In vitro Formation of Cochliobolus nisikadoi, the Perfect State of Bipolaris coicis (Bipolaris coicis의 완전세대인 Cochliobolus nisikadoi의 배양기내 형성)

  • The perfect state of Bipolaris coicis, causing leaf blight of Job's tears, was in duced under in vitro conditions. Eighty nine isolates of the fungus were collected from 17 locations in Korea from 1994 to 1996. They were crossed on Sach's nutrient agar, on which a piece of rice straw was placed, and incubated at $25^{\circ}C$. Pseudothecia were produced only by certain combinations of compatible isolates. Although pseudothecia were usually produced on rice straw two weeks after incubation, asci and ascospores were observed only in a few pseudothecia examined. The pseudothecia were black and globose with protruding ostiolar beaks. The locules were filled with a mass of hyaline and filamentous pseudoparaphyses. Asci were cylindrical to clavate and straight or slightly curved. The ascus wall was bitunicate with short stipes. Ascospores were filiform, hyaline, and arranged parallel to slightly coiled in the asci, measuring $143-166.4{\times}2.6-3.8\;{\mu}m$. Perfect state of the fungus was identified as Cochliobolus nisikadoi (Tsuda, Ueyama & Nishihara Alcorn), based on the morphological characteristics.

Friction of calcium phosphate brackets to stainless steel wire (인산칼슘재 브라켓과 강선사이의 마찰저항에 관한 연구)

  • Esthetic brackets which resemble the color of natural teeth have been widely used. But the frictional resistance of ceramic brackets, a typical esthetic bracket, is greater than that of metal brackets. The purpose of this study was to measure the frictional resistance of the new calcium phosphate brackets (CPB) which were recently developed and to evaluate its clinical usability by comparing the frictional differences of CPB with metal brackets and metal slot inserted ceramic brackets. Methods: Experimental groups were CPB (Hyaline II, Tomy, Tokyo, Japan), metal bracket (Kosaka, Tomy, Tokyo, Japan) and metal slot inserted ceramic bracket (Clarity, 3M Unitek, Monrovia, CA, USA). All of the brackets had 0.022-inch slot sizes. The brackets were tested with $0.019\;{\times}\;0.025$ inch stainless steel wire (3M Unitek, Monrovia, CA, USA). A biologic model was used to simulate the situation which would occur during orthodontic treatment with fixed appliances. Retraction force was applied at a speed of 5 mm/min for 30 seconds. The frictional resistance was measured on a universal testing machine (Instron 4467, Instron, Norwood, MA, USA). Results: CPB showed significantly higher friction than metal brackets (p < 0.05) and lower friction than metal slot inserted ceramic brackets (p < 0.01). Conclusions: CPB can be considered to be a useful orthodontic esthetic bracket with respect to frictional resistance, as its friction is remarkably lower than that of metal slot inserted ceramic brackets.

Morphological Characteristics and Molecular Identification of Calonectria ilicicola Causing Soybean Red Crown Rot (콩 검은뿌리썩음병균(Calonectria ilicicola)의 형태적 특징과 유전자 수준에서의 동정)

  • The 18 isolates of soybean red crown rot pathogen(Calonectria ilicicola) were isolated from infected soybean plants showing chlorosis symptom and red perithecia sign in 2002 and 2003 season in Korea. The morphological characteristics of the typical isolate SC03-15, such as macroconidia and macrovesicles in anamorph as well as perithecia, asci and ascospores in teleomorph, were observed from red crown rot pathogens. The perithecia were red-brown globose to ovoid, and $357.9(312.5{\sim}391.5.7){\mu}m$ high and $474.3(419{\sim}535){\mu}m$ diameter. Asci were hyaline and clavate and included 8 ascospores, which were straight to slightly curved, 1(-3)-septate, and $36.9(31.1{\sim}49.4){\times}5.2(4.5{\sim}6.1){\mu}m$. Macroconidia in anamorph were cylindrical, hyaline and in size $73.2(68.1{\sim}77.6){\times}5.8(4.9{\sim}6.8){\mu}m$. Further identification process was performed through molecular method, such as a phylogenetic analysis by rDNA sequencing of ITS and $\beta$-tubuline genes of all isolates tested in this study. All isolates were identified as Calonectria ilicicola by DNA sequencing of ITS and $\beta$-tubuline genes, and morphological characteristics.

Gray Mold Rot on Fruit of Cucumis melo var. reticulatus Caused by Botrytis cinerea (Botrytis cinerea에 의한 멜론 잿빛곰팡이병)

  • A new disease on mask melon grown under plastic film houses was found in Namhae area in May of 1999. Gray to dark brown mold were grown on the surface of matured fruits and infected inside tissues were discolored and rotten. Basal part of the fruit and blossom-end were frequently infected and colonized by fungi. About 2.2% of matured fruits were infected in the surveyed plastic film houses. The causal organism was isolated from the lesion and identified as Botrytis cinerea. The conidia in mass were hyaline or gray, 1-celled, mostly ellipsoid or ovoid and sized $8.8{\sim}21.2{\times}6.5{\sim}13.1\;{\mu}m$. Hyaline or pigmented conidiophores were tall, slender and determinated and, sometimes branched irregularly in upper part. Enlarged or rounded apical cells bear conidial cluster and sized $18.4{\sim}81.1{\times}4.3{\sim}11.4\;{\mu}m$. Optimum temperature for mycelial growth was recorded at $15{\sim}25^{\circ}C$. This is the first report on gray mold of melon caused by Botrytis cineria in Korea.

A Taxonomic Study on Korean Allium L. Based on the Morphological Characters (형태학적 형질에 기초한 한국산 부추속의 분류학적 연구)

  • For 20 taxa of Korean Allium, including 16 species and 5 varieties, examined were morphological characters (i.e. sexuality, structure and shape of underground part, types and growing patterns of leaf and scape, and shapes of perianth, filament and pistil). The specialization and the evolutionary trends of taxonomic characters were inferred from morphological examination. Taxonomic relationships and system of Korean Allium were also studied. The characters showing evolutionary trends were the structure and shape of underground part including rhizome and bulb, leaf, scape, inflorescens, filament and ovary. It seemed that thin and short rhizome developed from thick and long one, and the membranous simple bulb tunic evolved into fibrous reticulate one. The presence of hyaline sheath in A. monanthum of sect. Microscordum was apomorphic. Both angular leaf blade with 2-rowed vascular bundle and flat blade with 1-rowed vascular bundle were developed from terete one with 2-rowed vascular bundle. The base of filament have differentiated from entire to toothed, and 2-ovuled ovary as well as erect scape before flowering was plesiomorphic type. In addition, sexuality, structure of underground part, the presence of hyaline sheath and cross-section structure of leaf were taxonomic characters with the level of the subgenus in this genus. Shape of rhizome, bulb, leaf, scape and pedicel discriminated each section from other ones in the subgenera easily. The shape and arrangement of perianth and filaments were the diagnostic characters for species level along with shape of ovary and stigma.

A Case of Acute Fibrinous and Organizing Pneumonia (급성 섬유소성 기질화 폐렴 1예)

  • Acute fibrinous and organizing pneumonia (AFOP) is a histological pattern consisting of prominent intra-alveolar fibrin and organizing pneumonia, with out hyaline membranes or prominent eosinophilia. The clinical manifestations of AFOP resemble those of acute lung injury such as acute interstitial pneumonia (AIP). However, the classic histological patterns of AFOP differ from diffuse alveolar damage (DAD), bronchiolitis obliterans with organizing pneumonia (BOOP) or acute eosinophilic pneumonia (AEP). The characteristic intra-alveolar fibrin ball and lack of classic hyaline membrane are the predominant histological features of AFOP. Although some reports suggest that its clinical course is less catastrophic than DAD, the clinical entity that distinguishes AFOP from DAD has not been established. We present a case of pathologically demonstrated AFOP in a 79-year-old man. The radiological findings of our case were similar to those of DAD, presented with diffuse bilateral lung infiltrations. However, despite the rapid development of respiratory failure, the patient had a better response and outcome to steroid therapy than what would be expected for DAD.

Identification of Infectious Fungus in the Artificial Cultivation Rooms of Paecilomyces tenuipes (눈꽃동충하초 재배상 내에 발생된 오염균의 동정)

  • A infectious fungus (J131) recently isolated from artificial cultivation farms of Paecilomyces tenuipes is known to cause a great yield loss and deterioration of products. In this study, its occurrence and characterization were made. The isolate (J131) was identified as Paecilomyces farinosus (Holm ex S. F. Gray) Brown & Smith. Colonies of the isolate on PDA grew moderately fast, attaining a diameter of 19mm within 14 days at 24${\pm}$1$^{\circ}C$. Vegetative hyphae are smooth-walled and hyaline. Conidiophores mainly arose from the submerged mycelium, usually measuring 1.3-1.5$\mu\textrm{m}$ wide and consisted of verticillate branches with whorls of 2 to 4 phialides. Phialides of 3.8-5.8${\times}$1.5-1.8 $\mu\textrm{m}$, with a swollen basal portion tapered into a distinct neck of about 0.6-0.8 $\mu\textrm{m}$ wide. Conidia arisen from the phialides, were ellipsoidal to fusiform, smooth-walled, hyaline, 1.8-2.4${\times}$1.1-1.4$\mu\textrm{m}$. .When silkworm larvae were injected with concentration of 1${\times}$10$\^$8/ conidia/$m\ell$ of infectious fungus J131, infection rate was 41.3% and synnemata formation rate was 23.6%. Black spots were appeared on the surface of silkworm larvae in 5-9 days after injection. The surface of larvae was covered with hyphae which were gradually turned into yellow from white color during the incubation period of 3-4 days. 1 to several 4-6mm long synnemata of yellow or orange color were formed on the surface. The matured synnemata were ramified from its end and produced much conidia of white color.

  • Castleman's disease is a rare non-neoplastic lymphoproliferative disorder of unknown etiology. It is divided into three histologic subtypes; hyaline-vascular(HV), plasma cell(PC) type and mixed type (HV-PC). It has two clinical expressions. The localized form, which presents as a slow growing mass, has a relatively benign clinical course. The multicentric form is multilocated and holds significant morbidity. The mainstay of treatment of the localized form is surgical resection. The multicentric form requires medical treatment comprising prednisolone and other immunosuppressor drugs. The disease in children seems to have a more favorable course than in adults. We report a 13-year-old boy with Castleman's disease of multicentric form who was successfully treated with prednisolone and intravenous immunoglobulin.

BENTHIC FORAMINIFERA OF THE SUBTIDAL ZONE OF ASAN BAY, KOREA (牙山灣海底의 底棲有孔蟲)

  • Micropaleontological analyses of the bathic foraminifera of 132 subtidal surface samples of Asan Bay located in the middle of the western coast of Korea, fringing the Yellow Sea collected during late May-early June 1982, shows the results as follows; -Forainiferal numbers of total assemblages are influenced by hydrodynamic and ecologic factors as well as by the sedimentation in the study area. -Badly preserved faunas including replaced faunas seemed to be fossils, all having living counterparts in the study area and showing a somewhat similar type of wall structures with those of the total assemblages and showing no characteristic bathymetric occurrence compared to that of the biocoenose s.1. of the species, lead to infer theirprobable derivation from the environment without a remarkable environmental difference from the study area. But this inference demands further study in consideration of the physical conditions of the study area. -Living/total ratios show a strong negative correlation with the total foraminiferal numbers excluding replaced faunas. But this trend is somewhat deviated in the relatively high ratios. -Fisher-alpha diversity indices of the total assemblages excluding replaced faunas are lying between 1.7 and 3.7. Totals assemblages are mainly composed of hyaline forms with limited porcelaneous and agglutinated ones.

A HISTOPATHOLOGIC STUDY OF THE EFFECTS OF ARTHROGRAPHY ON THE TISSUES OF TEMPOROMANDIBULAR JOINT (악관절조영술이 악관절 조직에 미치는 영향에 대한 병리조직학적 연구)

  • This study was performed to observe the effects of arthrography on the tissues of temporomandibular joint histopathologically. Among 17 rabbits (34 joints), 2 (4 joints) were used as normal control group, others (30 joints) were experimentally arthrographed using 0.03㎖ diatrizoate meglumine (Hypaque meglumine 60, Wintrop, U.S.A). The rabbits were sacrificed at 2-hour, 1-, 2-, 4- and 7-day after experiment and the tissues of temporomandibular joint were prepared according to the usual method for light microscopic examination. The results were as follows: 1. There were no changes of the fibrous connective tissues of the condylar fossa, the fibrous connective tissues and hyaline cartilages of the condyle, and the articular dislc 2. In 2-hour experimental group, the proliferation of the surface synovial membrane cells, the congestion and hemorrage of the vessels were observed. 3. In 1 and 2-day experimental group, minimal lymphocytes infiltration was observed with the proliferation of the surface synovial membrane cells, the congestion and hemorrage of the vessels. 4. In 4-day experimental group, the diminution of the proliferation of the surface synovial membrane cells was seen, but there was no vascular changes. 5. In 7-day experimental group, the tissues showed similar appearance to the normal control group.
