Effects of Microbial Phytase Supplementation to Diets with Low Non-Phytate Phosphorus Levels on the Performance and Bioavailability of Nutrients in Laying Hens

  • An 8 week feeding trial was conducted with 864 ISA Brown laying hens, 48 weeks old, to determine if microbial phytase $(Natuphos^{(R)})$ supplementation can reduce non-phytate phosphorus (NPP) level in laying diets. The experiment consisted of four dietary treatments: T1, control diet with 0.26% NPP (0.55% total P) wand no supplementary phytase; T2, 0.21% NPP (0.50% total P) diet with 250 U of phytase/kg of diet; T3, 0.16% NPP (0.45% total P) diet with 250 U of phytase/kg of diet; and T4, 0.11% NPP (0.40% total P) diet with 250 U of phytase/kg of diet. T3 showed the highest egg production and egg weight and the lowest feed conversion while T4 gave the lowest egg production and the highest feed conversion and mortality. Daily feed consumption ranged from 130.4 g (T4) to 132.7 g (T2). T1 and T2 were not significantly different in the production parameters. Eggshell strength, egg specific gravity, and eggshell thickness were not significantly different among treatments. However, broken egg ratio was significantly lower in T2 and T4 than in T1. Retentions of Ca, P, Mg, and Cu were greater in phytase supplemented treatments (T2, T3, and T4) than the control (T1), and those in T3 and T4 were greater than in T2. Excretions of P in phytase supplemented treatments (T2, T3, and T4) were significantly (p<0.05) smaller than in T1 but excretions of N were not significantly different among the treatments. Contents of ash in tibiae were not significantly affected by treatments, but contents of Ca, P, Mg, and Zn was increased and that of Cu decreased by phytase supplementation. It is concluded that the NPP concentration in the diet of Brown layers consuming about 130 g/d of feed can be safely lowered from 0.26% (0.55% total P) to 0.16% (0.45% total P). The excretion of P was reduced by the inclusion of 250 U phytase/kg of diet.

Effects of three different diets on the fatty acid profile and sensory properties of fresh Pecorino cheese "Primo Sale"

  • Objective: This study aimed to evaluate and compare the effects of three different diets on the fatty acids (FA) profile and sensory properties of a characteristic Italian fresh cheese: Pecorino "Primo Sale" (PS). Methods: Fifty-four sheep were divided into three feeding groups: total mixed ration (TMR) enriched with extruded linseed (TL), control diet with TMR without any integration (TC), and pasture (P). During cheese production, six cheeses per experimental group were produced each week, stored for 10 days at 4℃, and then analyzed for chemical composition, FA profile, and sensory properties. Results: Saturated fatty acids (SFA) were significantly higher in PS from group TC (82.11%) than in cheese from other two groups (P 75.48% and TL 66.83%). TL and P groups presented higher values of polyunsaturated fatty acids, 4.35 and 3.65%, respectively, than that of TC group (2.31%). The lowest SFA/unsaturated fatty acid ratio was found in TL and P groups, while the highest was found in the TC group. Vaccenic acid and conjugated linoleic acid (CLA) were higher in group P (p<0.05) than in groups L and TC. Sensory properties of cheese from group P received the highest scores for odor intensity and friability, while control group had a greater chewing consistency. Overall, all cheeses received good scores for acceptability. Conclusion: In conclusion, this study showed how the integration of extruded linseed improved the FA profile of fresh pecorino cheese PS preserving its sensory properties. Levels of CLA in the PS group achieved using this approach was not higher than that in a grazing diet. Cheeses from groups P and L contained a higher value of FA, with nutritional implications for humans, when compared with un-supplemented diet. Linseed may be a good feeding strategy when pasture is not available.

Distribution of HPV Genotypes in Cervical Cancer in Multiethnic Malaysia

  • Background: Cervical cancer is the third commonest type of cancer among women in Malaysia. Our aim was to determine the distribution of human papilloma virus (HPV) genotypes in cervical cancer in our multi-ethnic population. Materials and Methods: This was a multicentre study with a total of 280 cases of cervical cancer from 4 referral centres in Malaysia, studied using real-time polymerase chain reaction (qPCR) detection of 12 high risk-HPV genotypes. Results: Overall HPV was detected in 92.5% of cases, in 95.9% of squamous cell carcinomas and 84.3%of adenocarcinomas. The five most prevalent high-risk HPV genotypes were HPV 16 (68.2%), 18 (40%), 58 (10.7%), 33 (10.4%) and 52 (10.4%). Multiple HPV infections were more prevalent (55.7%) than single HPV infections (36.8%). The percentage of HPV positive cases in Chinese, Malays and Indians were 95.5%, 91.9% and 80.0%, respectively. HPV 16 and 18 genotypes were the commonest in all ethnic groups. We found that the percentage of HPV 16 infection was significantly higher in Chinese (75.9%) compared to Malays (63.7%) and Indians (52.0%) (p<0.05), while HPV 18 was significantly higher in Malays (52.6%) compared to Chinese (25.0%) and Indians (28%) (p<0.05). Meanwhile, HPV 33 (17.9%) and 52 (15.2%) were also more commonly detected in the Chinese (p<0.05). Conclusions: This study showed that the distribution of HPV genotype in Malaysia is similar to other Asian countries. Importantly, we found that different ethnic groups in Malaysia have different HPV genotype infection rates, which is a point to consider during the implementation of HPV vaccination.

Effects of Fermented Garlic Powder on Production Performance, Egg Quality, Blood Profiles and Fatty Acids Composition of Egg Yolk in Laying Hens

  • The effects of fermented garlic powder on production performance, egg quality, blood profiles and fatty acid composition of egg yolk in laying hens were studied in a 35-d experiment. Two-hundred and forty (ISA brown) layers aged 41 weeks were randomly allocated into the following four treatments: i) CON (basal diet); ii) G1 (CON+fermented garlic powder 1.0%); iii) G2 (CON+fermented garlic powder 2.0%) and iv) G3 (CON+fermented garlic powder 3.0%). There were no differences (p>0.05) among treatments in egg production, egg weight, eggshell breaking strength and eggshell thickness throughout the whole experimental period. However, yolk height was increased significantly (p<0.05) by the addition of fermented garlic powder during the 5th week while yolk color was greater (p<0.05) in G2 and G3 than in CON and G1 in the 5th week. Compared with CON, Haugh unit was increased (p<0.05) in response to fermented garlic powder treatments during the 5th week. No significant effects on total protein, albumin and IgG were observed in response to any of the treatments over the experimental period (p>0.05). There was a significant (p<0.05) reduction in plasma cholesterol concentration when the dietary level of fermented garlic powder was increased from 0.0 to 3.0%. The levels of saturated fatty acids (SFA) were significantly decreased (p<0.05) in response to G2 and G3 while monounsaturated fatty acids (MUFA) were higher (p<0.05) in G2 and G3 treatment groups than in CON and G1. Compared with other treatments, polyunsaturated fatty acids (PUFA) and PUFA:SFA ratio were higher (p<0.05) in G3. In conclusion, this study demonstrated that addition of fermented garlic powder reduced plasma cholesterol concentration and did not cause adverse effects on production performance. Moreover, addition of 3.0% garlic powder decreased SFA but increased PUFA and PUFA:SFA ratio in egg yolk.

Nutritional Values of Rice Bran and Effects of Its Dietary Supplementations on the Performances of Broiler Chickens (생미강의 영양적 가치와 사료 내 첨가가 육계 성적에 미치는 영향)

  • Two experiments were conducted to evaluate the nutritional values of rice bran and to examine effects of its dietary supplementation on broiler performances. In the first experiment, true metabolizable energy(TME), nitrogen corrected true metabolizable energy(TMEn), and true amino acid availability(TAAA) values of the rice bran were determined by force-feeding sixteen roosters(ISA-Brown). In the second experiment, 3-week-old male broiler chickens(Avian) were divided into four groups and fed each one of four experimental diets containing 0, 5, 10 or 15% rice bran for 21 days. TME and TMEn values of the rice bran(dry matter basis) were 3.25 kcal/g and 3.12 kcal/g, respectively, and the average TAAA value of the 16 amino acids was 76.21%. The average feed intake and body weight gain of the birds fed diets containing rice bran were apparently greater than those of the control group although the differences were not significant statistically. From the results, it can be concluded that feed formulation using bioavailability values, such as TMEn and TAAA, is an effective method for protecting the high variation in growth performances and that rice bran can be used for broiler feeds to 15% without any significant negative effects.

Effect of Betaine on Immune Response in Laying Hens (비태인이 산란계의 면역 반응에 미치는 영향)

  • This experiment was conducted to determine the effect of betaine on immune response in laying hens. A total of 72 ISA-brown laying hens were divided into four groups of 18 hens each and fed corn-soybean meal based diets with addition of 0, 300, 600 and 1,200 ppm betaine for four weeks. The effect of betaine on splenocyte proliferations with mitogens, concanavalin A(Con A) and pokeweed mitogen(PWM), were assayed after incubation using [3H] thymidine uptake. Proliferations of splenocyte were significantly increased by activation of mitogen Con A or PWM. Mitogen effects of Con A were increased by Con A plus betaine injection(0.1 mM), whereas PWM effects did not affect in PWM plus betaine injection(0.1 mM) in vitro. Splenocyte of laying hens fed betaine tended to proliferate in the presence of PWM, but appeared to be slightly suppressed in the presence of Con A in vivo. Proliferation of splenocytes which were stimulated by Con A or Con A+betaine injection(0.1 mM) were increased in dietary 600 ppm betaine, but inhibited in dietary 1,200 ppm betaine supplementation. Spleen weights and sheep red blood cell(SRBC) titers of hens fed betaine tended to increase compared to those of control, but were not significantly different. These results suggested that betaine could increase splenocyte proliferation in vitro.

Comparison of Spatio-temporal Variations of Phytoplankton Communities in Lakes in the Boseong River Basin (보성강 유역에 위치한 호수에서의 식물플랑크톤의 시공간적 군집 비교 분석)

  • In this study, we compared the spatio-temporal differences of phytoplankton communities among 4 lakes in the Boseong River basin. Field research was conducted quarterly from 2014 to 2017 for this study. A total of 345 species of phytoplankton were identified including 107 Bacillariophyceae, 175 Chlorophyceae, 27 Cyanophyceae and 36 other phytoplankton taxa. Lake Boseong showed higher species numbers and density of phytoplankton than other lakes (Dunn's test, P<0.01). Bacillariophyceae such as Asterionella formosa, Aulacoseira granulata, Fragilaria crotonensis was dominated in most research periods, whereas Scenedesmus ecornis and Coelastrum cambricum belonging to Chlorophyceae were dominant species on August. The self-organizing map (SOM) classified 3 clusters with 10 × 7 grid and showed spatio-temporal variation of phytoplankton communities based on significant difference among each clusters. Total 31 species of phytoplankton were chosen as a indicator species using indicator species analysis(ISA) and reflected seasonal phytoplankton succession and diversity and density of phytoplankton according to nutrient concentration. Water temperature, Secchi depth, conductivity and DO were identified as important factors affecting the differences of phytoplankton communities in the studied lakes in Boseong River basin using non-metric multidimensional scaling (NMDS).

Effects of Dietary Monascus Culture on Cholesteral Content of Egg Yolk, Meat and Serum of Laying, Hens (Monascus 배양물의 첨가급여가 산란계의 난황과 계육 및 혈청의 콜레스테롤 함량에 미치는 영향)

  • This experiment was carried out to study the effect of dietary Monascus culture on the cholesterol contents of egg yolk, muscle and serum of layers with 180 Isa-Brown laying hens for 10 weeks. Control group(C) was fed the commercial laying hen diet and 2.67(T1), 5.33(T2) and 8.00(T3)% of Monascus culture which contained 0.6% monacolin-k added to control diet so as to supply the monacolin-k 20(T1), 40(T2) and 60(T3) mg respectively, per hen-day with 125g diet. Hen-day egg production and average egg weight were not affected by the dietary Monascus culture, but feed intake and feed conversion per kg egg were significantly decreased(P<0.05) as the dietary Monascus culture increased. Cholesterol contents of egg yolk measured 4~5 weeks after feeding the Monascus culture and those of thigh meat measured at the end of experiment were significantly decreased(P<0.05) as the dietary Monascus culture increased. Average cholesterol contents of serum showed a trend to decrease as the dietary Monascus culture increased without significant difference.

Effect of Dietary Xanthophylls Supplementation on Pigmentation and Antioxidant Properties in the Egg Yolks (Xanthophylls의 급여가 계란 노른자의 착색 및 항산화성에 미치는 영향)

  • This study was carried out to investigate the effects of dietary xanthophylls supplementation on pigmentation and antioxidant properties in the egg yolk. ISA Brown laying hens aged 50 weeks were fed five kinds of xanthophyll combination such as control(neither natural nor artificial xanthophylls in feed), T1(Commercial diet containing natural xanthopylls in grain + Lutein 10ppm + Capsantin 10ppm), T2(T1 + Capsanthin 65ppm), T3(T1 + Canthaxanthin 65ppm), T4(T1 + Capsanthin 10ppm + Canthaxanthin 65ppm), and T5(T1 + Capsanthin 65ppm + Canthaxanthin 10ppm). The pH values of all egg yolks were not significantly different during storage or feeding periods. CIE L$^{*}$(lightness) values of T2 ~ T5 egg yolks were lower than those of control and T1. Conversely, the CIE a$^{*}$(redness) value of T2 ~ T5 egg yolks showed significantly higher(P<0.05). Egg yolk from chicks fed xanthophylls increased CIE b$^{*}$ values. The CIE b$^{*}$(yellowness) values of T2 ~ T5 egg yolks were higher than control and T1 during storage at 37$^{\circ}C$ for 48 hours respectively. In the antioxidation experiment, dietary xanthophylls supplementation affected lipid antioxidation of egg yolk during storage. The TBARS(O.D.) of egg yolks from chicks fed xanthophylls were lower than that of control during incubation at 37$^{\circ}C$ for 10 hours. In conclusion, dietary xanthophylls supplementation influence to color difference and retardation of lipid oxidation in egg yolk.

Evaluation of the Nutritive Value of Local Defatted Rice Bran and Effects of Its Dietary Supplementation on the Performance of Broiler Chicks (국내산 탈지미강의 영양적 가치 평가와 사료 내 첨가가 육계 생산성에 미치는 영향)

  • Two experiments were conducted to evaluate the nutritive values of defatted rice bran (DRB) and examine the effects of its dietary supplementation on broiler performances. In Experiment 1, to measure true metabolizable energy (TME), nitrogen corrected true metabolizable energy (TMEn), and true amino acid availability (TAAA), 30g of DRB sample was forced-fed to each of 16 ISA-Brown roosters followed by a 48h collection of excreta. The TME and TMEn values for DRB were 2.19 kcal/g and 2.05 kcal/g, respectively. The mean value of TAAA of the 15 amino acids in DRB was 77.29%. In Experiment 2, a total of 72 Avian broiler chicks were divided into 4 groups with 3 replicates of 6 birds per replicate, and fed one of the experimental diets containing 0, 5, 10 or 15% of DRB. Feed intake, weight gain, feed conversion rate, and body composition were measured for 3 weeks. Although there were no significant differences (P$\geq$0.05), body weight gain and feed intake of chickens fed a diet containing 15% DRB were slightly higher than those of the other groups. From these results, it can be concluded that feed formulation using TMEn and TAAA of DRB is an effective method for assuring feed quality and DRB can be supplemented to broiler rations up to 15% level.
