing order of $CK_2> CK_3>CK_1$ Groihp D, E and F showed the same pattern with decreasing order of $CK_2>CK_3> CK_1.$ 5. Significance of CPK isoenzymes fractions in breed differance was only found at 8-week-old. $CK_1\;and \;CK_3$ in Maniker breed (P<0.05), and $CK_2$ in Manina breed were higher than that of the other breed (P < 0.01).
