유방 암세포에서 Protein Kinase C 동위효소의 전위 (Translocation of Protein Kinase C Isozymes in the Breast Cancer Cell Line)

  • Phorbol 12-myristate 13-acetate (PMA), bryostatin, dioctanoyl glycero1 (DiC8)과 같은 Protein Ki-nase C (PKC)의 활성제는 세포질로부터 막이나 핵으로 PKC 동위효소의 전위를 유도한다. 활성화된 PKC는 일반적으로 암을 유발시키는 역할을 하지만 그와 반대로 사람유방암세포의 성장을 약화시키는 기능을 가지고 있다. PKC의 항증식효과와 전위가 MCF-7 세포에서 조사되었다. PMA, bryostatin, DiC8로 활성화된 PKC 동위효소의 전위는 MCF-7 세포의 여러 장소에서 나타났다. PMA는 PKC $\alpha$$\beta$는 핵이나 핵막 그리고 PKC $\delta$$\varepsilon$은 세포막으로 일부 전위시켰고, 반면 DiC8과 bryostatin은 PKC $\alpha$$\beta$를 각각 핵과 핵막으로 전위를 유도하였다. PKC 활성제의 항증식 효과에 있어서 PMA ($IC_{50}$/ values of 1.2$\pm$0.3nM)와 DiC8 ($IC_{50}$/ values of 5.0$\pm$1.1$\mu$M)는 세포의 성장을 억제시켰다. Bryostatin 역시 세포의 성장을 억제시켰지만, PMA로 관찰된 것보다는 낮은 수준이었다. 즉 100nM bryostatin에 의해 16% 정도 성장이 감소되었다. 그러나 PMA는 bryo-stalin과 함께 처리하였을 때 PMA의 항증식 효과는 낮았으나, 10$\mu$M DiC8과 함께 처리하였을 때는 효과가 없었다. 이러한 결과들은 각 PKC 동위효소들이 다른 특이한 위치로 전위되었으며, 특히 PKC $\alpha$ 동위효소가 세포성장의 항증식 기능을 조절하는데 중요한 역할을 함을 시사한다.

Neuroprotective effects of antioxidant constituents isolated from Opuntia ficus-indica var. saboten Makino

  • Opuntia ficus-indicavar. saboten Makino (Cactaceae) is a tropical or subtropical plant that has been widely used as folk medicine for the treatment of diabetes, asthma, burn, edema and gastritis. The purposes of the present study were to identify antioxidant constituents from fruits and stems of the plant cultivated in Cheju island, Korea, and examine their in vitro neuroprotective activities. Using a chromatographic fractionation method, ten chemical constituents were isolated from ethyl acetate extracts. By means of chemical and spectroscopic methods, those were identified as eight flavonoids such as kaempherol (a), quercetin (b), kaempferol 3-methyl ether (c), quercetin 3-methyl ether (d), narcissin (e), dihydrokaernpferol (f), dihydroquercetin (g) and erioclictyol (h), and two terpenoids such as 3-oxo-${\alpha}$-ionol-${\beta}$-d-glucopyranoside (i) and roseoside (j). Among the isolated compounds, comrounds c~e and h~j were those reported for the first titre from the plant. Compounds b, d and g showed DPPH free radical scavenging activities with IC$\sub$50/ values of 28, 19 and 31, ${\mu}$M respectively. Compounds d and g also inhibited iron-dependent lipid peroxidation with IC$\sub$50/ values of 2.4 and 3.5 ${\mu}$M. In a primary rat cortical neuronal cell culture system, compounds b, d and g inhibited xanthine/xanthine oxidase-induced (IC$\sub$50/ values of 18.2, 2.1 and 54.6 ${\mu}$M) and H$_2$O$_2$-induced (IC$\sub$50/ values of 13.6, 1.9 and 25.7 ${\mu}$M) cytotoxicities. In addition, compounds d and g inhibited NMDA-induced excitotoxicity by 21 and 33%, and only compound d inhibited growth factor withdrawal-induced apoptosis by 31% at a tested concentration of 3 ${\mu}$M. The results suggest that the antioxidant constituents with in vitroneuroprotective activities may serve as lead chemicals for the development of neuroprotective agent.

Physiological Activities of Thiacremonone Produced in High Temperature and High Pressure Treated Garlic

  • To examine the possibility of using thiacremonone isolated from high-temperature-high-pressure treated garlic, this study investigated the physiological activities properties. The $IC_{50}$ values of hydroxyl, superoxide, hydrogen peroxide, and nitric oxide radical scavenging activities of thiacremonone were 92.50, 65.05, 12.60, and $81.53{\mu}g/mL$, respectively. On the other hand, the activities of vitamin C were 104.93, 99.43, 42.42, and $122.64{\mu}g/mL$, and the activities of butylated hydroxyanisole were 37.22, 68.45, 22.47, and $40.54{\mu}g/mL$, respectively. The $IC_{50}$ value of ACE inhibition activities of thiacremonone and captoprill were 0.265 and $0.036{\mu}g/mL$, respectively. The $IC_{50}$ value of xanthine oxidase inhibition activities of thiacremonone and allopurinol were 39.430 and $9.346{\mu}g/mL$, respectively. The $IC_{50}$ value of tyrosinase inhibition activities of thiacremonone and kojic acid were 101.931 and $65.648{\mu}g/mL$, respectively.

공정변수에 따른 초전도 튜브의 전기적 특성변화

  • High-temperature Superconductor(HTS) tubes were fabricated in term of different processing variables such as preheating temperature, speed of mold rotation and cooling rate by centrifugal forming method. For powder melting by induction the optimum range of melting temperatures and preheating temperature were $1050^{\circ}C{\sim}1100^{\circ}C$ and $550^{\circ}C$ for 30min, respectively. The mould roating speed was 1000rpm. A tube was annealed at $840^{\circ}C$ for 72hours in oxygen atmosphere. The plate-like grains were well developed along the roating direction and typical grain size was about more than $40{\mu}m$. It was found that Ic values increased with increasing the preheating temperature and speed of mold rotation. While Ic decreased with increasing the cooling rate. The measured Ic in $50mm{\times}70mm{\times}2.5mm$ tube was about 896Amp.

IC 패키지 마킹검사를 위한 적응적 다단계 이진화와 정합단위의 동적 선택 (An Adaptive Multi-Level Thresholding and Dynamic Matching Unit Selection for IC Package Marking Inspection)

  • 머신비전을 이용한 IC 패키지 마킹검사 시스템은 입력영상으로부터 검사할 요소들의 위치를 식별하고, 추출된 요소들을 학습된 표준 패턴과 비교하여 마킹의 불량 여부를 판단한다. 본 논문에서는 검사 대상 IC 패키지의 위치 판별, 마킹문자 추출, 핀원딤플 검출과 같은 일련의 작업들에 적합한 적응적 다단계 이진화 방법과 마킹문자의 국소적인 오류검출은 물론 잡영에 강건한 정합단위의 동적 선택 방법을 제안한다. 제안하는 이진화 방법은 이진화 대상 영역과 명도 값의 범위를 제한하여 Otsu의 이진화 알고리즘을 적용함으로써 특정 응용에 적응적인 이진화가 가능하다. 정합단위의 동적 선택 방법은 문자추출 및 배치분석에 대한 결과에 따라 정합단위를 선택한다. 그러므로 문자추출 및 배치분석 과정에서 발생하는 예기치 못한 부적절한 상황에서도 가능한 범위내에서 최소의 정합단위를 선택할 수 있다. 제안된 방법을 구현하여 8종의 IC 패키지, 총 280개의 영상에 대하여 실험한 결과, IC 패키지와 핀원딤플의 검출율은 100%였으며, 마킹상태에 대한 판정은 98.8%의 정확도를 나타내어 제안된 방법이 효과적임을 확인할 수 있었다.

1-Methyl Substituent and Stereochemical Effects of 2-Phenylcyclopropylamines on the Inhibition of Rat Brain Mitochondrial Monoamine Oxidase A and B

  • (E)-2-Phenylcyclopropylamine ((E)-TCP), (Z)-2-Phenylacyclopropylamine ((Z)-TCP), (E)-1-methyl-2-phenylcyclopropylamine ((E)-MTCP), and (Z)-1-methyl-2-phenylcyclopropylamine ((Z)-MTCP) were synthesized and used to determine to what extent 1-methylsubstitution and stereochemistry of 2-phenycyclopropylamines affect inhibition of monoamine oxidase (MAO). Inhibition of rat brain mitochondrial MAO-A and B by the compounds were measured using serotonin and benzylamine as the substrate, respectively and $IC_{50}$ values obtianed with 95% confidence limits by the method of computation. For the inhibition of MAO-A, (E)-MTPC ($IC_{50}$ = 6.2 * $10^{-8}$M) was found to be 37 times more potent than (Z)-MTCP ($IC_{50}$ = 7.8 * $10^{-8}$M), was 7 times more potent than (Z)-MTCP($IC_{50}$= 4.7 * $10^{-7}$M) and (E)-TCP($IC_{50}$ =7.8 * $10^{-8}$M),0.6 times as potent as (Z)- TCP ($IC_{50}$ = 4.4 * $10^{-8}$M). The results suggested that while without 1-methyl group, potency of a (Z)-isomer was comparable to that of (E)-isomer, the methyl group in its (Z)-position was very unfavorable to the inhibition of MAO and that in its (E)-position, the methyl group contributed positively to the potency as found by the fact that (E)-MTCP was 1-5 times more potent than (E)-TCP. In view of the selective inhibition of MAO-A- or B over MAO-A and 1-methyl substitution as well as the stereochemical factors did not significantly influence the selectivity.

Bioactive Constituents from the n-Butanolic Fraction of Aruncus dioicus var. kamtschaticus

  • Six compounds were isolated from the n-BuOH fraction of the aerial parts of Aruncus dioicus var. kamtschaticus including: sambunigrin (1), prunasin (2), aruncide A (3), aruncide C (4), 1-O-caffeoyl-${\beta}$-D-glucopyranose (5), and caffeic acid (6). Their structures were confirmed by comparing the spectral data with those reported in the literature. The isolated compounds (1 - 6) were then examined for their cytotoxic effects towards MCF-7, HL-60, and HeLa cancer cell lines, as well as their DPPH radical scavenging activity. The results indicated that compound 4 possessed the strongest inhibitory effect toward HeLa cell line with $IC_{50}$ value of $5.38{\pm}0.92{\mu}M$. Compound 3 possessed selective cytotoxic activity on HL-60 cells with $IC_{50}$ value of $6.27{\pm}0.17{\mu}M$, compound 5 was found as the best in inhibiting proliferation with $IC_{50}$ value of $2.25{\pm}0.09{\mu}M$, and the other compounds showed significant inhibition with $IC_{50}$ values ranging from 6.10 to $11.27{\mu}M$. Compound 5 also displayed the strongest cytotoxic effect toward MCF-7 cell line ($IC_{50}$ $4.32{\pm}0.15{\mu}M$). Both 5 and 6 demonstrated strong radical scavenging activity ($IC_{50}$ $6.87{\pm}0.03$ and $4.33{\pm}0.22{\mu}M$, respectively). Compounds 1 and 5 were isolated for the first time from this plant.

Comparison of Analysis Methods for Ammonia from Swine Production Facilities

  • This study was performed to evaluate the accuracy, validation and applicability of UV spectrophotometer (UV), Ion Chromatography (IC), and Detector tube (DT) methods for measuring ammonia (NH3) concentration in a swine confinement house and swine slurry storage tank. The mean values of $NH_{3}$ emitted from the house and slurry were 5.333 ppm and 42.192 ppm for the IC method; 4.13 ppm and 36.29 ppm for the Detector tube; and 5.417 ppm and 34.193 ppm for the UV method. The accuracy and the correlation of an ammonia level analyzed by the IC method compared to the UV method were 98% and 0.998($R^{2}$) in the swine confinement house and 94% and 0.997($R^{2}$) in the swine slurry storage tank. On the other hand, those of ammonia level measured by the Detector tube compared to the UV method were 77% and 0.957($R^{2}$) in the swine confinement house and 82% and 0.941($R^{2}$) in the swine slurry storage tank. This indicated that the accuracy and the correlation of the IC method compared to the UV method were higher than those of the Detector tube method compared to the UV method. Therefore, it was concluded that the IC method was more accurate in measuring ammonia concentration in a swine house and a swine slurry storage tank. The Detector tube method should not be applied to the swine slurry storage tank in which ammonia concentration is generally higher than 30 ppm because low accuracy is caused by a gross space between scales inscribed in the Detector tube.

The Effects of Alpinia katsumadai Extract on Anti-inflammation and Melanogenesis

  • Several solvent extracts from Alpinia katsumudai were prepared and their various anti-inflammatory activities were evaluated. Alpinia katsumadai extract showed high various anti-inflammatory effects among the 8 medicinal plant extracts, Butanol extract from Alpinia katsumndai showed a potent anti-oxidative and free radical scavenging activities, Free radical scavenging effect of butanol fraction of Alpinia katsumadai($IC_{50}$/ : 50$\mu$g/m$\ell$) was higher than butylated hydroxytoluene($IC_{50}$/ : 50$\mu$g/m$\ell$) and ascorbic acid($IC_{50}$/ : 22$\mu$g/m$\ell$). Alpinia katsumadai butanol fraction exhibited relatively high antioxidative activity($IC_{50}$/ : 335$\mu$g/m$\ell$) compared to ascorbic acid. The inhibitory effect of Alpinia katsumadal ethanol extract on elastase exhibited 10 to 78% at 100 to 1000$\mu$g/m$\ell$ concentration , tile $IC_{50}$/ values with 465.7$\mu$g/m$\ell$ for porcine pancreatic elastase(PPE) and 481.9$\mu$g/m$\ell$ for human leukocyte elastase(HLE), respectively The Alpinta katsumadai extract inhibited effectively hyaluronidase activity($IC_{50}$/ 335$\mu$g/m$\ell$), and showed inhibition in vitro on delayed hypersensitivity when it was topically applied. These results suggest that Alpinia katsumadai extract may reduce inflammatory skin trouble. The Alpinia katsumadai extract also showed higher Inhibitory effect of melanin biosynthesis on cultured melanoma cell compared to arbutin and kojic acid.
