Effects of Hwangryeonhaedoktang on Depression and c-Fos Expression in Paraventricular Nucleus of the Brain in the Chronic Mild Stress Treated Rats (황련해독탕(黃連解毒湯)이 우울증 모형 동물의 우울성향 및 PVN의 c-Fos 발현에 미치는 효과)

  • Objective : This study was designed to assess the protective effects of Hwangryeonhaedoktang on the animal model of depression, induced by chronic mild stress(CMS). Method : Male Sprague-Dawley rats were used for this experiment. The subjects were divided into 3 groups ( 1. CMS-drug: Hwangryeonhaedoktang administered during CMS treatment, 2. CMS-vehicle: water administered, 3. normal ). After 4 weeks of CMS treatment, they were executed forced swimming test(FST), open field test and c-Fos in paraventricular nucleus(PVN) were measured. Result : 1. In FST, immobility behavior decreased significantly in CMS-drug group. 2. There was no difference in the open field test between 3 groups 3. c-Fos expressed cell bodies in PVN were significantly less in CMS-drug than in CMS-vehicle group. Conclusion : These results suggest that Hwangryeonhaedoktang may have protective antidepressant effects in CMS model rats. And these effects could be explained by the elevated stress-copying behaviors which are related with PVN of hypothalamus.

Investigation of Neuroprotective Effect Using Herb Medicine against Dementia related Brain Damage in Organotypic Hippocampal Slice Culture (뇌해마배양조직을 이용한 치매 뇌손상 모델에 대한 소풍순기원(疏風順氣元)과 황련해독탕(黃連解毒湯) 처방약재의 신경보호효과 연구)

  • Objectives: This study was designed to assess neuroprotective effects of herb medicine against Alzhheimer's disease related brain damage in organotypic hippocampal slice culture. Methods: We induced dementia related brain damage in organotypic hippocampal slices by $\beta$-amyloid. Those slices were treated with herb medicines - Hwangryeonhaedoktang, Sopungsoongiwon. Using by PI staining, the extents of cell death were assessed. After that, we selected the best effective one among those herb medicines and the major components of that medicine were studied to reveal neuroprotective effects and related proteins by using PI stating. Results: In PI staining, Sopungsoongiwon is the best effective herb medicine between Hwangryeonhaedoktang and Sopungsoongiwon. Notopterygii Rhizoma, Corni Fructus, Areca Catechu, Aurantii Fructus Immaturus, Plantaginis Semen is the best effective one among the components of Sopungsoongiwon. Conclusions: We suggested that purgative effect would be the best effetive medicine on dementia related brain damage between clearing heat and toxic materials.

Effects of Microneedle Therapy System(MTS) and Hwangryeonhaedoktang Pharmacopuncture Solution on Hair Growth in an Alopecia Model of C57BL/6N Mouse (C57BL/6N 마우스 탈모모델에서 미세다륜침(MTS)과 황련해독탕 약침액 도포가 모발성장에 미치는 영향)

  • Objective : This study was carried out to investigate the effect of Hwangryeonhaedoktang pharmacopuncture solution (HRHDT) and microneedle therapy system (MTS) on hair growth in an alopecia model of C57BL/6N mice.Methods : Six-week old mice were depilated and separated in 4 groups ; CON (saline), MXD (3% Minoxidil), MTS and HRHDT+MTS. The treatments were applied twice a week for 17 days. The hair growth was determined photographically. The hair density, thickness and length were identified by Folliscope and the weights of body and organs were measured. In dorsal skin tissue, the expression of hair growth-related gene and protein was analyzed by RT-PCR. In addition, the hair follicles in the dermis were observed by H&E staining.Results : The promotion of hair growth was observed in HRHDT+MTS and MTS compared to CON. The hair density, thickness and length were also improved in HRHDT+MTS and MTS compared to CON. The mRNA expression of IGF-1, PRL and PL and the protein expression of VEGF and IGF-1 were increased in HRHDT+MTS and MTS compared to CON. The hair follicles and hair root growth were improved in HRHDT+MTS and MTS compared to CON. In the above results, HRHDT+MTS were more effective than MTS.Conclusions : These results suggest that HRHDT and MTS have a hair growth activity and can be useful for the treatment of alopecia.

  • There are total 403 prescriptions containing Radix Scutellariae in Donguibogam and among them, 106 prescriptions treat Radix Scutellariae as a principal medicine. This report describes the study on therapeutic range, symptom, pathology and composition of 106 Radix Scutellariae-main blended Prescriptions from Donguibogam. The frequency and percentage of Radix Scutellariae-main blended prescriptions for the therapeutic range are as follows; cold symptom-complex is the most frequent(11 prescriptions, 10.37%) and gynecology is second(10 prescriptions, 9.43%). The symptoms that Radix Scutellariae-main blended prescriptions are made out for are 70 prescriptions of 28 therapeutic ranges, for example, minor Eum physical appearance, fire of triple burners jaundice, etc. The main pathologies that Radix Scutellariae-main blended prescriptions are being affected by cold, fire triple burners, dampness and heat, wind and heat, heat of heart and lung, heat of liver, heat of spleen, etc. Through the research into the composition of Radix Scutellariae-main blended prescriptions, the kind of medicines and certain rules of compounding medicines centering on Radix Scutellariae are deduced as follows: The main combinations of herbal medicines are Radix Scutellariae and Fructus Gardeniae, Radix Scutellariae and Rhizona Rhei, and Radix Scutellariae, Rhizona coptidis and Fructus Gardeniae. The main combinations of prescriptions are Yihwanghwan(Radix Scutellariae, Rhizoma Cotidis, Radix Glycyrrhizae), Boanbaekchulsan(Radix Scutellariae. Rhizoma Atractylodis macrocephalae), Sambohwan(Radix Scutellariae, Rhizoma Cotidis, Cortex Phellodendrj), Samhwangwon(Radix Scutellariae, Rhizoma Cotidis, Rhizoma Rhei), Hwangryeonhaedoktang(Radix Scutellariae, Cortex Phellodendri, Rhizoma Cotidis. A Study on 'Dangdokyeok' Epidemics in the Early 17C of the Joseon -Focusing on Heojun's 『Byeokyeoksinbang』- (17세기 초 조선에서 유행한 '당독역'에 대한 연구 -허준의 『벽역신방』을 중심으로-)

  • Various aspect of epidemics broke out continually from the middle of Joseon Dynasty due to the famine and drought caused by abnormal climate of the sixteenth century and the war. Thus the Dynasty performed sacrificial rites, isolated the patients and published plenty of medical books related epidemics in order to cure of the patients, and Heojun edited 『Byeokyeoksinbang』 as 'Dangdokyeok' broke out at Gwanbuk(關北) districts in 1613, Heojun explained the cause of Dangdokyeok as meteorology under the feudal conditions, and concluded Simhwa(心火) by fever toxin, Therefore he selected the method of puting out Simhwa by attack of fever toxin. In addition he presented emergency treatment that can maintain the airway by bleeding. To treat Dangdokyeok, Heojun presented lots of prescriptions so as Seungmagalgeuntang(升麻葛根湯), Cheongyeolhaedoksan(淸熱解毒散), Yeongyopaedok-san(連翹敗毒散), Bangpungtongsaongsan(防風通聖散), Jowiseunggitang(調胃升氣湯) and Hwangryeonhaedoktang(黃連解毒湯) etc. And he proposed Samdueum (三豆飮), Realgar(石雄黃) and so on to prevent infection from that. They presume from 120 to 150 years as the period of human adaptation to the first epidemics. Dangdokyeok put a large number of people to death at first, but it wasn't referred at the history any more after Byeokyeoksinbang. So we can say that the treatment of Heojun may be effective. Common cold and dyspeptic cold broke out in our country differently from 'Shanghan(傷寒)' in the China, so we had settled 'pestilence infectious epidemic disease(瘟疫)' while 'epidemic febrile disease(溫病)' of the China. Dangdokyeok of Heojun is similar to 'Scalet fever' belonging to 'virulent heat pathogen(溫毒)', 'newly epidemic febrile disease(新感溫病)'. As a cure of Dangdokyeok, the Korean medicine uses the treatment of removing fever state whereas the western medicine uses the antibiotics to kill the streptococcus. The symptoms of Dangdokyeok are remarkably similar to those of the Scarlatina, so this occupies a high position on the world history of medicine in aspects of the period and details of symptoms. These days we have the problems that the tolerance of antibiotics increases and disease of unknown cause is prevalent. It means the western medicine get to limits. So if we progress epidemiography based on Heojun's medicine, we may contribute to the world history of medicine.
