수경재배 인삼잎 첨가 개성주악의 항산화 활성 및 품질 특성 (Antioxidant Activity and Quality Characteristics of Gaeseong-Juak prepared with Hydroponic-cultured Ginseng Leaf Powder)

  • This study aimed to evaluate the utilization of the top of a low intake of root, through the analysis of the antioxidant activity of the powder of hydroponic-cultured ginseng. Quality characteristics and antioxidant activity were compared and analyzed with Korea's traditional dessert Gaeseong-Juak, which is made of the powder of hydroponic-cultured ginseng's leaf by adjusting the added volume. DPPH radical scavenging activity of the hydroponic-cultured ginseng by the part powder was in the following order: highly stem, leaf and root. It was measured as 67.9%, 42.9%, and 25.9% at the 1 mg/m level. Gaeseong-Juak was prepared by adding hydroponic-cultured ginseng leaf powder at 0%, 0.3%, 0.6%, and 0.9% of the rice powder. As the content of hydroponic-cultured ginseng leaf powder increased, moisture content, L-value, and a-value were significantly decreased, while the b-value was significantly increased. The texture profile analysis of Gaeseong-Juak was not significantly different among the samples. In the sensory test, the sample containing 0.6% hydroponic-cultured ginseng leaf powder achieved good scores. The DPPH radical scavenging activity of Gaeseong-Juak was significantly increased, as the addition level increased, compared to the original. Based on the above results, hydroponic-cultured ginseng leaf was verified to be a possible natural antioxidant. It can increase food's nutritional values and possibilities when made of hydroponic-cultured ginseng, using leaf which is added to the traditional dessert Gaeseong-Juak.

발효에 의한 수경재배 인삼 잎의 항산화 및 간 보호 효과 (Antioxidant and Hepatoprotective Effects of Hydroponic-cultured Ginseng Folium by fermentation)

  • Objectives : Positive effects of Ginseng has great research attentions such as anticancer, anti-diabetic, antiaging, liver, immune function, CNS, etc. In this study, we investigated Hydroponic-cultured Ginseng Folium fermented byBacillus subtilisto establish fermentation conditions for enhancing functionality.Methods : Ginseng Folium were cultivated hydroponic-cultured and were extracted with methanol. We inoculateBacillus subtilisfor fermentation by adding to 0%, 3% and 5% sugar respectively and checked antioxidant activities, total phenolic content and total saponin content in 2 days intervals during 11 days. The antioxidant activities were studied by the 1,1-diphenyl-2-picryl hydrazyl(DPPH) radical, 2, 2'-Azino-bis(3-ethylbenzothiazoline-6 sulfonic acid) diammonium salt(ABTS) radical scavenging assay and Reducing power assay. We analyzed the Total phenol content, crude saponin content and ginsenoside content. Moreever, Hepatoprotective effects by Glutamic oxaloacetic transaminase(GOT) and Glutamic pyruvic transaminase(GPT) in Sprague-Dawley rat.Results : The results of DPPH and ABTS were 66.89% and 96.72%, respectively. The reducing power was resulted in optical density of 0.7312 with 3% sugar after 9 days of fermentation. and the concentration at 200 ㎍/㎖. Total phenol content was 36.92㎎/g with 3% sugar after 9 days of fermentation, in which crude saponin content wasn't changed, and ginsenoside content such as Rg3, Re and Rb was increased. Activities of GOT and GPT concentration were decreased in rat.Conclusions : This study suggests that hydroponic-cultured Ginseng Folium fermented byBacillus subtilisin 9 days showed significant efficacy of hepato-protection as well as antioxidant compared to the others. In addition, it shows not only improved value but also utilized hydroponic-cultured Ginseng Folium by fermentation.

Optimization of the extraction process of high levels of chlorogenic acid and ginsenosides from short-term hydroponic-cultured ginseng and evaluation of the extract for the prevention of atopic dermatitis

  • Background: Short-term hydroponic-cultured ginseng (sHCG), which is 1-year-old ginseng seedlings cultivated for 4 weeks in a hydroponic system, is a functional food item with several biological effects. However, the optimal extraction conditions for sHCG, and the bioactivity of its extracts, have not been evaluated. Methods: Chlorogenic acid (CGA) and ginsenoside contents were evaluated in sHCG, white ginseng (WG), and red ginseng (RG) using high-performance liquid chromatography. Response surface methodology (RSM) was used to optimize the extraction conditions (temperature and ethanol concentration) to maximize the yield of dry matter, CGA, and four ginsenosides (Re, Rg1, Rb1, and Rd) from sHCG. The optimal extraction conditions were applied to pilot-scale production of sHCG extracts. The expression levels of tumor necrosis factor (TNF)-α/interferon (IFN)-γ-induced thymic and activation-regulated chemokines (TARC/CCL17) were measured after treatment with sHCG, WG, and RG extracts, and the effects of their bioactive compounds (CGA and four ginsenosides) on human skin keratinocytes (HaCaTs) were evaluated. Results: CGA and four ginsenosides, which are bioactive compounds of sHCG, significantly inhibited TNF-α/IFN-γ-induced TARC/CCL17 expression. The optimal sHCG extraction conditions predicted by the RSM models were 80 ℃ and 60% ethanol (v/v). The sHCG extracts produced at the pilot scale under optimal conditions greatly alleviated TNF-α/IFN-γ-induced TARC/CCL17 production compared with WG and RG extracts. Conclusions: Pesticide-free sHCG extracts, which contain high levels of CGA and the ginsenosides Re, Rg1, Rb1, and Rd as bioactive compounds, may have therapeutic potential for atopic diseases.

팽이 및 수경인삼 분말 및 요구르트 발효에 의한 γ-Aminobutyric acid (GABA)의 전환효율 증진 (Effect of Conversion Rate of γ-Aminobutyric acid (GABA) by Yogurt Fermentation with Addition of Nanoparticle Winter Mushroom and Hydroponic Ginseng)

  • 팽이버섯 내에 존재하는 GAD 효소를 발효를 통해 활성화 시켜 MSG를 GABA로의 전환율을 높이고자 하였다. 효과적인 고농도 GABA를 생산하기 위해 나노분말 팽이버섯에다가 수경재배한 인삼을 첨가하여 야쿠르트발효기에서 발효한 결과 GABA 전환율은 팽이나노인삼분말 발효군(88%) > 팽이분말 발효군(52%) > 팽이나노분말 발효군(44%) 순으로 나타났다. 이러한 결과는 MSG를 기질로 첨가하는 식품에서 활용할 가치가 있으리라 사료된다.

Hydroponic and Organically Grown American Ginseng

  • A new cultural practice for commercial production of American ginseng by organically grown in a shaded greenhouse under controlled environment conditions has been developed. This new procedure will shorten the dormant period and reduce the cultivation period from 4 years in conventionally grown ginseng to 2 years.

Biocontrol of Korean Ginseng Root Rot Caused by Phytophthora cactorum Using Antagonistic Bacterial Strains ISE13 and KJ1R5

  • In this biocontrol research, we evaluated disease suppressive effects of antagonistic bacterial strains ISE13 and KJ1R5 against Korean ginseng root rot caused by P. eaetorum. We also examined the effects of nutrient solution in the hydroponic culture system for Korean ginseng on biological activity of the bacterial strains. As results of dual culture tests of the bacterial strains on $V_{8}$ juice agar, the strain ISE13 showed antifungal activity against P. eaetorum and other plant pathogenic fungi, but the strain KJ1R5 did not. When their inhibitory effects against infection of P. eaetorum on the roots grown in either nutrient solution or water were tested, the strains ISE13 and KJ1R5 inhibited the disease severity of Korean ginseng roots only grown with water, compared to buffer-treated, inoculated controls. However, the nutrient solution used for hydroponic cultures of ginseng in pots caused higher levels of disease severity by the strains ISE13 and KJ1R5 from 418.8\%$ to 40.0\%$ and from 24.3\%$ to 45.0\%$, respectively. In this study, the bacterial strains ISE13 and KJ1R5 could be potentially biocontrol agents to suppress Korean ginseng root rot caused by P. eaetorum. However, more attention using nutrient solution in hydroponic cultures for Korean ginseng production should be applied in biocontrol of plant diseases using the antagonistic microorganisms.

Glycosyl glycerides from hydroponic Panax ginseng inhibited NO production in lipopolysaccharide-stimulated RAW264.7 cells

  • Background: Although the aerial parts of hydroponic Panax ginseng are reported to contain higher contents of total ginsenosides than those of roots, the isolation and identification of active metabolites from the aerial parts of hydroponic P. ginseng have not been carried out so far. Methods: The aerial parts of hydroponic P. ginseng were applied on repeated silica gel and octadecylsilane columns to yield four glycosyl glycerides (Compounds 1-4), which were identified based on nuclear magnetic resonance, infrared, fast atom bombardment mass spectrometry, and gas chromatography/mass spectrometry data. Compounds 1-4 were evaluated for inhibition activity on NO production in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. Results and conclusion: The glycosyl glycerides were identified to be (2S)-1-O-7(Z),10(Z),13(Z)-hexadecatrienoyl-3-O-${\beta}$-$\small{D}$-galactopyranosyl-sn-glycerol (1), (2S)-1-O-linolenoyl-3-O-${\beta}$-$\small{D}$-galactopyranosyl-sn-glycerol (2), (2S)-1-O-linolenoyl-2-O-linolenoyl-3-O-${\beta}$-$\small{D}$-galactopyranosyl-sn-glycerol (3), and 2(S)-1-O-linoleoyl-2-O-linoleoyl-3-O-${\beta}$-$\small{D}$-galactopyranosyl-sn-glycerol (4). Compounds 1 and 2 showed moderate inhibition activity on NO production in LPS-stimulated RAW264.7 cells [half maximal inhibitory concentration ($IC_{50}$): $63.8{\pm}6.4{\mu}M$ and $59.4{\pm}6.8{\mu}M$, respectively] without cytotoxicity at concentrations < $100{\mu}M$, whereas Compounds 3 and 4 showed good inhibition effect ($IC_{50}$: $7.7{\pm}0.6{\mu}M$ and $8.0{\pm}0.9{\mu}M$, respectively) without cytotoxicity at concentrations < $20{\mu}M$. All isolated compounds showed reduced messenger RNA (mRNA) expression of interleukin-$1{\beta}$ (IL-$1{\beta}$), IL-6, and tumor necrosis factor-${\alpha}$ in LPS-induced macrophage cells with strong inhibition of mRNA activity observed for Compounds 3 and 4.

온도 및 생육시기에 따른 수경재배 인삼의 생장특성과 수량에 관한 연구 (Studies on Growth Responses and Yields of Panax ginseng C. A. Meyer Grown under Hydroponic Culture with different Temperatures and Growth Stages)

  • This study was carried out to have the basic and applied informations relating to increase the productivity and quality of ginseng. 2 years-old ginseng was cultivated under hydroponic culture with the controlled environment conditions in a greenhouse. Major growth characters and yields were investigated with two different temperatures and several growth stages. The plant height and stem diameter were higher at low temperature than those at high temperature. They were not clearly different with six different growth stages. The root length was not clearly different between two temperatures; however it was continuously grown from June until August. The root diameter was higher at low temperature than that at high temperature. It was rapidly increased from June until August. The length, width, and area of leaf were higher at low temperature than those at high temperature. The fresh and dry weights of different plant tissues were also heavier at low temperature than those at high temperature. The moisture content of ginseng root was continuously decreased from June until August. The yield of ginseng was higher at low temperature compared to that at high temperature. The cultivating conditions in hydroponic culture of ginseng, especially temperature, would be an important factor to have better growth and production.

토직, 상토 및 양액육묘에 의해 생산된 묘삼의 본포 이식 후 생장특성 (Growth Characteristics of Ginseng Seedling Transplanting by Self Soil Nusery, Nursery or Hydroponic Culture on Main Field)

  • Background: The production method of ginseng seedlings for ginseng cultivation is very important to ensure healthy rooting system as well as high quality, and yield of the resultant plants. This study was carried out to compare the growth characteristics of 2-year-old ginseng plants that were produced from seedlings grown in self soil nursery (SSN), nursery soil (NS) or hydroponic culture (HC). Methods and Results: The shading prop used was composed of four-layered 4 polyethylene (blue 3 + black 1) shade screen. The management of main field was done by inserting oil cake (1,200 kg/10 a) and then allowing Sudan grass to grow for a year. Seedling transplantation was carried out on April 6. Root growth was measured on October 25. Root weight was observed to be excellent at 6.0 g, following SSN transplantation. Root length was 21.2 cm for HC seedlings, but these plants had a physiological disorder (i.e., rusty root), in 83.5% plants of this treatment. The ratio of PD/PT (protopanaxadiol saponins / protopanaxatriol saponins) was higher in NS seedlings. Plant analysis revealed that Fe content was lower in HC seedlings with high rustiness. The growth of 2-years-old ginseng was different following these varying seedling cultivation methods, but seedlings from NS were not different from those grown in SSN. Conclusions: For the propagation of 2-year-old ginseng plants, NS seedlings may be a good substitute for SSN seedlings.

Comparison of Ginsenoside and Phenolic Ingredient Contents in Hydroponically-cultivated Ginseng Leaves, Fruits, and Roots

  • In this study, hydroponically-cultivated ginseng leaves, fruits, and roots were respectively extracted with ethanol. The contents of 12 ginsenosides and three phenolics in the extracts were quantitatively analyzed and the free radical scavenging activities were measured and compared. Hydroponically-cultivated ginseng leaves contained higher levels of gensenosides (Rg1, Rg2+Rh1, Rd, and Rg3) and p-coumaric acid than the other parts of the ginseng plants. The 2,2'-azino-di-(3-ethylbenzothiazoline)-6-sulfonic acid radical scavenging activities of leaves were also the highest. Accordingly, hydroponically-grown ginseng leaves were shown to hold promise for use as an environmentally-friendly natural anti-oxidant.
