소에서 정자활성, 수정 양상 및 착상전 지속적 수정란 발달에 있어서 환삼덩굴 추출액의 효과 (Effect of Humulus japonicus Extract on Sperm Motility, Fertilization Status and Subsequent Preimplantation Embryo Development in Cattle)

  • Humulus japonicus is an ornamental plant in the Cannabaceae family. Although the mode of action of Humulus japonicus is not fully understood, a strong relationship was observed between anti-inflammatory and anticancer in some types of cells. Recent studies also have shown that Humulus japonicus possesses anti-inflammatory activities and may significantly improve antioxidant potential in Raw 264.7 macrophage cells. Thus, the aim of this study was evaluated the effect of Humulus japonicus extract on sperm motility and subsequent preimplantation developmental competence of the bovine embryos. After in vitro maturation, the oocytes with sperms were exposed in in vitro fertilization (IVF) medium supplemented with Humulus japonicus extract (0.01, 0.05, $0.1{\mu}g/mL$, respectively) for 1 day. In our results, exposure of IVF medium to Humulus japonicus extract did not affect sperm motility and percentage of penetrated oocytes but ROS intensity was significantly decreased by $0.01{\mu}g/mL$ compared with other groups (p< 0.05). Moreover, treatment with $0.01{\mu}g/mL$ of Humulus japonicus extract was higher the frequency of blastocyst formation than the any other groups (p<0.05). Otherwise, treatment with $0.01{\mu}g/mL$ of Humulus japonicus extract not increased the total cell number but reduced apoptotic-positive nuclei number. In conclusion, our results indicate that supplementation of Humulus japonicus extract in IVF medium may have important implications for improving early embryonic development in bovine embryos.

환삼덩굴 메탄올 추출물의 in vitro 항산화 및 항암 활성 (Antioxidant and Antitumor Activities of Methanolic Extracts from Humulus japonicus)

  • To evaluate the antioxidant and potential of Humulus japonicus, total polyphenol and flavonoid content, radical scavenging activities, and antitumor activities were measured. The total polyphenol and flavonoid contents of the methanol extracts from Humulus japonicus were $30.13{\pm}1.13$ and $13.61{\pm}0.49$ mg gallic acid equivalent/g extract, respectively. DPPH radical and hydroxyl radical scavenging activities of methanol extracts of Humulus japonicus were 60% and 35%, respectively. The Humulus japonicus higher activities of anticancer activities on liver cancer cell lines compared to other cancer cell lines.

  • 1989년(年) 9월(月) 경남 북(慶南.北) 수로변(水路邊)에서 조사(調査)된 초종(草種)은 각각(各各) 33과(科) 88종(種) 및 33과(科) 96종(種)이었으며, 2차원적(次元的) 분류방법(分類方法)에 의(依)하여 경남지역(慶南地域)에 3개(個) 군락형(群落型)과 경북지역(慶北地域)의 5개(個) 군락형(群落型)이 조사(調査)되었다. 경남지역(慶南地域)의 군락(群落)은 피(Echinochloa crusgalli)군락(群落), 여뀌(Persicaria hydropiper)군락(群落) 및 고마리(Persicaria thunbergii)군락(群落) 등(等)이었고, 경북지역(慶北地域)은 환삼덩굴-미국가막사리(Humulus japonicus-Biden frondosa)군락(群落), 까마중(Solanum nigrum)군락(群落), 고마리-환삼덩굴(Persicaria thunbergii-Humulus japonicus)군락(群落), 환삼덩굴(Humulus japonicus)군락(群落) 및 고마리(Persicaria thunbergii)군락(群落) 등(等)이었다. 경남 북지역(慶南 北地域) 공(共)히 고마리, 여뀌 군락(群落)이 조사(調査)되었고 수생잡초(水生雜草)인 고마리, 여뀌가 수로변(水路邊)이 대표적(代表的)인 초종(草種)으로 조사(調査)되었다.

환삼덩굴(Humulus japonicus) 추출물의 항돌연변이 효과와 Flavonoid 성분의 분리 (Antimutagenic Effects and Isolation of Flavonoids from Humulus japonicus Extract)

  • 환삼덩굴의 생리활성물질을 구명하기 위하여 환삼덩굴을 물과 메탄올로 추출하여 돌연변이원성과 항돌연변이 효과를 조사하고 TLC와 HPLC로 flavonoid 성분을 분리하였다. 환삼덩굴의 물 추출물과 메탄올 추출물은 Salmonella typhimurium TA98과 TA100을 이용한 Ames test에서 돌연변이원성을 나타내지 않았다. 환삼덩굴의 메탄올 추출물은 NPD와 NQO로 돌연변이를 유도한 Salmonella typhimurium TA98과 TA100에 대하여 항돌연변이 효과를 나타내었다. 분획별 항돌연변이 효과는 헥산분획에서 높게 나타났으며 50% 돌연변이 억제농도($IC_{50}$)는 TA98에 대하여는 2.O mg/plate, TA100에 대하여는 0.5 mg/plate 였으며, 4.O mg/plate의 농도에서 TA98에 대하여는 약 58%, TA100에 대하여는 약 88% 정도의 돌연변이 억제활성을 나타내었다. 환삼덩굴의 에칠아세테이트 분획에서 quercitrin과 luteolin을 확인하였다.

소아 키 성장에 미치는 한삼덩굴추출분말의 유효성 및 안전성을 평가하기 위한 무작위배정, 이중눈가림, 위약 대조 인체적용시험: 인체적용시험 프로토콜 (Randomized, Double-blind, and Placebo-controlled Human Trial to Evaluate the Efficacy and Safety of Humulus japonicus Extract Powder on Child Height Growth: Study Protocol)

  • Objectives We aimed to confirm whether Humulus japonicus Extract Powder can enhance child height growth significantly and safely compared with a placebo. Methods A total of 150 children between the 3rd and 25th percentiles in height and between the ages of 6 and 9 years will be recruited to participate in this randomized, double-blind, placebo-controlled clinical trial. The participants will be randomly assigned to the treatment or placebo group. Participants in the treatment group will take one pack per day (700 mg of Humulus japonicus Extract Powder) for 24 weeks. Participants in the placebo group will take one package of placebo per day (0 mg of Humulus japonicus Extract Powder) for 24 weeks. The primary outcome will be a change in height after 12 weeks, and the secondary outcomes will be the height after 24 weeks, growth rate, height standard deviation, growth hormone, insulin-like growth factor-1 (IGF-1), insulin-like growth factor binding protein-3 (IGFBP-3), bone alkaline phosphatase (BALP), and osteocalcin after 12 and 24 weeks. Results This protocol was approved by the Institutional Review Board (IRB) of the Korean Medicine Hospital of Busan University (IRB No. PNUKHIRB-2023-03-002). Research participants will be recruited from June 2023 to December 2023. Conclusions The results of this study provide clinical information regarding the effectiveness and safety of the Humulus japonicus Extract Powder in increasing child height.

환삼덩굴을 이용한 견직물의 천연염색에 관한 연구 (Natural Dyeing of Silk Fabrics with Humulus japonicus Extract)

  • The purpose of this study is to investigate the dyeability of silk fabrics with Humulus japonicus (Hwansam) extract which is a kind of weeds that grows wild in the suburbs. In the research, the general extract method was conducted with the extract from fresh herb of root-removed Humulus japonicus while we investigated the optimal dyeing conditions and dyeability depending on the changes of the kind of mordants, methods and temperature. The functional properties such as antibacterial activities, deodorization and UV protection were also reviewed. The ideal output, as a result, was shown at 100% of dye concentration, $90^{\circ}C$ of dyeing temperature and 40 minutes of dyeing time while in terms of dye uptake depending on the kind of mordants and methods, it was found that among four mordants of $SnCl_2$, $AlK(SO_4)_2$, $CuSO_4$, and $FeSO_4$ post-mordanting with $FeSO_4$ showed the best result. Color fastness to washing, dry cleaning and rubbing was found strong at grade 4-5 and the grade to light was as good as 3 while to perspiration was good at 4-5 as well. In aspects of functional properties, it showed excellent results of 99% deodorization rate, 98% UV protection rate and 99.9% bacterial reduction against Staphylococcus aureus.

Evaluation of Biological Activities of Invasive Alien Plants for Development of Functional Biomaterials

  • This study aims to confirm the possibility of using the invasive alien plants in Jeju as a functional biomaterial. To achieve this purpose, 70% ethanol extract and solvent fractions were prepared for five invasive alien plants (Hypochaeris radicata, Rumex acetosella, Humulus japonicus Siebold & Zucc., Solanum viarum, Lactuca scariolar) and their antioxidant, antibacterial anti-inflammatory and anti-obesity effects were investigated. The DPPH radical scavenging activity of ethanol extract from invasive alien plants was shown in the order of Rumex acetosella > Hypochaeris radicata > Humulus japonicus. Antimicrobial activity of ethanol extract against food poisoning bacteria (4 species) and oral cavity-induced microorganisms (6 species) was measured. As a result, the extract of Humulus japonicus showed high antibacterial effects against food poisoning bacteria (E. coli, V. parahaemolyticus) and oral microbes (L. casei, S. epidermidis, E. faecalis). In LPS-induced RAW 264.7 cells, the anti-inflammatory effect of ethanol extract from invasive alien plants was investigated. As a result, the NO production inhibition activity was highest in the Rumex acetosella and the Humulus japonicus Siebold & Zucc. ethanol extract, and the NO production inhibition activity was concentration-dependent. In addition, the Rumex acetosella and the Humulus japonicus Siebold & Zucc. ethanol extract showed a concentration-dependent inhibitory effect on cytokine (IL-6) production. These extracts also showed inhibitory activity of COX-2, an inflammatory protein. This suggests that NO production inhibition activity by the extract of invasive alien plants is the result of inhibition of iNOS and COX-2 expression. Currently, organic solvent fractions of crude extract are manufactured and the investigation of active ingredients is continuing along with evaluation of biological activity such as anti-inflammatory. These results are expected to be a major data for the study on the separation and utilization of active ingredients with antioxidant, antibacterial and anti-inflammatory effects using foreign plant crude extract and solvent fractions, and are highly likely to be applied to the development of functional food and cosmetics materials.

환삼덩굴로부터 분리한 Luteolin-7-O-$\beta$-D-Glucoside의 활성산소 소거능 (Active Oxygen Scavenging Activity of Luteolin-7-O-$\beta$-D-Glucoside Isolated from Humulus japonicus)

  • We investigated the active oxygen scavenging activity and active compound from Humulus japonicus. The ethyl acetate and butanol fraction exhibited strong scavenging effects on hydroxyl radical. Furthermore active compound was isolated and purified using Amberlite XAD-2 column chromatography and preparative HPLC from ethyl acetate fracton, and major compound was identified as a luteolin-7-O-$\beta$-D-glucoside by the MS, UV, 1H-NMR and 13C-NMR analyses. Luteolin-7-O-$\beta$-D-glucoside exhibited strong scavenging effect on active oxygens such as superoxide radical, hydroxyl radical and hydrogen peroxide.

Growth Rate and Nutrient Content Changes of Humulus japonicus

  • The growth pattern and the seasonal changes in nutrient contents of Humulus japonicus were investigated. Stem length of H. japonicus reached the maximum from 240 to 260 Julian date and the median value was found at 255 Julian date. The maximum leaf area was observed from 235 to 248 Julian date and the median value was at 240 Julian date. The maximum growth rate of the stem length ranged from 205 to 227 Julian date. The leaf area showed the maximum growth rate from 196 to 214 Julian date. The median date in the growth rate of the stem length and leaf area was 212 and 205 Julian date, respectively. The growth rate of H. japonicus was related to rainy season and precipitation. Phosphorus and sodium contents of H. japonicus were correlated with maximum potential rate of relative growth. Although stem biomass of H. japonicus was $ 1.5{\sim}3.5$ times larger than that of leaf, N content of the leaf ($4.48{\sim}5.27%$) was about 2 times higher than that of the stem ($2.00{\sim}3.62%$). High content of N might be responsible for the high growth rate of H. japonicus in summer. This result provides valuable information for appropriate timing for the removal of H. japonicus.

환삼덩굴의 용매분획별 항균성 및 항산화성 (Antimicrobial and Antioxidative Activities of Solvent Fraction from Humulus japonicus)

  • 환삼덩굴의 생리활성물질을 검색하기 위하여 물과 메탄올로 추출하여 항균성과 trypsin 저해활성 및 항산화 효과를 조사하였으며, 메탄올 추출물은 다시 계통 분획하여 분획별 활성을 조사하였다. 세균 6종과 효모 2종을 대상으로 항균성을 조사한 결과, 물 추출물 보다는 메탄올 추출물의 항균성이 우수하였으며, Escherichia coli를 제외한 모든 시험균주에 항균효과를 나타내었다. 분획별 항균효과를 비교한 결과, 부탄올 분획이 모든 시험균주에 항균효과를 나타내었으며, 균종별로는 Staphylococcus aureus와 Saccharomyces cerevisiae에 대한 항균효과가 우수하였다. 부탄올 분획의 미생물에 대한 최소저해농도(MIC)는 $0.1{\sim}0.4%$ 범위였다. 환삼덩굴 추출물의 trypsin 저해활성을 조사한 결과, 물 추출물은 저해활성을 나타내지 않았으나, 메탄올 추출물은 저해활성을 나타내었다. 분획별 저해활성은 클로로포름 분획에서 가장 높았으며 클로로포름 분획의 50% 저해농도($IC_{50}$)는 1mg/ml였으며, EI(enzyme-inhibitor) 복합체 형성시간은 20분에 90% 이상이었다. 환삼덩굴의 메탄올 추출물은 돈지와 대두유를 기질로한 항산화성 시험에서 과산화물의 생성을 억제하였다. 메탄올 추출물의 분획별 활성은 클로로포름 분획에서 가장 높게 나타났으며, 클로로포름 분획의 농도를 0.2% 첨가하였을 때, 유지의 유도기간은 돈지는 15일, 대두유는 9일로서 산화안정성이 연장되었다.
