Single and Dual Ligand Effects on Gene Expression Changes in Mouse Macrophage Cells

  • We identified differentially expressed genes in RAW264.7 cells in response to single and double ligand treatments (LPS, $IFN{\gamma}$, 2MA, LPS plus $IFN{\gamma}$, and LPS plus 2MA). The majority of the regulated transcripts responded additively to dual ligand treatment. However, a significant fraction responded in a non-additive fashion. Several cytokines showing non-additive transcriptional responses to dual ligand treatment also showed non-additive protein production/secretion responses in separately performed experiments. Many of the genes with non-additive responses to LPS plus 2MA showed enhanced responses and encoded pro-inflammatory proteins. LPS plus $IFN{\gamma}$ appeared to induce both non-additive enhancement and non-additive attenuation of gene expression. The affected genes were associated with a variety of biological functions. These experiments reveal both dependent and independent regulatory pathways and point out the specific nature of the regulatory interactions.

Effect of Bifidobacteria on Production of Allergy-Related Cytokines from Mouse Spleen Cells

  • To study the effect of bifidobacteria on preventing allergy response, levels of IFN-$\gamma$, IgG2a, IL-4, and IgG1 were investigated in splenocytes isolated from ovalbumin (OVA)­sensitized allergic mice and BGN4-administered allergy­suppressed mice in the presence of various bifidobacterial strains. Most of the bifidobacteria, except 2A, increased production of Th I-associated immune markers, IFN -$\gamma$ and IgG2a. In addition, most of the bifidobacteria, except 2A and 19A, decreased production of IL-4, whereas the differences in the production of IgG1 were less pronounced. These results suggest that some strains of bifidobacteria may have the potential to prevent the occurrence of allergy by switching Th1/Th2-type antibodies and/or related cytokines.

Cytotoxicity of lymphokine activated peritoneal macrophages against Trichomonas vaginalis (질트리코모나스에 대한 림포카인황성대식세포의 세포독성능)

  • Trichomonas vaginalis is a parasitic nagellate in the urogenital tract of human. Innate cytotonicity of macrophages against T. vaginalis has been recognized, but any report on the cytotoxicity of Iymphokine-activated macrophages to T vaginalis is not yet available. The present study aimed to elucidate the Iymphokine-activated cell mediated cytotoxic effect against T. vaginalis by mouse peritoneal macrophages. Cytotoxicity was measured by counting the release of $^3H-thymidine$ from prelabeled protozoa, and tested in U-bottom microtiter plates. Nitrite concentration in culture supernatants was measured by standard Griess reaction. The results obtained are as follows: 1, The cytotoxicity of macrophages was increased by addition of rIL-2 or $rIFN-{\gamma}$$. 2, Cytotoxicity of macrophages was reduced by addition of rIL-4 to rOM-CSV, rIL-2 or $rIFN-{\gamma}$. 3. Crude Iymphokine mixed with anti-lL-2 decreased the cytotoxity of macrophages. 4. In case of macrophages cultured with $rIFN-{\gamma}$ or rIL-4, the concentration of nitrite was related with cytotokity of macrophages against T. vaginalis, but the cytotoxicity of macrophages cultured with rIL-2 and $rIFN-{\gamma}$ was decreased in spite of its high production of llitrite. From the results obtained, it is assumed that rIL-2 and $rIFN-{\gamma}$ enhance the cytotoxicity of macrophages while rIL-4 inhibits the cytotoxicity against T. vaginalis, and that the production of nitrite does not relate with the cytotoxicity of macrophages, but nitric oxide may play a role as an inhibitory factor on the proliferation of T. vaginalis.

Induction of Nitric Oxide and TNF-$\alpha$ by Herbal Plant Extracts in Mouse Macrophages (일부 한약재의 생쥐 대식세포 일산화질소와 TNF-$\alpha$ 생산 유도)

  • In this experiment, we show the effects of herbal plant extracts on the production of nitric oxide (NO) and TNF-$\alpha$. The extracts of Angelica gigas, Astragalus membranaceus, Acanthopanax sessiliflorus and Houttuynia cordata had no effect on NO synthesis by itself in mouse macrophage cell line (RAW264.7). However, the stimulation with these extracts in the presence of murine interferon-${\gamma}$(mIFN-${\gamma}$) resulted in increased NO synthesis. When these extracts were used in combination with mIFN-${\gamma}$, there were a marked cooperative induction of NO and TNF-$\alpha$ synthesis in a dose-dependent manner. The same results were obtained in the mouse peritoneal macrophages used. The optimal concentration of these extracts on NO synthesis was shown at 100$\mu\textrm{g}$/mL with 100U/mL of mIFN-${\gamma}$. NO synthesis was inhibited by NG-monomethyl-L-arginine. When cell lines were treated with extracts, the expression of inducible NO synthetase (iNOS) was markedly increased in RT-PCR analysis. In addition, synergy between mIFN-${\gamma}$ and extracts was dependent on extracts-induced tumor necrosis factor-$\alpha$(TNF-$\alpha$). These results suggest that water extracts of herbal plants can induce iNOS, NO and TNF-$\alpha$ synthesis of mouse macrophage cell line (RAW264.7) and peritoneal macrophages in combination with mIFN-${\gamma}$.

Studies on the Regulatory Effect of Cytokine Production in Patients with Cerebral Infarction by Yuldahansotang (열다한소탕(熱多寒少湯)이 태음인(太陰人) 뇌경색증(腦硬塞症) 환자(患者)의 세포활성물질생성조절(細胞活性物質生成調節)에 미치는 영향(影響))

  • 1. Background and Purpose According to Sasang constitutional medicine, Yuldahansotang(YHT) is a useful prescription for Teaumin patients with a variety of neurologic disorders. Then, I investigated about certain relationships between the effect of YHT and the changes of immune system, especially cytokine network. 2. Methods We studied 8 Taeumin patients with Cerbral infarction. They were treated with YHT in constitutional clinic of Wonkwang Kwagnju Oriental Hospital. We investigated the changes of cytokine network of them. We also investigated cytokine release by lipopolysaccharide- activated peripheral blood mononuclear cells from healthy Taeumin controls. 3. Results The mean interleukin (IL)-2 plasma levels were slightly lower in the plasma of patients than in normal group, whereas the mean IL-4, IL-6 and IgE levels were significantly higher. But there were no significant differences in $interferon-{\gamma}$($IFN-{\gamma}$) levels between each group. After administration of YHT for two to four weeks, plasma levels of $IFN-{\gamma}$ and IL-2 derived from T helper (Th) 1 cells were elevated significantly, whereas plasma levels of IL-4 and IL-6 derived from Th2 cells were reduced significantly. Plasma levels of IgE were reduced significantly, too. During the period of YHT administration, other adverse effects are not shown. It is increased significantly to Cytokine release by lipopolysaccharide -activated peripheral blood mononuclear cells from healthy Taeumin controls. And the release of $IFN-{\gamma}$, IL-2 and IL-6 was progressively decreased in the plasma treated with YHT. It shows regulatory effects of YHT to cytokine production.

Biostatic activity of Coix lacryma seed extract on Toxoplasma gondii in macrophages (율무씨 수침 추출물이 대식세포내 톡소포자충에 미치는 영향에 관한 실험적 연구)

  • Water extract of Coix locrvmn seeds (Co-Ex) was separated into several components; dissolved with Tris-Cl buffer and the supernatant (WC 1), ammonium sulfate treatment supernatant (WC2) and the pellet nvc31,9AE column chromatography of WC 1 and the peak portions; WC4, WCS and WC6. Murine peritoneal macrophages in DMEM containing 10% heat-inactivated FCS were infected with tachyzoites of ToxopIQsmc gondii, RH strain, in uifo. By adding modulators such as Co-Ex, WC 1,2,3,4.5,6 and LPS or IFN-γ for 24 hrs . toxoplasmastatic activity of macrophages was examined in relation to nitrite production. Nitrite production of macrophages was enhanced especially in the series of WC2, WC1 and the combination sample (WC1 + WC2 + WC3) by order than other components or fractions (WC4, WC5, WC6) tested . Toxoplasmastatic actions such as percentage of the inacrophages infected by T. gonnii and fold increase of T gondii in macrophages showed retroverse relations with the amount of nitrite production; i.e. as nitric oxide (NO) increased the phagocytic index of macrophages and the fold increase of tachyzoites in macrophages decreased . Nitrite (NO-2) production was increased by adding IFN-γ in all cases together with enhancement of biostatic effects. Through the results obtained, it is speculated that some components other than the non-proteinous and defatted components in Coix lacrwmn seeds may contribute to activate macrophages through induction of NO for the biostatic activity.

The Effect of Cordyceps militaris on Adaptive Immune Responses in DBA2 Mice Immunized with Influenza Vaccine (밀리타리스 동충하초(Cordyceps militaris)의 인플루엔자백신 적응면역에 미치는 영향)

  • Cordyceps militaris has shown to have various pharmacological activities including an immune-modulatory effect. Previously, we reported that anti-influenza effect of C. militaris in DBA/2 mice was mediated by increased IL-12 and the activation of NK cells. In this study, we investigated the effect of C. militaris on adaptive immune responses using DBA2 mice immunized with influenza vaccine. To determine the effect of C. militaris on antigen presentation capability, we treated RAW 264.7 cells with various concentrations of ethanol extract of C. militaris, which showed a significant upregulation of CD86 (B7.2), CD284 (TLR4), CD40, H-2k (MHC I) and I-Ad (MHC II). To examine the direct effect of C. militaris on adaptive immune responses, we immunized DBA2 mice with influenza vaccine in presence or absence of C. militaris. After 2 or 4 weeks, influenza-specific T cell proliferation, HAI titers and IFN-${\gamma}$ production were measured in plasma or PBMCs isolated from animals. Influenza-specific T cell proliferation and HAI titers were not considerably increased in immunized mice in presence of C. militaris. However, the production of IFN-${\gamma}$ was much greater in immunized mice with C. militaris as adjuvant than only immunized mice.

Effects of Water Extract Acorn on Mouse Immune Cell Activation Ex Vivo (도토리 추출물의 경구 투여가 마우스 면역 세포 활성에 미치는 효과)

  • Acorns have been used as a traditional remed as well as food source. However, few studies on their immunomodulating effects have been reported. In this study, the combined immunomodulative effect of a water extract of acorns was tested on seven to eight weeks old mice(balb/c). The mice were fed ad libitum on a chow diet, and a water extract of the plant mixture was orally administered every other day for four weeks at two different concentrations(50 and 500 mg/kg B.W.). The production of cytokine(IL-$1{\beta}$, IL-6, IL-2, IL-10, IFN-$\gamma$), secreted by macrophages stimulated with LPS or not, detected by ELISA assay using cytokine kit. After 48 h of incubation with mitogen(ConA or LPS) ex vivo study showed that cytokine (IL-$1{\beta}$, IL-6, IL-2, IL-10, IFN-$\gamma$) was detected in both of the 50 and 500 mg/kg B.W. supplementation groups with LPS stimulation. The results of this study may suggest that supplementation with acorn water extract increase immune function by regulating cytokine production capacity by activated macrophages.

Effect of Tuna Extracts on Production of Nitric Oxide and Inflammatory Cytokines (참치 추출물의 일산화질소 및 사이토카인 생성에 미치는 영향)

  • The effect of tuna extracts on the production of nitric oxide (NO) and cytokines including interleukin-6 (IL-6), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), and interferon-${\gamma}$ (IFN-${\gamma}$), was investigated. All extracts and fractions from tuna significantly reduced NO production induced by lipopolysaccharide (LPS). The acetone+methylene chloride (A+M) extract, n-hexane and 85% aqueous methanol (MeOH) fractions had stronger inhibitory effects among them. The 85% aqueous MeOH fraction at a 10-${\mu}g$ concentration significantly decreased LPS-induced IL-6 and TNF-${\alpha}$ productions at 6 h of incubation. In the case of LPS-induced IFN-${\gamma}$ production, the 85% aqueous MeOH fraction at a 3-${\mu}g$ concentration showed significantly higher levels at 48 h of incubation. These results show that the 85% aqueous MeOH fraction inhibited the production of NO and pro-inflammatory cytokines (IL-6, TNF-${\alpha}$), suggesting that this fraction acts as a potent immunomodulator.

Effects of Senggang Gamcho-tang (SG), ZingberisRhizoma (ZR) and Glycyrrihizae Radix (GR) extracts in Senggang Gamcho-tang (SG) on Activity of Murine Splenocytes (생강감초탕(生薑甘草湯) 및 생강(生薑), 감초(甘草)가 생쥐 비장세포의 활성에 미치는 영향)

  • Objectives : Zingberis Rhizoma (ZR) and Glycyrrihizae Radix (GR) have been widely used to prevent or treat allergic diseases. However, relatively little research has been conducted on the immune response to Senggang Gamcho-tang (SG) in a mixture of ZR and GR. The purpose of this study was to determine antiallergic effects of Senggang Gamcho-tang (SG) extracts ZR and GR on activity of murine splenocytes. Methods : This study was performed to investigate the effect of SG in mice, using in vitro experiments. Cells were treated with SG extract (1 ${\mu}g$/ml, 10 ${\mu}g$/ml and 100 ${\mu}g$/ml) plus Con A (2 ${\mu}g$/ml) and ZR (6.6 ${\mu}g$/ml and 66 ${\mu}g$/ml), GR (3.3 ${\mu}g$/ml and 33 ${\mu}g$/ml) respectively for 48 hrs. The production of IFN-${\gamma}$ and IL-4 were determined by ELISA. Results : No toxicity was found in splenocytes treated with SG extract for 48 hrs at the concentration of 0 ${\mu}g$/ml, 1 ${\mu}g$/ml, 10 ${\mu}g$/ml and 100 ${\mu}g$/ml, respectively. The production of IFN-${\gamma}$ increased to 23,968.7 pg/ml (p<0.001) while that of IL-4 significantly decreased to 14.1 pg/m l(p<0.05) only at the SG concentration of 100 ${\mu}g$/ml. With the GR concentration of 33 ${\mu}g$/ml in SG extract, the production of IFN-${\gamma}$ rose to 32,102.2 pg/ml (p<0.001) while that of IL-4 (p<0.05) declined to 34.2 pg/ml. However, there was no significant observation with regard to the effect of ZR on production of IFN-${\gamma}$ and IL-4. Conclusions : This study suggests an antiallergic effect of Senggang Gamcho-tang, which leads to the implication that SG extract may be used to enhance human immune function.
