A Study on the High Content Ceramide Stabilization Formulation with Cyclodextrin (사이클로덱스트린을 함유한 고함량 세라마이드 안정화 제형 연구)

  • In this study, we would like to study the stabilization of the high content of ceramide formulation by containing cyclodextrin. Ceramide, which constitutes the intercellular lipid, a human skin barrier, is a very important ingredient in moisturizing maintenance by protecting moisture in the skin and strengthening the skin barrier. However, since ceramide is poorly soluble, even if it is included in the cosmetic formulation, it has a problem that it is slowly gelled or crystallized and deposited over time, making it difficult to containing a high amount of ceramide. Cyclodextrin is a cyclic oligosaccharide connected with glucose molecules and has a cylindrical structure with hydrophilic outer surface and hydrophobic inner surface, which is known to improve the physicochemical properties of drugs such as improving solubility and absorption of poorly soluble drugs. We demonstrated the stability of the formulation containing high amount of ceramide by measuring hardness and observing emulsion drops with polarized microscope. This study also demonstrated that the high-content ceramide formulation containing cyclodextrin has the effect of preventing gelation or crystallization of ceramide, thus having excellent environmental conditions stability and skin moisturization.

Targeting HSP90 Gene Expression with 17-DMAG Nanoparticles in Breast Cancer Cells

  • Background: Dysregulation of HSP90 gene expression is known to take place in breast cancer. Here we used D,L-lactic-co-glycolic acid-polyethylene glycol-17-dimethylaminoethylamino-17-demethoxy geldanamycin (PLGA-PEG-17DMAG) complexes and free 17-DMAG to inhibit the expression of HSP90 gene in the T47D breast cancer cell line. The purpose was to determine whether nanoencapsulating 17DMAG improves the anti-cancer effects as compared to free 17DMAG. Materials and Methods: The T47D breast cancer cell line was grown in RPMI 1640 supplemented with 10% FBS. Encapsulation of 17DMAG was conducted through a double emulsion method and properties of copolymers were characterized by Fourier transform infrared spectroscopy and H nuclear magnetic resonance spectroscopy. Assessment of drug cytotoxicity was by MTT assay. After treatment of T47D cells with a given amount of drug, RNA was extracted and cDNA was synthesized. In order to assess HSP90 gene expression, real-time PCR was performed. Results: Taking into account drug load, IC50 was significant decreased in nanocapsulated 17DMAG in comparison with free 17DMAG. This finding was associated with decrease of HSP90 gene expression. Conclusions: PLGA-PEG-17DMAG complexes can be more effective than free 17DMAG in down-regulating of HSP90 expression, at the saesm time exerting more potent cytotoxic effects. Therefore, PLGA-PEG could be a superior carrier for this type of hydrophobic agent.

Preparation and Characterization of Conducting Composites Impregnated with Thick Polyheterocyclic Polymers (전도성 복합소재의 합성과 특성연구)

  • Light-weight conductive polymer composites were prepared by incorporating polyheterocycles such as polypyrrole and polythiophene into pores of a highly porous cross-linked polystyrene, host polymer, to form a conductive network. The highly hydrophobic and porous host polymer was synthesized by concentrated emulsion polymerization method. Polypyrrole-based composites, prepared by employing ferric chloride-methanol system, showed a conductivity as high as 0.82 S/cm. Conductivity of polythiophene-based composites, prepared from ferric chloride-acetonitrile system, was 6.05 S/cm. Conductivity of compositivity was influenced by the initial molar ratio of oxidant to monomer as well. SEM micrographs of the composites showed that conducting polymer coated uniformly the inside wall of the porous host polymer. Shielding effectiveness of the polypyrrole-based composites and of the polythiophene-based composites were 15.2 dB and 22.5 dB at 2.0 GHz, respectively. In the temperature range from 20 to 300K, a polypyrrole impregnated composite exhibited seimiconducting behavior and followed the variable range hopping(VRH) model for charge transport.

Synthesis and Characterization of High Molecular Weight Biodegradable Polyoxalate (고분자량 생분해성 폴리옥살레이트의 합성과 특성분석)

  • Biodegradable polymers have gained enormous attentions in the pharmaceutical and biomedical applications, especially in drug delivery. In this work, we report the synthesis and characteristics of high molecular weight polyoxalate with ~75000 Da. Hydrolytic degradation kinetics and degradation products were characterized by nuclear magnetic resonance and gel permeation chromatography. Polyoxalate is a semicrystalline and thermally stable polymer with a glass transition temperature of ${\sim}35^{\circ}C$, which is suitable for drug delivery applications. The hydrophobic nature of polyoxalate allows it to be formulated into nanoparticles and encapsulate drugs using a conventional oil-in-water emulsion/solvent displacement method. Polyoxalate nanoparticles also exhibited excellent cytotoxicity profiles. It can be suggested that polyoxalate has great potential for numerous biomedical and pharmaceutical applications.

Influence of Graphene Oxide and Graphite Nanoplatelets on Rheological and Electrical Properties of Polystyrene Nanocomposites (산화 그래핀과 나노 흑연이 폴리스티렌 나노복합재료의 유변물성 및 전기적 물성에 미치는 영향)

  • Carbon-based nanoplatelets such as graphene oxide (GO) sheets and graphite nanoplatelets (GNPs) are frequently used as conductive nanofillers for polymer nanocomposites. In this study, polystyrene (PS)/GO and PS/GNP nanocomposites were prepared through a latex technology and investigated to compare the effect of nanofillers on rheological and electrical properties of the PS nanocomposites. PS particles were prepared by emulsifier-free emulsion polymerization and GO was synthesized by using the modified Hummers' method from graphite. Hydrophilic GO was dispersed in aqueous PS suspension, but hydrophobic GNPs were dispersed with the help of a surfactant. In comparison with PS/GO nanocomposites, the rheological properties of PS/GNP counterparts were not too high because GNP existed in aggregates of graphene layers. Conducting pathways of PS/GO and PS/GNP nanocomposites were achieved at the electrical percolation threshold of 0.50 and 5.82 wt%, respectively. The reason for enhanced electrical conductivity in PS/GO nanocomposites is that GO was thermally reduced during molding.

Characteristics of α-Tocopherol-loaded Nanostructured Lipid Carriers and their Stabilization Effect (α-Tocopherol을 함유한 Nanostructured Lipid Carriers의 특성과 안정화 효과)

  • Loading of hydrophobic ${\alpha}$-tocopherol into nanostructured lipid carrier (NLC) was performed for improving its oxidative stability. First, various NLCs with different constituents and mixing ratios were prepared and their characteristics were investigated. While the stable NLCs were made when cetyl palmitate (CP) or glyceryl monosterate (GMS) was used as a solid lipid, the phase separation occurred in the NLCs consisting of stearic acid. Particle sizes of the NLCs were several hundreds of nanometers and the size decreased with increasing the ratio of solvent to lipid. It was examined from DSC thermogram and anisotropy test that the degree of crystallinity of the lipid phase decreased and the lipid matrix became less ordered when octyldodecanol, a long chain fatty alcohol, was added into the solid lipid. The oxidative stability of ${\alpha}$-tocopherol in NLC was remarkably improved compared to that in solution or emulsion under high temperature ($45^{\circ}C$) and UV radiation, which was verified through DPPH test and peroxide value measurement.

REACTION OF PAPER PULP AND ALKYL KETENE DIMER BY AGING TREATMENT DURING PAPERMAKIN PROCESS

  • Alkylketene dimer was known as a cellulose reactive or alkaline size because it does not require to fix to the fiber as do the traditional rosin sizes. A proposed sizing mechanism of AKD was the formation of P -ketoester bond between AKD and cellulose which provides the permanent attachment and the orientation of the hydrophobic alkylchains outward. However, some questions about the reaction had arisen and thus, the sizing mechanism of AKD has been a subject of controversy for several decades. The major concern of the controversy is that AKD is really reactive with cellulose or not in the papermaking conditions. In this study, reaction between AKD and pulp fiber was investigated, in order to find out whether AKD forms P-ketoester with pulp fiber during aging under no catalyzed neutral condition with obvious spectroscopic evidence. In addition, effect of aging treatment on the sizing development was studied. It has been disclosed that, in absence of water, AKD reacted with cellulose to form P -ketoester linkage under no catalyzed neutral condition, while, in presence of water, most of AKD was hydrolyzed to a dialkyl ketone or P -ketoacid. In addition, during the aging treatment of AKD-sized paper, its typical IR spectra bands gradually were reduced, completely disappeared after 6hr aging, and formed new absorption bands at 1707cm-' and shoulder peak at 1700cm-' which refer to the typical dialkylketone absorption bands. Therefore, the formation of P -ketoester between AKD and pulp fiber is impossible in the practical papermaking process. It could be suggested that the sizing development of AKD-sized paper is obtained by next two mechanism: 1) formation of a thin-layer of AKD on the fiber surface through melting and spreading of AKD emulsion particles by heat and 2) the hydrolysis of AKD to dialkyl ketone which has higher melting point, during drying and storage of AKD sized papers.

Preparation and Characterization of Nano-sized Hydrogels (nanogels) Using Inverse-miniemulsion Polymerization Method for Protein Drug Delivery (단백질 약물 전달을 위한 Inverse-miniemulsion Polymerization 방법으로 제조하는 나노크기의 수화젤(나노젤)의 제조 및 특성평가)

  • Hydrogels are thought to be a promising delivery carrier for protein drugs because of their favorable aqueous environment compared with nano/micro-particles of hydrophobic polymer such as PLGA. In this study, nano-sized hydrogels (nanogels) were fabricated using inverse-miniemulsion polymerization method. The mean size of nanogels in range of 90-160nm and affected by the preparation parameters such as sonication time and concentration of monomer. While longer sonication time and lower concentration of acrylamide monomer showed a tendency to produce smaller nanogels and to have lower lysozyme activity, variation of bis-methylene acrylamide concentration made no difference. Although both longer soncaton time and lower acrylamide concentration increased in vitro release rate, acrylamide concentration was more effectively affected to the control of protein release rate, which indicated that the release rate of protein from nanogels affected by not only their size but also internal structure. In conclusion, nanogels prepared by inverse-miniemulsion can be a useful carrier for application of protein drug, because of simple process, minimum contact of organic solvent and high protein activity.

A Study on the Synthesis of Organic-Inorganic Hybrid Waterborne Polyurethane by Using Graft Type Siloxane Polyol (그래프트형 실록산 폴리올을 이용한 유-무기 하이브리드 수분산 폴리우레탄의 합성에 관한 연구)

  • Organic- inorganic hybrid waterborne polyurethane (PUD) is synthesized by using hybrid polyol consist of carbonate (PCD), ester (PCL), and siloxane (PDSBP) in order to enhance anti-scratch property of PUD film. The diameter of graft type PUD emulsion is bigger than that of linear type PUD due to the graft structure of hydrophobic siloxane chain. The glass transition temperature of linear type PUD increase and the decomposition temperature of linear type PUD decrease with the content of PCD polyol. While, the decomposition temperature of graft type PUD almost same with increasing PDSBP content. The anti-scratch property and pencil hardness of graft type PUD improves as adding PDSBP polyol in the hybrid polyol system. When 9 wt% of PDSBP polyol is mixed, PUD films shows excellent anti-scratch property (~3.3 N), and pencil hardness (> 9 H).

Formation of the multiamellar vesicles of AHAsomes and effect of removal on the horny layer (AHAsomes의 multilamellar vesicles형성과 각질제거 효과)

  • In this context, it should be mentioned that multilamellar vesicles can be prepared with the main compounds of the intercellular lipids, ceramides, cholesterol, cholesteryl ester, squalane, lecithin, wax ester by effect of the wetting. We investigated properties formation of MLV with use of the AHAsomes and Microfluidizer. The multilamellar vesicles are formed merely adding polyol and water phase, followed with the microfluidizer. Formation of a practically pure AHAsomes system, containing the active ingredients directly incorporated without need for preservatives. There were very good encapsulated properties of the active ingredients whether hydrophilic(malic acid, tartaric acid, lactic acid, allantoin, urea) and hydrophobic(vitamin-I acetate, vitamin-A palmitate). Optimum condition (ormatiom of MLV was passed three times in the microfluidizer, particle size of the vesicles should be within range 50-523nm (mean=163.5nm). As application, It was compared that horny layer of the sole of foots removal with the general OM emulsion and the AHAsomes cream. There was used for three months, those got recovery wrinkles about 151.8% and elasticity three times more the AHAsomes than O/W emulsions, It was confirmed with the Image Analyzer and the Cutometer.
