Effects of Reactive Oxygen Species on DNA Stability in Humnn Spermatozoa

  • This study was designed to investigate the effects of reactive oxygen species (ROS) on DNA stability in human spermatozoa. To verify human spermatozoa were incubated with xanthine-xanthine oxidase (X 100$\mu$M-XO 50 mlU ~ 400 mIU), $H_2O_2$ (125 $\mu$M ~ 1 mM), sodium nitroprusside (SNP 0.1 $\mu$M ~ 100 $\mu$M) or lymphocyte. Otherwise, spermatozoa were incubated under low $O_2$ (5%) condition. Damage of sperm DNA was analyzed by single cell electrophoresis (Comet assay) and flow cytometry after acridine orange staining. In the presence of ROS, there was increase in DNA damage. The rate of DNA single strand breakage (9.0$\pm$1.0% ~ 46.0$\pm$4.6%) and DNA fragmentation (7.51$\pm$1.0% ~ 29.5$\pm$4.6%) were similar regardless of the kinds of ROS and exposure time. DNA damage in the lower $O_2$ condition (5%) was lower than ambient $O_2$ condition (20%). Taken together, it suggested that sperm DNA might be damaged by ROS. In the presence of ROS, increase in DNA damage and chromatin instability was obvious in spite of short exposure. Although present study reconfirmed that sperm incubation in the low concentration of ROS have the benefit m the induction of capacitation and Ah, the increase in DNA damage by ROS and possible genetic problem should be considered before the human trials.

  • Human cardiomyocytes (CMs) cease to proliferate and remain terminally differentiated thereafter, when humans reach the mid-20s. Thus, any damages sustained by myocardium tissue are irreversible, and they require medical interventions to regain functionality. To date, new surgical procedures and drugs have been developed, albeit with limited success, to treat various heart diseases including myocardial infarction. Hence, there is a pressing need to develop more effective treatment methods to address the increasing mortality rate of the heart diseases. Functional CMs are not only an important in vitro cellular tool to model various types of heart diseases for drug development, but they are also a promising therapeutic agent for cell therapy. However, the limited proliferative capacity entails difficulties in acquiring functional CMs in the scale that is required for pathological studies and cell therapy development. Stem cells, human pluripotent stem cells (hPSCs) in particular, have been considered as an unlimited cellular source for providing functional CMs for various applications. Notable progress has already been made: the first clinical trials of hPSCs derived CMs (hPSC-CMs) for treating myocardial infarction was approved in 2015, and their potential use in disease modeling and drug discovery is being fully explored. This concise review gives an account of current development of differentiation, purification and maturation techniques for hPSC-CMs, and their application in cell therapy development and pharmaceutical industries will be discussed with the latest experimental evidence.

Effect of Keratin-Based Biocomposite Hydrogels as a RhBMP-2 Carrier in Calvarial Bone Defects Mouse Model

  • Recently, in human medicine and veterinary medicine, interest in synthetic bone graft is increasing. Among them, bone morphogenic protein (BMP) is currently being actively researched and applied to clinical trials. However, BMP has the disadvantage of being expensive and easily absorbed into surrounding tissues. Therefore, BMP requires the use of small amounts and rhBMP (recombinant human bone morphogenetic protein)-2 carriers that can be released slowly. Hydrogel has the property of swelling a large amount of water inside when it is aqueous solution, and when it is, it consists of more than 90 percent water. Using these properties, hydrogels are often used as rhBMP-2 carrier. The scaffold used in this study is a hydrogel made from which keratin is extracted using human hair and based on it. In this study, we wanted to see the effect of bone formation in the calvarial defect model by using keratin-based hydrogel made with human hair as a scaffold. The experiment was conducted by dividing 3 groups a total of 12 mice. Calvarial bone defect is set to all 4 mm diameters. Bone formation was evaluated by using gross evaluation, micro-computed tomography (micro-CT), immunohistochemistry. Groups using keratin-based hydrogel were significantly observed compared to Group 1s, and the most bone formations were found when rhBMP-2 and hydrogel were used. This represents the superiority of the functions of the rhBMP-2 carrier by a new material, keratin-based hydrogel. Through gross evaluation, micro-CT, and immunohistochemistry, we can confirm that keratin-based hydrogel is a useful rhBMP-2 carrier.

조등산(釣藤散)의 약리학적(藥理學的) 고찰(考察) (A review on pharmacological properties of Jodeungsan)

  • The span of life of human beings has lengthened because of medical progress and so on. Consequently, the old have begun to remark that quality of life (QOL) is as important as extension of the life expectancy. However, some diseases such as stroke, cancer induce decrease of QOL. In case of the stroke, it is followed by chronic headache and dementia as an aftereffect, causing serious decrease of QOL. Jodeungsan, traditional medicine, have been prescribed for headache and hypertension. Many clinical trials and laboratory experiments about pharmacological effects of Jodeungsan have been reported. Jodeungsan ameliorated chronic headache and improved recognitive deficit. Moreover, Jodeungsan lowered blood pressure in hypertensive condition which is one of a major cause of stroke. These effects of Jodeungsan on several diseases are partly attributed to antioxidant effect. This report reviews the pharmacological effect of Jodeungsan in the view of stroke-related diseases.

노인성 음성에 대한 최신 연구동향 (Current Researches on Vocal Fold Aging)

  • Aging causes a variety of changes in the structure of the vocal fold (VF), resulting in aging-induced dysphonia (presbyphonia). Several studies have investigated the structure of the VF of elderly people from autopsy as well as animal studies. There is an increasing evidence on correlation of structural changes of VF with deteriorated voice in elderly. Although the cellular mechanisms of aging VF have only partially been elucidated, there are many recent advances in biological treatment on aging VF using bioactive molecules such as growth factors. In this study, I'd like to address aging-related structural, biological, and physiological changes in previous literature about human and rodent aging VFs, to provide further insight into the mechanisms responsible for presbyphonia and to translate the basic researches for future clinical trials.

  • Fuzzy control has been researched for application of industrial processes which have no accurate mathematical model and could not controlled by conventional methods because of a lack of quantitative input-output data. Intelligent control approach based on fuzzy logic could directly reflex human thinking and natural language to controller comparing with conventional methods. In this paper, the tested system is constructed for sending a ball to the goal position using wind from two DC motors in the path. This system contains non-linearity and uncertainty because of the characteristic of aerodynamics inside the path. The system used in this experiment could be hardly modeled by mathematic methods and could not be easily controlled by linear control manners. The controller, in this paper could control the system containing non-linearity and uncertainty because it is designed based on the input-output data and experimental knowledge obtained by trials.

  • The effects of task characteristics on the relative efficiency of visual display dimension were studied using a simulated tele-robotic task. Through a conventional method of task analysis. the tele-robotic task was divided into two categories: the task element requiring focused attention (FA task) and the task element requiring global attention (CA task). Time-ta-completion data were collected for a total of 120 trials involving 10 participants. For the CA task. there was no significant difference between the multiple two-dimensional (20) display and the three-dimensional (3D) monocular display. For the FA task. however. the multiple 20 display was superior to the 3D monocular display. The results suggest that the characteristics of a given task have a considerable effect on the choice of display dimensionality and the multiple 3D display is better for human operators to effectively judge depth if the task requires frequent use of focused attention.

Targeting Multidrug Resistance with Small Molecules for Cancer Therapy

  • Conventional cancer chemotherapy is seriously limited by tumor cells exhibiting multidrug resistance (MDR), which is caused by changes in the levels or activity of membrane transporters that mediate energy-dependent drug efflux and of proteins that affect drug metabolism and/or drug action. Cancer scientists and oncologists have worked together for some time to understand anticancer drug resistance and develop pharmacological strategies to overcome such resistance. Much focus has been on the reversal of the MDR phenotype by inhibition of ATP-binding cassette (ABC) drug transporters. ABC transporters are a family of transporter proteins that mediate drug resistance and low drug bioavailability by pumping various drugs out of cells at the expense of ATP hydrolysis. Many inhibitors of MDR transporters have been identified, and though some are currently undergoing clinical trials, none are in clinical use. Herein, we briefly review the status of MDR in human cancer, explore the pathways of MDR in chemotherapy, and outline recent advances in the design and development of MDR modulators.

변증시치(辨證施治)와 정병전방(定病專方) (A therapeutic following confirming symptom and a therapeutic regimen prescription)

  • A therapeutic following confirming symptom(辨證施治) has a proper characteristic in oriental medicine, which can be grasped but by pathological body reflection not by histological or molecular level etiology. It is also able to be selected a prescription by means of clinical trials which promote human body's natural resurgent. Whereas, a therapeutic regimen prescription(定病傳方) has a merit in prescribing a disease to the point. which has been developed and established a therapy method naturally for thousands years. Then, which of the two is the better method in clinic? The answer is a co-exist of the two therapy methods in a matter of developing deciding therapeutic cooperation. To tie a knot, two methods need the opposite one.

Antisense DNAs as Targeted Genetic Medicine to Treat Cancer

  • Nucleic acid therapies represent a direct genetic approach for cancer treatment. Such an approach takes advantage of mechanisms that activate genes known to confer a growth advantage to neoplastic cells. The ability to block the expression of these genes allows exploration of normal growth regulation. Progress in antisense technology has been rapid, and the traditional antisense inhibition of gene expression is now viewed on a genomic scale. This global view has led to a new vision in antisense technology, the elimination of nonspecific and undesirable side effects, and ultimately, the generation of more effective and less toxic nucleic acid medicines. Several antisense oligonucleotides are in clinical trials, are well tolerated, and are potentially active therapeutically. Antisense oligonucleotides are promising molecular medicines for treating human cancer in the near future.
