Background: Ginsenoside compound-Mc1 (Mc1) is a member of the deglycosylated ginsenosides obtained from ginseng extract. Although several ginsenosides have a cardioprotective effect, this has not been demonstrated in ginsenoside Mc1. Methods: We treated H9c2 cells with hydrogen peroxide (H2O2) and ginsenoside Mc1 to evaluate the antioxidant effects of Mc1. The levels of antioxidant molecules, catalase, and superoxide dismutase 2 (SOD2) were measured, and cell viability was determined using the Bcl2-associated X protein (Bax):B-cell lymphoma-extra large ratio, a cytotoxicity assay, and flow cytometry. We generated mice with high-fat diet (HFD)-induced obesity using ginsenoside Mc1 and assessed their heart tissues to evaluate the antioxidant effect and the fibrosis-reducing capability of ginsenoside Mc1. Results: Ginsenoside Mc1 significantly increased the level of phosphorylated AMP-activated protein kinase (AMPK) in the H9c2 cells. The expression levels of catalase and SOD2 increased significantly after treatment with ginsenoside Mc1, resulting in a decrease in the production of H2O2-mediated reactive oxygen species. Treatment with ginsenoside Mc1 also significantly reduced the H2O2-mediated elevation of the Bax:Bcl2 ratio and the number of DNA-damaged cells, which was significantly attenuated by treatment with an AMPK inhibitor. Consistent with the in vitro data, ginsenoside Mc1 upregulated the levels of catalase and SOD2 and decreased the Bax:B-cell lymphoma-extra large ratio and caspase-3 activity in the heart tissues of HFD-induced obese mice, resulting in reduced collagen deposition. Conclusion: Ginsenoside Mc1 decreases oxidative stress and increases cell viability in H9c2 cells and the heart tissue isolated from HFD-fed mice via an AMPK-dependent mechanism, suggesting its potential as a novel therapeutic agent for oxidative stress-related cardiac diseases.
For the induction of arthropathy, 4% hydrogen peroxide ($H_2O_2$) was injected for 4 weeks into the intra-articular space of the 25 New Zealand white rabbits to damage articular cartilage. The verification of arthropathy induction and the effect of scan-bio laser treatment were determined by measuring superoxide dismutase (SOD) activity, by observing gross and histopathologic findings. The SOD activity increased by about 40% in arthropathy group, as compared to controls. Although SOD activity in arthropathy group was not significantly different from the 2-week group, it was significantly different from the 4-week control and treatment groups. There was also a significant difference between the 4-week control and treatment groups. Grossly, erosions formed on the articular cartilage surface, and the lateral femoral condyle was damaged in arthropathy group. In comparison, there was slight, but not significant, progression of the lesion in the 2-week control group, and no difference between the 2-week treatment and control groups. Conversely, severe erosions damaged the articular cartilage in the 4-week control group. Cartilage proliferation was seen in gross observations in the 4-week treatment group, suggesting a treatment effect. Histopathologically, there was slight articular surface damage and apoptosis in arthropathy group, and serious cartilage damage, despite slight chondrocyte proliferation, in the 4-week control group. By contrast, the 4-week treatment group showed chondrocyte replacement, with close to normal articular cartilage on the articular surface. There was significant cartilage proliferation with regeneration of the articular cartilage on the articular surface in the group treated with low-level laser, as compared to control group, when arthropathy was induced by $H_2O_2$ injections. Therefore, low-level laser was effective in the treatment of chemically induced arthropathy.
쑥부쟁이는 국화과에 속하는 다년생 식물로서 다양한 질병의 예방 및 치료에 오랫동안 사용되어 왔다. 최근 연구에서 쑥부쟁이 잎 추출물이 항산화 및 항염증 효과가 있는 것으로 알려져 있지만, 정확한 효능 평가에 관한 연구는 여전히 미비한 실정이다. 본 연구에서는 RAW 264.7 대식세포에서 쑥부쟁이 잎 에탄올 추출물(EEAY)의 항산화 효능이 항염증 효능과 연관이 있는지의 여부를 조사하였다. 본 연구의 결과에 의하면, EEAY는 $H_2O_2$ 처리에 의한 RAW 264.7 세포의 세포 독성을 유의적으로 억제시켰으며, 이는 Nrf2 및 HO-1의 발현 증가와 관련이 있음을 보여 주었다. 또한 EEAY는 $H_2O_2$에 의한 apoptosis를 유의적으로 억제하였으며, 이는 caspase-3의 활성 억제에 따른 PARP의 분해 차단과 연관성이 있었다. 그리고 EEAY는 대표적인 항 염증성 사이토카인인 IL-10의 발현 및 생산을 증가시켰으며, 이는 전사 및 번역 수준에서의 TLR-4 및 Myd88 발현 증가와 관련이 있었다. 아울러 EEAY는 LPS에 의한 염증성 매개인자인 NO의 생성 증가를 현저히 억제하였으며, EEAY에 의한 NO 생성의 억제 효과는 HO-1 유도제인 hemin에 의해 더욱 증가되었다. 따라서 본 연구의 결과는 EEAY에 의한 산화적 및 염증성 스트레스에 대한 RAW 264.7 대식세포의 보호 효과에 최소한 Nrf2/HO-1 신호 경로의 활성화가 관여할 가능성을 보여주었다.
본 연구는 은행외종피 추출물의 항산화 및 DNA 손상 보호용 조성물에 관한 것으로 HaCaT 세포에서 은행외종피 물과 메탄올 추출물의 과산화수소의 공격에 대한 항산화 효과 및 DNA 손상보호에 대한 것이다. 이를 위해 은행외종피 물 추출물(GOSWE)와 은행외종피 메탄올 추출물(GOSME)의 항산화력을 위해서는 DPPH와 과산화수소 소거능을 실시하였다. GSOWE와 GOSME는 DPPH 소거능에서 다소 차이를 보였다. 더욱이, GOSME는 과산화수소 소거능에서 GOSWE보다 탁월한 효과를 발휘하였다. 1 mM $H_2O_2$ 처리된 HaCaT 세포의 세포생존실험에서 GOSME는 세포생존력을 향상시켰다. GOSME는 1 mM $H_2O_2$에 의한 플라스미드 DNA 단편화 저해능과 HaCaT세포에서 게놈 DNA 단편화 억제능이 있는 것으로 확인되었다. 이러한 결과는 GOSME는 활성산소제거를 통한 산화적 스트레스관련 세포손상 억제를 통한 세포사멸억제 및 과산화수소 유발 DNA 손상을 억제 할 수 있다는 것을 제시하였다. 결론적으로 GOSME는 산화적스트레스에 의한 피부질환에 대한 치료 및 방어제로 이용 될 수 있는 신소재로 평가 할 수 있다.
Objectives : Jasminum officinale L. var. grandiflorum is used as a traditional or folk remedy in China to treat arthritis, hepatitis, duodenitis, conjunctivitis, gastritis, and diarrhea. In this study, we aimed to study the hepatocyte protective activity and molecular mechanism of Jasminum officinale L. var. grandiflorum aqueous extract (JGW) using HepG2 hepatocyte cell lines. Methods : HepG2 cells were pretreated with diverse concentrations of JGW, and then the cells were exposed to tert-butyl hydroperoxide (tBHP) for inducing oxidative stress. Hydrogen peroxide (H2O2) production, glutathione (GSH) concentration, mitochondrial membrane potential (MMP) and cell viability were measured to investigate hepato-protective effects of JGW. Phosphorylation of AMP-activated protein kinases (AMPK), acetyl coenzyme A carboxylase (ACC) and effects of compound C on cell viability were examined to observe the role of AMPK on JGW-mediated cytoprotection. Results : Pretreatment with JGW (10-300 ㎍/mL) significantly suppressed cytotoxicity induced by tBHP in a concentration dependent manner and reduced the expression of cleaved PARP and cleaved caspase-3 proteins related to apoptosis in HepG2 cells. In addition, pretreatment with JGW significantly prevented the increase in H2O2 production, GSH depletion, and lower MMP induced by tBHP. Treatment with JGW (30 minutes of incubation and concentrations of 100 and 300 ㎍/mL) increased the phosphorylation of AMPK and ACC and treatment with compound C, a chemical inhibitor of AMPK, inhibited the cytoprotective effect of JGW. Conclusions : Our results demonstrated that JGW may protect hepatocytes from oxidative stress via activation of AMPK.
산화적 스트레스에 의한 망막 색소상피 세포의 손상 및 퇴화는 시력 소실을 포함한 다양한 망막질환의 원인으로 알려져 있다. 본 연구에서는 천연물 유래 14종 단일 성분과 합성 화합물 2종을 대상으로 망막질환 개선 가능성을 평가하기 위해, 사람 유래 망막 색소상피 세포를 대상으로 산화적 스트레스에 의한 세포 손상에 대한 보호 효능을 평가하였다. 그 결과, 16종의 후보물질 중 H2O2에 의해 유발된 세포독성을 개선시키는 효능을 보인 5종(auranofin, FK-509, hemistepsin A, honokiol 및 spermidine)을 선별하였다. 이를 기반으로 H2O2에 의한 미토콘드리아 기능 손상 및 활성 저하 억제능을 평가하였으며, auranofin을 제외한 4종에서 유의한 세포 보호 효능이 관찰되었다. 더불어 H2O2에 의해 유도된 DNA 손상에 미치는 영향을 조사한 결과, FK-506, honokiol과 spermidine이 DNA 손상을 개선시켰음을 확인하였다. 그러나 5종의 후보물질이 활성 산소종의 생성에는 뚜렷한 억제 효능을 나타내지 않았으며, 이는 5종의 후보 물질이 ROS-비의존적 기전을 통해 세포 보호효과를 나타낼 것으로 사료된다. 이상의 결과를 바탕으로, 조사 대상 후보물질 중에서 spermidine이 산화적 스트레스로부터 망막 색소상피 세포의 보호 효능이 가장 우수한 천연물 유래 단일 물질임을 규명하였으며, 동시에 합성물질로는 FK-506이 망막 세포 보호 효능이 우수한 것으로 나타났다. 비록 작용 기전에 대한 추가 연구가 필요하겠지만, 본 연구 결과는 spermidine과 FK-506이 산화적 스트레스에 의한 망막질환의 위험을 억제할 가능성이 있음을 시사한다.
다양한 질병의 발병 및 진행을 예방하기 위해 오랫동안 사용되어 온 대계는 항산화 활성을 포함하여 광범위한 생리 활성을 갖는 것으로 보고되었으나, 간세포에서의 효능에 대한 연구는 여전히 미비한 실정이다. 본 연구에서는 간세포에서 대계 에탄올 추출물(EECJ)의 항산화 활성 및 관련 기전을 조사하기 위해 인간 간세포암종 HepG2 세포주를 선택하였고, 산화적 스트레스를 모방하기 위해 H2O2를 처리하였다. 본 연구 결과에 따르면 EECJ는 H2O2가 처리된 HepG2 세포에서 세포 생존율의 감소와 LDH의 방출을 유의적으로 억제하였다. EECJ는 또한 세포 형태학적 변화의 관찰, 유세포 분석 및 LC3의 발현 결과에서 입증된 바와 같이 H2O2에 의해 유도된 자가포식을 유의하게 감소시켰으며, H2O2에 의해 유도된 세포사멸과 세포주기의 교란을 약화시켰다. 이는 H2O2에 노출된 HepG2 세포가 EECJ에 의하여 지속적인 세포 분열과 증식이 유지되고 있음을 의미한다. 또한, EECJ가 강력한 항산화 활성을 가지고 있음을 세포 내 ROS 및 미토콘드리아 슈퍼옥사이드 생산의 차단으로 확인하였으며, 이는 H2O2에 의해 감소된 세포 내 GSH 함량의 회복과 MnSOD 및 GPx의 발현 및 활성을 보존과 연관성이 있었다. 비록 EECJ에 함유된 활성 성분의 분석과 in vivo 동물 모델에서의 검증이 요구되지만, 본 연구의 결과는 EECJ가 산화적 스트레스로 인한 간세포의 손상을 예방하고 치료하기 위한 잠재적인 후보로 사용될 수 있음을 의미한다.
세포기반 치료제에 사용되는 줄기세포는 재생능력과 다양한 세포로의 분화능력으로 인해 재생 의학 분야에서 광범위하게 관심을 끌었으며, 많은 불치병에 적용된다. 하지만, 이러한 줄기세포는 여전히 치료 전 세포증식 및 질병 투여부위에서의 낮은 생존률로 인해 충분한 치료효과가 나타나지 않는 단점이 있다. 이것을 해결하고자, 우리는 세포부착능과 항세포자살 기능을 가지고 있는 carcinoembryonic antigen (CEA) gene family의 하나인 CEACAM 6를 사용하였다. 이것을 줄기세포에 적용 전, 먼저 세포별로 이 단백질이 발현되는지를 확인하였고, 이 유전자가 발현되는 벡터를 줄기세포에 삽입시키기 위한 최적 조건을 선정하였다. 그 후, 도입된 CEACAM 6발현벡터로부터 줄기세포에서 이 유전자가 발현되는지를 확인하였다. 그리고 인체투여 시 발생되는 산화적 스트레스와 유사한 조건에서의 이 유전자의 기능을 평가하기 위해 과산화수소(H2O2)를 처리하였다. 산화적 스트레스 조건하에서 CEACAM 6가 발현되는 줄기세포는 그렇지 않은 세포에 비해 세포의 생존률이 현저히 증가하는 것을 확인하였다. 이를 통해, 이 CEACAM 6는 줄기세포의 치료효능과 세포증식을 강화시킬 수 있는 다른 선택지로서의 가능성이 있음을 확인하였다.
