Geraniol이 L1210 세포와 ICR 생쥐 대식세포의 증식,Superoxide Dismutase(SOD)와 Inducible Nitric Oxide Synthase(iNOS) 효소활성에 미치는 영향 (Effect of Geraniol on the Proliferation of L1210 Cells and ICR Mouse Macrophages, and the Activities of Superoxide Dismutase (SOD) and Inducible Nitric Oxide Synthase ( iNOS) Activities)

  • The present investigation was undertaken to find out the anticancer activity of monoterpene compounds. Monoterpenes showed generally the inhibitory effect on the proliferation o f L1210 cancer cells (cytotoxicity). Geraniol was found to exibit the most potent cytotoxic effect on L1210 cells with an IC50 values of $0.67{\mu}g/ml$. On the other hand, geraniol proved to be capable of stimulating the macrophage proliferation (135% of control). When the life prolonging activity of geraniol by daily oral administration of 0.1~10${\mu}g/10{\mu}l/20$ g body weight to Sarcoma 180 bearing ICR mouse was examined, there was also a significant elevation of survival (best result of 134% of control). The contradictory effects of geraniol on the proliferation of L1210 cells and macrophages proved to be accompanied by the coincident alterations of RNS (reactive nitrogen species) related enzymes activities such as inducible nitric oxide synthase (Inos) in macrophages and ROS (reactive oxygen species) related enzymes activities such as superoxide dismutase (SOD) in L1210 cells, respectively.

마우스 T세포 증식인자(interleukin-2)에 의한 마우스 T림프구의 장기배양 및 그 특성 (Proliferation and Characteristics of Murine T Lymphocytes in A Mitogen-Induced Conditioned Media(Interleukin-2))

  • Balb/c mouse spleen cells in vitro sensitized against ICR spleen cells were cultured in conditioned media(CM). The CM was produced by ICR spleen cells stimulated with Concanavalin-A(Con-A), and sensitized lymphoid cells were grown in CM. ICR mouse spleen cells were appeared to be a good generator of IL-2. Optimal growth was seen in growth medium containing 20% fetal calf serum. and 25% CM. When cultures were initiated at 1, 5, $10{\times}10^4\;cells/ml$, the cells were increased in numbers by about 20, 13, 5-fold, respectively, every 9 days. Such growth pattern was sustained for about 4-6 weeks and thereafter the cell growth was diminished gradually. Direct immunofluorescence indicated that 93% of the lymphoid cells grown in CM(for 10 days) expressed Thyl surface antigen. And the cells grown in CM were cytotoxic to the sesitizing ICR mouse spleen cells though cytotoxicity level was not high. According to these results, the cells grown in CM were considered to be cytotoxic T lymphocytes. The lymphoid cells grown for 20 days were nearly unresponsive to Con-A and therefore dependent only IL-2 to be used for IL-2 assay.

ICR 마우스 위장관 내분비 세포의 부위별 분포 및 출현 빈도 : 면역조직화학적 연구 (The regional distribution and relative frequency of gastrointestinal endocrine cells of the ICR mice: An immunohistochemical study)

  • ICR 마우스 위장관 8개 부위(위저부, 유문부, 십이지장, 공장, 회장, 맹장, 결장 및 직장)에서 위장관내분비 세포의 부위별 분포 및 상대적 빈도를 somatostain, serotonin, glucagon, chloecystokinin(CCK)-8, secretin, pancreatic polypeptide(PP) 및 gastrin 등 총 7종류의 항혈청을 이용한 면역조직화학적 방법으로 관찰하였던 결과 somatostain, serotonin, glucagon, CCK-8, secretin, 및 gastrin 면역반응세포의 7종류의 내분비세포가 관찰되었다. 본 실험의 결과 장관부위에서는 주로 타원형 또는 방추형의 개방형 세포(open-typed cell)들이 관찰된 반면 위저부와 유문부에서는 주로 원형의 폐쇄형 세포(close-typed cell)을이 관찰되었다. 이들 면역반응세포들의 부위별 분포는 위장관 각 부위에 따라 매우 다양하게 관찰되었다. Somatostain 면역반응세포들은 대장을 제외한 위장관에서 전 부위에서 관찰되었고, serotonin 면역반응세포들은 전 위장관에 걸쳐 관찰되었으며, ICR 마우스에서 가장 높은 빈도를 나타내었다. Glucagon 면역반응세포들은 위저부와 직장에 국한되어 관찰되었으며, 각각 중등도 및 소수의 빈도를 나타내었다. CCK-8 면역반응세포들은 유문부, 십이지장 및 회장에서 각각 다수, 중등도 및 극소수의 빈도로 관찰되었다. 한편 secretin 면역반응세포들은 각각 소수 및 극소수의 빈도로 십이지장과 회장에 국한되어 출현하였고, gastrin 면역반응세포들은 유문부에 국한되어 다수 관찰되었다. 그러나 PP 면역반응세포들은 전 위장관에 걸쳐 관찰되지 않았다. 결론적으로 ICR 마우스의 위장관내분비세포의 부위별 분포 및 상대적 빈도는 다른 포유동물과 유사하게 관찰되었으나, 일부 특이한 양상을 나타내기도 하였다.

동결보존한 마우스 집합배의 생존성과 chimera의 생산에 관한 연구 (Developmental potential of aggregated mouse embryos and production of chimeras after freezing)

  • The present study was carried out to investigate the viability of frozen-thawed aggregated mouse embryos and to produce the chimeras. Different phenotypic embryos were obtained by mating ICR female mice with ICR or CBA male mice. The aggregated morulae were produced following aggregation of embryos at 4-, 8- and 12- to 16-cell stage. The desirable stage for the aggregation of the mouse embryos was 8- to 16-cell stage. The post-thawed in vitro survival rates of aggregated embryos in glycerol, DMSO and ethylene glycerol were 51.5, 78.6 and 69.4%, respectively. Although the higher survival was obtained in DMSO, there were no significant differences in the survival rates among the three cryoprotectants. A total of 155 frozen-thawed aggregated embryos were transferred to 11 recipient mice, 3 out of 7 offsprings were born to overt chimera. These experiments have proven that mouse chimeras can be obtained from frozen-thawed aggregated embryos.

노화촉진모델마우스(SAM)와 정상 마우스(ICR)에서 타우린의 혈액-뇌 관문 투과성의 비교 (The Blood-brain Barrier Permeability of Taurine in Senescence-accelerated Mouse and Normal Mouse (ICR))

  • This study compared the blood-brain barrier permeability of [$^3H$] taurine in senescence-accelerated mouse (SAM) and normal mouse with common carotid artery perfusion (CCAP) method and intravenous injection technique to establish a possible relation between aging and changes in tissue levels of taurine. The SAM strains show senescence acceleration and age-associated pathological phenotypes similar to geriatric disorders seen in humans. In the result of this experiments, the plasma clearance of [$^3H$]taurine in SAM was almost comparable with that of normal mice by intravenous injection technique, but the brain volume of distribution ($V_{D brain}$) of [$^3H$]taurine in SAM by CCAP method reduced by 85% compared with that in normal mice. These results suggest that aging may have an effect on the brain transport activity of taurine in disease state model animal.

마우스에서 Naegleria fowleri에 의한 뇌수막염 발생에 관한 실험적 연구 (Experimental Meningoencephalitis by Nuegleria fowleri in Mice)

  • Naegleria fowleri를 마우스 비강을 통하여 감염시킨 뒤 P.A.M. 발생율 및 발병후 생존기간을 마우스 주별 체중별, 성별, 접종아메 바수에 따라 비교하여 다음과 같은 성적을 얻었다. 1. N. fewleri를 감염시킨후 P.A.M. 발생은 ICR마우스보다 BALB/C마우스에서 많았으며 감염후 평균생존기간은 차이가 없었다. 2. 마우스 성별에 따른 N. fowleri감염후 P.A.M.발생율 및 발병후 평균생존기간은 차이가 없었다. 3. 체중이 다른 여러 실험군에서 P.A.M. 발생율 및 사망율은 차이가 없었으나 발병후 마우스의 평균생존 기간은 체중이 가벼운 실험군에서 짧았다. 4. 감염시킨 아메 바수에 따른 P.A.M. 발생율은 영양형 Ix104이상 감염 군에서는 차이가 없었으나 0.5x104인 경우 P.A.M 발생이 현저히 감소하였다.

  • This research was conducted to observe developmental capacity of the early embryos aggrigated to phytohemagglutinin-M(PHA-M) in the culture of mouse embryos in vitro. The results showed that the development of blastocyst increased to 2-celT >< 2-cell : 68. 9%, 4-cell $\times$4-cell : 92.5% and 8-cell $\times$8-cell : 97.3% in the aggrigated embryos of ICR mouse, and 2-cell $\times$ 2-cell : 90.0%, 4-cell $\times$4-cell : 93.9% and 8-cell $\times$ 8-cell : 100% in the aggrigated embryos of two different strains (ICR $\times$ CBA/J mouse). ICR Mouse의 아급성 알코올 대사에 보이차(Camelia sinensis L) 추출물이 미치는 효과 (Effects of Water Extracts of Camelia sinensis L on Blood Alcohol Concentration and Activities of Sub-acute Alcohol Metabolic Enzymes in ICR Mouse)

  • An eight-week-old male ICR mouse, which was induced with acute alcohol and sub-acute alcohol poisoning condition, was administered with bohee tea(Camelia sinensis L) extract. Under the inducement of the sub-acute alcohol poisoning condition, no considerable differences could be found in the blood alcohol concentration of the positive control group and the bohee tea group(p<0.05). The GOT activity of the three groups: bohee tea, Drink, and Alcodex decreased than that of the normal control group(9.064±4.687 unit)(p<0.05). In addition, the blood GOT activity of the dark green tea group dropped by 81.44% compared with that of the positive control group. On the other hand, the blood GTP activity of the bohee tea group decreased by 5.2% as opposed to that of the positive control and the Drink that decreased by 7.5% as opposed to that of the positive control. The hepatic ADH activity of the bohee tea increased by 22.7%, as opposed to that of the positive control group. The Drink, however, had an increase rate of 33.6%. In the case of the hepatic ALDH activity of the liver, no significant differences were ever recorded among all groups, except for the positive control group. Due to an intake of bohee tea extract, the hepatic ALDH activity decreased by 77.27% which could not be seen in the positive control group. However, Drink and A1codex had a decrease could be seen(p<0.05).

  • An eight-week-old male ICR mouse, which was induced with acute alcohol and sub-acute alcohol poisoning condition, was administered with bohee tea(Camelia sinensis L) extract. After oral administration of bohee tea and inducement of acute alcohol poisoning condition, the mouses blood alcohol concentration became as low as that of the normal control group. Its decrease rate was 87.26%, in comparison with that of the positive control group. Moreover, its blood GOT activity decreased with a rate of 93.1 % until it reached the normal level, as opposed to that of the positive control group. In addition, the GOT activity, despite rising after the alcohol intake, decreased(p<0.05) significantly after administration of each sample and reached the normal level. The bohee tea group experienced a significant decrease in the GOT activity, compared with the A1codex group and the Drink group. The GPT activity of the Alcodex group decreased by 11 % compared with that of the positive control group. The CTP activity of the bohee tea group decreased by 8.2%, while that of the Drink group decreased by 6.5%(P<0.05). However, there were no significant differences between the results in the control group and those of the test group. The bohee tea group's hepatic ADH activity increased by 22.7% compared with that of the positive control group. On the other hand, the hepatic ADH activity of the Drink group increased by 33.6% while that of the A1codex group increased by 20.4%. On the contrary, the bohee tea extract, the hepatic ALDH did not manifest any significant difference as compared with the normal control group. However, its decrease rate was about 16.67% as compared with that of the positive control group. The Drink group, meanwhile, obtained a decrease rate of about 21.59%.

난관체류시간에 따른 생쥐초기배의 체외발생능력 (Dependence of Mouse Embryonic Development in vitro on the Exposed Period to Oviductal Environment)

  • Development in vitro of 2-cell mouse embryos was examined after appropriate exposure to oviductal milieu to demonstrate biological activity present in the oviducts. ICR and ($C57Bl/6{\times}Balb/c$) $F_1$ hybrid mice were superovulated and mated for the recovery of early embryos. Embryos were recoverd at every 2h intervals from 32h post-hCG(hph) to 56 hph. The proportions of developmental stages were determined in the recovered embryos. Development in vitro of 2-cell embryos was more rapid in $F_1$ hybrid than in ICR, showing high proportions of 4-cell embryo and blastocyst at 120 hph. 100% of blastocyst development was obtained at 38hph in $F_1$ hybrid and at 50 hph in ICR when 2-cell embryos were cultured upto 120hph in vitro. Moreover, in vitro culture of oviducts containing 2-cell embryos in ICR mice for 12h from 34hph to 46hph increased developmental capacity of ICR mouse embryo in vitro. The results indicate that oviductal environment contains substances having mitogenic activity and overcoming early cell block in vitro. The mitogenic activity is effective in vitro as well as in vivo.
