Objectives : TA, a polyherbal extract, typically is used for pharmacopuncture therapy on patients with traffic accident-related injuries and musculoskeletal diseases. This study was performed to evaluate the safety of the TA extract, using a micronucleus test. Methods : The dose range and sampling time were first established. An in vivo micronucleus test was then performed to determine the induction of micronuclei in mouse bone marrow cells after a single intramuscular administration of TA to 7-week-old ICR mice (0.2 ml/animal, at 24 hours post-dosing). Results : The incidence of micro-nucleated polychromatic erythrocytes (PCEs) in PCEs in the TA group was similar to that in the negative-control group, while that in the positive-control group was significantly greater. The positive- and negative-control groups did not differ in the ratio of PCEs to total erythrocytes. Conclusions : Our toxicity study indicates that the TA extract does not induce micronucleus formation in mouse bone marrow cells.
The development of isolated blastomeres from mammalian preimplantation embryos has been basically studied for the multiplication of embryos from superior animals. Therefore, this study was investigated the effect of co-culture with mouse fetal fibroblast cells(MFFC) on in vitro development of blastomeres from mouse preimplantation embryos. Mature female ICR mice were treated with hormone to induce superovulation and embryos were collected at each 2, 4, and 8-cell stage. Then, after removing zona pellucida with protease, blastomeres were isolated by micropipetting, or reconstituted with different stage blastomere, and incubated for 72 hrs either in T6 or TCM199 or on the monolayer of MFFC, which was prepared with fibroblast cells from 14∼14 day mouse fetus. After incubation, we examined their development rates every day and the nuclei numbers of each blastocyst by Hoechst-33342 staining. In the development rates of blastomeres, there were no significant differences between media but the higher rateswere found in the monolayer of MFFC, regardless of reconsititution. In addition, blastomeres cultured with MFFC had slightly greater number of nuclei than those cultured in single media. Generally, the higher development rates of blastomeres were found from earlier stage embryos than the later ones, regardless of culture conditions. Reconsitituted blastomeres had more nuclei but did not show the higher development rates, compared to the single blastomeres. Taken together, our results suggest that co-culture with MFFC have a beneficial effect on the in vitro development of blastomeres from mouse embryos.

In order to investigate the acute toxicity of autoxidized methyl linoleate(AOML) on the activity of serum enzymes in the mouse, we administered once 0.45ml of AOML to ICR strain mouse by using stomach tube. The following results were obtained: The total lactic dehydrogenase(LDH) activities in the serum of AOML group were generally increased than those of normal group. According to electrophoresis, the activities of LDH, were increased while those of LDH, were decreased. The activities of glutamic oxaloacetic transaminase(GOT), glutamic-pyruvic transaminase(GPT) and ${\alpha}-amylase$ in the serum of AOML group were increased more than those of normal group. The activities of alkaline phosphatase in the serum of AOML group were increased but those of isozyme were not confirmed in the normal and AOML group. In the serum protein of AOML group, albumin was increased, on the other hand ${\gamma}-globulin$ was decreased. At the peripheral blood slide smear, lymphocytes were significantly decreased but neutrophils were increased and the morphological change of erythrocytes was observed. From these results we conclude that the AOML fed to mouse influences on the activity of various serum enzymes and blood cells in the mouse.
Guh Sung Y.L.S.-95 is one of the polyacidic solution of which main component is acetic acid. We investigated the subchronic toxicity of the Guh Sung Y.L.S.-95 using SPF ICR mouse for 4 weeks. The Guh Sung Y.L.S.-95 was administered by gastric intubation, 1.0, 2.5, 5.0 g/kg body weight. The results are as follows: 1. There are no adverse effects on the clinical obserbation and body weight changes. Also, there are some significant changes in organ weight, but it was meaningless because of the absence of dose-response relationships. 2. In the hematological patterns of administered mouse, there are no significant changes between the treated groups. Also, there are no serological enzymatic changes in the treated mouse. In the 1.0 g/kg treated group, ASP activity was increased significnatly compared with control group. But, this level of activity was fall under the normal physiological range of control mouse. 3. Histopathological findings of the brain, liver, heart, spleen, kidneys, stomach, lung, testis, ovary, uterus and thymus were not observed in the treated mouse. From the above results, the Guh Sung Y.L.S.-95 has no toxicity upto the 5.0 g/kg/day of oral dose for 4 weeks.

Anticarcinogenic activity of astaxanthin-contatining egg yolk(designate AEY) was investigated for mouse ascites carcinogenesis induced by mouse Sarcoma-180(S-180) cells. Female ICR mice(8mice/treatment, 7∼8weeks of age, 25±1g) were injected, i.p. with S-180 cells(1×107cell/ml PBS). Two days later, each mouse was given 0.1ml PBS containing AEY(10, 25 or 50ug/g body weight) or control egg yolk (CEY; 50ug/g body weight) every other day for 7 times. Control mice were only given 0.1ml S-180 cells and 0.1ml PBS. Mice treated with 25ug/g body weight of AEY showed 24.8 days of life, which was equivalent to 138% of control mice's life(180.0 dyas). Based on dose-dependant experiment of AFY, mice treated with 10ug/g body weight showed slightly longer life(19.4 days) relative to mice treated with control mice, and mice treated with 50ug/g body weight exhibited 21.9 days of life. Mice treated with any dose of AEY exhibited longer life than mice with CEY 50ug/g body weight. Body weight of mice treated with AEY was reduced relative to that of control mice CEY-treated mice. These results suggest than AEY inhibits the carcinogenesis of mouse ascites induced by S-180 cells.
The change in bodyweight, organ weight and the amount of water intake were investigated to assess the effect on a range of toxicological parameters of varying dosages of polyacetylene compounds. Concerning inhibition of solid tumor growth induced by Sarcoma-180 cells in the ICR mouse strain, relytionships between the dosage of polyacetylene compoilnds used and several toxicological parameters were observed. When 40μmoles/kg b.w. of panaxydol tvas administered for 3 consecutive days, the suppression of body weight growth in Wistar rats was about 17% that of the control group, however, the body weight gain was normalized 4 days after stopping arlnlinistration of the polyacetylene compounds. When the dosage of polyacetylenes was reduced to 20μmoles/kg b.w., the change in body weight gain was alleviated to within l0% of the control and the suppremission of the body weight gain increased in order for Panaxydol, Panaxynol, and Panaxytriol. The organ weight did not show any significant change between the control and the polyacetylene administered groups. Electron micrographs of the liver tissues of ICR mice injected with 40Umoles/kg b.w. of panaxydol gave no further significant information about toxicological changes. One hundred and twenty imprinting control region (ICR) mouse with initial body weights of 26 ± 2 g (5 weeks old) were assigned to six treatments for a two-week feeding trial to determine the effect of Pediococcus pentosaceus strains (PpS) which were isolated from three different types of Kimchi in ICR mice infected with Escherichia coli (Ec) or Salmonella Typhimurium (ST). Six groups constituted a normal control group without Ec or ST orally administrated (NC-; n = 20), a normal control group (NC+; n = 20), a group for which Lactobacillus plantarum was orally administrated (LP; n = 20), a group for which PpS A was orally administrated (PSA; n = 20), a group for which PpS B was orally administrated (PSB; n = 20), and a group for which PpS C was orally administrated (PSC; n = 20), the latter five groups constituted the Ec infected groups and the ST infected groups of 10 mice each. LP and PSC showed significantly (p < 0.05) improved growth performance compared to the other groups, except for NC- in the Ec infected mice group. NC+ showed significantly lower (p < 0.05) growth performance compared to the other groups, except for NC- in the ST infected mice groups. Regarding the Ec and Salmonella counts in the intestine, the LP and PSC groups had significantly lower (p < 0.05) counts than the NC+ and PSB groups. In conclusion, LP and PSC strains isolated from Kimchi can act as probiotics by inhibiting Ec and ST.
To evaluate the effect of Kam Doo-extract (KDE) on organophosphorous (OP)intoxication in mouse, this research was conducted. KDE prescribed with the equal weights of both Padix Glycyrrizae and Simen Glycine was extracted in water at 100$^{\circ}$C for 2hr, and concentrated in a vacuum evaporator. Animmal used in this research was ICR-strained male mice (bodyweight: 20 ~ 25g), and induction material for OP intoxication was DDVP(Dichlovos). Toxicity parameters used to evaluate KDE-preventive effect on DDVP were cholinesterase activity, and protection index of KDE against LDso values of DDVP. As the results, KDE prevented the inhibition of cholinesteranse activity due to DDVP-treatment, and inhanced the protecion index. Consequently our experimental data show the KDE will be useful as an preventive agent in respect that KDE is safe and effective against OP intoxication in mouse.
Cryopreservation of mouse embryos biopsied at 4-cell stage was investigated by ultrarapid freezing. Four-cell embryos were obtained from ICR mice on 55h after hCG injection. Zona pellucida of the embryos were partially dissected with a cutting pipet, and then single blastomeres were biopsied from the embryos followed by incubation in $Ca^2$+ and $Mg^2$+-free M16 medium for 30min. Biopsied embryos cultured for lh or 15h were frozen by ultrarapid freezing method using 3M DMSO or 5M glycerol as a cryoprotectant, respectively. The developmental rate of biopsied embryos after ultrarapid freezing and thawing to blastocysts was 81 % in the group of biopsied embryos cultured for lb and 98% in the group of biopsied embryos cultured for 15h, respectively. When biopsied embryos after ultrarapid freezing and thawing were transferred to the uteri of pseudopregnant recipients, normal live young were born. These results suggest that this freezing method can efficiently cryopreserve biopsied mouse embryos.
The pathological aspects of purified ricin from the seeds of the castor oil plant, Ricinus communis, were examined, using light and transmission electron microscopy. ICR mice were exposed to ricin by peritoneal injection with 100 ng/1 $m\ell$ PBS(pH 7.0) mouse and histological observations on the liver, spleen, thymus, lung and heart were carried out at intervals up to 48 h after exposures. All the organs examined were damaged by ricin. Among the organs, the spleen and thymus; immune organs were the most sensitive to ricin, whereas the effect delayed in the liver, lung and heart. Furthermore, the immune cells in each organ were the most sensitive to ricin. Accordingly, the effect of ricin on the organs seems to be affected by the immune cells existed in each organ, In each organ, the immune cells showed apoptotic changes, while the capillary endothelial and parenchymal cells showed necrotic changes.
