신문(神門) 침 자극이 급성 코카인 투여로 인한 보행성 활동량과 뇌내 c-Fos 발현에 미치는 효과 (Effect of Acupuncture$(HT_7)$ on Acute Cocaine-induced locomotor Activity and Fos-like Immunoreactivity in the Brain of the Rats)

  • Substantial evidence suggests that behavioral and reinforcing effects of cocaine can be mediated by the mesolimbic dopaminergic system. Injections of cocaine can produce one of the immediate-early gene, c-fos expression in the brain and behavioral activation. Acupuncture as a therapeutic intervention is widely used for the treatment of many mental disorders such as drugs of abuse. In order to investigate whether acupuncture has an influence on cocaine-induced reinforcing and behavioral effects, we examined the effect of acupuncture on cocaine-induced locomotor activity and c-Fos expression in the nucleus accumbens and the striatum using Fos-like-immunoreactivity(FLI). Male SD rats received acupuncture for 1 min after injection of cocaine hydrochloride(1 mg/kg, i.v.). The employed acupuncture point, Shenmen$(HT_7)$, has been clinically used to treat mental and psychosomatic disorders. Injections of cocaine produced a marked increase in locomotor activity and FLI in the nucleus accumbens and the striatum. Acupuncture at $HT_7$, but not at control points($PC_6,\;TE_4$ or tail), significantly attenuated cocaine-induced increase in locomotor activity and Fos-like immunoreactivity. These results demonstrated that reduction in locomotor activity by acupuncture may be reflected by reduction of postsynaptic neuronal activity in the nucleus accumbens and the striatum. Our results suggest that acupuncture may have a therapeutic effect on cocaine addiction.

질환모델마우스 생산을 위한 체세포핵이식방법 개선; I. 탈핵 및 재조합난자 생산기법 최적화 (Improvement of Somatic Cell Nuclear Transfer Technology for the Production of Disease Model Mouse: I. Optimization of Oocyte Enucleation and Reconstruction)

  • This study was undertaken to optimize enucleation and reconstitution methods for the production of cloned mice by somatic cell nuclear transfer Outbred ICR mouse oocytes at the metapahse- II stage were retrieved from female mice superovulated by PMSG and hCG. In Experiment 1, oocytes were enucleated in medium supplemented with cytochalasin B (CCB) of 3 levels (0, 7.5 or 15 $\mu\textrm{g}$/mL), and higher rate of encleation was obtained at 7.5 and 15 $\mu\textrm{g}$/mL than at $\mu\textrm{g}$/mL. In Experiment 2, oocytes enucleated in 7.5 $\mu\textrm{g}$/mL CCB-containing medium were reconstituted with different types of somatic cell by following methods; 1) cumulus cells by direct cell injection, 2) cumulus cells by electric fusion (1.25 kV/cm, 2 pulses for each 70 $mutextrm{s}$) or 3) STO cells by the electrofusion. Electrofusion of STO cells with enucleated oocytes yielded the greatest (P<0.05) rate of reconstitution without lysis (76%) than any other combinations. Although significant decrease in the rate of somatic cell introduction was found, the electrofusion of cumulus cells yielded better rate of reconstitution than direct injection (0 vs. 18%). In Experiment 3, the duration of electric stimulation for the fusion was changed to either 50 $mutextrm{s}$ or 90 $mutextrm{s}$, but no significant improvement of reconstitution efficacy was obtained. In conclusion, this study showed that ICR mouse oocytes could be used for the production of reconstituted oocytes and a fusion method of 1.25 KV/cm with 2 pulses using 570 cell was the optimal.

고속액체크로마토그래피를 이용한 비타민 B5 및 B6의 정량 분석 (Quantitative Analysis of Vitamin B5 and B6 Using High Performance Liquid Chromatography)

  • 식품 함유 비타민 $B_5$$B_6$의 최적 HPLC 분석 조건을 검토한 결과 비타민 $B_5$의 경우 YMC-Pack ODS-AM($250{\times}4.6mm$ I.D.) 칼럼을 이용하고, A용매로 50 mM $KH_2PO_4$(pH 3.5)을, B용매는 아세토니트릴을 이동상 용매로 사용하는 A용매 95% 등용매용리 조건에서 200 nm의 파장으로 분석하는 HPLC/DAD법을 최적조건으로 확립하였다. 한편 비타민 $B_6$의 최적분석조건은 여기파장(excitation) 290 nm, 방출파장(emission) 396 nm로 분석하는 HPLC/FLD법으로써, 칼럼은 YMC-Pack Pro RS $C_{18}$($250{\times}4.6mm$ I.D.), 이동상 용매는 A용매 20 mM $CH_3CO_2Na$(pH 3.6), B용매 아세토니트릴을 A용매 97% 등용매용리 조건으로 사용하였다. 비타민 $B_5$$B_6$의 표준검량선은 $R^2$값이 각각 0.9998 및 0.9999로 고도의 직선성을 나타내었고, 검출한계 및 정량한계는 비타민 $B_5$의 경우 각각 0.4 mg/L 및 1.3 mg/L, 비타민 $B_6$의 경우 각각 0.006 mg/L 및 0.02 mg/L로 산출되었다.

젖소 과배란 처리시 PEG(Polyethylene Glycol) 처리가 수정란 생산에 미치는 영향 (Effects of PEG on Embryo Production in Superovulated Hostein Cows)

  • 본 연구는 젖소에 있어서 다량의 수정란을 생산하여, 고능력의 유전인자를 조기에 확산할 수 있도록, 젖소에 있어서 PEG를 이용하여 과배란 처리기법 확립을 위해 실시하였으며, 그 결과는 다음과 같다. 1. 성선자극호르몬의 처리 방법에 따른 황체반응은 Folltropin 단독처리시 $5.8\%$, Folltropin+EB는 $20.6\%$, Folltropin+PEG는 $24.0\%$로 나타났으며, 유의적인 차이를 보였다(p<0.01). 2. 성선자극호르몬의 처리 방법에 따른 이식 가능 수정란 수에 미치는 영향은 Folltropin V 단독처리시 3.6개, Folltropin +EB 처리시 3.3개, Folltropin +PEG 8,000 처리시 2.8개이었으며, 유의적인 차이를 나타내지 않았다(p<0.01). 3. 성선자극호르몬의 처리방법에 따른 퇴행란과 미수정 란 Folltropin+EB와 Folltropin+PEG 처리구에서 Folltropin 단독처리보다 많이 회수되었으며,배반포 수정란의 수에 있어서는 처리간에 유의적인 차이를 보이지 않았다. 이상의 결과는 임의의 발정주기에 CIDR 삽입후 Folltropin의 1일 2회 4일간의 분할 투여법 뿐만 아니라 Folltropin을 PEG로 용해하여 1회 투여법을 이용한 과배란 처리기법도 젖소 공란우에서 수정란의 생산이 가능함을 보여준다.

헬리코박터 파이로리 균의 진단시약 개발을 위한 요소의 체내동태 및 호기 중 배설 (Pharmacokinetics and Excretion into Expired Air of Urea, a Potential Diagnosis Reagent of Helicobacter pylori Infection)

  • Purpose: The purpose of the present study was to investigate the pharmacokinetics of urea, a new potential diagnosis reagent of Helicobacter pylori infection. Methods: Considering the mechanism of urea breath test, we determined the excretion of urea in expired air after its oral administration in rats and beagle dogs at the dose of 2 mg/kg (including 50 mCi/mmol $^{14}C$-urea 50 ${\mu}Ci/kg$ for rats and 13.5 ${\mu}Ci/kg$ for dogs). Results: Urea was rapidly disappeared from the blood circulation by 1 hr after its i.v. bolus injection, followed by a slow disappearance by 24 hr. The half-lives at the distributive phase ($t_{1/2{\alpha}}$) and post-distributive phase ($t_{1/2{\beta}}$) were 2 min and 6 hr, respectively. The bioavailability of urea was 64.3% after its oral administration. The values of the volume of distribution ($V_{dss}$) and the total body clearance ($CL_t$) after the oral administration were compatible with those after i.v. administration. The recovery of urea in the bile was about 0.1% of the dose by 24 hr after its oral administration. Urea was extensively eliminated in the urine by 48 hr. The recovery ratios of urea in the urine and expired air were about 86.8% and 2.99% of the dose by 48 hr, respectively. Moreover, urea was mostly distributed from the blood circulation to the kidney, followed by being eliminated in the urine without metabolism. The concentration of urea in the kidney was 4.0 times higher than that of plasma at 40 min after its oral administration. Conclusions: These findings indicated that oral route appears to be available for the administration of urea. Orally administered urea, thus, was considered to be useful for the diagnosis of Helicobacter pylori infection.

Studies on the Analysis of Benzo(a)pyrene and Its Metabolites on Biological Samples by Using High Performance Liquid Chromatography/Fluorescence Detection and Gas Chromatography/Mass Spectrometry

  • An analytical method the determination of benzo(a)pyrene (BaP) and its hydroxylated metabolites, 1-hydroxybenzo(a)pyrene (1-OHBaP), 3-hydroxybenzo(a)pyrene (3-OHBaP), benzo(a)pyrene-4,5-dihydrodiol (4,5-diolBaP) and benzo(a)pyrene-7,8-dihydrodiol (7,8-diolBaP), in rat urine and plasma has been developed by HPLC/FLD and GC/MS. The derivatization with alkyl iodide was employed to improve the resolution and the detection of two mono hydroxylated metabolites, 1-OHBaP and 3-OHBaP, in LC and GC. BaP and its four metabolites in spiked urine were successfully separated by gradient elution on reverse phase ODS $C_{18}$ column (4.6 mm I.D., 100 mm length, particle size 5 ㎛) using a binary mixture of MeOH/H₂O (85/15, v/v) as mobile phase after ethylation at 90 ℃ for 10 min. The extraction recoveries of BaP and its metabolites in spiked samples with liquid-liquid extraction, which was better than solid phase extraction, were in the range of 90.3- 101.6% in n-hexane for urine and 95.7-106.3% in acetone for plasma, respectively. The calibration curves has shown good linearity with the correlation coefficients (R²) varying from 0.992 to 1.000 for urine and from 0.996 to 1.000 for plasma, respectively. The detection limits of all analytes were obtained in the range of 0.01-0.1 ng/mL for urine and 0.1-0.4 ng/mL for plasma, respectively. The metabolites of BaP were excreted as mono hydroxy and dihydrodiol forms after intraperitoneal injection of 20 mg/kg of BaP to rats. The total amounts of BaP and four metabolites excreted in dosed rat urine were 3.79 ng over the 0-96 hr period from adminstration and the excretional recovery was less than 0.065% of the injection amounts of BaP. The proposed method was successfully applied to the determination of BaP and its hydroxylated metabolites in rat urine and plasma for the pharmacokinetic studies.

흰쥐에서 배측 봉선핵의 전기자극이 췌장의 외분비기능에 미치는 영향 (Exocrine Pancreatic Secretion in Response to Electrical Stimulation of Dorsal Raphe Nucleus in Rats)

  • The present investigation was performed to see a possible influence of the dorsal raphe nucleus (DRN) on pancreatic exocrine secretion in anesthetized rats since the DRN had been known to exert a regulatory mechanism on sympathetic activity which was known to be very important for pancreatic exocrine secretion, particularly in rats. Twenty-nine Sprague-Dawley rats fasted for 24 hours were anesthetized by i.p. injection of 1 g/kg of urethane. The pancreatic duct was cannulated to collect pancreatic juice while bile juice was diverted into the jejunum. The duodenopyloric junction was tightly ligated. After surgery for collection of pancreatic exocrine secretion and recording of carotid blood pressure, a coaxial electrode was stereotaxically inserted in the DRN with a guide of a brain atlas. And then, electrical stimulus of biphasic square wave with 2 v, 2 msec, 40 Hz was applied on the electrode for 10 minutes. Pancreatic volume flow and protein output secreted in 10 min were measured. Either bilateral cervical vagotomy or spinal cord transection at the level of $C4{\sim}C5$ was performed 20 min prior to stimulation of the DRN. 1) Electrical stimulation of the DRN resulted in significant (p<0.05) increase in pancreatic volume flow and protein output. These stimulatory effects were not affected by cervical vagotomy but completely abolished by cervical cord transection. 2) Electrical stimulation of the DRN also resulted in significant (p<0.05) rise of blood pressure of the carotid artery. The hypertensive effect was not affected by cervical vagotomy but completely abolished by cervical cord transection. The results strongly suggest that the DRN, a part of the central serotonergic system, could exert a stimulatory influence on pancreatic exocrine secretion by increasing the sympathetic activity in anesthetized rats.

방사선 조사후 백서 공장 점막의 재생과정에서 5-fluorouracil 투여가 phospholipsse C 와 ras 암유전자단백의 발현에 미치는 영향 (The Influence of 5-Fluorouracil Administration Mode on the Expression of Phospholipase C and Ras Oncoprotein Associated with Regeneration of Rat Intestinal Mucosa Following Radiation)

  • Purpose : Phospholipase C(PLC) isozymes play significant roles in transmembrane signal transduction. PLC-${\gamma}1$ acts as the intracellular effector in signal transduction for cellular proliferation and differentiation. Ras oncoprotein is also involved in cell growth. We determined the biological significance of PLC and ras oncoprotein in regeneration following radiation and the effect of different modes of administration of 5-FU. Materials and Methods : To determine the effect of the administration mode of 5-FU on the regeneration of intestinal mucosa of rats following radiation, we compared the expression of PLC and ras oncoprotein in six groups. Group I had no treatment. Group II received radiation(8 Gy) only. Group III received radiation(8 Gy) and 5-FU(150mg/kg) continuous intravenous (iv) infusion for 12 hours. Group IV received radiation(8 Gy) and 5-FU(750mg/kg) iv bolus injection. Group V received only 5-FU(150mg/kg) continuous iv infusion for 12 hours, Group VI received only 5-FU (150mg/kg) iv bolus injection. Through immunoblotting and immunohistochemistry, we examined the expression of PLC and ras oncoprotein in rat jejunum at 96 hours after radiation or 5-FU administration and at 120 hours after radiation and 5-FU adminstration. We also investigated the histological findings using hematoxylin and eosin stain. Results : In the immunohistochemistry study, PLC-${\gamma}1$ expression was the highest in group III followed by groups II and VI in that order and was weakly positive in groups V and VI. PLC-${\gamma}1$ was hardly detected in the control group. The expression of ras oncoprotein was the same as the PLC-${\gamma}1$ expression for all groups. These results were confirmed by the histological findings regarding the mucosal regeneration. In the immunoblotting analysis, PLC-${\gamma}1$ expression was the highest in group III followed by group IV and II in that order. This difference between the immunoblotting and immunohistochemistry study was due to the high expression of PLC-${\gamma}1$ on the damaged surface epithelium rather than to its expression in the regeneration region as observed in the immunohistochemistry study for group IV. The expression of PLC-${\delta}1$ was positive only in group V and VI, which received both radiation and 5-FU, and the expression of PLC-${\beta}1$ was negligible for all groups. Conclusion : These results suggest that PLC-${\gamma}1$ mediated signal transduetion and ras oncoprotein may have a significant role in mucosal regeneration after radiation, and that continuous iv infusion of 5-FU may induce active regeneration in intestinal mucosa following radiation. In addition, the expression of PLC-${\delta}1$ in combined group of radiation and 5-FU implies that PLC-${\delta}1$ may be involved in signal transduction mediated by concerted action between radiation and 5-FU.

Capsaicin 적용 후 손상된 흰쥐 척수내 c-fos와 HSP70의 발현 (The expression of c-fos and HSP70 by the Capsaicin injection in the spinal cord(dorsal horn))

  • C-fos는 원종양유전자(proto-oncogene)인 v-fos의 세포 동족체로써, 성장인자나 신경전달 물질에 의해 수분 내에 다양한 형태의 세포에서 활성화된다. Fos단백질은 스트레스와 통증 과정의 신호전달기전에서 세포활동을 조절하는 3차전령으로 활동한다. 열충격 단백질(Heat shock protein : 이하 HSP)은 계통발생학적으로 초기 척추 동물에서부터 발현되며 생체방어체계의 중요한 인자로 세포가 고열, 외상, 허혈 등의 스트레스에 직면했을 때 발현이 증가하는 단백질로 알려져 있다. 본 연구에서는 캡사이신(capsaicin)으로 말초 신경병변을 유발시킨 후 통각신경활성의 지표로 이용되는 원종양 유전자인 c-fos의 발현과 열 또는 스트레스로 야기되는 손상에 대한 조직의 방어작용으로 발현되는 HSP 70의 발현을 동시에 관찰함으로서, 급성으로 유발된 말초 신경병변의 확인과 동시에 실험동물 체내에서 방어적인 역할을 밝히는 일환으로 이 실험을 실시하였다. 본 실험의 결과는 다음과 같다; 1. 척수 등쪽뿔 천층(Laminae I and II)에서 각각 c-fos와 HSP70을 항원으로 하는 면역조직화학적 방법으로 염색한 표본에서 0.9% NaCI 투여 2시간 후 c-fos와 HSP70의 양성을 나타내는 세포는 전혀 없음을 알 수 있었다. 2. 척수 등쪽뿔 천층에서 c-fos 단백질을 항원으로 하는 면역조직화학적 방법으로 염색한 표본에서 Capsaicin 투여 2시간 후 c-fos 단백질에 양성을 나타내는 세포가 많이 발현됨을 육안적 관찰로서 알 수 있었다. 3. 척수 등쪽뿔 천층에서 HSP70을 항원으로 하는 면역조직화확적 방법으로 염색한 표본에서 Capsaicin 투여 2시간 후 HSP7O의 양성을 나타내는 세포가 보통수준으로 발현됨을 육안적 관찰로서 알 수 있었다. 이 실험의 결과로 볼 때, 화학적인 신경병변 유발물질에 의한 손상을 방어하기 위해서 체내에는 내인성 물질이 형성될 것이라는 추측과 c-fos 가 다른 유전자의 발현을 유도한다는 점을 함께 고려 하였을때, Capsaicin에 의한 말초 신경병변에서 c-fos 발현이 많이 나타나는 것은 손상을 방어하는 물질의 생성에 관여하기 때문이며, 방어물질 중 이 실험에서 본 HSP70도 증가한 내인성 방어물질의 하나라고 할 수 있을 것이다.

원종 및 교잡종에 대한 미립자의 병원성에 관한 연구 (Studies on Pathogenicity of Nosema bombycis Naegeli to Parents and Hybrid in the Silkworm (Bombyx mori L.))

  • 미립자 병원체의 병원성에 대한 연구로서 그 결과를 요약하면 다음과 같다. 1. 품종별 감잠비율에 있어서 원종은 교잡종보다 훨씬 높았으나 원종간에는 차이가 없었다. 2. 첨식농도별, $10^{5}$ /ml<$10^{6}$/ml<$10^{7}$ /ml<$10^{8}$ /ml순으로 감잠비율이 높았다. 3. 영별로는 5령<4령<3령<2령 순으로 감잠비율이 높았다. 4. 품종과 농도 사이의 감잠비율에 대한 교호작용에 있어서 1) 고농도($10^{8}$ /ml)에서는 원종과 교잡종간에는 차이가 없었으며 2) 저농도($10^{5}$ /ml)에서는 교잡종은 원종보다 낮았으나 원종간에는 차이가 없었다. 3) 중간농도($10^{7}$ /ml, $10^{6}$/ml)에서는 교호작용이 엇갈려 원종의 $10^{6}$/ml과 교잡종의 $10^{7}$ /ml에서 교호작용이 같았다. 5. 면역에 있어서 N. bombycis포자의 면역방법은 항원 주사량, 면역기간 및 주사간격이 중요하며 특히 항원 주사량은 20ml($10^{8}$ spores/ml)이상이 필요한 것으로 생각되며 4처리구(A.B.C.D)중에서 D구가 제일좋았다. 6. N. bombycis의 생존포자를 면역원으로 해서 토끼에 면역시키면 간접법 염색에 의한 항체가에서 1 : 12, 800의 항체가 N. bombycis 면역혈청을 얻었으며, 잠체조직 및 N. bombycis의 포자는 자기 형광을 볼 수 있었으나 U.V. filter로 관찰하게 되면 FITC의 형광색을 명확히 식별할 수 있었다. 7. 미립자 병잠의 소직학적 관찰에서 미립자 포자는 누에 조직 중에서도 제일 먼지 중위피막조직에 기생하는 것을 알 수 있었다.
