Voltage-Dependent Ionic Currents and Their Regulation by GTP and Phorbol Ester in the Unfertilized Eggs of Mouse and Hamster

  • The present study was performed to investigate the properties of ionic currents elicited by voltage pulses in the unfertilized eggs of mouse and hamster by using the whole cell voltage clamp techniques and to find out if there are any differences in properties between eggs of the two rodents. In addition, the modulatory effect of G proteins and protein kinase C (PKC) on the ionic channels were observed. The inward current in hamster eggs was shown to be due to $Ca^{2+}\;current\;(i_{ca})$). The current voltage relations of these currents in hamster egg were analogous to those in mouse eggs. The amplitude of $i_{ca}$ in the hamster egg was larger than that in the mouse egg ($-3.12{\pm}1.07\;nA\;vs.\;-1.71{\pm}0.71\;nA,\;mean{\pm}\;SD$). These results suggest that the $Ca^{2+}$ channels in both kinds of eggs have similar channel properties but their density, and/or conduct ance per unit area is higher in hamster eggs than in mouse eggs. Outward currents in eggs of both mouse and hamster were carried by $K^+$. In hamster eggs, they appeared to comprise at least two components; a transient outward component ($i_{to}$) and a steady state component ($i_{\infty}.$ The $i_{to}$ was found to be dependent on intracellular $Ca^{2+}$ concentration; whereas on the other hand $i_{\infty}\;was\;Ca^{2+}$-independent. $Ca^{2+}$ currents were increased in eggs treated with GTP (or $GTP{\gamma}S$) or fluoroaluminate ($AIF_4^-$). In the hamster egg these increments were antagonized by GDP (or $GDP{\beta}S$) application. In contrast to the enhancement of $i_{ca},\;i_k$ was reduced following GTP (or $GTP{\gamma}S$) perfusion in mouse eggs. The transient component ($i_{to}$) in hamster eggs was increased by adding GTP but decreased by phorbol ester, TPA or dioctanoyl glycerol (DOG). Simultaneous application of $GTP{\gamma}S$ and DOG suppressed $i_{to}$ more effectively than a single application or DOG or TPA. From the above results, we have shown that ionic currents elicited by voltage pulses existed in the unfertilized eggs of mouse and hamster. There are at least two types of currents, $i_{ca}\;and\;i_k$ in mouse eggs, while three types, $i_{ca},\;Ca^{2+}$-dependent $i_k$ and $Ca^{2+}$-independent $i_k$ exist in hamster eggs. ionic channels in these eggs may be regulated either directly by GTP and PKC or indirectly by the substances linked with GTP and PKC.

  • Future communication systems are to be designed to support and serve multimedia and multipledata transmission. Nowadays, requirement of mobile subscribers for the various information such as movie, GPS(Global Positioning System) information, news-is increasing significantly. However, due to practical reasons, the capacity and number of capable channels are limited. To solve this problem, a large number of methods and schemes have been proposed and are under research. In this paper, we demonstrate how satellite DMB (Digital Multimedia Broadcasting) system works with hierarchical modulation scheme. By using hierarchical modulation, we can analyze the capacity. Meanwhile, system performance os evaluated and compared to conventional DMB system without using hierarchical modulation.

  • Recently some urban areas have been flooded due to heavy storm rainfalls. Though major causes of these floodings may be attributed to localized heavy rainfalls, other factors are related to urban flooding including deficiency of storm sewer network capacity, change of surface runoff due to covered open channels, and operational problems of storm drainage pump stations. In this study, hydrologic and hydraulic analysis of Sutak basin in Guri city were carried out to evaluate flooding problems occurred during the heavy storm in July, 2001. ArcView, a world most widely used GIS tool, was used to extract required data for the hydrologic analysis including basin characteristics data, concentration times, channel routing data, land use data, soil distribution data and SCS runoff curve number generation from digital maps. HEC-HMS, a GIS-based runoff simulation model, was successfully used to simulate the flood inflow hydrograph to Sutak pumping station.

  • A joint multipath routing algorithm and channel allocation and scheduling for wireless multihop and multichannel systems is discussed. In packet transmission, distribution of packets to multiroutes makes it possible to reduce the transmission cost of the channels. Cross-layer cooperation of routing, channel allocation, and scheduling is an effective method of packet distribution. As a framework for the cooperation, we propose a multiroute distance vector routing (MDVR) scheme. In the MDVR scheme, the routing table is logically placed in between the routing and link layers, and the table plays the role of a service access point between these two layers. To evaluate the performance of MDVR, simulation is performed in a multichannel, multihop environment. The simulation results show that the MDVR framework can be efficiently implemented in the form of a distributed routing algorithm. It is also shown that in MDVR, the system-wise channel efficiency is almost 25% higher than that in a conventional single-route routing approach.

  • In this work, we explore the feasibility of roll-pitch-yaw integrated autopilots for high angle-of-attack missiles. An investigation of the aerodynamic characteristics of a surface-to-air missile is presented, which reveals the strong effects of cross coupling between the longitudinal and lateral dynamics. Robust control techniques based on $H_{\infty}$ synthesis are employed to design roll-pitch-yaw integrated autopilots. The performance of the proposed roll-pitch-yaw integrated controller is tested in high-fidelity nonlinear five-degree-of-freedom simulations accounting for kinematic cross-coupling effects between the lateral and longitudinal channels. Against nonlinearity and cross-coupling effects of the missile dynamics, the integrated controller demonstrates superior performance when compared with the controller designed in a decoupled manner.

  • Recently, waste landfills built on seashores have been increased because of the easy availability of broad area near the urban communities. To evaluate the performance of the marine clay landfill liner numerical contaminant transport analyses are performed by selecting the typical section of a waste landfill built on seashores and using hydraulic conductivity data obtained from the site. Also, the laboratory electrical resistivity test and the in-situ corrosion test are performed in order to analyze the influence of the soil and leachates composing the landfills on the construct ion materials. From the results of contaminant transport analyses, it is shown that the leachates can be migrated faster through narrow pervious channels than the wide homogeneous pervious tedium and the importance of good quality barriers to prevent the contaminant migration is recognized. In the laboratory electrical resistivity test all the earth materials except the cover soils saturated with distilled water have small resistivities, which shows a high potential of corrosivity of soils composing landfills. However, the degree of corrosion of specimens buried in the landfills was not so severe except the zinc and carbon steel specimens. This apparently conflict results present the necessity of the investigation of other major factors and the long term in-situ corrosion test.

  • Flow field-flow fractionation (FlFFF) is a technology to separate the molecules by size in an open channel. Molecules with different size have different diffusivities and are located vertically in different positions when passing through an open channel. In this study, hollow fiber membranes instead of conventional rectangular channels have been used as materials for the open channel and this change would decrease the cost of manufacturing. FlFFF is a useful technique to characterize the biopolymeric materials. Retention time, diffusion coefficients and Stokes radius of analysis can be calculated from the related simple equations. Hollow-fiber flow field-flow fractionation (HF-FlFFF) has been used for the characterization and separation of protein mixture in a phosphate buffer solution and has demonstrated the potential to be developed into a disposable FlFFF channel. The important indexes for the analytical separation are selectivity, resolution and plate height. The optimized separation condition for protein mixture of Ovalbumin, Alcohol dehydrogenase, Apoferritin and Thyroglobulin is ${\dot V}_{out}/{\dot V}_{rad}=0.65/0.85\;mL/min$.

  • GMTI (Ground Moving Target Indicator) is a kind of airborne radar function that is used widely in military applications to detect the moving targets on the ground. In this paper, GMTI signal processing technique was presented and its performance was verified using sum and difference channels raw data obtained by the captive flight test.

  • Relaxin has been demonstrated to have regulatory functions on both the smooth muscle and extracellular matrix (ECM) of blood vessels and fibrotic organs. The diverse mechanisms by which relaxin acts on small resistance arteries and fibrotic organs, including the bladder, are reviewed here. Relaxin induces vasodilation by inhibiting the contractility of vascular smooth muscles and by increasing the passive compliance of vessel walls through the reduction of ECM components, such as collagen. The primary cellular mechanism whereby relaxin induces arterial vasodilation is mediated by the endothelium-dependent production of nitric oxide (NO) through the activation of RXFP1/PI3K, Akt phosphorylation, and eNOS. In addition, relaxin triggers different alternative pathways to enhance the vasodilation of renal and mesenteric arteries. In small renal arteries, relaxin stimulates the activation of the endothelial MMPs and EtB receptors and the production of VEGF and PlGF to inhibit myogenic contractility and collagen deposition, thereby bringing about vasodilation. Conversely, in small mesenteric arteries, relaxin augments bradykinin (BK)-evoked relaxation in a time-dependent manner. Whereas the rapid enhancement of the BK-mediated relaxation is dependent on IKCa channels and subsequent EDH induction, the sustained relaxation due to BK depends on COX activation and PGI2. The anti-fibrotic effects of relaxin are mediated by inhibiting the invasion of inflammatory immune cells, the endothelial-to-mesenchymal transition (EndMT), and the differentiation and activation of myofibroblasts. Relaxin also activates the NOS/NO/cGMP/PKG-1 pathways in myofibroblasts to suppress the TGF-β1-induced activation of ERK1/2 and Smad2/3 signaling and deposition of ECM collagen.

Characteristics of NMDA- and Glutamate-Induced Currents in Primary Cultured Rat Hippocampal Neurons (일차 배양 해마신경세포에서 NMDA- 및 Glutamate- 유도전류의 특성)

  • Objectives : This study was performed in cultured rat hippocampal neurons to investigate the acute electrophysiological features of ionotropic glutamate receptors which act as a major excitatory neurotransmitter in mammalian brain. Method : Glutamate receptor agonists were applied into the bath solution embedding in whole-cell patch-clamp recording of single hippocampal neuron. Results : In voltage-clamped at -60mV and the presence of 1mmol $Mg^{2+}$, extracellulary applied NMDA did not induce any inward current. Both the elimination of $Mg^{2+}$ and addition of glycine in bath, however, elicited a NMDAinduced inward current. $Mg^{2+}$ block current was increased gradually in more negative potentials from -30mV, showing a negative slope in I-V plot with $Mg^{2+}$. Glutamate-induced current represented an outward rectification. A non-NMDA receptor component occupied about 40% of glutamate-induced current in the voltage range of -80mV to +60mV. Conclusion : Present study suggests that glutamate activates acutely the non-NMDA receptors which induces an inward current in the level of resting membrane potential. This makes the membrane potential increase and can activate the NMDA receptors that permit calcium influx against $Mg^{2+}$ block. At the depolarized state of neuron, there may be recovery mechanisms of membrane potential to repolarize irrespective of voltage-dependent potassium channels in the hippocampal neurons.
