Effects of Auklandia Lappa on Dextran Sulfate Sodium-Induced Murine Colitis (당목향(唐木香)이 DSS(Dextran sulfate sodium)로 유발된 염증성 장질환 동물모델에 미치는 영향)

  • Objectives : Auklandia Lappa (ALE) is one of the herbs used frequently to treat abdominal pain and diarrhea and reported anti-inflammatory activity by suppressing proinflammatory cytokines. This study was designed to investigate whether ALE could show protective activities on experimental colitis induced by dextran sulfate sodium (DSS) models. Methods : Colitis was induced by DSS in Balb/c mice. ALE 10, 30, 100 and 300 mg/kg were orally administered twice a day for 7 days in DSS model. Mice weight was measured daily. Scoring of clinical findings was measured every other day. Colon length, edema, fecal blood and histological damages were assessed at day 7 in DSS model. In histological analysis, we checked cryptal glands, surface epithelium, submucosa, transmural, stroma and scored degree of inflammatory cell damage by modified histological scoring. We also calculated cytokines concentrations including IFN-${\gamma}$, TNF-${\alpha}$, IL-6, IL-$1{\beta}$, IL-17, IL-23, IL-10 and TGF-${\beta}1$ by Biometric Multiplex Cytokine Profiling method. Results : ALE showed the protective effects on DSS-induced experimental colitis. ALE inhibited shortening of colon length and histological damages of colon does-dependently, but it did not inhibit weight loss. ALE also inhibited IFN-${\gamma}$ and IL-6 expression, and upregulated cytokines (IL-10, TGF-${\beta}1$) related to regulatory T cell differentiation and proliferation. Conclusions : The current results demonstrate the clinical utility of ALE in traditional medicine and indicate the possibility of potent drug development of inflammatory bowel diseases from natural products. Further investigations for exact mechanisms will be needed.

The Inhibitory Effects of SoCheongRyongTang(SCRT) on Cytokine Production and Secretion of IgE in Highly Purified Mouse B cells (생쥐의 B 세포에서 IgE의 분비와 Cytokine 생산에 대한 소청룡탕(小靑龍湯)의 효과(效果))

  • Objective : We aimed to identify the inhibitory effects of SocheongRyoungTang on Cytokine Production and Secretion of IgE in Highly Purified Mouse B cells. This experiment was designed to investigate the effect of SoCheongRyongTang(SCRT) on Antiallergy. Materials and Methods : We measured the cytotoxic activity for cytokines transcript expression, production of $IL-1{\beta},\;IL-4,\;IL-6,\;IL-10,\;IFN-{\gamma},\;TNF-{\alpha},\;TGF-{\beta}$ proliferation of B cell in anti-CD40mAb plus rIL-4 plus HRF stimulated murine splenic B cells and histamine in anti-CD40mAb plus rIL-4 plus HRF stimulated mast cells. Results : 1. SCRT increased the gene synthesis of $IFN-{\gamma}(m-RNA)$, the appearance of IL-10, $IFN-{\gamma}$. 2. SCRT decreased the gene synthesis of $IL-l{\beta},\;IL-4,\;TGF-{\beta}(m-RNA)$ and the appearance of $IL-l{\beta},\;IL-4,\;TGF-{\beta},\;IgE$ significantly. Conclusion : SCRT decreased the proliferation of B cell significantly. According to the above results, it is suggested that SCRT extract might be usefully applied for prevention and treatment of Allergic disease.

Cytokine mRNA Expression in the Small Intestine of Weanling Pigs Fed Diets Supplemented with Specialized Protein or Peptide Sources

  • Cytokines play a central role in the mucosal immune response and are involved in regulation of nutrient absorption, metabolism and animal growth. This study investigated the effect of diet manipulation with specialized protein or peptide sources on expression of cytokine (IL-1, IL-6, IL-10, and TNF-${\alpha}$) mRNA abundance in different intestinal regions and at different ages post-weaning in piglets. A total of 48 (17 days of age, $6.16{\pm}0.34kg\;BW$) weanling pigs were fed either a corn-soy/whey protein basal diet, the basal diet supplemented with spray-dried plasma protein (SDPP), or the basal diet supplemented with $Peptiva^{(R)}$, a hydrolyzed marine plant protein. A fourth treatment group was fed the SDPP diet, but the feed intake level was limited (SDPP-LF). Pigs were killed at 3 and 10 d, and intestinal cytokine mRNA was measured by real-time PCR using the relative quantification method. The SDPP-LF group exhibited an increased TNF-${\alpha}$ mRNA abundance compared with the ad libitum SDPP group (p<0.05). The TNF-${\alpha}$ and IL-10 mRNA abundance increased from the proximal to distal part of the intestine, and the mRNA abundance was greater (p<0.01) in the distal intestine as compared with the proximal and middle intestine. The cytokines IL-1-${\beta}$, IL-10 and TNF-${\alpha}$ mRNA abundance also increased from d3 to d10 postweaning (p<0.01). In summary, restricted feeding increased the TNF-${\alpha}$ mRNA abundance in the small intestine, however neither SDPP nor peptide supplementation affected cytokine mRNA expression. Abundance of mRNA for most cytokines examined in this study increased with age post-weaning, suggesting that during 10 d after weaning the mucosal immune system is still under development.

IL-18 gene expression pattern in exogenously treated AML cells

  • IL-18 production may enhance immune system defense against KG-1 cells ; NB4 cells, which are associated with good prognosis, do not produce IL-18. In this study, we treated KG-1 cells with IL-18 and used microarray technology to assess subsequent effects on gene expression. In UniGene-array of 7488 human genes, expression of 57 genes, including stress related genes, increased at least 2-fold, whereas expression of 48 genes decreased at least 2-fold. Following exogenous exposure of KG-1 cells to IL-18, expression of CRYGC, $NF{\kappa}BIA$ and NACA gene were monitored. The latter is a transcriptional coactivator potentiating c-Jun-mediated transcription.$NF{\kappa}BIA$ is an inhibitor of $NF{\kappa}B$, and affects growth regulation, apoptosis and hypoxic stress. Studies, such as this one, are beginning to clarify the differences between cells associated with good and bad cancer prognoses, which may ultimately assist in medical treatment for acute myeloid leukemia.

A Critical Understanding of 'Juchegeonchuk' in North Korea in the Late 20th Century: Focused on The Art of Architecture (20세기 후반 북한의 '주체건축'에 대한 비평적 연구 - 『건축예술론』을 중심으로 -)

  • This research scrutinizes the concept of 'Juchegeonchuk' (self-reliance ideology in architecture), which is a key concept to understand the architecture and urbanism of North Korea in the 1970's and 80's. In the 1970's and 80's, Kim Jong-il began to appear in politics in earnest after Kim Il-sung appointed Kim Jong-il as Kim Il-sung's successor to the next generation. There have been many previous studies about 'Juchesasang' (self-reliance ideology) in various fields, except in architecture. On the other hand, the in-depth research into Juchegeonchuk, which was a method to strengthen Juchesasang, was not good enough in architecture. In this sense, this research investigates the concept of Juchegeonchuk based on the Geonchugyesullon (The Art of Architecture), which embodied the thought of architecture and culture of Kim Jong-il published in 1992. This research scrutinizes the idea of Juchegeonchuk on The Art of Architecture, and its representation in architecture and urbanism in Pyeongyang based on the idea of Juchegeonchuk. It enables to find a new interpretation of North Korea's architecture introduced by Kim, Jong-il in the late 20th century.

Anti-Inflammatory Effect of Essential Oils Extracted from Wood of Four Coniferous Tree Species (침엽수 4종 목부 정유의 항염증 효과 평가)

  • The aim of this study was to evaluate the anti-inflammatory effects of essential oils extracted from the wood of Chamaecyparis obtusa, Pinus densiflora, Pinus koraiensis, and Larix kaempferi. Essential oils were extracted by hydrodistillation, and their chemical components were determined by GC/MS. Major chemical components of these essential oils were ${\alpha}$-cadinol (19.25%), ${\tau}$-muurolol (14.20%), and ${\alpha}$-pinene (13.74%) in C. obtusa; ${\alpha}$-pinene (47.16%), longifolene (14.31%), ${\beta}$-phellandrene (11.78%), and ${\beta}$-pinene (11.02%) in P. densiflora; ${\alpha}$-pinene (13.49%) and longifolene (10.79%) in P. koraiensis, and geranyl linalool (23.58%) and ${\alpha}$-pinene (18.57%) in L. kaempferi. To evaluate the anti-inflammatory effects of essential oils, lipopolysaccharide (LPS)-induced RBL-2H3 mast cells were treated with these essential oils; then, the changes in the mRNA expression level of the cytokines IL-4 and IL-13 were examined. Further, degranulation was evaluated by measuring ${\beta}$-hexosaminidase release. After LPS-induced RBL-2H3 mast cells were exposed to $10^{-7}%$ of all types of essential oils, the gene expression levels of IL-4 and IL-13 within the cells remarkably decreased. The relative mRNA expression level of IL-4 was 69.6% in P. densiflora, 63.2% in P. koraiensis, 55.1% in C. obtusa, and 45.8% in L. kaempferi compared with that in the group treated with LPS. The mRNA expression level of L-13 should a similar trend. The inhibitory rate of IL-13 mRNA expression of P. densiflora, P. koraiensis, C. obtusa, and L. kaempferi was 57.8%, 57.1%, 51.1%, and 34.5%, respectively. ${\beta}$-Hexosaminidase release significantly decreased following the treatment with the four types of essential oils. The rate of ${\beta}$-hexosaminidase release were 38.1% C. obtusa; 33.0% P. densiflora; 27.4% P. koraiensis; and 9.1% L. kaempferi. Among all types of essential oils, that extracted from P. densiflora wood showed the highest anti-inflammatory activity. These results show that the tested essential oils exert an anti-inflammatory effect through the inhibition of degranulation and expression of cytokines.

Comparing the anti-inflammatory effect of nanoencapsulated lycopene and lycopene on RAW 264.7 macrophage cell line (RAW 264.7 대식세포주에서 나노입자화 리코펜의 항염증 증진 효과)

  • Purpose: We developed a method to load lycopene into maltodextrin and cyclodextrin in an attempt to overcome the poor bioavailability and improve the anti-inflammatory effect of this polyphenol. Methods: Nanosized lycopenes were encapsulated into biodegradable amphiphillic cyclodextrin and maltodextrin molecules prepared using a high pressure homogenizer at 15,000~25,000 psi. Cell damage was induced by lipopolysaccharides (LPS) in a mouse macrophage cell line, RAW 264.7. The cells were subjected to various doses of free lycopene (FL) and nanoencapsulated lycopene (NEL). RT-PCR was used to quantify the tumor necrosis factor (TNF-${\alpha}$), interleukin-$1{\beta}$ (IL-$1{\beta}$), IL-6, inducible nitric oxide synthase (iNOS), and cyclooxigenase-2 (COX-2) mRNA levels, while ELISA was used to determine the protein levels of TNF-${\alpha}$, IL-$1{\beta}$, and IL-6. Results: NEL significantly reduced the mRNA expression of IL-6 and IL-$1{\beta}$ at the highest dose, while not in cells treated with FL. In addition, NEL treatment caused a significant reduction in IL-6 and TNF-${\alpha}$ protein levels, compared to cells treated with a similar dose of FL. In addition, mRNA expression of iNOS and COX-2 enzyme in the activated macrophages was more efficiently suppressed by NEL than by FL. Conclusion: Overall, our results suggest that lycopene is a potential inflammation reducing agent and nanoencapsulation of lycopene can further improve its anti-inflammatory effect during tissue-damaging inflammatory conditions.

Effects of Saenghyeolbosu-Tang on the Morphometric Changes of Femur, and on the Factors Related with Bone Metabolism in Ovariectomized Rats (생혈보수탕(生血補髓湯)이 난소적출 흰쥐 대퇴골의 형태계측학적 변화 및 골대사 관련인자에 미치는 영향)

  • Purpose : This study was carried out to investigated the effects of Saenghyeolbosu-Tang on the morphometric changes of femur, and on the hormones and cytokines associated with bone metabolism in ovariectomized rats. Methods : Twenty-four female Sprague-Dawley rats were divided into sham operated group(normal), ovariectomized group(control), and treated with extract of Saenghyeolbosu-Tang group(treated). Each group was evaluated the changes of body weight at 0, 3, 6, 8, weeks after ovariectomy. Morphometric analysis(femur weight, femur/body weight, femur ash weight, femur ash/body weight, cross sectional area of compact bone and cancellous bone of femur) and histopathological examination were performed at 8 weeks after ovariectomy. Estrogen and cytokines($TNF-{\alpha},\;IL-1{\beta}$, IL-6) assay were performed at 8 weeks after ovariectomy. Results : 1. The femur weight(g) of treated group(1.59${\pm}$0.08) was significantly increased(p<0.05)compared with the control group(1.50${\pm}$0.07) at 8 weeks. 2. The femur/body weight(g) of treated group(5.27${\pm}$0.36) was significantly increased(p<0.05) compared with the control group(4.78${\pm}$0.37) at 8 weeks. 3. In the cross sectional area of cancellous bone of femur(${mm^2}$) the treated group(1.60${\pm}$0.20)was significantly increased(p<0.01) compared with the control group(1.19${\pm}$0.19) at 8 weeks. 4. The serum estrogen level(pg/ml) of treated(83.67${\pm}$27.44) and control group(800.49${\pm}$22.27)showed no significantly different changes at 8 weeks. 5. The serum $TNF-{\alpha}$ level(pg/ml) of treated(33.71${\pm}$6.43) and control group(41.35${\pm}$8.47) showed no significantly different changes at 8 weeks. 6. The serum $IL-1{\beta}$ level(pg/ml) of treated group(53.78${\pm}$10.83) was significantly decreased(p<0.001) compared with the control group(79.80${\pm}$11.40) at 8 weeks. 7. The serum IL-6 level(pg/ml) of treated group(55.52${\pm}$9.02) was significantly decreased(p<0.01) compared with the control group(69.69${\pm}$7.95) at 8 weeks. Conclusion : This study shows that Saenghyeolbosu-Tang inhibits bone resolution in ovariectomized rats. These results may be related to the inhibitory effect of Saenghyeolbosu-Tang on the secretion of $IL-1{\beta}$ and IL-6 in the Pathogenesis of osteoporosis.

The External Use Effects of Samwhangsejegami Extract on Atopic dermatitis of NC/Nga mice (삼황세제 가미방 외용이 NC/Nga 마우스의 아토피 피부염에 대한 효과)

  • Objectives : In this study, Samhwangsaejaegami extract was tested to prove its anti-atopic dermatitis effect on NC/Nga mice. Methods : In order to evaluate the external use effects of Samhwangsaejae extract on anti-atopic dermatitis, the expression of filaggrin, serine palmitoyltransferase(SPT), and COX-2 were analyzed. In vivo study, clinical skin severity score, IgE, IL-4, IL-5 and IL-6 level were analyzed through NC/Nga atopic mice model after 12 weeks external treatment. Results : In vitro study results showed the reduction in the expression of filaggrin, SPT, and COX-2. In vivo study results demonstrated the significant reduction in clinical skin severity score, IgE, IL-4, IL-5, IL-6 expression level. Conclusions : These results showed Samhwangsaejaegami extract can be a promising candidate for anti-atopic dermatitis treatment.
