Colletotrichum fioriniae에 의한 호박 과실 탄저병의 발생 보고 (First Report of Colletotrichum fioriniae Causing Anthracnose on Fruit of Pumpkin (Cucurbita moschata) in Korea)

  • 강원도 강릉시 사천면 비닐하우스에서 재배중인 호박 과실에 탄저병이 발생하였다. 병든 호박 과실에 분홍색의 분생포자 층이 동심원으로 나타나 점차 확대되어 과실이 무르는 증상을 나타내었다. 원인균을 규명하기 위하여 순수 분리 후 균학적 특성 및 ITS, GAPDH, CHS-1, HIS3, ACT, TUB2 염기서열 분석결과 Colletotrichum fioriniae로 동정하였다. 또한, 병원성이 확인되었고 접종시험에서 동일한 균이 반복적으로 분리되었다. 따라서, 이러한 결과를 바탕으로 C. fioriniae에 의한 호박 과실에 발생하는 탄저병의 발생을 국내 처음으로 보고한다.

대전 계족산과 충남 오서산 및 전북 백암산 주위 야생화들로부터 효모의 분리 및 동정 (Isolation and Identification of Yeasts from Wild Flowers in Gyejoksan, Oseosan and Beakamsan of Korea)

  • 국내 자연환경의 효모 균총을 확인함으로써 유용한 효모자원을 확보하기 위한 연구의 일환으로 대전 계족산, 충남 오서산 그리고 전북 백암산 지역에 서식하는 야생화들을 채집하여 이들로부터 효모들을 분리, 동정하였다. 대전의 계족산에서 수집한 야생화로부터는 10종에 속하는 12균주의 효모들을 분리하였고, 충남의 오서산에서 수집한 야생화로부터는 10종의 효모들 17균주를 분리하였다. 또한 전북 백암산의 야생화로부터 모두 24종에 속하는 38균주의 효모들을 분리하였다. 계족산, 오서산, 그리고 백암산에서 모두 발견된 효모는 Cryptococcus 속, Pseudozygoma 속, Rhodotorula 속, Sporobolomyces 속에 속하는 9종의 효모들이었다. 특히 3개 지역에서 수집한 야생화들로부터 Cryptococcus 속에 속하는 Cryptococcus aureus만이 모두 분리되었는데, 이 결과는 각 지역마다 독특한 효모 다양성을 보이고 있음을 시사하는 것이다.

  • Sequence analysis of a DNA region encompassing the site of hybridization to actl, the gene for type II minimal polyketide synthase (PKS) for actinorhodin biosynthesis, from Streptomyces ablus revealed three more complete open reading frames additional to the already found two genes, plausibly encoding ${\beta}-ketoacyl$ synthase/acyl transferase (KS/AT) and chain length determining factor (ClF). The open reading frames (ORFs) were named salA, salD, and salE, from the upstream. In the homology analysis of the deduced amino acid sequences, SalA resembles the Streptomyces glaucescens Tcml, decaketide cyclase, SalD resembles acyl carrier protein in type II PKS, and SalE resembles the Actlll ketoreductase, The whole 4.4 kb of DNA sequence obeys the same conservation pattern as other type II PKSs. Therefore, we suggest that the 4.4 kb DNA from Streptomyces albus encompasses genes encoding enzymes for polyketide biogenesis in the organism and its organization is type II. The exsitence of SaIA, an analogue of the aromatic cyclase, revealed a relatedness of the 4.4 kb DNA with the aromatic PKS.

  • Saccharomyces cerevisiae HIS 5 유전자는 histidinol phosphate aminotransferase (EC: 2, 6, 1,9)를 code하는 아미노산 합성유전자이다. 이 유전자는 plasmid pSH 530에 cloning되어 E. coli와 Saccharomyces cerevisiae 숙주에서 promoter로서 전사하였다. HIS 5 유전자의 총염기 수는 736개이였고 5' 상류영역에는 긴 reading frame, directed repeat, 전사개시점, 그리고 Pribnow box염기배열이 있었다. 특히 HIS 5 유전자의 ATG 주변 염기배열은 -A-A-A-T-T-A-C-A-C-T-A-T-G-G-T-T-T-T-T-G-A-T-였으며 C block은 없었다.

두툽상어(Scyliorhinus torazame)Metallothionein cDNA의 cloning 및 이의 분자적 특성 (Molecular Cloning and Characterization of a Novel Metallothionein Isoform Expressed in Tiger Shark(Scyliorhinus torazame))

  • 두툽상어의 간조직 cDNA library의 EST를 통해 중금속의 세포내 농도 조절과 환경으로부터 흡수한 유해 중금속의 해독작용 등의 기능을 수행하는 MT 유전자를 cloning하였다. 염기서열 분석 결과 두툽상어의 MT 유전자는 204bp의 coding 영역과 182bp의 3'UTR 영역으로 구성되어 있었으며, 종결코돈 이후 162bp의 polyadenylatin 신호서열과 이로부터 15bp 이후의 poly(A)서열 등이 확인되었다. 염기서열로부터 유추한 68개의 아미노산 서열에는 다른 척추동물에서와 같이 Cys 잔기가 전체의 29.4%(20/68)로 풍부하였으며, 아미노산 서열수준에서 포유류와는 43~54%, 어류들과는 41~45%의 상동성을 나타냈었다. 특히 20개의 Cys은 어류와 18개가 다른 척추동물과는 19개가 잘 보존되어 있었다. 두툽상어 MT는 특이하게 모든 척추동물에서 잘 보존된 $\beta$-domain 끝 쪽의 9번째 Cys앞에 5개의 아미노산을 더 갖고 있었으며, 경골어류 MT의 특징인 4번째 위치의 gap이 없고, 18번째 Cys의 위치가 어류와 달리 다른 척추동물들과 같았다. 또한 C 말단의 아미노산 잔기가 다른 생물체와는 모두 다른 Ser을 갖는 특징을 나타내었다. 이와 같은 두툽상어 MT유전자의 특징들은 연골어류의 분자적 진화과정을 알 수 있는 분자 표지유전자로 이용할 수 있을 것이다.

cDNA Cloning and Expression of Human Rotavirus Outer Capsid Protein VP7 in Insect Cells

  • Rotavirus is a major cause of severe gastroenteritis in young children and animals throughout the world. The VP7 of rotavirus is thought to induce the synthesis of neutralizing antibodies and to be responsible for determining viral serotypes. The cDNA coding for the VP7 capsid protein of human rotavirus, obtained from Korean patients (HRV-Y14), was cloned and its nucleotide sequence was determined. Comparative analysis of the nucleotide sequences between VP7 of Y14 and that of other foreign isolates showed $92.7~95.2\%$ homology to G1 serotypes (RV-4, KU, K8, WA), $74.2\%$ homolgy to G2 serotype HU-5, $76.4\%$ homology to G3 serotype SA-11, and $77.6\%$ homology to G4 serotype A01321. These data suggest that HRV-Y14 can be classified as a G1 serotype. cDNA coding for VP7 of HRV-YI4 was subcloned into the baculovirus vector and the VP7 glycoprotein was expressed in insect cells. The expressed proteins in Sf9 cell extract and tissue culture fluid were separated on SDS-PAGE, and Western blot analysis with monoclonal antibody raised against the synthetic peptide containing 21 amino acids within the VP7 conserved region was performed. The molecular weight of recombinant VP7 was estimated to be 36 kDa which is about the same size as the native VP7. Addition of tunicamycin in the culture media caused a reduction of the molecular weight of the recombinant VP7 indicating that the expressed protein was glycosylated.

  • 진핵세포 유전자의 기초대사발현의 조절계를 밝히기 위한 일환으로, 효모의 histidine생합성계 효소의 구조유전자 HIS5를 이용하였다. HIS5 유전자는 충분한 아미노산 조건하에서는 발현이 억제되어 비교적 높은 기초발현만을 하나, 어떤 아미노산이 결핍되면 탈억제되어 높은 발현량을 보이며 탈억제는 cis의 작용인자인 promoter상의 5'-TGACTC-3' 및 trans 작용인자 GCN4와 GCD17 GCN2등이 관여한다. trans 작용인자들에 의한 HIS5 유전자의 발현량의 변화를 간단하게 측정하기 위하여, HIS5 promoter와 repressible acid phoshates(APase)의 구조유전자중 promoter를 제거한 DNA단편을 연결시켜 HIS5-PHO5 융합유전자를 이용하였다. gcn2 및 gcn4 변이주의 APase 활성은 야생주와 비교하여 3내지 4배 낮았으며, gcn2변이주와 gcn2 gcn4 이중변이주의 APase 활성은 유사하였다.

Five New Wood Decay Fungi (Polyporales and Hymenochaetales) in Korea

  • The wood decay fungi are a diverse taxonomic group that plays a pivotal role in forest carbon cycling. Wood decay fungi use various enzymatic pathways to digest dead or living wood in order to obtain carbon and other nutrients and these enzymatic systems have been exploited for both industrial and medical applications. Over 600 wood decay fungi species have been described in Korea; however, the recent application of molecular markers has dramatically altered the taxonomy of many of these wood decay fungi at both the genus and species levels. By combining molecular methods, specifically sequences of the internal transcribed spacer region, with traditional morphological characters, this study identified five new species records for Korea in five genera: Aurantiporus, Favolus, Neofavolus, Loweomyces, and Hymenochaetopsis. Three of these genera (Aurantiporus, Favolus, and Loweomyces) were previously unknown in Korea. The relatively simple morphology of the wood decay fungi often leads to ambiguous taxonomic assignment. Therefore, molecular markers are a necessary component of any taxonomic or evolutionary study of wood decay fungi. Our study highlights the need for a more robust and multifaceted approach in investigating new wood decay fungi in Korea.

First Record of Paramphistomes Fischoederius cobboldi and Paramphistomum epiclitum Detected in Bovine Rumen from a Local Market of Savannakhet Province, Lao PDR

  • In the present study, we report on the occurrence of paramphistomes, Fischoederius cobboldi and Paramphistomum epiclitum, in Lao PDR with the basis of molecular data. Parasite materials were collected from bovines bred in Ban Lahanam area, Savannakhet Province, Lao PDR at Lahanam public market. Morphological observations indicated 2 different species of paramphistomes. The mitochondrial gene cox1 of the specimens was successfully amplified by PCR and DNA sequencing was carried out for diagnosis of 11 specimens. Pairwise alignment of cox1 sequences were performed and confirmed F. cobboldi and P. epiclitum infecting bovines in Laos. Although there were many limiting points, as the small number of worm samples, and the restricted access of the animal host materials, we confirmed for the first time that 2 species of paramphistomes, F. cobboldi and P. epiclitum, are distributed in Lao PDR. More studies are needed to confirm the paramphistome species present in Savannakhet and its hosts to clear the natural history of these parasites of ruminants in the region and measure the impact of this parasite infection in the life and health of the local people.

Alternative Sigma Factor HrpL of Pectobacterium carotovorum 35 is Important for the Development of Soft-rot Symptoms

  • A bacterial artificial chromosome library of Pectobacterium carotovorum 35 was constructed to characterize the genome and to sequence its hrp region. The hrp cluster of P. carotovorum 35 consisted of 26 open reading frames in five operons. A promoter-based green fluorescent protein technology was used to identify the genes regulated by the alternative sigma factor, HrpL, in P. carotovorum 35. The majority of the selected clones contained the hrpJ operon promoter sequence, which harbors a hrp box, but no putative hrp boxes were detected within the promoter sequences of two other hrpL-regulated genes encoding for pectate lyase and large repetitive protein. Although the promoters of five other hrp operons also contained hrp boxes, their expression was not HrpL-dependent in the promoter-based selection in E. coli. However, transcriptional analysis showed that expression from all operons harboring hrp boxes, except for the hrpN operon, was reduced significantly in the hrpL mutant. The severity of soft-rot symptoms when the hrpL mutant was applied to the surface of tobacco leaves, mimicking natural infection, was greatly attenuated. These results indicate that the hrpL gene of P. carotovorum 35 may be involved in the development of soft-rot symptoms.
