장기저장을 위해 제조한 동결건조 산채 블록의 항산화 활성 변화 (Change of Antioxidant Activities in Preparing Freeze Dried Wild Vegetable Block for the Long-term Storage)

  • 곰취(Ligularia fischeri, GC), 고추잎(Capsicum annuum L., GCY), 취나물(Aster scaber, CNM), 머위대(Petasites japonicus S. et Z. Max, MYD) 및 고구마순(Ipomoea batatas L. (Lam), GGM)과 같은 산채들의 물 추출물의 항산화능력을 평가하고 이들 동결건조 블록 물 추출물들의 항산화력과 비교하였다. 산채 물 추출물들과 그들의 동결건조 블록물 추출물들의 항산화력 측정은 2,2-diphenyl-1-picrylhydrazyl(DPPH) radical 소거작용, hydroxyl radical 소거작용 및 아질산염 소거작용과 같은 방법에 의해 알아보았다. 산채 물 추출물은 그들의 동결건조 물 추출물보다 총 페놀함량이 더 높았다. GC, GCY, CNM, MYD 그리고 GGM 물추출물들의 총 페놀 함량은 각각 $471.66{\pm}3.52\;{\mu}g/mg,\;141.33{\pm}2.51\;{\mu}g/mg,\;177.33{\pm}2.88\;{\mu}g/mg,\;238.66{\pm}9.50\;\mu}g/mg\;그리고\;122.67{\pm}3.51\;{\mu}g/mg$이었다. 1000 ppm GC, GCY, CNM 그리고 GGM 물 추출물의 DPPH radical 소거작용은 그들의 동결건조 블록 물 추출물보다 더 높았고, 1000 ppm CNM, GC, GCY, MYD 그리고 GGM의 물 추출물의 DPPH radical 소거작용은 각각 90.9%, 89.9%, 76.6%, 71.1% 그리고 57.4%였다. 10000 ppm GC, GCY, CNM, MYD 그리고 GGM 물 추출물들은 hydroxyl radical 소거작용을 각각 38.8%, 33.4%, 35.9%, 34.3% 그리고 33.8%까지 증가시켰고, GCY, CNM 그리고 GGM의 물 추출물은 동결건조 블록 물 추출물과 유사한 활성을 나타내었으나 GC와 MYD의 물 추출물이이들 동결건조 블록들의 물 추출물의 hydroxyl radical 소거작용보다 약간 더 영향력이 있었다. 산채 물 추출물들과 이들 동결건조 블록 물 추출물들은 실험된 모든 농도에서 DPPH radical 소거작용 및 hydroxyl radical 소거작용을 나타내었다. GC 물 추출물의 아질산염 소거작용은 현저하게 농도 의존적으로 증가하였고, GC 물 추출물의 아질산염 소거작용이 그것의 동결건조 블록 물 추출물의 아질산염 소거작용보다 높았다. 이상의 결과들로부터 동결건조블록이 산채와 비교하여 산채가 가지고 있는 항산화력을 유지하고 있다는 것을 알수 있었다.

환삼덩굴로부터 분리한 Luteolin-7-O-$\beta$-D-Glucoside의 활성산소 소거능 (Active Oxygen Scavenging Activity of Luteolin-7-O-$\beta$-D-Glucoside Isolated from Humulus japonicus)

  • We investigated the active oxygen scavenging activity and active compound from Humulus japonicus. The ethyl acetate and butanol fraction exhibited strong scavenging effects on hydroxyl radical. Furthermore active compound was isolated and purified using Amberlite XAD-2 column chromatography and preparative HPLC from ethyl acetate fracton, and major compound was identified as a luteolin-7-O-$\beta$-D-glucoside by the MS, UV, 1H-NMR and 13C-NMR analyses. Luteolin-7-O-$\beta$-D-glucoside exhibited strong scavenging effect on active oxygens such as superoxide radical, hydroxyl radical and hydrogen peroxide.

Radical Scavenging Activities of Korean Traditional Rice Wine, Takju

  • The principal objective of this study was to assess the radical scavenging activities and total polyphenols, flavonoids, and flavonol contents of takju, a Korean traditional rice wine. The antioxidant properties of the wine and takju samples were evaluated using five distinct assays: specifically, 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2-azino-di-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS); hydroxyl; superoxide anion; and nitric oxide (NO) radical-scavenging activity assays. In this study, the takju evidenced strong scavenging activities against the hydroxyl, superoxide anion, and NO radicals. Furthermore, the total polyphenol contents of T-2 were similar to those previously observed in white wine (p<0.05). However, the flavonoids and flavonol contents of all takju samples were significantly (p<0.05) lower than that of white wine. The results of this study show that takju possesses a powerful radical scavenging activity against a variety of oxidative systems. The findings of this study also indicate that takju should be considered a useful antioxidant, and that their functional compound reduces oxidative stress.

Effect of Schizandra chinensis Extracts on Oxidative Damage

  • In this study, we evaluated the protective effects of supercritical extracts and two step ethanol extracts after supercritical extraction from Schizandra chinensis on antioxidant activities and oxidative DNA and cell damages. Supercritical extracts removed DPPH (1,1-diphenyl-2-picryldrazyl) radical by 85.5% at 200 ${\mu}g$/ml, but showed low activities of scavenging and chelating the hydroxyl radical and ferrous iron. However, two step ethanol extracts showed low activities of scavenging the DPPH radical, but removed the hydroxyl radical by 86% at 200 ${\mu}g$/ml. In addition, we tested the activities of extracts for reducing hydroxyl radical-induced DNA and cell damage. Two step ethanol extracts showed protective effect against the oxidative DNA damage by reducing DNA segmentation, inhibiting DNA migration and decreasing the expression of phospho-H2AX. Also, two step ethanol extracts showed protective effect against the oxidative cell damage by inhibiting lipid peroxidation and increasing the expression of p21 protein. Taken together, we suggest that two step ethanol extracts from S. chinensis have a role as useful inhibitors against oxidative damages.

연자육(Lotus Nelumbo nucifera Seed) 단백질로부터 항산화 펩타이드 분리 정제 및 특성 (Purification and Characterization of Antioxidant Peptides from Lotus Nelumbo nucifera Seed Protein)

  • Lotus Nelumbo nucifera seed protein (LSP) was isolated by alkaline solubilization after removing fat and phenolics by hexane and ethanol treatment. Antioxidant peptides from LSP were produced with Alcalase® and pepsin and hydroxyl radical scavenging activities were determined. LSP-Alcalase® hydrolysates showed higher hydroxyl radical scavenging activity than LSP-pepsin hydrolysates. To purify antioxidant peptides, LSP-Alcalase® hydrolysates were subjected to high performance liquid chromatography (HPLC) separation on the C18 column and the active fraction was further purified using a SuperdexTM peptide 10/300 GL column. Finally, the active fraction (F8-2) was evaluated for antioxidant activities by 2,2-diphenyl-1-picrylhydrazyl (DPPH), hydroxyl radical scavenging, and oxygen radical absorbance capacity (ORAC) assays. The EC50 values of the F8-2 were 105.81±0.02 ㎍/mL for DPPH and 32.26±0.02 ㎍/mL for hydroxyl radical and the F8-2 exhibited 7.22 μM trolox equivalent (TE)/100 ㎍ F8-2. Glutathione (GSH), which is a positive control, showed EC50 values of 19.87±0.01 ㎍/mL for DPPH and 15.95±0.03 ㎍/mL for hydroxyl radical and an ORAC value of 14.17±0.03 μM TE/100 ㎍ GSH. Finally, sixteen peptides were identified by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Among them, Ile-Tyr and Leu-Tyr showed higher antioxidant scores.

인삼 및 산양삼의 항산화 효능 비교 -Superoxide radical과 Hydroxy radical 소거활성을 중심으로- (A study on the comparison of antioxidant effects among cultivated ginseng, and cultivated wild ginseng extracts -Using the measurement of superoxide and hydroxy radical scavenging activities-)

  • Objectives : The objective of this study was to compare the antioxidant effects among cultivated wild ginseng and ginseng extracts. Methods : In vitro antioxidant activities were examined by superoxide and hydroxyl radical scavenging activities of ginseng and cultivated wild ginseng extracts. Results : 1. In the superoxide radical scavenging activities of ginseng and cultivated wild ginseng extracts, antioxidant activities of cultivated wild ginseng extracts was showed higher than cultivated ginseng in the concentration of 0.25 and $0.50mg/m{\ell}$. 2. In the hydroxyl radical scavenging activities of ginseng and cultivated wild ginseng extracts, antioxidant activities of cultivated wild ginseng extracts was showed higher than cultivated ginseng in the concentration of 1.0, 2.5, and $5.0mg/m{\ell}$. Conclusions : In summary, the results of this study demonstrate that cultivated wild ginseng extracts had higher antioxidant activities to cultivated ginseng.

동양 면요리 육수의 항산화 활성의 비교 (Comparison of Antioxidant Activities of Various Meat Broths Served with Oriental Noodles)

  • 본 연구에서는 한국, 중국, 일본 등의 대표적 면요리 육수의 항산화 활성을 검토하였다. 이를 위하여, 한국식, 중국식, 일본식 육수를 각각 제조하여, ESR과 비색법을 이용하여 DPPH, hydroxyl, peroxyl, superoxide, ABTS 등의 radical 소거 활성을 측정함으로써 항산화력을 비교하였다. 한국식 육수, 중국식 육수 및 일본식 육수의 DPPH radical 소거 활성은 2 $mg/m{\ell}$의 농도의 실험에서 2.78%, 25.37% 및 40.74%의 소거 활성을 보였다. 한국식 및 중국식 육수는 hydroxyl radical 소거 활성은 매우 낮았으나, 일본식 육수는 0.5 $mg/m{\ell}$에서 59.06%로 소거 활성이 높았다. 한국식, 중국식 및 일본식 육수의 peroxy radical 소거 활성($IC_{50}$)은 각각 0.121 $mg/m{\ell}$, 0.222 $mg/m{\ell}$ 및 0.013 $mg/m{\ell}$으로 일본식 육수가 peroxy radical 소거 활성이 높았다. 한국식, 중국식 및 일본식 육수의 ABTS radical 소거 활성의 $IC_{50}$ 값은 1.2 $mg/m{\ell}$, 1.8 $mg/m{\ell}$ 및 1.249 $mg/m{\ell}$였다.

발아 특수미의 항산화 활성 (Antioxidative Activity of Germinated Specialty Rices)

  • 발아에 의한 거대배아미와 유색미의 기능성 변화를 항산화 활성에 중심을 두고 조사하였다. 전반적으로 환원력은 일반현미보다 거대배아미와 유색미가 높았고, 발아처리는 환원력을 증가시키는 경향이 있었다. 지질과산화에 대한 억제효과를 조사한 결과, in vitro의 linoleic acid peroxidation system 및 ex vivo의 토끼 적혈구 막지질 과산화 system 모두에서 억제효과도 유색미가 가장 높았고, 그 다음이 거대배아미, 일반현미의 순서로 나타났다. Superoxide radical에 대한 소거활성은 유색미>거대배아미>일반현미의 순서로 낮아졌으며, 무발아 조건과 비교할 때 발아처리는 라디칼에 대한 소거활성을 증진시켰다. Hydroxyl radical 소거활성도 유색미가 가장 높았으며, 그 다음이 거대배아미, 일반현미의 순서였다. 발아에 의하여 일반현미의 hydroxyl radical 소거활성을 급격히 증가되었지만, 발아미간에 소거활성의 순위는 변하지 않았다. In vitro에서 조사된 ROS 소거활성과 같은 경향이 HL-60 세포에서 TPA로 자극으로 발생된 ROS에 대한 소거활성에서도 나타났다. 실험 결과, 유색미와 거대배아미에서는 발아와 연관되어 세포독성의 증가가 발견되지 않는 차별적인 ROS 소거활성의 증가 현상이 발견되었다.

Antioxidant and Free Radical Scavenging Potential of Justicia gendarussa Burm. Leaves in vitro.

  • Antioxidant activity of 70% aqueous ethanolic extract of leaves of Justicia gendarussa (EJ) was evaluated. EJ was prepared by cold maceration method. The antioxidant potency of EJ was investigated employing various established in vitro systems, such as DPPH radical scavenging, nitric oxide (NO) scavenging, ${\beta}-carotene$ linoleic acid module system (${\beta}$ CLAMS), hydroxyl (OH) radical scavenging, anti lipid peroxidation. $IC_{50}$ values were determined in each experiment. Also, ferric ion reduction capacity of extracts in presence and absence of chelating agent (EDTA) and total antioxidant capacity were determined. Preliminary phytochemical investigation was carried out to know the nature of constituents present in the leaves and correlate it with antioxidant activity. Further total phenolic content was determined in EJ. $IC_{50}$ values of EJ were 123.09 ${\pm}$ 3.01, 643.0 ${\pm}$ 61.10, 132.3 ${\pm}$ 6.03, 68.5 ${\pm}$ 11.5 and 68.13 ${\pm}$ 1.38 ${\mu}g/mL$ in DPPH radical scavenging, NO scavenging, ${\beta}$ CLAMS, OH radical scavenging and anti lipid peroxidation activity respectively. In total antioxidant capacity assay, ascorbic acid equivalent value was found to be 205.56 ${\pm}$ 4.69 ${\mu}g/mg$ of extract. Total phenolic content was found to be 43.76 ${\pm}$ 4.27 ${\mu}g$ equivalent of gallic acid per mg of extract. Phytochemical investigation reveals the presence of flavonoids. The results indicate that EJ possess antioxidant activity and flavonoids are responsible for this activity.

해조류 추출물 및 활성성분의 라디칼 소거능 (Radical Scavenging Effect of Methanol Extracts from Seaweeds and Their Active Compounds)

  • The radical scavenging activity of methanol extracts of seaweeds and their active compounds, alginic acid, fucoidan and phloroglucinol, were investigated under in vitro. Among methanol extracts of seaweeds (sea mustard, sea tangle, seaweed papulosa, fusiforme, sea lettuce, purple laver and chlorella), seaweed papulosa and sea tangle showed strong scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and hydroxyl radical (${\cdot}OH$). In addition, under in vitro, the scavenging activities on DPPH radical of alginic acid and fucoidan, which are active compounds of brown algae, and phloroglucinol, the active compound from Ecklonia species, were evaluated and compared. Fucoidan and phloroglucinol showed strong DPPH scavenging effect, in particular, phloroglucinol had strongest activity among the active compounds. On the other hand, alginic acid did not exert DPPH scavenging activity. From the present study, we could confirm the antioxidative activity of seaweeds and its active compounds.
