${\gamma}{\delta}$ T Cells in the Peripheral Blood of Pulmonary Tuberculosis (폐결핵환자의 말초혈액에서의 ${\gamma}{\delta}$ T 림프구에 관한 연구)

  • Background: T cell mediated immunity is important in the defense mechanism of tuberculosis. Since ${\gamma}{\delta}$ T cell receptor was found to react with 65 kD heat shock protein of M.tuberculosis, there have been some reports on the role of ${\gamma}{\delta}$ T cells in the defense against M.tuberculosis. But until now, the role of the ${\gamma}{\delta}$ T cells in tuberculosis is not clear. Methods: We therefore measured the percentage of ${\gamma}{\delta}$ T cells of peripheral blood by flowcytometry before and after stimulation with Con-A, PPD and H37Ra lysate and compared between tuberculosis patients and healthy controls. Results: ${\gamma}{\delta}$ T cells of pheripheral blood in pulmonary tuberculosis patients was $7.5{\pm}5.2%$, showing no difference compared with healthy control($10.0{\pm}4.8%$). But IL-2R(+) ${\gamma}{\delta}$ T cells were higher in tuberculosis compared with healthy control($4.8{\pm}5.1%$ vs. $1.8{\pm}2.8%$). After stimulation with PPD or Con-A, the percentage of ${\gamma}{\delta}$ T cells showed no significant change, but IL-2R(+) ${\gamma}{\delta}$ T cells increased significantly in both tuberculosis($17.9{\pm}13.4%,\;57.6{\pm}20.2%$ respectively) and healthy control group($11.5{\pm}9.1%,\;80.8{\pm}9.3%$). After stimulation with H37Ra lysate, percentage of ${\gamma}{\delta}$ T cells showed increasing tendency in healthy control group, but it was not statistically significant. Conclusion: In this study, we could not demonstrate the role of ${\gamma}{\delta}$ T cells in the peripheral blood of pulmonary tuberculosis. It is suggested that further studies will be needed in the regional sites of M.tuberculosis infection.

  • Purpose : 자하거(Hominis Placenta; HP)는 건강한 사람의 태반을 홍제(烘製)하여 건조한 것으로 한의학에서는 기혈(氣血)을 대보(大補)하고 신정(腎精)을 보익(補益)시켜 구병(久病)으로 인한 신체허약(身體虛弱)이나 혹은 체질허약(體質虛弱)과 혈기부족(氣血不足) 및 신허정휴(腎虛精虧) 등 등(證)을 치료(治療)하는데 단미(單味) 또는 복방(複方)에 배오(配伍)하여 쓰여왔다. 또한 자하거는 면역학적으로 골대사 활성이 있는 것으로 알려져 있어 본 연구에서는 자하거의 항골다공증 활성을 분자세포생물학적으로 검정하고자 하였다. Methods : Osteoblast cells에서 자하거가 COX-2 mRNA의 발현과 $PGE_2$ 생합성을 억제시키는지를 관찰하기 위해 먼저 TNF-${\alpha}$, IL-${\beta}$ 와 IL-6를 처리한 후 $PGE_2$의 생합성과 더불어 COX-2 mRNA의 발현을 확인하였다. 그 후 TGF-${\beta}$, 자하거(紫河車)와 이 둘의 조합인 자하거+TGF-${\beta}$가 COX-2 mRNA 발현과 $PGE_2$ 생합성을 저해시키는지 관찰하였다. 또한 자하거가 IL-1${\beta}$로 유발된 흰쥐의 과칼슘혈증을 감소시키는지를 확인하였다. Results : IL-6, IL-1${\beta}$와 TNF-${\alpha}$를 동시에 처리하면 이것을 단독으로 처리한 것과 비교해 볼 때 $PGE_2$의 생합성과 더불어 COX-2 mRNA의 수치가 상승작용을 일으키며 증가하였다. TGF-${\beta}$, 자하거와 이 둘의 조합인 자하거+TGF-${\beta}$은 COX-2 mRNA 발현, $PGE_2$ 생합성 및 골재흡수를 감소시켰다. 자하거(紫河車)는 IL-1${\beta}$, TNF-${\alpha}$와 IL-6 각각 또는 이들의 조합으로 인해 증가하는 COX-2 mRNA 발현과 $PGE_2$ 생성을 감소시키는 반면 COX-1 mRNA 발현에는 유의성 있는 영향을 미치지 않았다. 한편 자하거는 농도의존적으로 IL-1${\beta}$로 유발된 흰쥐의 과칼슘혈증을 감소시켰다. 이러한 결과는 흰쥐의 두개골 골아세포에서 $PGE_2$ 생산에 대한 IL-${\beta}$, TNF-${\alpha}$, IL-6의 상승작용이 COX-2의 유전자 발현 증가에 기인함을 보여주었다. Conclusions : 이러한 결과들로부터 자하거가 골대사과정중 골재흡수를 억제하는데 효과적임을 밝히게 되었으며, 자하거의 골다공증의 억제기전이 골재흡수관련 단백질들의 전사조절에 있음을 최초로 해명하게 되었다.

The Anti-inflammatory and Analgesic Activities of Artemisia capillaris Thunberg (인진 추출물의 소염진통작용)

  • Inflammatory mediators such as interleukin-1 (IL-1), tumor necrosis $factor-{\alpha}\;(TNF-{\alpha}),\;interferon-{\gamma}\;(IFN-{\gamma})$ and lipopolysaccharide (LPS) are thought to play major roles in joint diseases such as a rheumatoid arthritis (RA), and there is considerable evidence playing a role for these cytokines in osteoarthritis (OA). Therefore, we have studied the effects on anti-inflammation and analgesic by ethyl acetate fraction from 70% ethanol extract of Artemisia capillaries (EAC). As a positive control, apigenin, which is known as an anti-inflammatory agent as an iNOS inhibitor, was used and showed the dose-dependent inhibitory effect. EAC showed strong inhibitory efficacy against cytokine-induced proteoglycan degradation, $PGE_2$ production, nitric oxide (NO) production, and matrix-matalloproteinases (MMPs) expression in rabbit articular chondrocyte. In the writhing test induced by acetic acid, EAC $(200{\sim}400\;mg/kg)$ exhibited a dose-dependent inhibition of writhing. The results indicate that EAC have anti-inflammatory and analgesic activities, and could be a good herbal medicine candidate for curing of RA and/or OA.

  • 과거에는 하천 범람으로 인한 홍수피해가 많았으나 최근에는 도시화로 인한 불투수면적의 증가로 홍수도달시간의 단축 및 노면수의 배수불량으로 인한 내수 홍수피해가 많아졌다. 이러한 변화는 도시하천의 홍수예보에 밀접한 관련이 있으며 관련된 분석 모형 및 연계방안 또한 매우 중요하게 되었다. 일반적으로 하천에 대한 유출해석 모형으로 HEC-RAS((Hydrologic Engineering Center-River Analysis System)가 주로 사용되고 있으나 현재와 같이 도심지 하천에서는 내배수의 특성을 고려한 SWMM(Storm Water Management Model)을 사용한다. 또는 이 두모형의 연계를 통해 유출해석을 진행하기도 한다. 최근 HEC-RAS와 SWMM모형이 최신 버전을 공개하였다. HEC-RAS의 경우 2016년 9월 5.0.3버전을 출시하며 1D뿐만 아닌 2D의 모의도 가능하도록 기능을 개선하였으며 SWMM의 경우 2016년 09월 07일 5.1.011버젼이 공개되었다. 본 연구에서는 공개된 최신 모형을 도림천 지역에 적용하여 도림천 지역에 적합한 모형 및 연계 방법을 찾아보려 한다. 이를 통해 최적의 도시홍수예보 시스템을 구성하기 위한 모형 및 연계방안의 조사와 가장 합리적인 도시홍수 시스템의 구성방안을 제시하고자 한다.

Suppressive Effect of CYTG on Collagen Induced Arthritis(CIA) in DBA1/J Mice (CYTG 처방이 콜라겐 유발 관절염 모델에 미치는 효과)

  • To evaluate effect of CYTG on inhibiting the occurrence of arthritis, we performed the experiments including production of inflammatory cytokine and immunoglobulin in collagen arthritis model. The results were obtained as fellows. CYTG group showed inhibitory effect on arthritis incidence than control group for four weeks. Arthritis index of CYTG group reduced compared with control group. In CYTG group, production of cytokines which show suppressive effect on inflammation(IL-2, COX-2) was increased and which promotes inflammation(IL-10) was decreases in spleen. In CYTG group, production of immunoglobulin (IgG-RF) was reduced compared with control, and rate of CD3+CD69+T cell is lower in lymph node and CD4+CD25+ T cell is higher in lymph node and spleen. And synovial infiltration in the knee were observed in the controls (PBS-treated mice), whereas CYTG-treated mice exhibited significantly reduced histologic evidence of destruction and inflammation. So, the histopathological scoring average of CYTG group was 2.5 compared with control group(CIA mice) 4.5. It was thought that our data express high effect via immune system specially through the controling the inflammatory cytokines and immunoglobulins. CYTG could be usefully applied for the prevention and treatment of RA, and also is expected to be clinically helpful on the treatment of RA through modification.

  • Corilagin (CRN) isolated from Euphorbia helioscopia as rheumatoid arthritis drug. CRN was medicated to the abdominal cavity of collagen induced arthritis (CIA) mice that was an animal model for rheumatoid arthritis and its effects on incidence and arthritis index were studied. The results were as folllows; It was exhibited that medicating corilagin inhibited the infiltration of activated T lymphocytes into an inflammatory joint. The production of IgG and IgM that were RF (rheumatoid factor) and inflammatory cytokine, IL-6 and $TNF-{\alpha}$were reduced. After measuring $IFN-{\gamma}$and IL-4, it was found that it was shifted into Th2 immune response as increasing in IL-4. After liver function test, studies on liver poisoning of AST/ALT should be continued.

Interrelationship between Cell Differentiation and Expression of mRNA for Transferrin in HL-60 Leukemia Cell Line

  • The interrelationship between the differentiation and expression of mRNA for transferrin in the HL-60 leukemia cell line was studied. Transferrin mRNA was expressed in HL-60 leukemia cells and the amount was 50% of that in the positive control cell line, HepG-2 cells. The expression of $T_f$ mRNA in HL-60 cells was not regulated by IL-1, IL-6 and $TNF-{\alpha}$, respectively. The expression of $T_f$ mRNA in the differentiated cells into a granulocyte lineage by DMSO, or all-trans RA, was up-regulated (160-170% of control cells); whereas, the expression was not regulated in the differentiated cells into a macrophage lineage by PMA. These results suggest that the differentiation to a granulocyte lineage of HL-60 leukemia cells appear to be related with the upregulation of transferrin mRNA expression.

The regulatory effect of AST cream on atopic dermatitis-like skin disease.

  • In this study, we investigated an inhibitory effect of AST cream on atopic dermatitis (AD) using a 2,4-dinitrochlorobenzene-induced AD murine model. Topical treatment with AST cream ameliorated the severity of AD-like lesional skin through decreases in infiltration of inflammatory cells and time of scratching behaviors. Also, AST cream reduced histamine and IgE levels in serum. The protein levels of IL-4 and IL-6 in AD-like lesional skin were suppressed by AST cream. These findings suggest that AST cream would be an alternative therapeutic agent for AD-like skin diseases.

Inhibitory Effect of Gamihwalhyeol-tang on Inflammatory Cytokine and NF-kB, AP-1 Activation in Human Synovial Cells (가미활혈탕이 Rheumatoid arthritis 관련 싸이토카인 및 전사인자에 미치는 영향)

  • The present study was carried out to examine the effects of Kami-hwal-hyeol-tang(KHHT) on the immune responses of synoviocyte cells prepared from the rheumatoid arthritis patients, and also on the collagen-mediated arthritis in mouse model. Several experiments were performed in vitro and in vivo to analyse the immunomodulatory effects of KHHT, and the major findings are summarized below: 1. KHHT did not show the cytotoxicity against mLFCs and hFLSs. 2. KHHT inhibited gene expression of IL-1β, IL-6, TNF-α, COX-2, NOS and GM-CSF in hFLSs. Furthermore, KHHT-treated hFLSs showed reduced production of pro-inflammatory cytokines such as IL-1β and IL-6 compared to the control cells. 3. KHHT treatment of hFLSs inhibited the binding activity of NF-kB and AP-1 to their consensus DNA sequences. 4. KHHT treatment(400 ㎍/㎖) of hFLSs significantly inhibited hFLSs proliferations compared to the control cells. 5. KHHT significantly reduced the production of ROS in hFLSs compared to the control cells. The present data show that KHHT plays an important role for the regulation of AP-1 and NF-kB gene expression. Also, it was found that KHHT has anti-arthritis effect. Further studies of KHHT in relation to RA therapeutics may provide important information to develop drugs to treat this disease.

Comparison of Inhibitory Potency of Various Antioxidants on the Activation of BV2 Microglial Cell Lines Induced by LPS

  • Antioxidant properties have been proposed as a mechanism for the putative anti-inflammatory effects of phenolic compounds. To reveal the relationship between antioxidant activity and anti-inflammatory effects of various antioxidants, we measured 1, 1-diphenyl-2-picryhydrazyl(DPPH)-reducing activity and examined the inhibitory effects on LPS-induced inflammation-related gene expression in the BV2 microglial cell line. Lipopolysaccharide(LPS)(0.2 ${\mu}g/ml$) was used with or without antioxidants to treat cells, and the regulation of iNOS and cytokine gene expression was monitored using an RNase protection assay(RPA). Although, all tested antioxidants had similar DPPH-reducing activity and inhibited nitrite production, but the curcuminoid antioxidants(ferulic acid, caffeic acid, and curcumin) inhibited LPS-induced gene expression(iNOS, $TNF-\alpha,\;IL-1{\beta}$, IL-6, and IL-1 Ra) in a concentration-dependent manner. Other tested antioxidants did not exhibit the same effects; N-acetylcysteine(NAC) only began to suppress $IL-1{\beta}$ gene expression just below the concentration at which cytotoxicity occurred. Moreover, the antioxidant potency of curcuminoids appeared to have no correlation with anti-inflammatory potency. Only curcumin could inhibit LPS-induced microglial activation at a micromolar level. These data suggest that curcumin may be a safe antioxidant possessing anti-inflammatory activity.
