Isolation and Identification of Rumen Fungus and Its Produced Conjugated Linoleic Acid (반추위에서 분리한 곰팡이의 Conjugated Linoleic Acid 생산과 ITS-1 영역의 염기서열 해석)

  • The aim of this study was to isolate and identify men fungus which produces conjugated linoleic acid. IS-13 fungus hydrogenated conjugated linoleic acid and trans-11 vaccenic acid within 12 hr after addition of linoleic acid. The homology of IS-13 rumen fungus was compared with internal transcribed spacer 1 region (ITS1)sequences of twenty three men fungi. The length of ITS1 region of IS-13 isolate was 218 bp. IS-13 isolate has the most similar sequence (98% matched) with Orpinomyces species according to maximum-likehood and distance matrix results. The result supported that IS-13 isolate belonged to Orpinomyces genus.

Phylogenetic Analysis of Dendropanax morbifera Using Nuclear Ribosomal DNA Internal Transcribed Spacer (ITS) Region Sequences (Internal transcribed spacer (ITS) region의 염기서열 분석에 의한 보길도산 황칠나무의 분자 계통학적 연구)

  • Dendropanax morbifera is an endemic tree species of Korea, it is restricted to the southern parts of Korea. The internal transcribed spacer (ITS) region of nuclear ribosomal DNA (nrDNA) for Dendropanax morbifera grown at Bogil-do, Korea was determined. We investigated the sequence-based phylogenetic relationships of plants related and clarified its taxonomical position. The determined sequences consisted of 689 residues. ITS1 was 222 bp long while ITS2 was 233 bp long. The 5.8S rDNA was 160 bp long. The ITS region sequences of the Dendropanax species included in this study were obtained from GenBank. Oreopanax polycephalus was used as the outgroup. A pairwise alignment was calculated using the Clustal X program. A phylogenetic tree was constructed by the neighbor-joining method using the Tree view program. Sequence similarities among species including D. morbifera Bogil-do isolate showed the range 92.6 to 99.7% in sequence-based phylogenetic analysis using total 615 base pairs of ITS1, 5.8S rDNA and ITS2. D. morbifera Bogil-do isolate exhibited the highest degree of relatedness to D. chevalieri, sharing 99.7% ITS region similarity. D. morbifera Bogil-do isolate also showed to D. trifidus, sharing 99.4% ITS region similarity.

  • In this study, nucleotide sequence structures of intergenic transcribed spacer (ITS) 1, complete 5.8S ribosomal RNA gene and ITS 2 region were analyzed to identify three Peruvian entomopathogenic fungal isolates. The isolates had highly conserved sequence region in 5.8S rRNA gene and unique sequences in ITS 1 and 2 region among them. 5.8S rRNA gene regions were highly conserved and showed high homoloies among tested isolates. In contrast, ITS region showed species-specific sequence region, resulting in inter-genus differencies. Scanning electron microscopic images of these isolates supported the result of ITS-based identification. From these result, Peruvian entomopathogenic fungal isolate J270, J278, were identified as Beauveria bassiana and J271 was identified as Lecanicillium attenuatum.

Taxonomic Review of the Umbelliferous genus Sium L. in Korea: Inferences based on Molecular Data (분자생물학적 자료에 의한 한국산 개발나물속의 분류학적 고찰)

  • The taxonomy of umbel genus Sium L., Apiaceae in Korea was reviewed on the basis of molecular phylogenies derived from sequences of nuclear ribosomal DNA internal transcribed spacer (ITS) regions. The ITS sequence-derived phylogeny indicates that S. heterophyllum, endemic to Korea, is identical to S. tenue, which is known as endemic to Ussuri regions. Comparisons of sequence pairs across both spacer regions gave divergence values and revealed the identity between S. tenue and S. heterophyllum on Mt. Moonsoo. On the other hand, the ITS sequences support species delimitation of S. ternifolium, as reported recently as a new species that differs from other Sium species. The ITS sequence divergence values of 1.4 and 1.6% support species delimitation between S. serra and S. ternifolium.

Molecular Phylogeny of the Amynthas-complex (Oligochaeta: Megascolecidae) Inferred from ITS Nucleotide Sequences (Ribosomal DNA ITS 유전자를 이용한 왕지렁이(빈모강: 지렁이과) 그룹의 계통분류)

  • Phylogeny of the species mainly from the genus Amynthas in family Megascolecidae was inferred at the molecular level using ITS regions in rDNA. With 26 species of earthworms from 10 genera in 2 families, a stretch comprising the 3'-end of the 18S rRNA, ITS1, 5.8S rRNA, ITS2, and 5' end of 28S rRNA was amplified by applying the primers ITS-1, ITS-2. Phylogenetic analyses of nucleotide sequences with a help of MP, NJ, and QP yielded 5 groups similarly. Genus Amynthas was separated largely into two groups, Korean and Philippine origins. Species grouped into the 1st were Amynthas jirensis, A. agrestis, A. gucheonensis, A. sopaikensis, A. bubonis, A. multimaculatus, A. koreanus, A. dageletensis, A. heteropodus, A. odaesanensis, Pontoscolex sp., Pheretima sp. 1, and Dendropheretima banahawensis. Amynthas halconensis, A. isarogensis, A. mindrooensis, Pithemera sp. 2, Pithmera sp. 1, and Pleionogaster sp. clustered into one clade forming the 2nd group. Polypheretima sp. 1 and polypheretima. sp. 2 stayed closely together representing a separate monophyletic status, forming the 3rd group, apart from species in other genera. Archipheretima sp. falls into the 4th group. Distinct morphological characteristics from Archipheretima also coinsides with its branching away from others in the previously reported molecular analyses. Similar to Perionyx excavatus that has been selected as an outgroup, Aporrectodea tuberculata also showed a long branch in the phylogram, but it differed from other 24 species included in the analyses. Unlike others, for example, its habitat is very closely related to that of man.

PCR Detection and Sequence Analysis of the rDNA ITS Regions of Rhizina undulata (Rhizina undulata rDNA ITS 영역의 PCR 검정 및 염기배열 분석)

  • To investigate genetic diversity and PCR detection of Rhizina undulata, PCR detection and sequence analysis of rDNA ITS region of R. undulata in soil were analyzed and developed. The length of partial 18S rDNA from four R. undulata isolates were 1,375 nt. The sequence similarity of R. undulata isolates was 100%. The rDNA ITS regions of R. undulata isolates were 585 nt long. Nucleotide sequencing of the ITS regions showed that PDK-1, PTT-1 and PDJ-9 isolates had 100% sequence identity. But, PDS-5 isolate differed from the three isolates by two nucleotide substitution. R. undulata-specific primers designed by the sequence of ITS region were used in PCR detection of R. undulata. PCR products about 525 bp size, which is specific to R. undulata, were amplified from total DNAs of R. undulata isolates. To assay the sensitivity of PCR detection by R. undulata ITS-specific primer, purely cultured mycelial suspension of R. undulata was serially diluted and mixed with 100g of sterile sandy loam soil, respectively. And then, PCR products of total DNAs extracted from each mycelium-soil mixtures were analysed. The PCR protocol could detected up to 1ng mycelium of R. undulata within 100g of soil.

  • After the 1884 Home Insurance building in Chicago and 1889 Tower building in New York, 15 Park Row became the tallest building in the USA in 1899 and it held this record for nine years. Completed just before the arrival of the 20th century, this building deserves to be recognized for the sophistication of its architecture with respect to contextual understanding of the 19th century historic texture in Lower Manhattan, its uniquely economical floor plans, and its use of natural ventilation and daylight. The compositional techniques using the proportional analysis of its context reflect the highest level of architectural education of its time by its architect Robert H. Robinson. In addition, it also offers a unique glimpse to the state-of-the-art technology of its time regarding its structure, its use of movement systems that was capable of serving 4000 users.
