Production and Prophylactic Efficacy Study of Human Papillomavirus-like Particle Expressing HPV16 L1 Capsid Protein

  • To perform the prophylactic study of a vaccine derived from human papillomavirus (HPV) using Balb/c mice, we produced virus like particles consisting of HPV capsid protein L1 which has been reported to induce significant humoral and cellular immunity using various animal model systems. In order to produce HPV16 VLPs, the cDNA of L1 capsid protein in HPV type 16, obtained by polymerase chain reaction, was inserted into yeast expression vector, YEG$\alpha$-HIR525 under the control of GAL10 promoter. The transformation of YEG$\alpha$-HPV16 L1 was performed into the yeast Saccharomyces cerevisiae Y2805 by the lithium acetate method and the yeast clone expressing the highest level of L1 capsid protein of human papillomavirus type 16 was selected by Western blot analysis using anti-HPV16 L1 antibody. The purification of HPV16 VLP has been performed by the ultracentrifugation and gel-filtration methods. To validate the vaccine efficacy of the purified HPV16 VLPs and investigate the properties of HPV16 VLPs to induce humoral immunity, ELISA assay was performed. A significantly increased production of anti-HPV16 VLP antibodies was observed in sera from immunized mice. The neutralization activity of antibodies in the sera from the vaccinated mice was demonstrated by a rapid and simple assay to detect hemagglutihation inhibition activity.

Development of a Screening System for Drugs Against Human Papillomavirus-Associated Cervical Cancer: Based On E7-Rb Binding

  • The human papillomavirus E7 protein can form a specific complex with a retinoblastoma tumor suppressor gene product (p105-Rb) that results in the release of the E2F transcription factor, which is critical for the growth-deregulation and transforming properties of the viral E7 oncoprotein. In an attempt to apply interaction between the E7 oncoprotein and a target cellular protein Rb for an in vitro screening system for drugs against human papillomavirus infection, we primarily investigated the E7Rb binding through a pull down assay and enzyme-linked immunosorbent assay. The pull down assay showed that both glutathione S-transferase-tagged E7 and His-tagged E7 immobilized on resins specifically produced complexes with bacterially expressed Rb in a dose-dependent manner, as determined by immunoblot analyses. This result coincided with that of an enzyme-linked immunosorbent assay, which is a useful system for the mass screening of potential drugs. Taken together, this screening system (based on the interaction between E7 and Rb) can be a promising system in the development of drugs against cervical cancers caused by human papillomavirus infection.

The Aetiological Role of Human Papillomavirus in Colorectal Carcinoma: An Iranian Population- Based Case Control Study

  • Background: Human papillomavirus (HPV) is one of the most common sexually transmitted infections worldwide and the association between HPV infection and genital cancers has been well established. This study concerned the possible role of HPV infection in colorectal carcinoma (CRC) in the Iranian population. Materials and Methods: We examined 80 tissues obtained from patients with colorectal cancer consisting of 58 colon cancer samples and 22 rectal cancer samples and 80 tissues from patients with unremarkable pathologic changes as matched controls by sex, study center and anatomical sites. HPV infection and genotypes were detected using nested PCR and sequencing methods, respectively. Results: HPV DNA was detected in 5/80 (6.25%) cases including 1 of 22 (4.54%) patients with rectum cancer and 4 of 58 (6.9%) patients with colon cancer and 1/80 (1.25%) of controls. Furthermore, HPV-18 was detected as the most frequent type and we found no significant correlation between prevalence of HPV infection and anatomical sub- sites. Conclusions: Although a causal relation between human papillomavirus and colorectal cancer was not found through this study, analysis of medical records pointed to a possible role for high- risk types of HPV in increasing the potential of aggressiveness in colorectal cancer. This study shows a particular frequency of HPV genotypes in patients with colorectal cancer in Iran. Since HPV vaccines are limited to a few types of virus, using cohort studies in different geographical zones to screen for patterns of HPV infection in different organs might increase the efficacy and optimization of the current vaccines.

여성 청소년 어머니의 인유두종바이러스 예방접종 예측요인 (Predictors of Human Papillomavirus Vaccination of female Adolescent mothers)

  • 본 연구는 건강신념모델을 근거로 인유두종바이러스(human papillomavirus: HPV) 예방접종 행위 모형의 적합도를 검증하고자 시도되었다. 연구 대상자는 여성 청소년 어머니 285명이며, 자료 수집은 2017년 6월부터 9월까지 하였다. 자료는 SPSS 및 AMOS 프로그램을 이용하여 분석하였다. 연구결과, 여성 청소년 어머니의 지각된 유익성은 의도를 완전 매개하여 예방접종 행위에 영향을 주고, 지각된 장애성은 의도를 부분 매개하여 예방접종 행위에 영향을 주는 것으로 나타났다. 예방접종 의도는 예방접종 행위를 예측하는 중요한 변수였고 지각된 장애성은 예방접종 행위에 가장 큰 영향요인으로 나타났다. 이러한 결과는 여성 청소년 어머니의 HPV 예방접종 과정의 이해를 증진시키고 예방접종률을 높이는 새로운 전략 개발에 도움이 될 것이다.

성별에 따른 고교생의 인유두종 바이러스 백신 접종실태와 관련 건강신념 및 지식의 차이 (Comparison of Human Papillomavirus Vaccination Status, Associated with Health Belief and Knowledge between Male and Female Highschool Students)

  • Purpose: The purpose of this study was to investigate and compare human papillomavirus (HPV) vaccination status associated with HPV health belief and knowledge between male and female highschool students. Methods: In the winter of 2012, 183 students responded to self-administered questionnaires, and t-test, ${\chi}^2$-test, and ANOVA were used for data analysis. Results: There were significant differences in the experiences of hearing about HPV vaccination, source of HPV information, experience of being vaccinated with HPV vaccination, reason for HPV unvaccination and vaccination intention by gender. The mean scores for health belief of HPV vaccination were 1.72 (total score: 4) in men and 2.31 in women. There were significant differences in HPV health belief (t=-5.14, p<.001). The mean scores for knowledge of HPV vaccination were 0.08 (total score: 1) in men and 0.12 in women, but there were no significant differences in HPV knowledge (t=-1.62, p=.108). There were positive associations between HPV health belief and knowledge in women (p=.02). Conclusion: This study showed different HPV vaccination status, health belief and knowledge in gender. The results indicate a need to consider levels of health belief and knowledge of HPV vaccination in target populations of highschool students when planning a HPV related education program.

Human Papillomavirus Genotypes Associated with Mucopurulent Cervicitis and Cervical Cancer in Hangzhou, China

  • Background: To investigate the infection status and predominant genotype distribution of human papillomavirus (HPV) infection among Chinese patients with mucopurulent cervicitis (MPC) or cervical cancer (CC) in Hangzhou. Methods: Initially, 217 cases of healthy cervix controls (n=50), acute MPC (n=89), and CC (n=78) were included; samples were collected between January 1, 2010, and January 30, 2013. Cervical specimens were screened for HPV using a nested polymerase chain reaction assay and DNA sequencing. Results: Overall prevalence of HPV infection was 16.7% in the control group, 51.9% in the MPC group, and 84.4% in the CC group. The predominant genotype detected in all 3 groups was the oncogenic variant HPV 16 (55.8%, 17.3%, and 6.3% in the CC, MPC and control specimens, respectively), HPV58 was the second most predominant HPV type in CC (9.1%), MPC (8.6%), and control group (4.2%). Most or all of the genotypes were oncogenic in the three groups. Conclusions: Infection with HPV was found to be prevalent among Chinese women with MPC or CC and oncogenic variants were in the majority. Therefore, peoples who suffered MPC with HPV DNA positive should be regularly followed-up, for prevention and early treatment of cervical cancer.

No Significant Association between Human Papillomavirus and Breast Cancer, Sanandaj, Iran

  • Background and Objective: Any role of human papillomavirus (HPV) in the development of breast cancer is conjectural. The aim of this study was to investigate possible links between HPV and breast cancer in women, Sanandaj, Iran. Methods: In this case-control study, 70 formalin fixed and paraffin embedded blocks of breast malignant tumors as a case group and 70 blocks of lesions without malignancy were selected as controls. Sections about $10{\mu}m$ thick were prepared. After removing the paraffin, DNA was extracted. Samples were tested by PCR using general and high-risk specific HPV primers. Results: All 70 malignant breast tumors (cases) were invasive ductal carcinomas, and of the 70 controls, 17 (24.3%) were fibrocystic tumors and 53 (75.7%) fibroadenomas. The age range of women in the case group was 25-72 years old and in the control group It was13-66 years. Using HPV general primers two samples were positive in the case group, confirmed to be HPV-18 using high-risk specific primers. Conclusion: No statistically significant association was found between breast cancer and HPV. It is necessary to confirm this result by further investigations in other populations.

Whole Genome Analysis of Human Papillomavirus Genotype 11 from Cervix, Larynx and Lung

  • The prevalence of human papillomavirus genotypes differs in various target organs. HPV16 is the most prevalent genotype in the cervix while genotypes 6 and 11 are highly prevalent in skin and aero-digestive tract infections. In this study HPV11 positive specimens were selected from cervix, larynx and lung biopsy tissue to analyze the whole genome by PCR and direct sequencing. Five HPV11 whole genomes were characterized, consisting of two cervical specimens, two laryngeal specimens and one lung specimen. The results showed high homology of HPV11 in these organs. Phylogenetic analysis showed that all HPV11 derived from various organs belonged to the same lineage. Molecular characterization and functional studies can further our understanding of virulence, expression or transmission. Additional studies on functional protein expression at different organ sites will also contribute to our knowledge of HPV infection in various organs.

일부 여대생의 자궁경부암 지식과 인유두종바이러스 지식 및 감염 예방행위의도에 관한 연구 (Study on the Knowledge of Cervical Cancer and Human Papillomavirus and Preventive Behavior Intention of Female University Students)

  • Purpose: This study was conducted to find out how much female university students knew about cervical cancer and Human papillomavirus (HPV) and to develop preventive behavior strategies for HPV infection. Method: In total, 511 students were involved and the assessment was done by means of cervical cancer knowledge questions, HPV knowledge questions and Alert questions. Result: The value regarding the knowledge of cervical cancer and of HPV turned out lower than the median while the degree of HPV prevention behavior intention was approximately mean. The values of the three variables, i.e. the knowledge of cervical cancer, knowledge of HPV, and the degree of preventive behavior intention were higher proportionately, showing a statistically significant difference, as the age of the subjects went higher, as long as they were nursing majors, and were more knowledgeable about HPV (p<.05). The three variables correlated positively (p<.05). Conclusion: Since the lack of knowledge with regard to cervical cancer and HPV is seen to cause a certain portion of female students to be vulnerable to the disease and lead them to a low degree of prevention, it is highly recommended to organize systematic sex education and thereby increase the willingness for preventive actions.

Cervical Pathology in High-Risk Human Papillomavirus-Positive, Cytologically Normal Women

  • This study was undertaken to evaluate the prevalence of significant cervical pathology among women who are high-risk human papillomavirus (HR-HPV)-positive/cytology negative, the most common combination of positive co-tests. The records of 244 women HR-HPV-positive/cytology-negative who had undergone colposcopy at Srinagarind Hospital, Khon Kaen University during January 2010 and April 2014 were reviewed. Mean age was 46.4 years. Of these 224 women, 75 were positive for HPV types 16/18 (33.5%) and 123 were positive for non-16/18 types (54.9%). HR-HPV was not genotyped in the remaining 26 women (11.6%). Prevalence of significant lesions for the entire cohort was 2.4%, and 2.6% and 3.3%, respectively, for those with HPV 16/18 and other oncogenic HPV types. One woman with HPV 16/18 (1.3%) had invasive cervical cancer. Multiparous women were more likely to be infected with HPV 16/18 compared to nulliparous women (36.3% versus 17.6%, respectively). In conclusion, the prevalence of significant cervical lesion among our study population was 2.4%. Multiparous women were more likely to be infected with HPV 16/18 compared to nulliparous women.
