Sensing the Stress: the Role of the Stress-activated p38/Hog1 MAPK Signalling Pathway in Human Pathogenic Fungus Cryptococcus neoformans

  • All living organisms use numerous signal-transduction pathways to sense and respond to their environments and thereby survive and proliferate in a range of biological niches. Molecular dissection of these signalling networks has increased our understanding of these communication processes and provides a platform for therapeutic intervention when these pathways malfunction in disease states, including infection. Owing to the expanding availability of sequenced genomes, a wealth of genetic and molecular tools and the conservation of signalling networks, members of the fungal kingdom serve as excellent model systems for more complex, multicellular organisms. Here, we employed Cryptococcus neoformans as a model system to understand how fungal-signalling circuits operate at the molecular level to sense and respond to a plethora of environmental stresses, including osmoticshock, UV, high temperature, oxidative stress and toxic drugs/metabolites. The stress-activated p38/Hog1 MAPK pathway is structurally conserved in many organisms as diverse as yeast and mammals, but its regulation is uniquely specialized in a majority of clinical Cryptococcus neoformans serotype A and D strains to control differentiation and virulence factor regulation. C. neoformans Hog1 MAPK is controlled by Pbs2 MAPK kinase (MAPKK). The Pbs2-Hog1 MAPK cascade is controlled by the fungal "two-component" system that is composed of a response regulator, Ssk1, and multiple sensor kinases, including two-component.like (Tco) 1 and Tco2. Tco1 and Tco2 play shared and distinct roles in stress responses and drug sensitivity through the Hog1 MAPK system. Furthermore, each sensor kinase mediates unique cellular functions for virulence and morphological differentiation. We also identified and characterized the Ssk2 MAPKKK upstream of the MAPKK Pbs2 and the MAPK Hog1 in C. neoformans. The SSK2 gene was identified as a potential component responsible for differential Hog1 regulation between the serotype D sibling f1 strains B3501 and B3502 through comparative analysis of their meiotic map with the meiotic segregation of Hog1-dependent sensitivity to the fungicide fludioxonil. Ssk2 is the only polymorphic component in the Hog1 MAPK module, including two coding sequence changes between the SSK2 alleles in B3501 and B3502 strains. To further support this finding, the SSK2 allele exchange completely swapped Hog1-related phenotypes between B3501 and B3502 strains. In the serotype A strain H99, disruption of the SSK2 gene dramatically enhanced capsule biosynthesis and mating efficiency, similar to pbs2 and hog1 mutations. Furthermore, ssk2, pbs2, and hog1 mutants are all hypersensitive to a variety of stresses and completely resistant to fludioxonil. Taken together, these findings indicate that Ssk2 is the critical interface protein connecting the two-component system and the Pbs2-Hog1 pathway in C. neoformans.

Distance measurement System from detected objects within Kinect depth sensor's field of view and its applications (키넥트 깊이 측정 센서의 가시 범위 내 감지된 사물의 거리 측정 시스템과 그 응용분야)

  • Kinect depth sensor, a depth camera developed by Microsoft as a natural user interface for game appeared as a very useful tool in computer vision field. In this paper, due to kinect's depth sensor and its high frame rate, we developed a distance measurement system using Kinect camera to test it for unmanned vehicles which need vision systems to perceive the surrounding environment like human do in order to detect objects in their path. Therefore, kinect depth sensor is used to detect objects in its field of view and enhance the distance measurement system from objects to the vision sensor. Detected object is identified in accuracy way to determine if it is a real object or a pixel nose to reduce the processing time by ignoring pixels which are not a part of a real object. Using depth segmentation techniques along with Open CV library for image processing, we can identify present objects within Kinect camera's field of view and measure the distance from them to the sensor. Tests show promising results that this system can be used as well for autonomous vehicles equipped with low-cost range sensor, Kinect camera, for further processing depending on the application type when they reach a certain distance far from detected objects.

A Pursuit of Innovation in the Korean Genetics-Genomics Research System through a Culturalist Strategy (문화적 전략을 통한 한국 유전학-유전체학 연구체계의 혁신 모색)

  • The Korean genetics and genomics reveal a firm willingness to participate in and contribute to the production of creative scientific knowledge at a world level at present, though they have short past histories of introduction from the Western counterparts and those of education for the next generations. But the Korean genetics and genomics have been developed in a fragmented and biased manner. By reconfiguring the various research projects of genomics into the Genome Project of Korea, which reflect a worldly trend in life science, but have been established in a scattered fashion in Korea, and incorporating some neglected areas of genetics, such as human genetics and theoretical and population genetics which can be reconstructed in a new way, a genetics-genomics research system can be formulated on the multi-tiered perspective of concept, knowledge, and institution, while the system being a subsystem of the national research system of life science in Korea. Innovation can be pursued in the systematic practice through a culturalist strategy. The culturalist strategy with the practice based on the research system consists of 1) intensification of fundamentalness of genetics and genomics, 2) advancement of communitarianism in geneticist-genomicist community, 3) research on the cultural bio-species along with the promotion of scientific arts and culture, and 4)formation of the Korean science studies of genetics-genomics and the diffusion of the knowledge produced. The first two strategy components are the ones that intends to bring out changes in the structural aspect of the scientist community in Korea. The third is the one that attempts to magnify the interface between the scientist community and the Korean society at large and increase its connectivity between both, while the fourth is the one that has an intentionality toward the Korean society outside of the scientist community. This culturalist strategy is intended to increase the cultural constructivity of the genetics-genomics research system in Korea.

  • In large scale environments like airport, museum, large warehouse and department store, autonomous mobile robots will play an important role in security and surveillance tasks. Robotic security guards will give the surveyed information of large scale environments and communicate with human operator with that kind of data such as if there is an object or not and a window is open. Both for visualization of information and as human machine interface for remote control, a 3D model can give much more useful information than the typical 2D maps used in many robotic applications today. It is easier to understandable and makes user feel like being in a location of robot so that user could interact with robot more naturally in a remote circumstance and see structures such as windows and doors that cannot be seen in a 2D model. In this paper we present our simple and easy to use method to obtain a 3D textured model. For expression of reality, we need to integrate the 3D models and real scenes. Most of other cases of 3D modeling method consist of two data acquisition devices. One for getting a 3D model and another for obtaining realistic textures. In this case, the former device would be 2D laser range-finder and the latter device would be common camera. Our algorithm consists of building a measurement-based 2D metric map which is acquired by laser range-finder, texture acquisition/stitching and texture-mapping to corresponding 3D model. The algorithm is implemented with laser sensor for obtaining 2D/3D metric map and two cameras for gathering texture. Our geometric 3D model consists of planes that model the floor and walls. The geometry of the planes is extracted from the 2D metric map data. Textures for the floor and walls are generated from the images captured by two 1394 cameras which have wide Field of View angle. Image stitching and image cutting process is used to generate textured images for corresponding with a 3D model. The algorithm is applied to 2 cases which are corridor and space that has the four wall like room of building. The generated 3D map model of indoor environment is shown with VRML format and can be viewed in a web browser with a VRML plug-in. The proposed algorithm can be applied to 3D model-based remote surveillance system through WWW.

  • In the IoT environment, various things could be connected. Those connected things learn and operate themselves, by acquiring data. As human being, they have self-learning and self-operating systems. In the field of IoT study, therefore, the key issue is to design communication system connecting both of the two different types of subjects, human being(user) and the things. With the advent of the IoT environment, much research has been done in the field of UI design. It can be seen that research has been conducted to take complex factors into account through keywords such as multi-modality and interusability. However, the existing UI design method has limitations in structuring or testing interaction between things and users of IoT environment. Therefore, this paper suggests a new UI prototyping method. In this paper, the major analysis and studies are as follows: (1) defined what is the behavior process of the things (2) analyzed the existing IoT product (3) built a new framework driving personality types (4) extracted three representative personality models (5) applied the three models to the smart home service and tested UI prototyping. It is meaningful with that this study can confirm user experience (UX) about IoT service in a more comprehensive way. Moreover, the concept of the personality of things will be utilized as a tool for establishing the identity of artificial intelligence (AI) services in the future.

  • Harmful air pollutants are exhausted from the various industrial facilities including the coal-fired thermal power plants and these substances affects on the human health as well as the nature environment. In particular, nitrogen oxides ($NO_x$) and sulfur dioxide ($SO_2$) are known to be causative substances to form fine particles ($PM_{2.5}$), which are also deleterious to human health. The integrated system composed of selective catalytic reduction (SCR) and wet flue gas desulfurization (WFGD) have been widely applied in order to control $NO_x$ and $SO_2$ emissions, resulting in high investment and operational costs, maintenance problems, and technical limitations. Recently, new technologies for the simultaneous removal of $NO_x$ and $SO_2$ from the flue gas, such as absorption, advanced oxidation processes (AOPs), non-thermal plasma (NTP), and electron beam (EB), are investigated in order to replace current integrated systems. The proposed technologies are based on the oxidation of $NO_x$ and $SO_2$ to $HNO_3$ and $H_2SO_4$ by using strong aqueous oxidants or oxidative radicals, the absorption of $HNO_3$ and $H_2SO_4$ into water at the gas-liquid interface, and the neutralization with additive reagents. In this paper, we summarize the technical improvements of each simultaneous abatement processes and the future prospect of technologies for demonstrating large-scaled applications.

  • Conventional MRI methods using T1-, T2-, diffusion-, perfusion-weighting, and functional imaging rely on characterizing the physical and functional properties of the tissue. In this review, we introduce an imaging modality based on measured the mechanical properties of soft tissue, namely magnetic resonance elastography (MRE). The use of palpation to identify the stiffness of tissue remains a fundamental diagnostic tool. MRE can quantify the stiffness of the tissue thereby providing a objective means to measure the mechanical properties. To accomplish a successful clinical setting using MRE, hardware and software techniques in the area of transducer, pulse sequence, and imaging processing algorithm need to be developed. Transducer, a mechanical vibrator, is the core of MRE application to make wave propagate invivo. For this reason, considerations of the frame of human body, pressure and friction of the interface, and high magnetic field of a MRI system needs to be taken into account when designing a transducer. Given that the wave propagates through human body effectively, developing an appropriate pulse sequence is another important issue in obtaining an optimal image. In this review paper, we introduce the technical aspects needed for MRE experiments and introduce several applications of this new field.

Eye Tracking Using Neural Network and Mean-shift (신경망과 Mean-shift를 이용한 눈 추적)

  • In this paper, an eye tracking method is presented using a neural network (NN) and mean-shift algorithm that can accurately detect and track user's eyes under the cluttered background. In the proposed method, to deal with the rigid head motion, the facial region is first obtained using skin-color model and con-nected-component analysis. Thereafter the eye regions are localized using neural network (NN)-based tex-ture classifier that discriminates the facial region into eye class and non-eye class, which enables our method to accurately detect users' eyes even if they put on glasses. Once the eye region is localized, they are continuously and correctly tracking by mean-shift algorithm. To assess the validity of the proposed method, it is applied to the interface system using eye movement and is tested with a group of 25 users through playing a 'aligns games.' The results show that the system process more than 30 frames/sec on PC for the $320{\times}240$ size input image and supply a user-friendly and convenient access to a computer in real-time operation.

Big Data based Tourist Attractions Recommendation - Focus on Korean Tourism Organization Linked Open Data - (빅데이터 기반 관광지 추천 시스템 구현 - 한국관광공사 LOD를 중심으로 -)

  • Conventional exhibition management information systems recommend tourist attractions that are close to the place in which an exhibition is held. Some recommended attractions by the location-based recommendation could be meaningless when nothing is related to the exhibition's topic. Our goal is to recommend attractions that are related to the content presented in the exhibition, which can be coined as content-based recommendation. Even though human exhibition curators can do this, the quality is limited to their manual task and knowledge. We propose an automatic way of discovering attractions relevant to an exhibition of interests. Language resources are incorporated to discover attractions that are more meaningful. Because a typical single machine is unable to deal with such large-scale language resources efficiently, we implemented the algorithm on top of Apache Spark, which is a well-known distributed computing framework. As a user interface prototype, a web-based system is implemented that provides users with a list of relevant attractions when users are browsing exhibition information, available at http://bike.snu.ac.kr/WARP. We carried out a case study based on Korean Tourism Organization Linked Open Data with Korean Wikipedia as a language resource. Experimental results are demonstrated to show the efficiency and effectiveness of the proposed system. The effectiveness was evaluated against well-known exhibitions. It is expected that the proposed approach will contribute to the development of both exhibition and tourist industries by motivating exhibition visitors to become active tourists.

A Remote Control System for Home Appliance using Low Speed Network (저속 네트워크 기반의 가전기기를 위한 원격 제어 시스템)

  • Home networking and Internet control technologies must satisfy three requirements with a view to being applied to home for raising the qualify of human life. Firstly, the cost of adding home networking ability to the traditional appliance must be minimized. Secondly, the mutual accessibility between home network and Internet must be guaranteed in the various Internet service environment of home. Finally, communication must be possible between the home network and the Internet that use different communications protocols. This paper proposes the ARCS (Appliance Remote Control System) that controls appliances based on LnCP (Living network Control Protocol) via the Internet. LnCP is the most cost-effective network and control protocol for home network. ARCS guarantees the accessibility from the Internet to home network and provides users with the virtual interface designed for controlling appliances using the Internet application program. In addition, this paper proposes AICS (Appliance Internet Control Specification). AICS, is based on LnCP, defines the message structure and control commands for the Internet control communication. ARCS and home gateway are developed to meet AICS.
