Newly recorded chlorophytes, Monoraphidium subclavatum, Deuterostichococcus epilithicus, and Pseudostichococcus monallantoides in Korea

  • Monoraphidium subclavatum (FBCC-A409) of this study clustered in the same clade as Messastrum gracile strains in the 18S rDNA phylogeny. Compared to Messastrum gracile, Monoraphidium subclavatum did not form a colony, and the curvature of the cell was slightly curved or slightly crescent-shaped. This result means that the genus Monoraphidium is still based on the morphospecies concept, and was not monophyletic and not distinguishable as a separate genus. Two Stichococcus-like strains of this study (NIBRCL0000114567, NIBRCL0000114571) belong to Deuterostichococcus epilithicus and Pseudostichococcus monallantoides respectively in phylogenetic analysis using 18S rDNA sequences. These two species are consistent with recent research in the morphology and the genetic analyses using 18S and ITS rDNA sequences. We reported M. subclavatum, D. epilithicus, and P. monallantoides as newly recorded species in Korea.

Characterizations of five heterotrophic nanoflagellates newly recorded in Korea

  • Heterotrophic nanoflagellates (HNFs, 2-20 ㎛ in size) are substantially capable of controlling bacterial abundance in aquatic environments, and microbial taxonomists have studied ecologically important and abundant HNFs for a long time. However, the classifications of HNFs have rarely been reported in Korea on the basis of morphology and 18S rDNA sequencing. Here, previously reported five HNFs from non-Korean habitats were isolated from Korean coastal seawater or intertidal sediments for the first time. Light microscopic observations and 18S rDNA phylogenetic trees revealed that the five isolated species were Cafeteria burkhardae strain PH003, Cafeteria graefeae strain UL001, Aplanochytrium minuta (formerly Labyrinthuloides minuta) strain PH004, Neobodo curvifilus strain KM017 (formerly Procryptobia sorokini), and Ancyromonas micra (formerly Planomonas micra) strain IG005. Being morphologically and phylogenetically indistinct from its closest species, all isolates from Korea were therefore regarded as identical species detected in other countries. Thus, this result indicates an expansion of known habitats that range from those of the five isolates in natural ecosystems on Earth.

느티만가닥버섯 균주의 형태 및 유전적 유연관계 분석 (Analysis of Morphological and Genetic Relationships amomg Isolates of the Artificially Cultivated Mushroom, Hypsizygusmarmoreus)

  • Hypsizygus marmoreus의 형태적 특성과 유전적 유연관계를 조사하기 위해 한국 및 다른 국가에서 111개의 균주를 수집하였다. RAPD (Random amplified polymorphic DNA) 및 ITS rDNA 염기서열 분석을 이용하여 H. marmoreus 균주간의 유전적관계를 확인하였다. URP-PCR을 이용한 RAPD 분석결과, H. marmoreus의 모든 균주는 크게 3개의 그룹으로 분류되었으나, 90% 이상의 높은 유사성을 보였다. 또한 형태적 및 지리적으로 분류가 가능하였으나 갈색 균주 및 흰색 균주는 구별되지 않았다. 따라서 형태적, 지리적 차이가 있는 16개의 균주를 선별하여, 640 bp 길이의 ITS 염기서열을 정렬하고 비교하였다. ITS 염기서열의 유사성은 공시균주와 GenBank의 H. marmoreus 균주 사이에서 94.8-99.1 %였다. 수집지역과 형태에 차이가 있었지만, 실험에 사용된 공시균주는 모두 H. marmoreus에 가까운 유연관계를 형성하였다.

Multiplex PCR을 이용한 4 종류 목향(木香)의 감별 (Identification 4 kinds of Muxiang using Multiplex PCR)

  • Objectives : Aucklandiae Radix (Muxiang) one of important herbal medicines in oriental medicine, is defined as the dried root of Aucklandia lappa (Asteraceae). Owing to the similarities in the morphology and name, Inulae Radix (Tu-Muxiang) and Vladimiriae Radix (Chuan-Muxiang) as well as Aristolochiae Radix (Qing-Muxiang) originated from other medicinal plants are often used as substitutes and/or adulterants of Aucklandiae Radix. Therefore, a reliable authentication of these herbal medicines is necessarily for the public health and prevention of misuse. Methods : 32 samples of medicinal plants supplying Aucklandiae Radix, Inulae Radix, Vladimiriae Radix, and Aristolochiae Radix were collected in Korea and China. The ITS (Internal transcribed spacer) nucleotide sequences of samples were determined. The PCR primers to amply DNA marker of each herbal medicine were designed basing on the specific ITS regions showing differences in the sequences among medicinal plants. Results : Primer set Al R/IS F designed in this work amplified 220 bp PCR product only in samples of Aucklandiae Radix. In contrast, primer set Ih F/IS R, Vs R/IS F, and AcR F1/Ac R amplified 250 bp product, 356 bp prouct, and 516 bp product respectively to identify Inulae Radix, Vladimiriae Radix, and Aristolochiae Radix. Conclusions : The primers designed basing on the nucleotide sequences of ITS regions appearing differenced in the sequences among medicinal plants amplified the DNA markers for the identification of Aucklandiae Radix, Inulae Radix, Vladimiriae Radix, and Aristolochiae Radix. These herbal medicines were more efficiently identified by multiplex PCR method using all primers in a single PCR process.

Taxonomy of $Ganoderma$ $lucidum$ from Korea Based on rDNA and Partial ${\beta}$-Tubulin Gene Sequence Analysis

  • In the present study, a phylogenetic analysis was undertaken based on the internal transcribed spacer (ITS) rDNA and partial ${\beta}$-tubulin gene sequence of the $Ganoderma$ species. The size of the ITS rDNA regions from different $Ganoderma$ species varied from 625 to 673 bp, and those of the partial ${\beta}$-tubulin gene sequence were 419 bp. Based on the results, a phylogenetic tree was prepared which revealed that Korean $Ganoderma$ $lucidum$ strains belong in a single group along with a $G.$ $lucidum$ strain from Bangladesh.

PCR 다형성 분석에 의한 한국산 잣버섯의 유전적 다양성 및 유연관계 (Genetic Diversity and Phylogenetic Relationship in Korean Strains of Lentinus lepideus Based on PCR Polymorphism)

  • 잣버섯(Lentinus lepideus)은 외국에서 철도의 침목을 부후시켜 기차가 탈선되는 원인을 제공하는 곰팡이로 알려져 있으나 예로부터 맛과 영양이 좋고 약리효과도 높아서 야생에서 채취하여 이용해왔다. 최근 이 버섯의 새로운 품종과 인공 재배법이 개발되면서 한국산 잣버섯의 유전적 다양성과 유연관계의 규명이 필요하게 되었다. 이에 우리나라의 여러 지역에서 수집한 14개의 잣버섯 균주와 3개의 표고균주를 대상으로 rDNA의 ITS region 염기서열과 genomic DNA의 RAPD-PCR을 수행하였다. ITS1과 ITS2 영역의 염기의 수는 각각 173에서 179와 203에서 205 염기쌍으로 계통에 따라 변이가 있었는데 ITS1 영역의 염기서열이 ITS2의 영역보다변이가 많았고 5.8S 지역은 염기의 수가 156쌍으로 모든 균주가 동일하였다. 각각의 균주 간 유연관계를 알아보기 위해 ITS 영역의 염기서열을 이용하여 분지도를 작성해 본 결과 실험에 사용한 균주는 4개의 클러스터로 나눠지는 것으로 나타났고 실험에 사용한 3개의 표고는 잣버섯과는 다른 새로운 클러스터로 나눠졌다. 또한 20 종류의 primer를 이용하여 RAPD를 수행한 결과 10개의 primer가 효과적으로 염색체 DNA를 증폭하는 것으로 나타났다. 증폭의 양상은 잣버섯의 계통과 primer의 종류에 따라 변이가 있었는데, 증폭된 DNA의 단편 수는 적은 것은 5개에서 많은 것은 37개, 단편의 크기는 0.2 kb에서 2.6 kb까지 다양하였다. 본 실험 결과 실험에 사용한 한국산 잣버섯의 계통과 품종간의 유연관계는 높았으나 유전적 다양성은 낮았다.

PCR-Based Sensitive Detection of Wood-Decaying Fungus Phellinus linteus by Specific Primer from rDNA ITS Regions

  • Based on the rDNA ITS sequences data, specific primer set for PCR detection of wood-decaying fungus Phellinus linteus was designed. The length of PCR products using designed primer set(SHF and SHR) was about 540 bp. Among 11 species, 17 isolates of Phellinus spp. including Phellinus linteus, P. pomaceus, P. spiculosus, P. baumi, P. pini, P. igniarius, P. gilvus, P. biscuspidatus, P. weirii, P. johnsonianus, P. robutus, and P. igniarius, seven isolates of Phellinus linteus showed about 540 bp-sized single band. This molecular technique could offer a useful tool for detecting and identifying Phellinus linteus.

Identification of a Regulatory Region within the luxR Structural Gene in a Marine Symbiotic Bacterium, Vibrio fischeri

  • The light-organ symbiont of pine cone fish, Vibrio fischeri, senses its presence in the host and responds to environmental changes by differentially expressing its symbiosis-related luminescence genes. The V. fischeri luminescence genes are activated by LuxR protein in the presence of an autoinducer. In an effort to elucidate the mechanism of regulation of luxR, a plasmid containing luxR was mutagenized in vitro with hydroxylamine and a luxR mutant plasmid was isolated by its ability to activate luminescence genes cloned in E. coli in the absence of the autoinducer. The specific base change identified by DNA sequencing was only single base transition at +78 from the transcriptional start of luxR. Based on a Western immunoblot analysis, the nucleotide change directed the synthesis of much higher level of LuxR protein without any amino acid substitutions. The results suggest that the region including the +78th base is presumably internal operator required for autorepression of luxR, and the increased cellular level of LuxR results in activation of luminescence genes by autoinducer independent fashion.

First Record of Acrobeloides nanus (Cephalobidae: Rhabditida: Nematoda) from Korea

  • Acrobeloides nanus (de Man, 1880) Anderson, 1968 belonging to the family Cephalobidae Filpijev, 1934 (Cephalobomorpha) is newly reported from South Korea. This species is distinguished from other Acrobeloides species by its low and blunt labial probolae, five lateral incisures with middle incisure extending to the tail tip, and bluntly rounded tail. In this study, details of morphological characters of A. nanus is described and illustrated based on optical and scanning electron microscopy. In addition, molecular sequence data of the D2-D3 region of 28S rDNA, 18S rDNA and mitochondria DNA cox1 region from this species are provided as DNA barcode sequences.

A new species and a new record of Meghimatium Slugs (Pulmonata: Philomycidae) in Korea

  • Until now, five species (Meghimatium bilineatum, M. fruhstorferi, Limax flavus, L. marginatus, Deroceras reticulatum) in three genera of families Philomycidae and Limacidae have been reported in Korea. Philomycidae is a family of air-breathing land snails. Meghimatium hongdoensis sp. nov. is described based on its middle size (60-80 mm in body length), its body coloration (dark-red yellow), no dorsal with streaks, its genitalia, and 16rDNA sequence analysis. Specimens of this new species were collected from Hongdo island, Sinan-gun, Jeollanam-do, Korea. This species is only known from the type locality at Hongdo to date. It could be found in high-humidity sites. Meghimatium uniforme (Laidlaw 1937) was the first one reported in Korea. It was also collected from Gageodo island, Sinan-gun, Jeollanam-do, Korea. In this study, morphological characteristics including their radula and genital structures of these two species were described. Preliminary results of mitochondrial 16S rDNA sequencing and phylogenetic analyses indicated that these species belonged to the Korean clade.
