연교산(連蘿散)이 아토피 동물 모델에 미치는 영향 (Effect of Yeonkyo-san in Atopic Dermatitis Animal Model)

  • The purpose of this study is to examine closely effect that YKS used to atopic dermatitis disease patient get in atopy eruption control experimentally. We analyzed the expression of IgE, IL-6, IL-4, IL-5 and IL-13's level in serum, and IFN-${\gamma}'$ production by YKS extract. We also analyzed YKS extract get to NC/Nga mice's skin establishment analyzes neck-back skin after biopsy, and H&E method measured about epidermis and dermis part in comparison with control group. In this research YKS extract as treatment result to a NC/Nga mice, IgE and IL-6 content in serum decreased remarkably than control group. And decreased than result control group which measure IL-4, IL-5, IL-13's level in serum, and IFN-${\gamma}'$ production secreted in Th1 cell displayed increase by YKS extract. IL-4 and IFN-${\gamma}'s$ gene revelation amount displayed marked decrease than control group in result that observe effect that get in skin of a NC/Nga mice. Also, In culture supernatant which cultivate for 14 day after separate skin cell, IL-13 and IL-6 production decreased than control group. YKS extract get to NC/Nga mice's skin establishment analyzes neck-back skin after biopsy, and dye by H&E method decreased about epidermis and inflammation of dermis part remarkably than control group. These results suggest that Th1 cell and Th2 cell observe to be shifted by secretion amount of IL-4 and IFN-$\gamma$ by YKS extract could know that YKS extract can use usefully in allergy autoimmune disease.

Endotoxin Induces Late Increase in the Production of Pulmonary Proinflammatory Cytokines in Murine Lupus-Like Pristane-Primed Modelp

  • Lupus-like syndrome is characterized by multiple organ injuries including lungs and kidneys. Endotoxin induces a transiently intent systemic inflammatory response and indirectly transient acute lung injury in normal condition. However, whether endotoxin may trigger the persistent development of lung injury in chronic, inflammatory lupus-like syndrome compared with normal condition remains unclear. We examined the pulmonary vascular permeability and production of proinflammatory cytokines, such as TNF-${\alpha}$, IL-6, IL-10 and IFN-${\gamma}$, which play prominent roles in the pathogenesis of lupus-like tissue injury, 6 hand 72 h after i.p. lipopolysaccharide (LPS; endotoxin) injection in pristane-primed chronic inflammation ICR mice characterized by a lupus-like syndrome. These results demonstrated that levels of serum IL-6, IL-10 and IFN-${\gamma}$ and bronchoalveolar lavage (BAL) IL-6 and IFN-${\gamma}$ were remarkably increased 6 h in LPS-exposed pristane-primed mice compared with pristane-primed controls, while pulmonary vascular permeability and levels of serum and BAL TNF-${\alpha}$ were not. And levels of BAL TNF-${\alpha}$, IL-6 and IL-10 were significantly enhanced 72 h in LPS-exposed pristane-primed mice compared with pristane-primed controls. Also, LPS significantly induced the increased in vitro production of TNF-${\alpha}$, IL-6 and IL-10 by lung cells obtained from LPS-exposed pristane-primed mice compared with LPS-exposed normal mice. Our findings indicate that LPS may trigger persistent progression of lung injury through late overproduction of BAL TNF-${\alpha}$, IL-6, and IL-10 in lupuslike chronic inflammation syndrome compared with normal condition.

Comparative Study of the Endotoxemia and Endotoxin Tolerance on the Production of Th Cytokines and Macrophage Interleukin-6: Differential Regulation of Indomethacin

  • Endotoxin tolerance reduces the capacity of monocytes to produce proinflammatory cytokines, results in cellular immune paralysis, and down-regulates the production of helper T (Th)1 type cytokines with a shift toward a Th2 cytokine response. Prostaglandin (PG)E$_2$ in the immune system also results in macrophage inactivation and the suppression of Th1 activation and the enhancement of Th2 activation. However, the inhibitory effects of PGE$_2$ on the altered polarization of the Th cell and macrophage interleukin (IL)-6 production characterized in part by cellular immune paralysis in a state of endotoxin tolerance is unclear. This study was undertaken, using indomethacin, to investigate the role of endogenous PGE$_2$ on the Th cytokines and macrophage IL-6 production in a state of endotoxin tolerance compared to those with endotoxemia mice, wherein, in this latter case, the increased production of proinflammatory cytokines and PGE$_2$ is exhibited. Endotoxemia was induced by injection of lipopolysaccharide (LPS; 10 mg/kg in saline) i.p. once in BALB/c mice, and endotoxin tolerance was induced by pretreatment with LPS (1 mg/kg in saline) injected i.p. daily for two consecutive days and then with LPS 10 mg/kg on day 4. Splenocytes or macrophages were obtained from endotoxemia and endotoxin tolerance models pretreated with indomethacin, and then cytokine production was induced by Con A-stimulated splenocytes for the Th cytokine assays and LPS-stimulated macrophages for the IL-6 assay. Our results showed that endotoxemia led to significantly reduced IL-2 and IL-4 production, to significantly increased IL-6 production, whereas interferon $(IFN)-{\gamma}$ production was not affected. Indomethacin in the case of endotoxemia markedly attenuated $IFN-{\gamma}$ and IL-6 production and didnt reverse IL-2 and IL-4 production. Endotoxin tolerance resulted in the significantly reduced production of IL-2 and $IFN-{\gamma}$ and the significantly increased production of IL-4 and IL-6. Indomethacin in endotoxin tolerance greatly augmented IL-2 production, significantly decreased IL-4 production, and slightly attenuated IL-6 production. These findings indicate that endogenous PGE$_2$ may mediate the suppressed Th1 type immune response, with a shift toward a Th2 cytokine response in a state of endotoxin tolerance, whereas endotoxemia may be regulated differentially. Also, endogenous PGE$_2$ may mediate macrophage IL-6 production in the case of endotoxemia to a greater extent than in the case of endotoxin tolerance.

보중익기탕의 T세포 증식 유도 효과 (Effect of Bu-Zhong-Yi-Qi-Tang on Proliferation of T Cells)

  • 먼저 OVA항원에 대해 특이적으로 증식반응을 나타내는 T세포주를 수립하였고, 수림된 세포주는 세포 표면 단백질이 CD4$^{+}$CD8$^{-}$이며 IL-2와 IFN-${\gamma}$를 분비하는 Type I에 속하는 보조 T세포(Thl)인 것을 확인하였다. 보중익기탕의 total 분획은 OVA항원에 대해 특이적으로 반응하는 Thl세포의 증식반응을 증가시키는 효과를 나타내지 않았으며 고농도에서 오히려 증식반응을 억제하였다. 그러나, 보중익기탕의 polysaccharide 분획은 전반적 인 농도에서 T세포의 증식반응을 유의하게 증가시키는 효과가 있는 것으로 나타났다. 보중익기탕의 polysaccharide 분획을 첨가하였을 때 T세포의 IL-2 분비량은 대조군보다 약간 적었지만, IFN-${\gamma}$ 분비량은 대조군보다 증가하였다. 그리고, 분비된 IL-2와 결합하는 T세포의 IL-3 수용체 발현양도 증가하는 것으로 나타났다. 또한, 항원제시세포의 MHC class II의 발현양도 증가시켰다. 이상의 결과로 보중익기탕의 polysaccharide분획은 T세포의 IL-2수용체 발현양을 증가시키고, 항원제시세포의 MHC classs II의 발현양을 증가시켜서 T세포의 증식반응을 증가시키는것으로 생각된다 그리고 보중익기탕이 생체 면역반응에 미치는 보다 정확한 효과를 평가하기 위해서는 직접 살아있는 실험동물에 투여하는 in vivo 실험이 필요하다.

결핵균이 폐포대식세포의 기능에 미치는 영향에 관한 연구 -H37Ra 결핵균종에 의한 사람 몇 백서 폐포대식세포의 Superoxide 생성의 변화- (The Effects of Mycobacterium Tuberculosis on Alveolar Macrophages -The Alterations of Superoxide Production in both Human and Rat Alveolar Macrophages Exposed to Mycobacterium Tuberculosis H37Ra Strain-)

  • 연구배경 : 산소반응성 대사물에 의한 결핵균 살해능은 대식세포의 활성화 상태에 의해 좌우되는데 체외에서의 이러한 효과를 priming 이라 한다. priming 효과를 유도하는 물질에는 IFN-$\gamma$와 LPS가 대표적인 물질인데 이들 자체가 superoxide와 같은 산소반응성 물질의 생성을 증가시키지는 않지만 식균작용이나 PMA와 같은 화학적 물질에 반응하여 증강된 산소반응성 물질의 생성을 유도한다. 혈액단구세포와는 달리 폐포대식세포는 일상적인 외부환경에 노출되어 있기 때운에 priming 효과에 대한 논란이 있고 결핵균 세포벽의 각 성분이 대식세포의 활성화에 상반되는 결과를 보인다는 보고들이 있어 본 연구에서는 사람의 폐포대식세포와 혈액단구세포, 그리고 백서의 폐포대식세포에서 IFN-$\gamma$에 의한 priming 효과를 비교 관찰하였고 결핵균증 H37Ra 균증이 폐포대식세포에 노출되었을 때 나타나는 superoxide의 생성능의 변화를 비교하여 보았다. 방법 : 사람과 백서에서 얻은 기관지폐포세척액을 Petri dish에 부착시키고 cold shock 방법으로 부착된 대식세포를 분리하여 24시간 IFN-$\gamma$로 전처치한 후의 priming 효과와 H37Ra 결핵균종을 노출시켰을 시의 superoxide의 생성능을 ferricytochrome reduction 방법으로 측정하여 비교하였다. 결과 : 1) 사람 폐포대식세포에서는 고농도의 IFN-$\gamma$ 전처치로도 priming 효과가 관찰되지 않은 반면 혈액단구세포와 백서 폐포대식세포에서는 priming 효과를 관찰할 수 있었다. 2) 폐포대식세포를 비독력 결핵균종인 H37Ra 생균에 노출시킨 결과 사람과 백서 공히 triggering 효과를 나타내었고 그 분해물에 의한 노출 역시 유사한 결과를 나타내었다. 결론 : 사람의 폐포대식세포는 다른 대식세포와는 달리 일상적인 외부환경에 노출되어 있으으로 priming 효과가 관찰되지 않았으며 비독력 결핵균종인 H37Ra에 의해서는 폐포대식세포의 활성화를 억제하는 효과를 관찰할 수 없었다.

Evidence of hydrolyzed traditional Korean red ginseng by malted barley on activation of receptor interacting proteins 2 and IkappaB kinase-beta in mouse peritoneal macrophages

  • Red ginseng, which has a variety of biological and pharmacological activities including antioxidant, anti-inflammatory, antimutagenic and anticarcinogenic effects, has been used for thousands of years as a general tonic in traditional oriental medicine. Here, we tested the immune regulatory activities of hydrolyzed red ginseng by malted barley (HRG) on the expressions of receptor interacting proteins (Rip) 2 and $I{\kappa}B$ kinase-beta (IKK-${\beta}$) in mouse peritoneal macrophages. We show that HRG increased the activations of Rip 2 and IKK-${\beta}$ for the first time. When HRG was used in combination with recombinant interferon-${\gamma}$ (rIFN-${\gamma}$), there was a marked cooperative induction of nitric oxide (NO) production. The increased expression of inducible NO synthase from rIFN-${\gamma}$ plus HRG-stimulated cells was almost completely inhibited by pre-treatment with pyrrolidine dithiocarbamate (PDTC), an inhibitor of nuclear factor-${\kappa}B$ (NF-${\kappa}B$). In addition, the treatment of peritoneal macrophages with rIFN-${\gamma}$ plus HRG caused significant increases in tumor necrosis factor (TNF)-${\alpha}$ mRNA expression and production. Because NO and TNF-${\alpha}$ play an important role in the immune function and host defense, HRG treatment can modulate several aspects of the host defense mechanisms as a result of the stimulations of the inducible nitric oxide synthase and NF-${\kappa}B$. In conclusion, our findings demonstrate that HRG increases the productions of NO and TNF-${\alpha}$ from rIFN-${\gamma}$-primed macrophages and suggest that Rip2/IKK-${\beta}$ plays a critical role in mediating these immune regulatory effects of HRG.

소에서 Interferon이 동결-융해 정자의 운동성과 인공 수정 임신율에 미치는 효과 (Effect of Interferon Supplementation on the Motility of Frozen-thawed Spermatozoa and the Pregnancy Rate after Artificial Insemination in Bovine)

  • The increase in the meat quality and milk production of cows, which breed Korean Native Cow (KNC) and Holstein cow, is not improving reproductive efficiency. In this study, we examined the effect of interferon (IFN) supplementation on motility of frozen-thawed semen and pregnancy rate after artificial insemination of KNC and Holstein cow. In experiment 1, we investigated the effect of IFN-tau concentration (10,000 IU and 20,000 IU) on the percentage of total motility (TM) and progressive motility (PM) of frozen-thawed spermatozoa. In experiment 2, KNC were infused 20,000 IU IFN-tau at insemination or after insemination. In experiment 3, KNC or Holstein cow were inseminated with frozen-thawed semen and infused 20,000 IU IFN-gamma or -tau after insemination. In experiment 1, the average of TM (23.9% to 30.9%) and PM (18.5% to 21.9%) were similar between control and treatments. In experiment 2, the pregnancy rates of IFN infusing times were not different from 45.8% to 53.6%. In experiment 3, the pregnancy rates of Holstein cow infused different kinds of IFN were similar (control, IFN-gamma, IFN-tau; 42.9%, 40.5%, 48.0%). In the case of KNC, however, the pregnancy rate of control was 55.6%, which was significantly lower than that of IFN-gamma (68.9%; p<0.05). Thus, IFN is effective on the improvement of reproductive efficiency, but further study is needed.

Screening of KMU-4, 6, 7 on inflammatory responses in IFN-γ and LPS-induced mouse peritoneal macrophages

  • 본 논문은 IFN-$\gamma$ 및 LPS로 유도된 생쥐 복막 대식 세포에서 염증 반응에 대한 KMU-4, 6, 7 검색에 관한 것으로, 주요 내용은 다양한 질병 치료에 사용되는 생약에서 추출한 한국 해양 식물 (KMU-4, 6, 7)에 대한 연구를 중심으로 한다. 이 논문에서는 쥐 복막 대식 세포를 이용하여, KMU-4, 6, 7가 IFN-$\gamma$ 및LPS로 유도된 산화질소, COX-2 발현, 세포 생존력에 미치는 영향을 평가하였다. KMU-6는 IFN-$\gamma$ 및 LPS로 유도된 산화질소를 억제하고 COX-2 발현에 거의 영향을 미치지 않았다. 이는 KMU-6가 대식 세포 매개 염증성 질환 조절에 사용될 수 있음을 의미한다. 위 결과로부터 KMU-6는 LPS로 자극한 쥐 복막 대식세포에서 COX-2 발현을 억제시켜 산화질소 생성을 억제하는 효과가 있다는 내용이다.

비유기간에 따라 murine 림프조직에서 생산되는 lymphokine의 양상에 관한 연구 (Murine lymphokines production in lymphoid organs during the various stages of lactation)

  • 비유중기, 건유초기 및 임신말기의 C3H/HeN 마우스의 비장, 장간막림프절, 말초림프절(액와, 상완 및 샅림프절)유래 림프구에서 생산되는 lymphokine의 양을 비교하였다. 비장에서의 IL-2 생산양은 임신말기와 비유중기에 낮았지만, IL-4, IL-6와 $IFN_{\gamma}$의 생산양은 임신말기와 건유초기에 높았다. 말초림프절에서의 IL-4, IL-6 및 $IFN_{\gamma}$ 생산양은 임신말기와 건유초기에 높았지만, IL-2의 생산양은 임신말기, 비유중기 및 건유초기에 각각 감소하였다. 장간막림프절에서의 IL-4 생산양은 임신말기와 비유중기에 각각 증가하였으나, IL-2의 생산양은 임신말기에 감소하였다. 이와같이 전반적으로 건유초기와 임신말기에 IL-2의 생산양은 낮지만, IL-4, IL-6와 $IFN_{\gamma}$의 생산양이 높은 결과는 이 시기에 유방염의 발생비율이 높은 것과 연관성이 있는 것으로 추정된다.
