Sustainable Production of Dihydroxybenzene Glucosides Using Immobilized Amylosucrase from Deinococcus geothermalis

  • The amylosucrase encoding gene from Deinococcus geothermalis DSM 11300 (DgAS) was codon-optimized and expressed in Escherichia coli. The enzyme was employed for biosynthesis of three different dihydroxybenzene glucosides using sucrose as the source of glucose moiety. The reaction parameters, including temperature, pH, and donor (sucrose) and acceptor substrate concentrations, were optimized to increase the production yield. This study demonstrates the highest ever reported molar yield of hydroquinone glucosides 325.6 mM (88.6 g/l), resorcinol glucosides 130.2 mM (35.4 g/l) and catechol glucosides 284.4 mM (77.4 g/l) when 400 mM hydroquinone, 200 mM resorcinol and 300 mM catechol, respectively, were used as an acceptor substrate. Furthermore, the use of commercially available amyloglucosidase at the end of the transglycosylation reaction minimized the gluco-oligosaccharides, thereby enhancing the target productivity of mono-glucosides. Moreover, the immobilized DgAS on Amicogen LKZ118 beads led to a 278.4 mM (75.8 g/l), 108.8 mM (29.6 g/l) and 211.2 mM (57.5 g/l) final concentration of mono-glycosylated product of hydroquinone, catechol and resorcinol at 35 cycles, respectively, when the same substrate concentration was used as mentioned above. The percent yield of the total glycosides of hydroquinone and catechol varied from 85% to 90% during 35 cycles of reactions in an immobilized system, however, in case of resorcinol the yield was in between 65% to 70%. The immobilized DgAS enhanced the efficiency of the glycosylation reaction and is therefore considered effective for industrial application.

Effects of Aqueous Extracts from Naturalized and Korean Wild Plants on Seed Germination and Seedling Growth of Zoysiagrass (자생식물과 귀화식물의 수용 추출액이 Zoysiagrass의 발아와 유근생장에 미치는 영향)

  • The soil pH favored by Korean wild plants is 5.33~7.20, while naturalized invader species prefer soils of pH 3.95~6.48. The germination rate of Zoysia japonica was inhibited sharply, up to 60%, in extracts of naturalized plants of concentrations over 50%. Erigeron canadensis extract most strongly inhibited germination, while the Korean wild species, Cassia mimosoides var. nomame increased germination rates in concentrations of 30% and 50%. The seedling growth of Zoysia japonica in extracts of Korean wild species and naturalized species did not show differences, but was inhibited strongly, up to 60%, in the extract of Cassia mimosoides var. nomame concentration of over 15%. Analysis of the extract from Cassia mimosoides var. nomame identified procatechuic acid, $\rho$-hydroquinone, $\rho$-coumaric acid and ferulic acid at 254nm; and vanillic acid, hydroquinone, benzoic acid and cinnamic acid were discovered at 284nm. The seed germination and seedling growth of Zoysia japonica were investigated under different concentrations of phenolic compounds. Ferulic acid and vanillic acid were associated with an increased germination rate, while $\rho$-coumaric acid appeared to inhibit seedling growth.

Determination of Reorganization Energy from the Temperature Dependence of Electron Transfer Rate Constant for Hydroquinone-tethered Self-assembled Monolayers (SAMs)

  • The temperature dependence on the electron transfer rate constant $(k_{app})$ for hydroquinone redox center in $H_2Q(CH_2)_n$SH-SAMs (n = 1, 4, 6, 8, 10, and 12) on gold electrode was investigated to obtain reorganization energy $(\lambda)$ using Laviron’s formalism and Arrhenius plot of ln $[k_{app}/T^{1/2}]$ vs. T^{-1} based on the Marcus densityof-states model. All the symmetry factors measured for the SAMs were relatively close to unity and rarely varied to temperature change as expected. The electron tunneling constant $(\beta)$ determined from the dependence of the $k_{app}$ on the distance between the redox center and the electrode surface gives almost the same $\beta$ values which are quite insensitive to temperature change. Good linear relationship of Arrhenius plot for all $H_2Q(CH_2)_n$SH-SAMs on gold electrode was obtained in the temperature range from 273 to 328 K. The slopes n Arrhenius plot deduced that $\lambda$ of hydroquinone moiety is ca. 1.3-1.4 eV irrespectively of alkyl chain length of the electroactive SAM.

Electrodeposition of Graphene-Zn/Al Layered Double Hydroxide (LDH) Composite for Selective Determination of Hydroquinone

  • A graphene-Zn/Al layered double hydroxide composite film was simultaneously prepared by electrochemical deposition on the surface of a glassy carbon electrode (G-LDH/GCE) from the mixture solution containing GO and nitrate salts of $Zn^{2+}$ and $Al^{3+}$. The modified electrode showed good electrochemical performances toward the simultaneous electrochemical detection of hydroquinone (HQ), catechol (CA) and resorcinol (RE) due to the unique properties of graphene (G) and LDH such as large active surface area, facile electronic transport and high electrocatalytic activity. The redox characteristics of G-LDH/GCE were investigated with cyclic voltammetry and differential pulse voltammetry. The well-separated oxidation peak potentials, corresponding to the oxidation of HQ, CA and RE, were observed at 0.126 V, 0.228 V and 0.620 V respectively. The amperometric response of the modified electrode exhibited that HQ can be detected without interference of CA and RE. Under the optimized conditions, the oxidation peak current of HQ is linear with the concentration of HQ from 6.0 ${\mu}M$ to 325.0 ${\mu}M$ with the detection limit of 0.077 ${\mu}M$ (S/N=3). The modified electrode was successfully applied to the direct determination of HQ in a local tap water, showing reliable recovery data.

Effects of Reducing Agents on Textural Changes and Protein Modification in Extruded Wheat Gluten (압출성형시 환원제 첨가에 의한 밀가루 글루텐의 조직 변화와 단백질의 변성)

  • Addition of reducing agents during extrusion markedly affected physical and chemical properties of wheat flour and gluten extrudates. Expansion at the die was increased for wheat flour and gluten extrudates. Organic materials containing sulfur were evaporated as a flavor from gluten at the die and total sulfur contents were decreased. Physical shape was different for gluten extrudates without reducing agents. It was difficult to form the long strand of gluten extrudate without cooling die. Hydroquinone accelerated cell breakdown and produced more irregular shape of extrudate. However, addition of cysteine decreased the cell breakdown and produced the long strand of gluten extrudates. Chemical reactions of reducing agents such as cysteine and hydroquinone were different for high content (<80%) of wheat gluten. It was assumed that reducing agents donated hydrogen to inhibit the formation of disulfide crosslinking, decreased the dough strength and produced the broken cell and irregular shape of extrudates. Whereas, cysteine reacted as a binder as well as reducing agent and formed long strands. The evidence of reaction of reducing agents was shown from the fact that non-protein disulfide was increased and protein disulfide was slightly decreased from cysteine added gluten extrudate.

Effect of Thermal Treatment on the Electrocatalytic Activities and Surface Roughness of ITO Electrodes

  • The electrocatalytic activities and surface roughness of indium-tin-oxide (ITO) electrodes have been investigated after thermal treatment at 100, 150, or $200^{\circ}C$ for 30 min, 2 h, or 8 h. To check electrocatalytic activities, the electrochemical behavior of four electroactive species (p-hydroquinone, $Ru(NH_3){_6}^{3+}$, ferrocenemethanol, and $Fe(CN){_6}^{4-}$) has been measured. The electron transfer rate for p-hydroquinone oxidation and ferrocenemethanol oxidation increases with increasing the incubation temperature and the incubation period of time, but the rate for $Ru(NH_3){_6}^{3+}$ is similar irrespective of the incubation temperature and period because $Ru(NH_3){_6}^{3+}$ undergoes a fast outer-sphere reaction. Overall, the electrocatalytic activities of ITO electrodes increase with increasing the incubation temperature and period. The surface roughness of ITO electrodes increases with increasing the incubation temperature, and the thermal treatment generates many towering pillars as high as several tens of nanometer.

Amperometric Detection of Hydroquinone and Homogentisic Acid with Laccase Immobilized Platinum Electrode

  • DeniLite$^{TM}$ laccase immobilized platinum electrode was used for amperometric detection of hydroquinone (HQ) and homogentisic acid (HGA) by means of substrate recycling. In case of HQ, the obtained sensitivity is 280 nA/ ${\mu}$M with linear range of 0.2-35 ${\mu}$M ($r^2$ = 0.998) and detection limit (S/N = 3) of 50 nM. This high sensitivity can be attributed to chemical amplification due to the cycling of the substrate caused by enzymatic oxidation and following electrochemical regeneration. In case of HGA, the obtained sensitivity is 53 nA/ ${\mu}$M with linear range of 1-50 $[\mu}M\;(r^2$ = 0.999) and detection limit of 0.3 ${\mu}$M. The response times ($t_{90%}$) are about 2 seconds for the two substrates and the long-term stability is 60 days for HQ and around 40-50 days for HGA with retaining 80% of initial activities. The very fast response and the durable long-term stability are the principal advantages of this sensor. pH studies show that optimal pH of the sensor for HQ is 6.0 and that for HGA is 4.5-5.0. This shift of optimal pH towards acidic range for HGA can be attributed to the balance between enzyme activity and accessibility of the substrate to the active site of the enzyme.

A Study on the Synthesis and Curing Characteristics of PPG-type Polyurethane Methacrylates (PPG계 Polyurenthane Methacrylates의 합성과 경화특성)

  • PPG and BPA-type polyurethane prepolymers(NCO terminated) were obtained from bisphenol A and four types of PPG, having different molecular weight and numbers of functional groups. PPG and BPA-type polyurethane dimethacrylates were synthesized by reacting PPG and BPA-type polurethane prepolymer with 2-HEMA respectively. PPG-type polyurethane dimethacrylates were formulated with initiator(CHP), inhibitor(hydroquinone) and reactive diluent(TEGDMA). The effect of formulation on the torque changes was studied. Four kinds of PPG-type polyurethane dimethacrylates, having different molecular weight and numbers of functional group, were formulated with the same amount of CHP, TEGDMA and hydroquinone. The effects of the molecular weight of PPG-type polyurethane dimethacrylate and functional group numbers of PPG-type polyurethane dimethacrylates on the torque were investigated. These results showed that the torque of PPG-type polyurethane dimethacrylates, having same numbers of functional group, increased with decreasing molecular weight of dimethacrylates and torque of PPG-type polyurethane dimethacrylate, having similar molecular weight, was increased with increasing the number of fuctional group. The glass-transition temperature(Tg) of gels obtained by thermosetting cure for the four kinds of PPG-type polyurethane dimethacrylates were measured by DSC and molecular weights between cross-links(Mc) were calculated from Tg changes.
