Transformation of Pentachlorophenol by Oxidoreductive Catalysts (산화환원 촉매에 의한 Pentachlorophenol의 Transformation)

  • Pentachlorophenol(PCP), which is very persistent in soil and water environment, was tried to detoxify with oxidoreductive catalysts(peroxidase, laccase, tyrosinase and birnessite). To find out detoxification of PCP, the transformation of PCP through oxidative coupling was investigated in the presence of various oxidoreductive catalysts. PCP incubated with peroxidase was significantly transformed, however, in case of tyrosinase, the transformation was negligible. Using peroxidase, the optimal reaction condition was pH 5.6 and $16^{\circ}C$. The transformation of PCP was very fast in initiation step until 30 min but, that was not observed after 180 min. The transformation of PCP was increased by increasing peroacidase amount. When the effect of humic monomer was investigated as co-substrate on the transformation of PCP, the transformation of PCP was mostly decreased in the incubation with peroxidase, laccase, and birnessite. The transformation of PCP, however, was slightly increased by the incubation with tyrosinase in the presence of humic monomers as co-substrate, except catechol. On the basis of the results obtained, it may be suggested that PCP is able to be effectively detoxified through oxidative coupling mediated with oxidoreductive catalysts.

Enhanced Phosphorous Uptake and Growth Improvement of Creeping Bentgrass after Application of Liquid Fertilizer Containing Humic acid and Saccharomyces cerevisiae Broth (부식산 및 효모균 배양액 함유 액상비료 처리에 따른 크리핑 벤트그래스의 인 흡수 및 생육 증대 효과)

  • This study was conducted to evaluate the effect of liquid fertilizer containing humic acid and Saccharomyces cerevisiae broth (LHS) on changes of turfgrass growth by investigating visual quality, chlorophyll content, dry weight of clipping, and nutrient content in leaf tissues. Treatments were designed as follows; control fertilizer (CF), HS-1 ($CF+1.0mL\;m^{-2}\;LHS$), HS-2 ($CF+2.0mL\;m^{-2}\;LHS$), and HS-3 ($CF+4.0mL\;m^{-2}\;LHS$). After treatment of LHS on creeping bentgrass, soil pH in the treated plots was decreased than that of CF. As compared to CF, visual quality, chlorophyll content and content of N, P and K were not significantly different in the LHS treatments. However, clipping yield and phosphorus uptake of HS-2 were significantly increased by 22% and 33%, respectively. These results showed that application of LHS improved the phosphorus uptake and growth of creeping bentgrass, which would be an alternative management tool for the cool season turfgrass under stress conditions.

Growth and Quality Changes of Creeping Bentgrass by Application of Liquid Fertilizer Containing Humic Acid (부식산 함유 액상비료 시비에 따른 크리핑 벤트그래스의 생육과 품질 변화)

  • This study was conducted to evaluate the effect of liquid fertilizer containing humic acid (LFHA) on changes of creeping bentgrass quality and growth. Treatments were designed as follows; control fertilizer (CF), HA-1 ($CF+1ml\;m^{-2}LFHA$ LFHA), HA-2 ($CF+2ml\;m^{-2}LFHA$ LFHA), and HA-3 ($CF+4ml\;m^{-2}LFHA$). As compared with CF, soil chemical properties and chlorophyll content of clipping of LFHA treatments were not significantly different. Visual quality in both of HA-2 and HA-3 treatments was higher than that of CF treatment from September to November, and clipping yield on October 27. Shoot density, root length, and the content of nitrogen, phosphorus or potassium were increased by application of LFHA. The clipping yield was positively correlated with phosphorus content, potassium content or shoot density. Similarly, LFHA level was proportionate to clipping yield of creeping bentgrass, and N, P, K contents in the leaf tissue. These results generally demonstrated that the application of LFHA improved the growth and quality of creeping bentgrass by increase of N content or P in leaf tissue.

Effects of Compost Application and Plastic Mulching on Soil Carbon Sequestration in Upland Soil (밭토양에서 퇴비시용과 비닐멀칭이 토양탄소 축적에 미치는 영향)

  • BACKGROUND: In most studies, soil carbon sequestration has been evaluated simply with change of soil organic carbon content. So far, information regarding stability of soil organic carbon is limited. METHODS AND RESULTS: This study was conducted to determine changes in soil organic carbon (SOC) content and stability of carbon in response to compost application rates and plastic mulching treatment during the hot pepper growing season. Under the pot experiment condition, compost was mixed with an arable soil at rates corresponding to 0, 10, 20, and 40 Mg/ha. To determine effects of plastic mulching on soil carbon sequestration, plastic mulching and no mulching treatments were set up in soils amended with the application rate of 20 Mg/ha. The SOC content did not significantly increase with application of compost and plastic mulching at harvest time. No significant changes in bulk density with compost application and plastic mulching was found. These might result from short duration of experiment. While hot water extractable organic carbon content significantly decreased with compost application and plastic mulching, humic substances increased. Belowground biomass of hot pepper was biggest at the recommended application rate (20 Mg/ha) of compost. CONCLUSION: From the above results, continuous application of compost at the recommended application rate could improve increase in SOC content and stability of carbon in long term aspect.

Amino Acid Contents in the Hydrolysates of Fulvic Acids Extracted from Decomposing Plant Residues (부숙식물유체(腐熟植物遺體)에서 추출(抽出)한 Fulv 산(酸) 가수분해(加水分解) 용액중(溶液中)의 Amino 산함량(酸含量))

  • Sixteen amino acids in the hydrolysates of fulvic acid fraction from 7 plant materials were determined. Analyzed amino acids were aspartic acid, glutamie acid, arginine, histidine, lysine, glycine, alanine, valine, leucine, isoleusine, phenylalanine, tyrosine, serine, threonine, proline, and methionine. Four crop residues, wild grass cuttings and forest tree litters were put under investigation. 1. The content of amino acids in fulvic acid fractions extracted after 90 days of compositing ranged from 0.15% to 0.53% by dry weight. The highest value was found in the fulvic acids of wild grass cuttings and the lowest in those of wheat straw, being equivalent to 1/5-1/31 of those found in humic acids. 2. The group of neutral amino acids shared the largest portion followed by acidic and basic amino acids. 3. Arginine was not detected in fulvic acid fractions from well decomposed residues. 4. Aromatic amino acids, phenylalanine and tyrosine, were virtually absent in fulvic acid fractions. 5. Glycine, glutamic acid and aspartic acid were the 3 major amino acids contained in fulvic acids of well decomposed residues. With glutamic acid and aspartic acid excluded, the decreasing order of concentration of amino acids was roughly in parallel with the increasing order of molecular weight.

Reductive Degradation of 4-Chlorophenol Compound by Nickel-Coated Zero Valent Iron (니켈로 코팅된 염가금속을 이용한 4-염화페놀의 환원제거율 평가)

  • Reductive dechlorination of chlorophenols by nickel coated iron was investigated to understand the feasibility of using Ni/Fe for the in situ remediation of contaminated groundwater. Zero valent iron (ZVI) was amended with Ni(II) ions to form bimetal (Ni/Fe). Dechlorination of 4-chlorophenol and formation of intermediates was studied using Ni/Fe. Effects of initial contaminant concentration, bimetal loading, presence of humic acid, and solution chemistry were also evaluated. Experimental results showed that Ni/Fe bimetal was so effective that more than 95% of 4-CP degradation was achieved within 240 minutes. Pseudo first-order rate constant for the dechlorination reaction was well correlated with bimetal loading. Humic acid competed for the reactive sites on the nickel coated iron with chlorophenols, lowering the dechlorination efficiency. No significant changes in solution pH were observed in the dechlorination of chlorophenols with Ni/Fe in the absence of buffer, indicating that reactivity of bimetal (Ni/Fe) could be prolonged. Phenol was found as a dechlorination intermediate of the conversion of 4-chlorophenol compound by Ni/Fe.

A Study on Fractions and Leaching Potential of Heavy Metals in Abandoned Mine Wastes (휴ㆍ폐광산지역에서 폐재내 중금속의 존재형태 및 용출특성에 관한 연구)

  • This study investigates the fractional composition and the leaching characteristics of heavy metals in polluted soils due to mining activities. The fractionated composition of heavy metals is classified into five fractions; adsorbed, carbonate, reducible, organic and residual fraction. The status of humic substances in mine wastes of most sites are polyhumic except tailing from Sangdong mine. According to the sequential extraction procedures (SEPs), leaching probabilities of Cd in coal wastes and tailing are relatively low due to high percentage of residual fraction. 46.4% of Ni in tailings from Sangdong mine is probably leached under oxidized environment, and 39.4% of Cu in these tailings is readily extracted under strongly oxidized environment by organic fraction. According to leaching condition of pH 3.0 and pH 5.6, the amount of heavy metals leached out of coal wastes and tailing increases to 1/2 hours. At pH 3.0 and pH 5.6, concentration of Ni in tailing increases up three times of the initial value. Heavy metals released from coal wastes and tailing were not influenced significantly by leaching time.

Isolation of Methylotrophic Actionmycetes Capable of Producing Antagonistic Activity Against Oral Resident Bacteria and Screening of Mutants (구강상재균을 중심으로 항균력을 나타내는 메탄올 자화방선균의 분리 및 변이주 생산)

  • In order to select an effective antibiotic substance against oral resident bacteria, we were isolated from soil and texonomically analyzed. Seven hundred and eighteen strains were isolated on humic acid- vitamin agar(HV agar) and 220 strains were on methanol medium from three each paddy forest, field and riverside soil samples. So, during the screening of antibiotics from soil, we isolated microorganisms showing powerful antagonistic activity against oral resident bacteria. Microorganism was tested against 25 strains of bacteria, yeast and fungi. Among them, No. 248 strain exhibited the most strongly growth inhibition. So, the taxonomical analysis the isolated strain was found to be unknown Actinomyces sp. and was named No 248. A production of the antibiotics from No. 248 begins at the early exponential phase developed at the 72th hour under the optinum conditions. The property of No. 248 antimicrobial compound was very stable under acid(pH 3.0) and alkali(pH 10.0) treatment, but it was instable in heat treatment at $120^{\circ}C$. For the improvement of antibiotic activity, two mutants were isolated from strain No. 248 by the treatment of mutagenic agents, NTG and hydroxylamine. As a result, the mutant strains excreted the potent antibiotics to inhibit the growth of Candida albicans.

  • This study was designed to evaluate the characteristics, land use and genesis of well adapted paddy soils. They were mostly classified as the Haplaquepts in the U.S.D.A soil taxonomy and as the Grey soils in the Japanese soil classification system. The proportion of these soils in the total acreage of paddy lands was thirty three percents. The fifty four percents in average of these soils were distributed on the local valley and fans on gentle slopes developed from granite, granite gneiss and shale parent materials. The rests were on the fluvio-marine deposits and alluvial deposits. The soils were characterized with prominant development of gleized horizons and clayey or fine loamy textured category. The available soil depth and the ground water level were relatively deep. The base saturation percent were high with weak acidic pH. The potential productivity of these soils was high.
