Studies on the Development of Antihyperlipidemic Drugs from Oriental Herbal Medicines(II) - Antihyperlipidemic Effects of Oriental Herbal Medicines - (한방약물로부터 항고지혈증 치료약물개발(2) - 수종 한약재의 항고지혈증 효과 -)

  • In the previous reports, we selected 80% MeOH extract of 7 herbs including Scutellariae Radix(SR), Paeoniae Radix Rubra(PRR), Moutan Cortex(MC), Angelicae Gigantis Radix(AGR), Crataegi Fructus(CF), Bambusae Caulis in Taeniam(BCT) and Cinnamomi Ramulus(CR), which exhibited the inhibitory effect on HMG-CoA reductase and DPPH free radical scavenging effect in vitro, and antihyperlipidemic effects on antihyperlipidemic rats induced by Triton WR 1339 in vivo. Among them, SR, MC, AGR and BCT showed significant suppression of elevated serum LDL-cholesterol level, and AGR and CF showed significant liver weight increase on high cholesterol diet induced hyperlipidemic mice. And, SR, PRR, AGR, BCT and CR significantly suppressed the elevated serum total cholesterol and triglyceride levels on corn oil induced hyperlipidemic rats. Then, in order to research new antihyperlipidemic agents from the oriental medicinal herbs, we chose SR, AGR, CR and BCT which have the antihyperlipidemic effect in vitro and in vivo, and those herbs were systematically fractionated with organic solvent. EtOAc fraction of SR, hexane fraction of BCT, AGR and chloroform fraction of CR exhibited remarkably inhibitory effect on HMG-CoA reductase activity.

Effects of Feeding the Mixture of Linseed and Sunflower Seed Oil on the Fatty Acid Composition of Serum Lipoprotein in Dietary Hyperlipidemic Rats (아마인유와 해바라기 종자유의 혼합급이가 식이성 고지혈증 흰쥐 혈청 지단백의 지방산조정에 미치는 영향)

  • This study was carried out to investigate the effect of the feeding mixture of linseed oil, rich in n-3 PUFA and the sunflower seed oil, rich in n-6 PUFA on the lipid metabolism in the dietary hyperlipidemic rats. After male Sprague-Dawley rats were induced hyperlipidemia by feeding the diet containing lard, butter and cholesterol for 3 weeks, then they were fed with the diet containing lard 3 .0% and butter 12.0% for control, the mixture in different proportion of both linseed oil and sunflower seed oil and antihyperlipidemic drugs for 2 weeks. Analysis of the fatty acid composition of the serum lipoprotein fractions showed following results. In the fatty acid composition of serum lipoprotein , the proportion of C18:2 was dominant in all fractions, C20:5 in LDL and HDL fraction and C22:6 in chylomicron fraction. The ratio of n-3P/n-6P tended to increase gradually as it of the test lipid increased in groups 5 to 9 group and was affected by the fatty acid composition of the test lipids.

  • This study was carried out to investigate the effect of the feeding mixture of linseed oil, rich in n-6 PUFA on the lipid metabolism in the dietary hyperlipidemic rats. After male Sprague-Dawley rats were induced hyperlipidemia by feeding the diet containing lard, butter and cholesterol for 3 weeks. then they were fed with the diet containing lard 3.0% and butter 12.0% for control, the mixture in different proportion of both linseed oil and sunflower seed oil, and antihyperlipidemic drugs for 2 weeks. Analysis of the fatty acid composition of the brain and heart lipids showed following results. In the fatty acid composition of brain lipids, C20:4 and C22:6 were the major fatty acids but showed little difference among the groups. In the fatty acid of heart lipids,C18:2 was the major fatty acid. The proportion of C20:4 decreased gradually as n-3P/n-6P ratio of the test lipids increased in groups 5 (linseed oil 12.0%) to 9 (sunflower seed oil 12.0%) while the proportion of C22:6 was not affected by the fatty acid composition of the test lipids.

Effects of Kamibangpungtongsungsan on Hypertension in Spontaneous Hypertensive Rats Fed a Hyperlipidemic Diet (고cholesterol 식이 자발성 고혈압 백서(白鼠)에서의 가미방풍통성산(加味防風通聖散)이 고혈압에 미치는 영향)

  • This experiment was performed to determine the effects of Kamibangpungtongsungsan on hypertension in spontaneous hypertensive rats fed a hyperlipidemic diet (H-SHR). The results are summarized as follows : 1. Kamibangpungtongsungsan significantly decreased the blood pressure and pulse of rats in H-SHR. 2. Kamibangpungtongsungsan significantly decreased the levels of Aldosterone in H-SHR. 3. Kamibangpungtongsungsan significantly decreased the levels of dopamine and epinephrine in H-SHR, but did not significantly reduce the levels of norepinephrine, in H-SHR. 4. Kamibangpungtongsungsan did not significantly reduce the levels of electrolytes in H-SHR. 5. Kamibangpungtongsungsan significantly decreased the TNF-$\alpha$ and IL-6 levels in H-SHR but the IL-10 level increase no significant. These results suggest that the Kamibangpungtongsungsan might be usefully applied for the treatment of hypertension with hyperlipidemia.

Effects of Onion and Naringin Extract Mixed Products on Antihyperlipidemic Levels of Bio FIB Hamster (FIB 햄스터에서 양파즙과 나린진 추출 혼합물의 항고지혈증 효과 -투과전자현미경적 관찰-)

  • This study investigated the effects to lowering hyperlipidemic levels in hamsters by feeding them naringin and onion extracts. The hyperlipidemic levels on all of the hamsters were raised by feeding them with high fat atherogenic diet (HFAD) containing 10% coconut oil and 0.05% cholesterol. The experimental groups were randomly assigned to 5 groups (normal, control, OEN, OES, and OESN) each 6 hamsters over a 40 days period. The control group (HFAD) received a 10% coconut oil and 0.05% cholesterol with basal diet, the OEN group which received an onion extract mixture contaning 50% water and 50% onion extract, the OES group which received 100% onion extract, the OESN group which received 100% onion extract combined with 1% naringin. Serum cholesterol and triglyceride levels significantly decreased in the naringin treated group while the HDL cholesterol level increased. On the other hand, the morphology of the mitochondria and nuclei found in the livers of the treated group given naringin were siginficantly different when compared to those group given high fat diets. In conclusion, onion extract with naringin decreased cholesterol levels in hamster's livers.

Anti-hyperlipidemic activity of Rhynchosia nulubilis seeds pickled with brown rice vinegar in mice fed a high-fat diet

  • The abnormal content of blood lipids often results in metabolic diseases, such as hyperlipidemia and obesity. Many agents, including natural sources from traditional food, have been developed to regulate the blood lipid contents. In this study, we examined the anti-hyperlipidemic activity of Rhynchosia nulubilis seeds pickled with brown rice vinegar (RNSpBRV), a Korean traditional pickled soybean food. Since RNSpBRV is made of R. nulubilis seeds (RNS) soaked in brown rice vinegar (BRV), we compared the anti-adipogenic activity between RNS, BRV and solid fraction of RNSpBRV (SF-RNSpBRV), liquid fraction of RNSpBRV (LF-RNSpBRV). For this, the inhibitory effect of lipid accumulation in 3T3-L1 adipocyte was checked by adding methanol extracts of mixed RNS and BRV, LF-RNSpBRV, and SF-RNSpBRV. The addition of each methanol extract up to 1 mg/ml showed no cytotoxicity on 3T3-L1 adipocyte, and approximately 20% of the lipid droplet formation was suppressed with the methanol extract of BRL or SF-RNSpBRV. The highest suppression (42.1%) was achieved with LF-RNSpBRV. In addition, mice fed a high fat diet (HFD) supplemented with 5% RNSpBRV powder led to increased high density lipoprotein (HDL) cholesterol and lower blood glucose, triglyceride, and total cholesterol compared to mice fed with a HFD diet only. Interestingly, the size of the epididymis cells gradually decreased in HFD + 1% RNSpBRV and HFD + 5% RNSpBRV-fed mice if compared those of HFD-fed mice. Taken together, these results provide evidence that RNSpBRV has a regulatory role in lipid metabolism that is related to hyperlipidemia.

Lactic Acid Bacteria Increases Hypolipidemic Effect of Crocin Isolated from Fructus of Gardenia jasminoides

  • The fructus of Gardenia jasminoides Ellis (GF) has been widely used as a natural colorant in Asian countries, and also as a Chinese traditional medicine for its homeostatic, antiphlogistic, analgesic, and antipyretic effects. In the present study, its main component, crocin, was fermented with lactic acid bacteria (LAB) and their antihyperlipidemic activity was measured. The GF extract, fermented GF (F-GF), crocin, and fermented crocin (F-crocin) significantly inhibited the increase of serum triglyceride (TG) level in corn oil feeding-induced triglyceridemic mice, as well as that of serum TG and total and LDL cholesterol levels in Triton WR-1339-induced hyperlipidemic mice. These agents also showed hypolipidemic activity in hyperlipidemic mice induced by high fat diet for 5 weeks. F-GF and F-crocin exhibited more potent hyperlipidemic effects than GF and crocin. The results suggest that the hypolipidemic effect of GF and crocin can be synergistically activated by LAB, and that F-GF and F-crocin may improve hyperlipidemia in clinic, compared with GF and crocin.

Effect of Brassica rapa L. extracts and ${\beta}-sitosterol$ on hyperlipidemic rats (순무와 ${\beta}-sitosterol$의 고지혈증 억제에 대한 연구)

  • To evaluate the anti-hyperlipidemic effect of Brassica rapa L. and its major compound, $\beta$-sitosterol, the present study was undertaken, hypercholesterolemia was induced in rats with poloxamer-407, Triton WR-1339, 30% corn oil high cholesterol diet. The ethanol extract of Brassica rapa L. significantly decreased total cholesterol (TC), phospholipid, triglyceride at doses of 200 mg/kg, and $\beta$-sitosterol significantly exerted anti-hyperlipidemic activity at a dose of 15 mg/kg in rats with hyperlipidemia. Taken together, Brassica rapa L. and $\beta$-sitosterol can be useful agents for the prevention or treatment of hyperlipidemia.

Effects of Schisandra Fructus hexane fraction on high fat diet induced hyperlipidemic mice (오미자(五味子) 헥산 분획 추출물이 고지방 식이에 의한 고지혈증 생쥐의 지질대사 및 간 조직 유전자 발현에 미치는 영향)

  • ■ Objectives The berries of Schisandra chinensis (Schisandra Fructus) are given the name Omiza in Koreane(五味子), and have been used asremedies for many ailments: to resist infections, increase skin health, and combat insomnia, coughing, and thirst. This study was designed to investigate the effects of Schisandra Fructus hexane fraction (SFH) on serum lipid levels in Hyperlipidemic mice. ■ Methods In this experiment, effects on total cholesterol, HDL-cholesterol, triglyceride, AST, ALT, fasting blood glucose in serum were measured. And in addition, histopathological and gene expression changes in liver tissue was also observed. ■ Results SFH did not affects weight gain, serum AST and ALT in hyperlipidemic mice. Oral administration of SFH lowered levels of total cholesterol and triglyceride, which were elevated by induction of hyperlipidemia. Finally, administration of SFH lowered fasting blood glucose significantly. And SFH also ameliorates anti-oxidative stress systems in internal organs which play key role in disease prevention. ■ Conclusion Results in our study suggest that SFH can prevent obese through regulation of dyslipidemia and hyperglycaemia.

Effects of Mahwangimun-tang in Cholesterol Diet and Triton WR-1339 -induced Hyperlipidemic Rats (마황이문탕(麻黃二門湯)이 흰쥐의 고지혈증 병태모델에 미치는 영향)

  • This study was designed to investigate the hyperlipidemia-healing effect of aqueous extract from Mahwangimun-tang(麻黃二門湯) and it was performed on the hyperlipidemia of rats induced by high cholesterol diet and Triton WR-1339. We prepared two types of hyperlipidemia model in rats induced by high cholesterol diet and Triton WR-1339. The amount of total cholesterol, HDL-cholesterol, LDL -cholesterol, triglyceride, phospholipid, free fatty acid, the activity of superoxide dismutase and glutathione were measured. The aqueous extract from Mahwangimun-tang showed significant decreasing effects on total cholesterol, LDL- cholesterol, triglyceride in the both model. And it showed significant increasing effects on HDL-cholesterol, the activity of SOD in both model. These results suggest that aqueous extract from Mahwangimun-tang has healing efficacy on hyperlipidemia induced by high cholesterol diet and Triton WR-1339.
